A 2-year old female African hedgehog (Atelerix albiventris) weighing 645 grams was presented with a well circumscribed mass in the subcutaneous of the left shoulder. It measured $2{\times}2cm$ in size and had a hard texture. There were no clinical signs. In a fine-needle aspiration (FNA) cytological evaluation, mast cells were identified with Wright-Giemsa stain. Mast cell tumor was confirmed and characterized by surgical sampling and histopathological evaluation. Four stains (hematoxylin-eosin, toluidine blue, periodic acid-Schiff, and Safranin O) were examined in order to find an applicable stain for a mast cell tumor histopathological evaluation in hedgehog. This case showed that periodic acid-Schiff and toluidine blue staining were the most useful for assessment of a mast cell tumor in a hedgehog.
In an effort to obtain basic data of carbohydrate metabolism during spermiogenesis of the sexually-matured Jindo dog, the glycogen distribution in the testis was investigated by light and transmission electron microscopy. Periodic acid thiocarbohydrazide silver proteinate physical development(PA-TCH-SP-PD) staining method provided better results in the detection of glycogen granules from Sertoli cells and germ cells than the periodic acid schiff(PAS) staining method did. Pre-treatment of the tissue sections with ${\alpha}$-amylase elicited a significant decrease in PA-TCH-SP-PD stained granules, which suggested that the stained granules were of glycogen origin. High concentration of the glycogen granules were observed in the Sertoli cells, especially in its column, sheet-like processes, club-like processes, and tubular processes. The glycogen granules were unevenly distributed in some Sertoli cell columns. These results strongly indicated that the Sertoli cells of Jindo dogs showed vigorous activity of carbohydrate metabolism.
The olfactory organs of two African lungfishes, Protopterus amphibius and P. dolloi, were investigated using a stereo microscope and a compound light microscope and were described anatomically, histologically, and histochemically. Like other lungfishes, these species present the following general features: i) elongated olfactory chamber (OC), ii) anterior nostril at the ventral tip of the upper lip, iii) posterior nostril on the palate of the oral cavity, iv) lamellae with multiple cell types such as olfactory receptor neurons, supporting cells, basal cells, lymphatic cells, and mucous cells (MC), and vi) vomero-like epithelial crypt (VEC) made of glandular epithelium (GE) and crypt sensory epithelium. Some of these features exhibit differences between species: MCs are abundant in both the lamellar and inner walls of the OC in P. amphibius but occur only in lamellae in P. dolloi. On the other hand, some between feature differences are consistent across species: the GE of both P. amphibius and P. dolloi is strongly positive for Alcian blue (pH 2.5)-periodic acid Schiff (deep violet coloration), and positive with hematoxylin and eosin and with Masson's trichrome (reddish-brown staining), unlike the MCs of the two species which stain dark red with both Alcian blue (pH 2.5)-periodic acid Schiff and Masson's trichrome but respond faintly to hematoxylin and eosin. The differing abundance of MCs in the two lungfishes might reflect different degrees in aerial exposure of the olfactory organ, while the neutral and acid mucopolysaccharide-containing VEC, as indicated by staining properties of the MCs, is evolutionary evidence that P. amphibius and P. dolloi are the closest living relatives to tetrapods, at least in the order Dipnoi.
A 7-year-old, female snow goose (Anser caerulescens hyperboreus) with history of decreased activity for 2 month died in Daejeon Zoo Land in September 2006. At necropsy, granulomatous pneumonia and hepatomegaly with multiple cysts were observed. Small masses were found in the spleen. Microscopically, fibrinous pneumonia distributed in most of the lung lobe with pulmonary edema and congestion. Especially, granulomatous inflammation with numerous multinucleated giant cells was observed around the dilated bronchi. To confirm the diagnosis, acid-fast (Ziehl-Neelsen method) and periodic acid-Schiff (PAS) staining was performed. Acid-fast staining showed red bacterial colony indicating tuberculosis. PAS staining was also positive enough to diagnose aspergillus spp. co-infection that was an opportunistic fungi occurring in immuno-compromised animals. Based on the above results, we confirmed that the case submitted was diagnosed as avian tuberculosis.

Formaldehyde use is associated with serious health risks, which can affect medical personnel and technicians. Therefore, we investigated the efficacy of an alternative fixative, with respect to two types of formalin fixatives, by hematoxylin and eosin (H&E) staining, periodic acid Schiff (PAS) staining, immunohistochemical (IHC) staining, and RNA extraction. For H&E staining, the circular nucleus was stained dark blue by the basic dye hematoxylin and the cytoplasm was stained red by the acid dye eosin in all three fixative samples. No difference was found in the Duksan General Science (DGS), Sigma-Aldrich, and Core-Fix fixative samples (Corebiotech) used to fix kidney tissue, after PAS staining. IHC staining showed that CD4 was significantly increased in the lippolysaccharide (LPS)-treated group compared to the control group (vehicle), confirming the changes in specific molecules. The quantity and quality of RNA from tissues fixed in the three types of fixatives were evaluated. The average concentration of RNA was 106 ng/µL and average purity at A 260/280 ratio was 1.7~2.0, regardless of fixative used. For quality of protein, glyceraldehyde 3-phosphate dehydrogenase (GAPDH) protein was confirmed by Western blotting. In conclusion, Core-Fix can be used as a fixative for pathological tissues, in histological and molecular diagnoses.
A leucine aminopeptidase has been isolated from the culture medium of the soil fungus, Aspergillus flavus. The enzyme was found to be a glycoprotein, as judged by electrophoresis analysis and the subsequent staining by the periodic acid-Schiff's reagent. Carbohydrate moieties could be cleaved by N-glycosidase, but not by O-glycosidase, indicating that the glucans are linked to the asparagine residue in the protein. Removal of N-glucans was observed without prior denaturation of the protein, implying that the N-glycosidic linkage is exposed and accessible to glycosidase. When the activity of native or deglycosylated enzyme was measured in the presence of various metal ions, removal of carbohydrates increased the aminopeptidase activity of the enzyme.
A female slaty-backed gull (Larus schistisagus) died suddenly without apparent clinical signs. At necropsy, well demarcated 7 to 10 mm yellow to white nodules were presented at the lungs and thoracic cavity. Microscopically, multifocal necrotic granulomas were observed in the lung tissue and amorphous acidophilic fibrin were accumulated in the granuloma and normal alveolar space. Periodic acid-Schiff (PAS) staining demonstrated numerous pinkish red branched hyphae embedded in the center of these granulomas. According to fungal culture using Sabouraud's dextrose agar plate, Aspergillus fumigatus was isolated from lung lesions. This is the first report for pulmonary aspergillosis of wild slaty-backed gull in Korea.
Histochemical observations on the effect of gold alloys crown margin in the periodontal membrane were caried out by use of rabbits. The animal were setted with gold alloys crown on the incisor teeth. The cervical margins were given extence into gingival sockets. The animals were sacrificed at the 30 days experimented period. Specimens of tissue were obtained from the mandibular incisor teeth with jaw. All tissues were fixed immediately in 10% neutral formalin solution. Different histochemical staining methods for the determination of fibrous components were the Mallory-Heidenhain Bielschowsky-Gomori, Gomori's aldehyde fuchsin. Periodic acid-Schiff reaction and Hematoxylin eosin stanin method. The results were as follows. The periodontal membrane is composed primarily of collagenous fibers and fibrous component run functional arrangement but in lower gold content groups, horizontal fiber groups were irregularly run. Elastic fibers were obligue run in experimental groups. There were no significant differences in reticularfibers in distribution.

A renal oncocytoma was diagnosed in an 8-year-old female Maltese dog with a history of renal cysts. Tumor cells were not detected until six months after observation of renal cysts. Nephrectomy was performed to treat the neoplasia. Tumor-like masses with numerous nodules were observed in the inner surface of cysts present in the caudal part of the left kidney. Histologically, the tumor consisted of cells with abundant eosinophilic cytoplasm. The diagnosis was based on histological features, periodic acid-Schiff reaction, and immunohistochemical cytokeratin staining. Based on a literature review, this is the first canine renal oncocytoma case reported in Korea.
We report large tumors of the left thoracic cavity, which arose from the ribs, were diagnosed as chondrosarcoma in a 4-year-old male mixed dog. The dog was presented with swelling in the left side of the chest wall and lameness. The masses were found to be circumscribed, whitish-grey, large and firm. The cut surface revealed whitish-grey lobules of varying size with cartilaginous consistency. Those were subsequently metastasized to the lung and mandibular lymph node. Histologically, the tumors were made up of clusters of chondroid cells having pleomorphic hyperchromatic nuclei with occasional mitosis. A special and immunohistochemical staining was performed to confirm the diagnosis. Chondroid matrix in tumor showed a positive reaction for alcian blue-periodic acid-Schiff staining. Immunoreactivity to S-100 proteins was present in the cytoplasm and/or nucleus of chondrocytes in tumor. The final diagnosis was grade III chondrosarcoma in the rib, considering histological features in grading criteria.
