  • 포플라 세포(細胞)의 Sucrose gradient 원심분리(遠心分離)에 나타난 세포(細胞)분획은 6개의 band 및 침전으로 분리(分離) 되었다. Peroxidase의 활성변화(活性變化)는 휴면기(休眼期)에 높아졌으며, 자발적(自發的) 휴면(休眠)이 타파되면서 낮아지기 시작하였다. 휴면기(休眠期)에 급격히 높아진 Peroxidase 활성(活性)은 Peroxidative 활성(活性)을 지닌 세포내(細胞內) Microbody에 존재(存在)하는 것으로 사료된다. Catalase의 활성(活性)은 11월(月), 12 월(月) 1월(月)의 휴면기(休眼期)에 활성(活性)이 낮았으며. IAA의 농도는 휴면초기(休眼初期)에 감소하기 시작하여, 12월(月)에 최저(最低)가 되었다. 자연휴면(自然休眠)이 타파된 1월(月)부터는 IAA 농도가 증가하기 시작하여 4월(月)의 발아기(發芽期)까지 계속되었다. 이와같은 Peroxidase, Catalase를 비롯한 IAA의 변화(變化)는 봄철의 발아(發芽) 및 재생장(再生長)과 가을철 휴면조절(休眠調節)에 영향을 미치는 것으로 사료된다.

  • Horseradish peroxidase(HRP) in organic solvent can be reactivated by evaporation. In order to measure the evaporation effect, the enzyme solutions were obtained by evaporation and dilution of organic solvent, respectively. Although two situations were thermodynamically identical, the activity from evaporation was higher than that from dilution. From the UV absorbance and the fluorescence intensity mesurements, it can be explained that reactivation of enzyme activity might be caused by reversible folding, and the enzyme obtained by evaporation was more refolded than that obtained by dilution.

  • Investigations of the effects of pickle product ingredients on lipoxygenase (LOX) and methemoglobin (MHG, a nonenzymatic oxidant) catalyzing oxidation of linolenic acid were conducted. In addition, activities of LOX, peroxidase (POD) and catalase (CAT) in dry spices used in pickle products were determined. Some commercial pickle brines were observed to inhibit oxidation of linolenic acid by LOX and MHG. The ingredients in pickle products, such as dill oil emulsion, onion concentrate, oil cassia, polysorbate 80 and turmeric acid, reduced LOX and MHG catalyzed oxidation. Lipoxygenase activity was present in garlic, mustard seed and red pepper. Only in mustard seed, peroxidase activity was observed. Catalase activity was observed in garlic, black pepper, allspice and red pepper.

  • The Isozyme activities of Lentinula edodes were studied as a preliminary study for genetic analysis after protoplast fusion. The presence of peroxidase, esterase, superoxide dismutase, acid phosphatase, alkaline phosphatase, alcohol dehydrogenase and ${\alpha}-amylase$ was examined. An intracellular buffer-soluble protein from the mycelia was used for enzyme analysis on nondenaturing polyacrylamide gels. The auxotrophs of Lentinula edodes were positive for peroxidase, esterase, superoxide dismutase and acid phosphatase. However, alkaline phosphatase, alcohol dehydrogenase and ${\alpha}-amylase$ were not detected. The esterase and peroxidase were not affected by the various culture age. Isozyme identification may be a useful tool after protoplast fusion.

  • Enzyme activities and phenolic compound were compared to investigate the physiological characteristics during shoot formation from diploid and haploid of Nicotiana tabacum w BY4. The Nakata medium with 1.0 mg/L IAA, 0.5 mg/L Kinetin and 3 g/L activie carbon was excellent to induce the haploid plants from the middle size anther within 30 days after culture. The MS medium with 0.5 mg/L 2,4-D was good for callus induction from leaf explants of diploid and haploid, and a lot of plane were regenerated from calli on the MS medium supplemented with 2.0 mg/L BAE Activities of peroxidase for both of diploid and haploid plane were the highest at 2.0mg/L BAP Activities of IAA oxidase and catalase of haploid Plants were high or than those of diploid plants. On the other hand, activity of peroxidase of haploid plants were lower than those of diploid plants.

  • This study was carried out to investigate the changes of enzymes and micro inorganic nutrients that is associated with flower senescence during flower development in tulip cultivars, ‘Apeldoorn’ and ‘Golden Apeldoorn’. Ribonuclease, peroxidase and protease activities were gradually increased from the stage of early flowering to later Polyphenol oxidase showed the highest activity at stage 5, which the flower was in full bloom indicating that it acts at an initial stage of flower senescence. The protease activity was different in the petal extracts during flower development between the cultivars ‘Apeldoorn’ (red petal) and ‘Golden Apeldoorn’ (yellow petal). This result suggested that protease might relate to pigment biosynthesis in petal of tulip. In contrast to the decrease of inorganic nutrients K, Mn, Zn and P contents during floral development, Ca, Mg and Fe showed the gradual increasement that is similar with ribonuclease, peroxidase and protease. It suggests that they have some interaction during flower senescence.

  • The white rot fungus Coriolus hiysutus decolorized several recalcitrant dyes. Four different types of dyes, including azo, triphenyl methane, heterocyclic, and polymeric dye, were treated by the mycelial preparation. Triphenyl methane dye, bromophenol blue lost over 95% of its color. Congo red and Poly R-478 were decolorized less than bromophenol blue, 57 and 55%, respectively. However, heterocyclic dye, methylene blue was not decolorized significantly and only red shift was observed. Extracellular laccase and peroxidase activities were appeared maximally in high level of dye decolorization media. In electrophoretic experiments, common active bands of laccase and peroxidase were found in all dye decolorized medium. These results indicated that the culture conditions which yield high levels of laccase and peroxidase activity lead to high levels of dye decolorization, and these two enzymes might be play an important roles in dye decolorization.

  • This study carried out to investigate activities of the superoxide dismutase(SOD), peroxidase(POD) and diterpene cyclase of rice plant treated by environmental stress conditions. The rice leaves of Ilpum was shown to have the most highest specific activity of POD. POD activity in the susceptible cv. Ilpum and Hwasung during the disease progress show a increase, while for a decreasing activity in the resistant cv. Dasan. SOD activity of rice plants treated by chitosan were increased as the treatment concentration increased. At the chitosan application, the SOD activity in the susceptible cv. Hwansung at the rice leaves 14days after treatment was higher in treatment than chitosan-untreated control. The activity of POD in the susceptible cv. Ilpum decreased as long as UV-B irradiation, while that in the resistant cv. Dasan increased. Futhermore, activity of that in Dasan were similar to as time on UV irradiation, Hwasung showed two times higher than untreated control at 40 minutes after UV-B exposure. The activity of diterpene cyclase in rice plants inoculated with Pyricularia oryzae has much higher tendency than UV-B irradiation. The activity of diterpene cyclase in the rice leaves gradually increased until 5 days after inoculation with Pyricularia oryzae.

  • The present study was carried out to investigate the change of peroxidase activity of soybean seed infected with Cerrospora kikurhii. The protein content, polyphenol oxidase activity and peroxidase isozyme pattern in health and infected soybean seed were also compared. 1. The peroxidase activity was substantially higher in the infected soybean seeds than that in the healthy seeds either cracked or not. No significant differences in protein content were recognized among the seeds tested. 2. No significant differences in peroxidase activities and protein contents were notified between healthy and infected seeds from the measurements on each parts of dissected seeds, cotyledon and seedcoat, however the peroxidase activity in the seed coat of the stained seed was 2.5 times to healthy seed. 3. The activities of polyphenel oxidase were undectable in both healthy and diseased seeds. 4. The electrophoretic Patterns of the Peroridase isozyme were the same between healthy and in footed seed. 5. Therefore, the increase of peroxidase activity in infected soybean seedcoat was mainly due to the biochemical reaction against the pathogen.

  • The experiments were conducted to investigate the activity changes of peroxidase, existence of isoenzyme and the changes of reserved substances in tomato under subatmospheric pressure storage condition. The results obtained were as follows: 1. Soluble fraction showed the highest peroxidase activity and followed by cell wall fraction, mitochondria fraction and ribosome fraction in that order. 2. Peroxidase activity was decreased during the ripening and senescence period in tomato. Especially, peroxidase activity in tomato was higher at a room temperature ($25^{\circ}C$) than at a low temperature ($15^{\circ}C$). The decreasing inclination was similar in both treatment. The peroxidase activity was higher in 380 Torr, than in 570 Torr. 3. At least, two isoperoxidases(Soluble or solubilized) were identified from different extraction procedures. Three of four isoenzymes were recognized from a vertical slab of polyacrylamide gel electrophoresis. 4. The changes of components in tomato under SAP were generally affected by temperature and pressure. Especially, quality of tomato stored at a low temperature ($15^{\circ}C$) and SAP (380 Torr.) was best during storage.
