For the complete and accurate amino acid determination of protein and food samples, 3 different hydrolysis procedures have been conducted in parallel for each sample, which include the alkaline hydrolysis for tryptophan determination, performic acid oxidation prior to the acid hydrolysis for the determination of cysteine and cystine, and the 6N HCl hydrolysis for the determination of the rest of amino acids. In the present study, amino acid concentrations obtained from the modified single hydrolysis procedure were compared with the values from the conventional hydrolysis procedures in casein and nine food and composite dish samples. In most of the samples tested, the modified single hydrolysis procedure gave significantly higher values of cysteins and cystein compared to the performic acid oxidation method, but resulted in a considerable destruction of tryptophan in food and composited dish samples. There was no consistent difference in the rest of amino acid concentrations between the two hydrolysis systems. Therefore, for complete amino acid determination of various foods and composite dishes, the single hydrolysis method may replace the 6N HCl hydrolysis and performic acid oxidation methods, and thereby reduces 3 hydrolyses to 2 steps with much higher recoveries of the sulfur containing amino acids.
Soluble keratose(S-keratose) could be obtained by oxidizing the wool keratin with performic acid. S-keratose/PVA solutions were made by dissolving them in the formic acid and S-keratose/PVA web of sub-micron size was made by electro-spinning technique. SEM, elemental analysis, FT-IR, X-ray diffractometry, and TGA were used to characterize the properties of S-keratose/PVA solutions and fibers. As the S-keratose content in S-keratose/PVA solution increased, viscosity of solution decreased while electrical conductivity has increased. The fiber diameter has decreased with increasing the S-keratose content. S-keratose/PVA ratio in fibers were similar to the ratio in the solutions. The crystalline structures of S-keratose and PVA existed separately in the electrospun webs. Thermal stability of the web increased with the PVA content increasing.
Oxidation method was used to extract S-keratose from wool. Wool was treated with performic acid and soluble fraction(S-keratose) was collected by evaporating the solvent. S-keratose and Nylon 6 were dissolved in formic acid at the ratio of 100/0, 80/20, 50/50, 20/80, and 0/100, and S-keratose/Nylon 6 web of sub-micron size was made by electro-spinning technique. SEM, EA, FT-IR, XRD, and TGA were used to characterize the properties of S-keratose/Nylon 6 solutions and electrospun fibers. As the Nylon 6 content increased, viscosity, conductivity of the electrospinning solution and the diameter of spun fiber increased. Electrospun nonwoven webs have the same S-keratose/Nylon 6 ratios of the spinning solutions. The crystalline structures of S-keratose and Nylon 6 existed separately in the electrospun webs. Thermal stability of the webs increased due to Nylon 6 content.
TFormic acid-hydrogen peroxide (or performic acid) pulping process needs milder reaction condition than other chemical pulping process. Two-step formic acid-hydrogen peroxide pulping process can produce the chemical pulp with similar pulp yield and lignin content compared with soda-anthraquinone process. Formic acid-hydrogen peroxide pulp can be produced less xylan content than other alkaline pulps, which favor for dissolving pulp production. Formic acid-hydrogen peroxide pulp showed better response beating than soda-anthraquinone(AQ) pulps with reaching target freeness with less beating. Also, formic acid-hydrogen peroxide pulp had better tensile index at similar freeness level compared with soda-AQ pulps.
다관능성 에폭사이드 PPE(Polyglycerol Polyglycidyl Ether)를 양모섬유에 처리하였을 때 부가반응의 특성을 고찰하였다. 에폭사이드 PPE에 의한 부가반응이 촉매작용을 하는 NaSCN 존재하에서 이관능성 EGDE보다 높은 부가율을 얻을 수 있었다. 에폭사이드 부가반응은 고온에서 반응이 잘 일어나며 에폭사이드 처리욕의 pH는 반응 초기 급격히 상승했다가 이후 점차 초기 값까지 안정화된다. 처리한 양모섬유를 FTIR의 KBr-pellet법과 ATR법에 의해 분석하여 absorbance ratio를 비교하였을 때 부가반응은 표면으로의 그라프트 반응이 우세함을 알 수 있었으며 이러한 결과는 SEM 관측을 통하여 확인할 수 있었다. Performic acid/Ammonia solubility를 에폭사이드 처리 양모섬유에 적용하여 용해도 변화를 보았을 때 부가된 양이 많을수록 용해도 값이 감소하며 특히 15.6%를 중심으로 용해도 변화가 둔화되었다. 아미노산 분석으로 에폭사이드가 반응한 아미노산이 cystine임을 알 수 있었으며 단백질 사슬간에 가교반응이 이루어졌음을 확인할 수 있었다.
