Changes of Vitamin C and Superoxide Dismutase(SOD)-like Activity of Persimmon Leaf Tea by Processing Method and Extraction Condition (감잎의 처리방법과 추출조건에 따른 감잎차의 Vitamin C와 Superoxide Dismutase(SOD) 유사활성의 변화)

  • The persimmon leaf tea was produced from persimmon leaves by three different methods (conventional, steamed, fermented) and the changes of total vitamin C and superoxide dismutase(SOD)-like activity were investigated. The amount of total vitamin C was the highest in fermented persimmon leaf tea which was 47% of total vitamin C of raw persimmon leaves. The SOD-like activity of conventional and fermented persimmon leaf teas were a little higher than that of steamed. The total vitamin C of steamed persimmon leaf tea was decreased slowly as the extraction time was increased. That of fermented persimmon leaf tea was increased generally at $60^{\circ}C\;and\;70^{\circ}C$, and increased until 5min and then decreased at $80^{\circ}C\;and\;90^{\circ}C$, The optimum extraction condition of total vitamin C in fermented persimmon leaf tea was 3min and 5min at $80^{\circ}C\;and\;90^{\circ}C$, 5min and 10min at $60^{\circ}C$. The total vitamin C of conventional persimmon leaf tea was so little that could not be measured by DNP method. The SOD-like activity of conventional and steamed persimmon leaf teas were increased and that of fermented showed the trend of increasing-decreasing-increasing. Fermented persimmon leaf tea had higher SOD-like activity than conventional and steamed at all tested conditions, and the optimum extraction condition of SOD-like activity was 3min and 10min at $80^{\circ}C\;and\;90^{\circ}C$.

Antimutagenic Effects of Persimmon Leaf Tea Extract (PLTE) in Mice Using Micronucleus Induction (MN) Test (마우스 소핵(Micronuclei, MN) 시험방법을 이용한 감잎차 추출물의 돌연변이 억제효과)

  • The antimutagenic effects of persimmon leaf tea extract (PLTE) at concentration levels consumed by human were examined in mice using micronuleus induction with MMC(mitomycin C) or 4-NQO (4-nitroquinoline-1-oxide). When mice received oral gavage of 10 equivalent to PLTE 24 hr and 6 hr before, and 5 equivalent to PLTE 6 hr before and 3 hr after intraperitoneal injection of MMC, a significant decrease in the frequency of micronuclei were observed. The induction of micronuclei by 4-NQO was suppressed by oral dosage of PLTE at 5 equivalent to PLTE 6 hr before and 3 hr after, 10 equivalent to PLTE 3 hr before and 3 hr after intraperitoneal injection of MMC. Though the components of PLTE have not been analyzed so far, our present results suggest the existence of several bio-antimutagens and/or desmutagens in PLTE, beside catechin, well-known antimutagen.

The Chemical Composition of Persimmon (Diospyros kaki, Thumb) Leaf Tea (감 (Diospyros kaki, Thumb) 잎차의 화학 성분)

  • Chemical components relevant to the characteristic taste of the Korean native persimmon(Diospyros kaki, Thumb) leaf tea were analyzed. Samples were processed by using three different methods ; SHT(steamed and then hot-air dried), DHT(dried in the shade, steamed and then hot-air dried) and RHT(roasted and then hot-air dried). The components analyzed were general compositions of dried perisimmon leaves and extracted solution. The composition of moisture, ash, crude lipid and total nitrogen did not show significant variation among different processing methods of the persimmonleaf tea. The contents of caffeine, tannin and vitamin C in persimmon leaf tea were in the range of $178.4~209.8{\mu}mol/g$, 29.1~38.5mg% and 325.3~2084.7mg%, respectively. The vitamin C content was significantly higher in the RHT than other treatments. The contents of caffeine, tannin and vitamin C in the tea extracted solution were in the range of $101.5~130.1{\mu}mol/g$, 15.4~25.9mg% and 111.0~1274.3mg%, respectively. The vitamine C in the tea solution was the highest in the RHT treatment and 61.1% of vitamin C in the leaf tea was extracted out in these processing methods. The major amino acids contained in the leaf tea were in decreasing order glutamic acid, aspartic acid, leucine and phenylalanine, these four amino acids consisting 38.9~39.8% of the total amino acid contained in the persimmon leaf tea. The major amino acids contained in the tea solution were glutamic acid, proline, histidine and arginine. Six kinds of 5'-nucleotides, CMP, AMP, UMP, IMP, GMP and hypoxanthine were detected and CMP was the most abundant component in fresh leaf, leaf tea and tea solution. The second highest 5'-nucleotides in both leaf tea and tea solutions were GMP, AMP and UMP in all processing method. The highest free sugar contained in the fresh leaf tea and tea solution was sucrose.

Effects of Persimmon leaf on the Photoaging Skin Improvement(2) (감잎의 광노화 피부 개선에 미치는 효과(2))

  • This study was performed to investigate the anti-photoaging effects of Persimmon leaf tea(PLT) in hairless mice(SKH-1) exposed to UVB radiation. The animals were divided into non-treated group (normal, N) and UV-radiated groups. UV-radiated groups were divided into only UV-radiated group(control, C) and UV-radiated and PLT treated experimental groups[first extraction treated group(PLT-I), second extraction treated groupe(PLT-II), and third extraction treated group(PLT-III)]. Three PLT treated experimental groups of mice were treated with both oral administration(300mg/Kg B.W./day) and topical application (100 ul of 2% conc./mouse/day) for 4 weeks. Anti-photoaging effects of Persimmon leaf were evaluated by MTT assay, anti oxidative reaction, MMP immunohistochemistry, gelatin zymography assay and RT-PCR observations. Treatment with Persimmon leaf tea(PLT)-I, and -III groups decreased immunohistochemical density of matrix metalloproteinases(MMP)-3 and -9 related to degradation of extracellular matrix in skin. Especially, immunohistochemical density of MMP-2 decreased in PLT-I, -II and -III groups in skin. On the effects of antioxidant function on the treatment with Persimmon leaf tea(PLT), treatment of HaCaT cells with extracts of PLT-I and PLT-II had also significantly reduced intracellular ROS produced by UVB irradiation in a dose dependent manner(PLT-I, p<0.05, p<0.01, p<0.001; PLT-II, p<0.01, p<0.001). Gelatin zymography assay revealed that PLT-II and PLT-III (200 ug/ml) had inhibitory effect on MMP-9 expression in UVB-radiated HaCaT cells. Western blot analysis revealed that PLT-1, -II and -III groups down-regulates the expression of inflammatory associated genes(IL-$1{\beta}$) and PLT-1 and -II groups down-regulates the expression of TNF-${\alpha}$ in a dose dependent manner. Our study suggests that Persimmon leaf tea(PLT) extracts participates in inhibitory effects on the morphological and molecular experiments related to photoaging skin on UVB irradiated hairless mice.

The Leaf of Diospyros kaki Thumb Ameliorates Renal Oxidative Damage in Mice with Type 2 Diabetes

  • Diabetic kidney disease is the most common and severe chronic complication of diabetes. The leaf of Diospyros kaki Thumb (persimmon) has been commonly used for herbal tea and medicinal purposes to treat a variety of conditions, including hypertension and atherosclerosis. However, the effect of persimmon leaf on kidney failure has not been investigated. This study aimed to examine the role of persimmon leaf in protecting the diabetes-associated kidney damage in a mouse model of type 2 diabetes. Mice were fed either a normal chow diet with or without powered persimmon leaf (5%, w/w) for 5 weeks. In addition to kidney morphology and blood markers of kidney function, we assessed levels of oxidative stress markers as well as antioxidant enzymes activities and mRNA expression in the kidney. Supplementation of the diet with powered persimmon leaf not only decreased the concentration of blood urea nitrogen in the plasma but also improved glomerular hypertrophy. Furthermore, the persimmon leaf significantly decreased the levels of hydrogen peroxide and lipid peroxide in the kidney. The activities of superoxide dismutase, catalase, and glutathione peroxidase and the mRNA expression of their respective genes were also increased in the kidney of persimmon leaf-supplemented db/db mice. Taken together, these results suggest that supplementation with the persimmon leaf may have protective effects against type 2 diabetes-induced kidney dysfunction and oxidative stress.

Anti-photoaging effects of Persimmon leaf were evaluated by anti oxidative reaction, stereomicroscopic and microscopic observations. The expression of photoaging skin related factors including mast cell tryptase, proliferating cell nuclear antigen (PCNA) and vascular endothelial growth factor (VEGF) was examined by immunohistochemical staining. Treatment of PLT-I, -II, -III prevented the wrinkle formation as well as epidermal hyperplasia, inflammatory cells, disruption of collagen in photoaged skin induced by UVB radiation. It also reduced the PCNA and VEGF expression in the UVB irradiated dorsal skin. Furthermore, it significantly decreased the number of mast cells in the UVB irradiated dermis(p<0.05 and p<0.01). On the effects of oxidative stress and antioxidant function on the treatment with water extract from Persimmon leaf tea(PLT), the activity of superoxide dismutase(SOD) was significantly increased in PLT-III group(p<0.05), and catalase(CAT) was significantly increased in PLT-I and PLT-III groups(p<0.05), and PLT-II group(p<0.001). These extracts showed relatively antioxidant activity and protective effect on UVB-induced oxidative stress in hairless mice(SKH-1). Our results suggest that Persimmon leaf tea may serve as an useful radical scavenging antioxidant and anti-photoaging skin agents in the UVB irradiated skin.

Effects of Persimmon Leaf Tea Extract, Green Tea Extract and Oolong Tea Extract on the Frequencies of Mutagen-Induced Sister Chromatid Exchange in Chinese Hamster Ovary Cells (감잎차, 녹차, 우롱차 추출물이 돌연변이 물질로 유발된 Sister Chromatid Exchanges 빈도에 미치는 영향)

  • The suppressing effects of crude extracts of three Korean teas, persimmon leaf tea extract (PLTE), green tea extract (GTE) and oolong tea extract (OTE), were studied on the induction of sister chromatid exchange (SCE) in cultured Chinese hamster ovary cells. When cells were treated with tea extract after mitomycin C (MMC) treatment, the frequency of MMC-induced SCEs were decreased at the high concentration $(1000\;{\mu}g/mL)$ of PLTE in the presence of S9 mix and at low concentrations $(20{\sim}80\;{\mu}g/mL)$ of PLTE in the absence of S9 mix, Whereas GTE and OTE showed suppressing effects on the MMC-induced SCEs at low concentrations $(10{\sim}20\;{\mu}g/mL)$ for OTE and $160\;{\mu}g/mL$ for GTE only in the presence of S9 mix. MMC-induced SCEs were decreased by post-treatment with each tea extracts with S9 mix in the G1 phase of the cell cycle. These results suggest that PLTE, GTE and OTE could have bio antimutagenic activities, and also suggest that PLTE might have unknown antimutagenic components which would be responsible for the inhibitory effect against direct acting mutagenicity.

Effects of extraction conditions on color quality and antioxidant properties of persimmon (Diospyros kaki Thunb.) leaf tea (침출 조건이 감잎차의 색 및 항산화 특성에 미치는 영향)

  • This study was carried out to investigate the effect of the extraction conditions for persimmon (Diospyros kaki Thunb.) leaf tea (PLT) on its color quality and antioxidant properties. The amount of persimmon leaf (PL) powder and pH influenced the PLT's color and antioxidant capacity. As the amount of PL powder in tea increased, lightness decreased while yellowness increased. The PLT with the highest amount of PL (10 mg/mL) exhibited the highest 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging activity and ferric reducing antioxidant power (FRAP). In addition, the PLT with the highest PL showed the highest total polyphenol and flavonoid contents. Subsequently, PLT was prepared using 10 mg/mL PL powder under varying pH conditions. As pH increased from 4 to 7, lightness decreased while redness and yellowness increased. Antioxidant capacity was determined using 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity and FRAP assay showed that lower pH exhibited higher antioxidant capacity. The PLT extracted under the lowest pH of 4 showed higher polyphenol and flavonoid contents than that extracted under higher pH conditions. Overall, PLT extraction using a solvent with pH 4.0 showed better antioxidant activities and higher amounts of polyphenolic compounds. Simultaneously, lesser lightness, redness, and yellowness were detected in PLT extracted under pH 4 conditions. In conclusion, to acquire a better functional health benefit in terms of antioxidant capacity, preparing PLT under pH 4 conditions is suggested.

Determination of Antioxidants Contents in Various Plants Used as Tea Materials (다류소재 식물류 중의 황산화물질 함량 분석)

  • Forty plants used as tea materials were chosen for determining the contents of total phenolics, selenium (Se), ${\beta}-carotene$, ${\alpha}-tocopherol$ and ascorbate. Total phenolics and ascorbate contents were analyzed colorimetrically. The Se contents were measured by hydride-atomic absorption spectrometry. The contents of ${\beta}-carotene$ and ${\alpha}-tocopherol$ were simultaneously determined by high performance liquid chromatography using separate detectors, UV for ${\beta}-carotene$ and FL for ${\alpha}-tocopherol$ analyses. The contents of these antioxidants were as follows (per 100 g dry plant); Contents of total phenolics in green tea leaf, black tea leaf, oolong tea leaf and instant coffee were about 7 g and the Se contents in corni fructus and arrowroot were found to be about $4{\mu}g$, which were the highest among all plants used. Contents of ${\beta}-carotene$ in eucommiae cortex, persimmon leaf and green tea leaf were 8587, 6222 and $3652\;{\mu}g$ respectively. The persimon leaf contained the highest ${\alpha}-tocopherol$ content (33 mg) and then followed by eucommiae cortex (26 mg), green tea leaf (16 mg) and black tea leaf (13 mg) in order. Ascorbate contents were found to be high in green tea leaf (199 mg) and black tea leaf (117 mg).

Physiological activities of water extracts from sweet persimmon leaves (단감 잎 추출물의 생리 활성)

  • This study was conducted to evaluate several physiological activities of 4 sweet persimmon cultivar leaf water extracts. One new cultivar ('Hongchu' (HC)]) and three traditional cultivars ('Sangseojosaeng' (UW), Japanese Uenishiwase; 'Seochonjosaeng' (NW), Japanese Nishimurawase; and 'Buyu' (FY), Japanese Fuyu) were used in this study. The HC extract showed significantly higher DPPH and ABTS radical scavenging activities, tyrosinase inhibitory activity, and acetaldehyde dehydrogenase activity than the other extracts, while the FY extract exhibited a relatively higher alcohol dehydrogenase activity. The HC extract contained higher amount of phenolics and ascorbic acid. These results suggest the possibility of high-quality persimmon leaf tea development using the new sweet persimmon cultivar, HC.
