Priming Effects on Germination of Aged Tobacco Seeds

  • Tobacco (Nicotiana tabacum L. cv KF109) seeds were artificially aged in a controlled environment of 45$^{\circ}C$ and 80% relative humidity condition for different duration up to 14 days before priming in polyethylene glycol 6000 solution of equivalent osmotic potential of -0.8 MPa for 8 days at $25^{\circ}C$. The seeds aged only and primed after aging were germinated at 15$^{\circ}C$ and $25^{\circ}C$ to observe the priming effects on the germination of aged seeds at different temperature. The germination percentage of the aged seeds was rapidly dropped starting from 8 days of aging and mean germination time (T$_{50}$) was greatly increased, particularly in germination at 15$^{\circ}C$. The germination capacity was greatly restored in the primed seeds after aging, particularly in the seeds of longer aging and germinating at 15$^{\circ}C$.>.

The Variation of Cone, Seed and Germination Characteristics of Picea jezoensis (Siebold & Zuccarini) Carriere Populations in Korea (한국산(韓國産) 가문비나무 자생집단(自生集團)의 구과(毬果), 종자(種子) 및 발아특성(發芽特性) 변이(變異))

  • This study was conducted to investigate the variation of cone, seed and germination characteristics among populations and among individuals within populations of Picea jezoensis(Siebold & Zuccarini) Carriere distributed in Korea. Cone collected from 25 trees in two natural populations(Mt. Jiri and Mt. Dokyu) and their thirteen cone and seed characteristics as well as three germination behaviors were analyzed. Statistical analysis showed significant differences among populations and among individuals within populations in 9 traits except for seed breadth, seed weight, index of seed wing and mean germination time. Generally, morphological variation of cone and seed wings from Mt. Jiri population has smaller and longer than those of Mt. Dokyu population. Percentage of sound seeds and Tetrazolium test from Mt. Jiri population showed 1.79 and 1.87 higher values than Mt. Dokyu population, respectively. The maximum germination percentage was obtained at the optimum temperature of $20^{\circ}C$ and in this case, mean germination time and germination rate showed 7.5 days and 2.9 ea./day, respectively. In seed germination behaviors, percentage(40.7%) and rate(0.90 ea./day) of germination in Mt. Jiri population were more higher and faster than those of Mt. Dokyu population(17.7%, 0.37 ea./day). According to correlation analysis, P. jezoensis populations with small restricted distribution may have been reduced because seed qualities were correlated with increased levels of inbreeding and disproportion flowering.

Effect of KNO3 Priming on Various Properties of Kenaf Seed under Non-Saline and Saline Conditions

  • The main objective of this study was to increase the germination percentage of kenaf seeds with less number of times under non-saline and saline conditions. Therefore, the first goal was to assess the response of kenaf seeds to NaCl. The second goal was to evaluate the effects of $KNO_3$ on kenaf seed germination. The germination percentage exhibited a decreasing tendency in germination rate. Plant dry weight was approximately 0.2 g in all treatments at 5 days after germination. As time passed, the electrical conductivity (EC) value of hydro-priming (HP) consistently increased by 8.7 mS/cm at 24 hours of immersion. However, seeds primed with $KNO_3$ showed no difference in EC values even as times passed. Regarding the priming effect, priming in 100 mM $KNO_3$ concentration for 12 hours increased germination up to 85% in $H_20$ solution and in 0 mM $KNO_3$ concentration upto 73.8% under 0.3% NaCl solution, compared to that of Control. Germination synchronization, shoot length, and leaf unfolding of primed seeds were greater than those of the Control. In addition, main root and hair roots appeared more rapidly in the treated seeds and were more abundant compared to that of the Control. The T50 (times to reach 50% of the final germination percentage) of the Control in both $H_20$ and 0.3% NaCl solutions was 18 and 22 hours, respectively. However, when treated $KNO_3$ priming (0 to 100 mM) in $H_20$ and 0.3% NaCl solution, 9 hours was sufficient to reach T50. Primed (hydro-priming and $KNO_3$) seeds had a lower MDG (mean days untill germination; 0.6-0.62) compared to that of the Control (1.13-1.31) in $H_20$ and 0.3% NaCl solutions. Regarding dry weight of plants after priming, an increasing tendency after the priming treatment in the H20 solution was observed. Furthermore, no significant difference in plant dry weight under 0.3% NaCl stress was observed between the Control and primed seeds. Taken together, the results suggest that 50-100 mM $KNO_3$ priming for 24 hours optimize seed germination rate in less number of times of exposure with great vigor. Therefore, it is recommended for kenaf seed invigoration before planting.

Effect of ultrasonication, light and liquid smoke treatment on germination of lettuce seeds

  • Seed priming leading to improved emergences, germination speed and uniformity under unfavorable conditions without loss of longevity are the best level of techniques. We studied the possibility to obtain primed seed with reduced mean germination time (MGT) and uniformity but with simple, fast and effective methods. The experiments were carried out at $15^{\circ}C$ for 16 hours with priming treatment. To compare the effects of each treatment, we used 200mM $CaCl_2$ priming or hydro priming or no imbibition after ultrasonication or red light treatment. The light treatment was performed by irradiating 2000 lux of red light for 15, 30, 60, and 120 minutes. Addition, in order to investigate the effect of the priming solution, treated at 200mM $CaCl_2$ for 8 hours. Ultrasonication treatment was performed for 5, 10, and 20 minutes at exposures of 13.0, 17.3, and 21.6 kHz during priming. For liquid smoke (LS) treatment, seed soaked in 0%, 0.5%, 1.0%, 5.0% and 10.0% of diluted water and 200mM $CaCl_2$ solution. After each treatment, the seeds were dried to moisture content ranged 5-8% at $25^{\circ}C$ for 24 hours. The effect of seed treatment was evaluated with germination percentage (GP), MGT, germination index(GI), germination rate(GR), Germination Uniformity(GU) and heath seed percentage(HS). For several factors tested, we found that the desired germination improvement could be obtained by treating the seed with ultrasonication at 17.3 kHz for 5 minutes in water or red light exposure at 2000 lux for 120 min in water that resulted very similar to those used to 200mM $CaCl_2$ priming for 16 hrs. However, LS treatment showed no improvement in all diluted solution. Therefore, the methods applied ultrasonication and red light treatment showed high potential for fast and easy treatment avoiding pollution of salt solution waste.

Changes of Chemical Composition in Hybrid Seed during Germination and Seedling Growth of Hybrid Rice (1대잡종 벼의 발아시 종실내 성분함량의 변화와 발아율 및 초기생육)

  • This study was carried out to investigate changes of chemical composition in seed, germination percentage, and seedling growth of two F$_1$ hybrid rices(V20A/Cheongcheongbyeo, IR62829A/Cheongcheongbyeo) during germination compared with their parents (V20B, IR62829B and Cheongcheongbyeo) and check variety(Samgangbyeo). F$_1$ hybrid rice showed higher content of free amino acid, inorganic elements, ($P_2O_5$, $K_2$O, MgO) and total sugar, and higher a-amylase activity in brown rice than those of their parents and check variety during germination. Therefore$F_1$hybrid rices showed hybrid vigor in germination percentage at 33 hour and 48 hour after sowing. $F_1$ Hybrid rices also showed /heterosis in seedling growth was different between V20A/Cheongcheongbyeo and IR 62829A/Cheongcheongbyeo. There were positive significant correlation between a-amylase activity and germination percentage, and seedling growth.

The Effects of Water Level and Temperature on Seed Germination and Early Seedling Development of Rice (Oryza sativa I.)

  • The application of direct seeding cultivation reduces time, labor, and cost. However, this application often has poor seedling establishment and leads to lower yield as compared to transplanting system. The tolerance to anaerobic and low temperature germination is important to improve seedling establishment and the wide-spread application of direct seeding method. This study was carried out to evaluate the responses of three japonica cultivars to different temperatures (15℃, 18℃, 21℃, 24℃, and 27℃) and different flooding levels (1 cm, 4 cm, 7 cm, 10 cm) during germination and seedling development. The mean survival percentage significantly increased (P<0.05) when the flooding level decreased and when temperature increased. There were significant effects of the interaction between temperature and water depth on survival percentage and seedling height. When temperature decreased from 27℃ to 15℃, the germination duration significantly increased from 6.4 days to 16.3 days while the germination speed, survival percentage, and seedling height decreased from 5.3 seeds day-1, 61.9% and 190.6 cm to 2.2 seeds day-1, 33.2%, and 47 cm, respectively. The increase in temperature under submergence condition was associated with the increased expression of Amy3D and ALDH2a but the decreased expression of ADH1 and PDC1. The results of this research would be used for further studies and breeding programs to improve rice seedling establishment and the application of direct seeding cultivation.

Effect of Seed Priming on the Enhancement of Seed Germination in Cool Season Turfgrass (Priming 처리가 한지형 잔디류의 발아 증진에 미치는 영향)

  • This study was carried out to identify the optimum condition for priming, as a series of experiments for the enhancement of germination percentage and germination synchronization in cool-season turfgrass seeds. The optimum priming conditions to maximize the germinability in both Kentucky bluegrass and Tall fescue was a 1 day treatment of 100 mM $K_3PO_4$ at $25^{\circ}C$. The seeds treated with the optimum priming condition enhanced the germinability compared to control, and shortened the time for germination with reduced $T_{50}$ and mean daily germination (MDG). However, as the concentration and treatment period of NaOH or KOH increased, significantly decrease in germination percentage was observed. Germination percentage in Creeping bentgrass and Bentgrass was below 10%, regardless of priming treatment. However, the priming treatment with 200 mM $KNO_3$ for three days improved the germination up to 6%, which was not a statistically significant level. The most effective priming period for Kentucky bluegrass and Tall fescue was one day treatment, but the germinability was suppressed as the treatment period was extended until six days. For priming chemical treatment, germinability was improved with the concentration of 100 mM, while it was reduced at higher concentrations of 200 mM and 300 mM.

Effect of $GA_3$, Kinetin and Physical Treatment on the Seed Germination of Zanthoxylum piperitum A.P. DC. ($GA_3$, Kinetin 및 물리적 처리가 초피나무 종자의 발아에 미치는 영향)

  • This study was conducted to improve germination ratio of Zanthoxylum piperitum A.P. DC. seeds. Stratification for 60 days after scarification of seed with sand was germination percentage to 5.4% and $GA_3$, 50ppm for 24 hrs after scarification of seed with sand showed 8.9%. Soaking the seeds in $GA_3$, 50ppm for 24 hrs after 40 to $70^{\circ}C$ hot water treatment for 10 minutes showed low germination of 4.4%. Based on $H_2SO_4$, NaOH and $HNO_3$, treatments, germination percentage did not improve at all regardless of soaking time. The highest germination of 91.1% was observed when seed was soaked in $GA_3$ 100ppm for 48 hrs after stratification for 60 days at $4^{\circ}C$. Kinetin treatment at 50ppm for 24 hrs had the greatest germination percentage of 31.7% but it did not improve germination ratio compared to $GA_3$ treatment.

Priming Conditions to Improve Germination of Salvia (Salvia splendens F.) Seeds (샐비아(Salvia splendens F.) 종자의 발아촉진을 위한 Priming 조건에 관한 연구)

  • This experiment was conducted to determine the effect of chemicals and their concentrations, priming temperature and duration, and different germination temperature on germinability of salvia seeds. The highest percentage of germination was obtained with 50 or 100 mM $KH_2PO_4$, or with -0.50 or -0.75 MPa PEG 8000. When number of days to attain 50% of the final germination percentage (T50) and mean number of days to germination (MDG) were taken into account, 50 mM $KH_2PO_4$ or -0.50 MPa PEG was most effective for early germination. No seeds germinated when primed in $K_3PO_4$ or NaOH solution. Priming the seeds at $20^{\circ}C$ was better than priming at $15^{\circ}C$ or $25^{\circ}C$. Priming at $20^{\circ}C$ for 4 or 6 days reduced the MDG by 2.3 days compared with nonprimed seeds. Seeds primed with -0.50 MPa PEG at $20^{\circ}C$ showed a high germination percentage with reduced T50 and MDG. When seeds were primed in a mixture of -0.50 MPa PEG and 50 mM $KH_2PO_4$ solution and germinated at $30^{\circ}C$ or $35^{\circ}C$, percent germination was lower than nonprimed seeds. However, the combined treatment retained the priming effect for reducing T50 and MDG.

Effect of chlorella culture solution using animal liquid manure on improving seed germination in perennial ryegrass

  • This experiments were conducted to evaluate the influence of Chlorella culture solution on seed germination of perennial ryegrass seeds. Chlorella are known to contain different bioactive compounds. In present research work, Chlorella culture solution using liquid manure as medium have been used to study their effects on germination and root length. The study conducted a germination experiment in petri-dishes. Four treatments were compared: non-treated control treated with distilled water, Chlorella culture solution and Chlorella culture filtrate, and liquid manure. The germination percentage of perennial ryegrass seeds was highest in the Chlorella culture solution treatment. Days required for 50, 70% seed germination was the fast in Chlorella culture solution and the Chlorella culture filtrate treatment. Root length of perennial ryegrass seeds was long by 1~2cm in the Chlorella culture solution compared with no treated control. The germination index of perennial ryegrass seeds was high by 180~202% in the Chlorella culture solution treatment compared to no treatment. Chlorella culture solution and the Chlorella culture filtrate have shown stimulatory effects in germination and development of root. Use of Chlorella culture solution and the Chlorella culture filtrate can be recommended to farmers as a ecofriendly practice for better germination and growth. Present research work reveals that Chlorella contain certain growth promoting substances which enhances seed germination.
