들깨 조직으로부터 callus 유기에 따른 지질 및 단백질 조성의 변화 (Changes of Protein and Lipid During Callus Induction and Plant Regeneration from Perilla frutescens)

  • The biochemical change during regeneration of perilla callus were investigated by comparing total protein and lipid contents, protein band pattern in SDS-PAGE, and fatty acid composition in the calli cultured for various period(0, 1, 3, 5 and 6 weeks) Calli were induced from cotyledon and hypocotyl explants of peplants of perilla on perilla on MS medium containing BA(0.5 mg/L) and NAA(0.5mg/L). The protein contents reached the peak at 3 weeks after induction of calli, and then was decreased. Total lipid contents was decreased as the culture period increased. The band pattern of polypeptides showed that 30KD and 45KD polypeptides and 22KD and 45KD polypetides were major proteins in the cotyledon and hypocotyl explants, respectively. However increase of culture period only 30KD protein was highly accumulated.

Determination of Seed Lipid and Protein Contents in Perilla and Peanut by Near-Infrared Reflectance Spectroscopy

  • Near-infrared reflectance spectroscopy (NIRS) was used to estimate the lipid and protein contents in ground seed samples of perilla (Perilla frutescens Brit.) and peanut (Arachis hypogaea L.). A total of 46 perilla and 80 peanut calibration samples and 23 perilla and 46 pea. nut NIRS validation samples were used for NIRS equation development and validation, respectively. Validation of these NIRS equations showed a range of very low bias (-0.05 to 0.13 %) and standard error of prediction corrected for bias (0.224 to 0.803%) and very high coefficient of determination ($R^2$) (0.962 to 0.985). It was concluded that NIRS could be adapted as a mass screening method for lipid and protein contents in perilla and peanut seed.

  • 들깨묘는 재배가 용이하며 건강채소로 애용되고 있다. 본 연구에서는 발아 후 2일째의 들깨묘에 대하여 세포 상해를 일으킬 수 있는 NaOH/SDS 용액을 처리하고 페놀 화합물을 처리한 재조합 Agrobacterium 세포를 이용하여 형질 전환을 수행하였다. 형광분석에 의한 GUS 유전자발현 분석에서 50 mM acetosyringone 처리 및 0.5% NaOH /0.01% SDS 처리의 복합처리에 의하여 가장 높은 GUS활성이 나타났으며 상승효과를 제시할 수 있었다. 이러한 결과는 hepatitis B virus antigen (HBsAg) 단백질 발현으로 성공적으로 확인할 수 있었다.

참깨와 들깨 단백질의 기능성에 관한 연구 (Studies on the Functional Properties of Sesame and Perilla Protein Isolate)

  • 참깨와 들깨 단백질을 알칼리에서 추출시킨 후 참깨는 pH6.0, 들깨는 pH4.0에서 침전시켜 분리 단백질을 제조하여 그들의 기능성 즉, 용해도, 유화성, 기포성, 유지 및 수분 흡착력 등을 pH와 염농도를 달리하여 측정하였다. 단백질 용해도는 참깨 단백질이 pH6.0에서, 들깨와 대두 단백질은 pH4.0에서 가장 낮아 등전점을 보이었고, pH2.0과 pH8.0 이상에서는 크게 증가하였다. 0.1M NaCl 첨가시에는 참깨 단백질과 들깨 단백질의 경우 전 pH 범위에서 단백질 용해도가 증가하였다. 유화력은 각 시료 단백질의 등전점 부근에서 가장 낮았으며 0.1M NaCl 첨가시에는 모든 시료들의 유화력이 증가되었으나 0.5M NaCl 농도에서는 오히려 감소되는 경향이었다. 유화안정성은 유화력과 비슷한 크기로 나타나, $80^{\circ}C$에서 30분 가열에서도 이들 단백질이 열에 안정함을 보이었다. 기포 형성력은 참깨와 들깨 단백질이 대두 단백질에 비해 현저히 낮았으며, 0.1M NaCl과 0.5M NaCl 첨가시에는 모두 그 값이 증가되는 경향을 보이었다. 기포 안정성은 대두 단백질이 10분 이내에 크게 감소된 반면 참깨와 들깨 단백질은 30분까지 완만하게 감소하였다. 유지 흡착력은 들깨 단백질이 가장 높은 값을 보였고, 수분 흡착력은 시료간에 차이가 없었다.

마우스에 있어서 부패들기름 식이가 면역반응에 미치는 영향 (The Effect of Rancid perilla oil diet on the Immune Response in Mice)

  • The effect of rancid perilla oil on the immune response in mice was studied. ICR male mice were divided into 5 groups and were fed on the experimental diets for 4 weeks. Mice were sensitized and challenged with sheep red blood cell. Immune responses were evaluated by antibody production, Arthus reaction, delayed type hypersensitivity (DTH), Rosette forming cell and macrophage activity. Biochemical items were measured by serum protein and serum albumin. The weight of spleen, thymus and liver were measured. The rancid perilla oil diets decreased humoral and cellular immune responses, the number of peripheral circulating white blood cells and total protein and serum albumin. These results showed that the high rancid perilla oil diet decreased more humoral and cellular immune response, the number of peripheral circulating white blood cells, and total protein and serum albumin than the low rancid perilla oil diet did.

들깨류사 종.속 수집 유전자원의 잎품질 및 지방산 조성 (Leaf Quality and Fatty Acid Composition of Collected Perilla Related Genus and Species Germplasm)

  • 들깨관련 종·속 수집 유전자원 들깨(Perilla frutescens var. japonica Hara), 자소(Perilla frutescens var. acuta Kudo), 축면자소(Perilla frutescens var. crispa Decaisne), 청자소(Perilla frutescens var. Kudo for viridis Makino), 들깨풀(Mosla punctulata Nakai), 산들깨(Mosla japonica Maxim) 및 쥐깨풀(Mosla dianthera Maxim)에 대한 잎품질 및 지방산 조성을 조사한 결과를 요약하면 아래와 같다. 1. 수집한 들깨관련 종·속 가운데 Perilla속 유전자원이 Mosla속 유전자원보다 주경엽수가 많았고 길이에서 3배 정도, 폭에서 약 5배 정도 컸었다. 2. 들깨관련 종·속 모든 수집 유전자원이 들깨보다 방향성 정도가 높아 방향성 개량육종의 교배재료로서 활용가치가 기대된다. 3. 잎의 유연성 면에서는 Perilla속 유전자원이 Mosla 속 유전자원보다 부드러웠다. 4. 종실성분함양에서 들깨에 비하여 들깨관련 수집 종·속의 기름과 단백질함량이 낮았으나 리놀렌산 함량이 높아서 공업용 또는 의약용을 위한 고리놀렌산 들깨품종개량의 교배재료로 활용가치가 기대된다. 5. 수집 Perilla속 유전자원의 불포화지방산인 리놀렌산과 기름함량, 단백질함량 그리고 포화지방산과는 부의 상관을 보였다.

들깨 종자의 발아에 따른 단백질 및 지방의 조성 변화 (Changes of Protein and Lipid Composition During Germination of Perilla frutescens Seeds)

  • To investigate changes in protein and total lipid contents, seed storage protein pattern, and fatty acid composition of germination perilla(Perilla frutescens) seeds. Also, the corresponding value components in cotyledons, hypocotyles and roots were measured according to germination stage. The results were summarized as follows ; During germination, pertein and total lipid contents of Yepsilldalggae and Kwangyang cultivar were decreased continuously. In particular, protein contents rapidly decreased to the 3 days after germination(DAG), and then total lipid contents rapidly decreased between 3 DAG and 10 DAG. In changes of protein and total lipid contents of cotyledons, protein contents of Yeupsildalggae was increased during the germination, but Kwangyang cultivar was decreased during the same periods. The total lipids contents of Yeupsildalggae and Kwangyang cultivar were decreased during the germination. According to SDS-PAGE analysis, there was no detectible polypeptide bands on the gel before seed germination suggesting that this may be due to the rapid degradation of the storage proteins in the mature seed by hydrolyttic enzymes during the stage. During germinatation , the polypeptide band with 27$\sim$28KD of Yeupsildalggae and Kwangyang cultivar were accumulated gradually. In changes of fatty acid composition of total lipid of Yeupsildalggae and Kwangyang cultivar , saturated fatty acids such as palmitic acid and stearic acid increased during the germination. On the other hand, unsaturated fatty acid such as linoleic acid and linolenic acid decreased during the same periods. However, oleic acid increased to the 5 DAG, and then was repidly decreased.

흰깨, 검은깨, 들깨 중의 무기질 및 Saponin 함량 (Mineral and Saponin Component in white Sesame, black Sesame and Perilla Seed)

  • the purpose of this paper is to study on the proximate composition, mineral and saponin content in white sesame, black sesame, and perilla seed. For this purpose, we have compared six different materials: white-raw-sesame, white-roast-sesame, black-raw-sesame, black-roast-sesame, raw-perilla seed and roast perilla seed, and have come to the following results. The crude fat content was the highest in white-raw-sesame(55.3%). In all the six samples, the crude fat content in raw seeds was all higher than that in roast seeds. The crude protein content was the highest in the roast perilla seed(24.6%), and in the six samples, the crude protein content in roast seeds all higher than that in roast seeds. The total sugar content was found to be the highest in the roast perilla seed(8.29%). The reducing sugar content was higher in raw perilla seed(1.57%) than in other sample materials. The ash content was the highest in black raw-sesame(5.93%), and that percentage rates was the same as that of FAO and of Japan. Minerals like Cd. Mn. Cu. Na. Mg. Pb. and Ca. were found to be contained more in black sesame than in other sample materials. The minerals contained most in white sesame were Zn.(61.6ppm) and Fe(49.4ppm), and K was contained a little more in perilla seed than in the others. The sample materials which contain saponin most were white-roast-sesame(0.34%) and black-roast-sesame(029%).

밀양지역 들깨품종별 잎의 아미노산 조성 및 향기성분 비교 (Comparative Studies on the Amino Acids and Flavor Compounds Among Some Varieties of Perilla Leaves Cultivated in Miryang Area)

  • 우리나라 들깻잎의 주산지인 밀양지역에서 재배하고 있는 4종의 품질 특성을 조사하고자 아미노산과 휘발성 향 성분을 분석하여 비교하였다. 단백질구성아미노산에서 주요 아미노산은 aspartic acid, glutamic acid, glycine, alanine, arginine, 및 threonine 등 이었고, 그 외에 lysine, leucine, isoleucine 등도 다소 검출되었으나, tyrosine과 함황아미노산인 cysteine과 methionine의 함량이 가장 적 게 나타났으며, 유리 아미노산은 4종 모두에서 약34종이 검출되었다. 그리고 유리아미노산의 조성과 단백질구성아미노산의 조성에서 품종간의 차이는 거의 없는 것으로 나타났다. 들깻잎의 휘발성 향기성분을 GC-MSD로 분석한 결과, 잎들깨1호는 52종, 엽실들깨는 48종, 그리고 밀양9호와 YCPL은 47종이 분리되어 검출되었다. 깻잎의 휘발성 화합물중에서 가장 많은 함량으로 검출된 perilla ketone은 엽실들깨 $551.42\;{\mu}g/g$, 잎들깨1호 $301.59\;{\mu}g/g$, YCPL $187.00\;{\mu}g/g$, 그리고 밀양9호는 $145.75\;{\mu}g/g$이었으며, 그 외 3-hexen-1-ol, trans-2-hexenal, $\beta-caryophyllene$ 그리고 $\alpha-farnesene$등이 검출되었으나 이 화합물들의 함량은 $20\;{\mu}g/g$을 초과하지 않았다.

Purification and Partial Characterization of a Peroxidase from Perilla Callus

  • Cotyledons of perilla6 were cultured on MS medium containing 0.5 mg/l NAA and 0.5 mg/l BA for 7 weeks. The activity of perilla peroxidase was observed to increase following culture stages as assessed by peroxidase assay. A peroxidase (POD) was purified from perilla tissue cultured on MS medium for 7 weeks. The peroxidase was purified using ion exchange and gel nitration chromatography. The perilla peroxidase had a molecular mass of 30 kDa by SDS-PAGE. We showed that the N-terminal amino acid sequence of this protein shared 67% identity with the tea peroxidase. As indicated by SDS-PAGE, the banding pattern of the 30 kDa polypeptide present in total soluble protein from perilla tissue was increased following culture stages. Immunoblot analysis indicated that perilla peroxidase protein appeared after 3 weeks of perilla tissue culture, and continued to increase with extended duration of tissue culture for at least 7 weeks.
