E. coli-S. cerevisiae shuttle vector인 plasmid YRp7을 이용하여 B. amyloliquefaciens의 $\alpha$-amylase gene을 E. coli 내에 cloning하였다. 이때 제한 효소 Sau 3 AI에 의해 얻어진 $\alpha$-amylase gene의 크기는 약 1.95kb정도였으며 E. coli내에서 비교적 안정하게 유지되고 발현되었다. 재조합 plasmid p-EA24를 함유한 E. coli는 B. amyloliquefaciens의 약 65% 정도의 $\alpha$-amylase를 생성하였으며, 최적온도, pH, CaCl$_2$의 영향등 $\alpha$-amylase의 효소학적인 성질을 비교 조사해 본 결과 B. amyloliquefaciens의 $\alpha$-amylase와 동일하였다. 또한 E. coli에서 생성된 $\alpha$-amylase로 70% 정도가 periplasmic space에 존재하였으며 나머지는 세포 내부에 존재함을 알았다.
A 16 kilobase (kb) HindIII fragment of alkalophilic Bacillus sp. YC-335 containing a gene for xylanase synthesis was inserted at the HindIII site of pBR322 and cloned in Escherichia coli HB101. After subcloning of recombinant plasmid pYS52, the 1.5 kb fragment was found to code for xylanase activity, and the hybrid plasmid was named pYS55. The DNA insert of the plasmid was subjected to restriction enzyme mapping, which showed that pYS55 had single site for PuvII and SstI in the 1.5 kb insert fragment. Southern hybridization analysis revealed that the cloned gene was hybridized with chromosomal DNA from alkalophilic Bacillus sp. YC-335. About 64% of the enzyme activity was observed in the extracellular and periplasmic space of E. coli HB10l carrying pYS55.
Paenibacillus macerans 유래의 cycloinulooligosaccharide fructanotransferase (CFTase) 유전자(cft)를 Saccharomyces cerevisiae SEY2102에 발현시키기 위해 대장균과 효모의 shuttle vector인 pYES2.0에 subcloning 하였다. 구축된 pYGECFTN (8.6 kb) plasmid를 S. cerevisiae SEY2102에 형질전환하였고, uracil이 결핍된 SD 배지에서 선별하였다. cft 유전자는 선별된 형질전환체(S. cerevisiae SEY2102/pYCECFTN)에서 GAL1 promoter 조절하에 성공적으로 발현되어 cyclofructan(CF)을 생성함을 TLC로 확인하였다. 그러나, 균체 외로의 효소 분비는 이루어지지 않았고 cytoplasm보다 periplasmic space에 많이 존재하였다 S. cerevisiae에서 발현된 P. polymyxa유래 CFTase보다 P. macerans 유래 CFTase의 CF 생성이 image analyzer로 확인한 결과, 더 많음을 알 수 있었다. 효소반응 5분째부터 CF가 생성됨을 확인하였고, 최적온도와 최적 pH는 각각 $45^{\circ}C$와 pH 8.0로 나타났으며, $55^{\circ}C$까지 효소활성이 안정적으로 유지되었다. Dahlia tubers, chicory root, Jerusalem artichoke 등의 inulin 기질에 따른 반응산물 분석 결과, 모든 기질로부터 CF가 생산되었으며, dahlia tubers와 Jerusalem artichoke로부터 가장 효과적으로 생성되었다.
Candida (Pseudozyma로도 알려짐) antarctica lipase B(CAL-B)는 학문적으로 그리고 산업적으로 많이 활용되고 있다. CAL-B 자체에 대한 연구는 많이 진행되어온 반면, CAL-B 상동체에 관한 연구는 그리 알려진 바가 없다. 본 연구에서는 단백질 유사성 검색을 통해서 CAL-B의 상동체 탐색을 수행하였고, 6종의 단백질 서열을 찾았다. 해당하는 유전자들을 대장균에 대한 코돈 최적화를 수행하였고, 이를 바탕으로 유전자 합성을 진행하였다. 이들 유전자를 대장균 발현용 벡터에 클로닝한 후, 대장균 내에서 단백질 발현을 시도하여 이들 중 4종의 단백질이 성공적으로 발현되었다. 이들 단백질들이 가수분해 효소로서의 활성이 있는지 확인하기 위해서, 4-nitrophenyl acetate와 4-nitrophenyl butyrate를 반응기질로 하여 가수분해 반응성을 확인하였다. 이들 단백질들의 비활성(specific activity)값은 $(1.3-30){\times}10^{-2}{\mu}mol/min/mg$로 측정되었고, 이는 CAL-B의 비활성 수치보다는 다소 낮은 값에 해당하였다. (${\pm}$)-1-phenylethyl acetate의 가수분해 반응에 대한 입체선택성은 이들 상동체 효소들 중에서 Pseudozyma hubeiensis SY62에서 유래된 효소만이 CAL-B의 입체선택성과 유사함이 확인되었다.
본 연구의 목적은 위치특이적 펩타이드 및 단백질을 사용하여 재조합 대장균에서 활성형 인간 상피세포성장인자(hEGF)를 고효율로 발현할 수 있는 방법을 찾아내는 데 있다. 재조합 대장균내 cytoplasm 및 periplasm 영역에서 hEGF의 발현을 위해 각각 세개의 응합 펩타이드 및 단백질을 선정하여 상호 비교하였다. 재조합 대장균에서 hEGF의 발현유도시 대부분 불용성 단백질로 생산되는 현상을 극복하기 위해 cytoplasm영역에서는 ATS, thioredoxin, 리파제를 융합파트너로 사용하였으며 periplasm 영역에서는 foldase인 DsbA와 DsbC, 용융성 고발현 단백질인 maltose binding protein을 선택하여 사용하였다. Periplasm영역에서 발현유도를 시키는 융합단백질의 경우 cytoplasm영역에서의 발현양도 용융성 형태로 고발현 되는 것을 알 수 있었으며 전체적으로 약 2배가량의 용융성 형태로 발현되었다. hECF의 발현율을 가장 높일 수 있는 융합단백 질은 maltose binding protein이었으나 발현된 융합단백질의 24%가 불용성 단백질로 형성되어 활성형 형태로 얻는 데 한계가 있었으며, 활성형 형태로 hEGF의 발현을 위해서는 DsbA를 응합단백질로 사용한 경우에 18.1 mg/L로 가장 높은 발현농도를 보였다. Cytoplasm 영역에서 발현유도를 한 경우에는 ATS와 thioredoxin을 응합파트너로 hEGF를 발현한 경우 용융성 형태로 높은 발현율을 보였다. 특히 ATS와 같은 펩타이드를 N-말단에 융합시킨 경우 불용성을 방지하는 효과를 보여 이황화결합의 불완전성이나 소수성으로 인해 불용성 단백질로 발현되는 기존의 단백질을 활성형 형태로 발현하는데 될 수 있음을 확인할 수 있었다.
Glucose oxidase (GOD) is an oxidoreductase catalyzing the reaction of glucose and oxygen to peroxide and gluconolacton (EC 1.1.3.4.). GOD is a widely used enzyme in biotechnology. Therefore the production of monoclonal antibodies and antibody fragments to GOD are of interest in bioanalytics and even tumor therapy. We describe here the generation of a panel of monoclonal antibodies to native and heat inactivated GOD. One of the hybridomas, E13BC8, was used for cloning of a single chain antibody(scFv). This scFv was expressed in Escherichia coli XL1-blue with the help of the vector system pOPE101. The scFv was isolated from the periplasmic fraction and detected by western blotting. It reacts specifically with soluble active GOD but does not recognize denatured GOD adsorbed to the solid phase. The same binding properties were also found for the monoclonal antibody E13BC8.
In vitro analyses of type I signal peptidase activities require protein precursors as substrates. Usually, these pre-proteins are expressed in vitro and cleavage of the signal sequence is followed by SDS polyacrylamide gel electrophoresis coupled with autoradiography. Radioactive amino acids have to be incorporated in the expressed protein, since the amount of the in vitro expressed protein is usually very low and processing of the signal peptide cannot be followed by SDS polyacrylamide gel electrophoresis alone. Here we describe a rapid and simple method to express large amounts of a protein precursor in E. coli. We have analyzed the effect of ionophors as well as of azide on the accumulation of expressed protein precursors. Azide blocks the function of SecA and the ionophors dissipate the electrochemical gradient across the cytoplasmic membrane of E. coli. Addition of azide ions resulted in the formation of inclusion bodies, highly enriched with pre-apo-plastocyanine. Plastocyanine is a soluble copper protein, which can be found in the periplasmic space of cyanobacteria as well as in the thylakoid lumen of cyanobacteria and chloroplasts, and the pre-protein contains a cleavable signal sequence at its N-terminus. After purification of cyanobacterial pre-apo-plastocyanine, its signal sequence can be cleaved off by the E. coli signal peptidase, and protein processing was followed on Coomassie stained SDS polyacrylamide gels. We are optimistic that the presented method can be further developed and applied.
The PAS factor, whose gene has been cloned from V vulnifcus, is a protein secretion factor. Although the role of the PAS factor in Vibrio is still unknown, it may be involved with the bacterial protein secretion. The PAS factor is a 76 amino acid polypeptide, and its expression in E. coli cells makes the host cell membrane leaky, resulting in the excretion of periplasmic proteins into the culture medium. Highly expressed PAS factor is harmful to the cell, this may be due to a disruption of the membrane structure or function.
