Production of Antihypertensive Constituents from Ganoderma lucidum IY005 by Fermentation Using Industrial Wastes (산업폐자원을 이용한 발효에 의한 영지의 항고혈압 성분의 생산)

  • Fruit bodies of Ganoderma lucidum have been used for most pharmacological studies, but pharmacological effects are likely variable because the habitats and strains of Ganoderma lucidum are different. Therefore, their fermentation is required to produce constant and reliable pharmacolo­gical constituents from Ganoderma lucidum. During the studies of medium for industrial application. it was found that ginseng root residues, remaining after being extracted with ethanol, were a good carbon source for a fermentation of Genoderma lucidum and a corn steep liquor was also economical for the nitrogen source. Yield of the mycelial cultured in ginseng root residues and corn steep liquor was 2.5 times higher than that in glucose and peptone, known as a conventional medium of Ganoderma lucidum. The polysaccharide content of the extracts from the cultured mycelia was higher than that from fruit bodies, but protein content was vice versa. Extracts of the cultured mycelia were more effective and lasting than extracts of the fruit bodies in decreased hypertention of spontaneously hypertensive rats (SHR).

Biosynthesis of Organic Germanium Using Cordyceps militaris (동충하초를 이용한 유기게르마늄의 생산)

  • In the present study, an attempt has been made to produce a high quality medicinal mushroom Cordycops militaris supplemented with organic Ge. Cordycops militaris was cultivated in SDAY liquid medium containing yeast extract 10 g, peptone 10 g, glucose 40 g per liter and chrysalis media supplemented with inorganic Ge at 100, 500, 1000, and 5000 ppm concentrations. The greatest organic Ge production of 442.4 ppm/g and 284 ppm/g were observed in SDAY liquid and Chrysalis media cultures supplemented with 100 ppm inorganic Ge respectively. Similarly, 4,509.7 ppm/g and 1,058 ppm/g of organic Ge were obtained from liquid and chrysalis media cultures supplemented with 5,000 ppm and 1,000 ppm inorganic Ge, respectively. In addition, higher concentration of organic Ge was obtained in mycelia than fruiting bodies. These results indicate that the concentration of organic Ge increase with decreasing inorganic Ge concentration in the medium. This is the first report on production of high valuable Cordyceps militaris contained with organic Ge.

  • Rhodosporidium fluviale 201-C-1 and Kluyveromyces thermotolerans 150-JE-2(1) were isolated from wild flowers in Geumsan-gun, Chungcheongnam-do and Wando-gun, Jeollanam-do, respectively. Rhodosporidium paludigenum 86-J-1 and Candida sp. 80-J-3 were also isolated from flowers in Jeju island, Korea. Characteristics for these yeasts were not reported before. Therefore, morphological and physiological characteristics for these yeasts were investigated in this study. R. fluviale 201-C-1 was halophilic asporogenous yeast and formed pseudomycelium. It also grew at $30^{\circ}C$ and pH 4.0-6.0. K. thermotolerans 150-JE-2(1) was sugar-tolerant and halotolerant asporogenous yeast. It formed pseudomycelium and showed positive urease activity. Candida sp. 80-J-3 and R. paludigenum 86-J-1 were asporogenous yeasts, and grew well in yeast extract-peptone dextrose(YPD) medium, potato-dextrose medium as well as vitamin-free medium and YPD medium containing 5% NaCl.

Isolation of Wild Yeasts from Humus-rich Soil in City Park of Daejeon Metropolitan City, Korea, and Characterization of the Unrecorded Wild Yeasts (대전광역시 도시 자연공원 부엽토와 주변 산림토양들로부터 야생효모의 분리, 동정 및 국내 미기록 야생효모들의 특성)

  • Totally 91 strains of wild yeasts were isolated from the humus rich soil in the Bogyong city park of Daejeon city, Korea. Majority of the strains belonged to Cryptococcus spp., which included 11 strains of Cryptococcus aureus. Among them, Kwoniella mangroviensis JSS0500, Candida corydalis JSS0501, Candida bombi JSS0503, Candida multigemmis JSS0504, Cryptococcus dimennae JSS0506, Cryptococcus saitoi JSS0507, Cryptococcus victoriae JSS0508, Metschnikowia pulcherrima JSS0502, Papiliotrema aurea JSS0505, Debaryomyces vanrijia JSS0509, Occultifur externus JSS0510, Filobasidium floriforme JSS0511 and Yamadazyma scolyti JSS0512 represented newly recorded yeast strains in Korea, and their microbiological characteristics were investigated. All of these unrecorded yeasts showed oval shape and also formed ascospores and pseudomycelia, except for Kwoniella mangroviensis JSS0500, Candida bombi JSS0503, and Metschnikowia pulcherrima JSS0502. Seven strains including Candida corydalis JSS0501 grew in vitamin-free medium, and 4 of the wild yeasts including Cryptococcus victoriae JSS0508 were halotolerant, i.e., capable of growing in 10% NaCl-containing yeast extract-peptone-dextrose (YPD) broth. Debaryomyces vanrijia JSS0509 was found to be a thermophilic yeast that grew at $37^{\circ}C$.

Comparison of Mycelial Growth Characteristics According to Culture Conditions of Ulleungdo Collection Strains (울릉도 수집 균주의 배양조건에 따른 균사 생장 특성 비교)

  • The collection of biological data of indigenous species must comply with the Nagoya Protocol. Fungi contain various bioactive substances making them an attractive source of several products, including food and medicines. In this study, we investigated the growth characteristics of five indigenous fungal strains, Fomitiporia punctata, Polyporus ulleungus, P. brumalis, Gymnopus subnudus, and Tyromyces kmetii, isolated from samples collected in the Ulleungdo Island. The growth rates for each strain were assessed across various temperatures (20 ℃ to 35 ℃), culture media (Potato dextrose agar, Malt extract & Yeast extract agar, Malt extract agar, Malt extract & peptone agar, Sabouraud dextrose agar, and Modified Melin-Norkrans agar), and pH conditions (4.0 to 8.0). Additionally, we assessed the mycelial growth characteristics in liquid culture. The mycelial growth in different media varied across species; specifically, F. punctata (in MMNA), G subnudus (in MMNA), and P. brumalis (in MEPA) showed rapid growth. Optimal growth temperatures ranged between 25 ℃ and 30 ℃ for most species, with the exception of T. kmetii and P. brumalis, which were able to grow across all the temperatures tested. P. brumalis showed the best growth rate, whereas P. ulleungus showed the lowest growth potential. The optimal pH conditions for mycelial growth ranged between 4.0 and 5.0. In experiments using culture flasks, the dry weight of the culture filtrates decreased with the increasing incubation time and showed a significant decrease between 1 and 6 months of incubation, indicating that the five strains take longer than a month to fully use the culture media. Our findings highlight and establish the optimal growth conditions for five different fungal species that can be used in future application studies.

Isolation of Wild Yeasts Obtained from Waters and Soils of Riversides in Sangjubo of Nakdong River and Daechung Dam of Geumgang Midstream, Korea and Characterization of Unrecorded Wild Yeasts (낙동강 상주보와 금강중류 대청댐 주변으로부터 야생효모의 분리 및 국내 미기록 효모들의 균학적 특성)

  • The goal of this study was to isolate wild yeasts from waters and soils in Sangjubo of Nakdong river and Daechung dam in Daejeon city, Korea and investigate characteristics of previously unrecorded novel wild yeasts. In total, 94 strains from 39 different species of wild yeasts were isolated from 47 soils and waters samples obtained from Sangjubo in Nakdong river. Among these, 3 strains, including Rhodosporidium azoricum JSL-GRU-009, represented novel yeast strains which were previous recorded. Two hundred nine strains from 105 different species of wild yeasts were isolated from 130 soils and water samples obtained from Daechungdaegyo and Daechung dam of Shintanjin in Daejeon city, Korea. Ten strains including Bullera pseudoalba JSL-GRU-005 also represented newly recorded strains in Korea. All the 13 previously unrecorded yeasts were oval or global in shape, and nine strains, including R. azoricum JSL-GRU-009, formed ascospores. Ten strains grew well in vitamin-free medium. R. azoricum JSL-GRU-009 grew well in 20% glucose-containing yeast extract-peptone-dextrose (YPD) medium and 3 strains, including Rhodotorula oryzicola JSL-GRU-003, grew well in 5% NaCl-containing YPD medium.

Patulin Producing Capacity in Broth Culture Media of Penicillium crustosum Isolated from Korean Apple (국내산 사과로부터 분리된 Penicillium crustosum의 액상배지에서의 Patulin 생성능 평가)

  • The patulin producing capicity of Penicillium crustosum, an isolate from Korean apple, in various broth culture media, was investigated, and compared with patulin production by the standard strain P. griseofulvum(ATCC 46037). The maximal patulin production capacity of the P. griseofulvum ATCC 46037 was 2,029-2,829 ppm in 5-GYEP, SY and MEB broth media. The patulin-producing capacity of the isolated fungus(P. crustosum) attained 2,794 ppm in a 5-GYEP broth medium, but was only 324 and 11 ppm in SY and MEB media, respectively. There were no significant correlations between mycelial growth levels and patulin-producing ability in either P. crustosum or P. griseofulvum. The patulin production of P. griseofulvum was induced in the wide pH range of pH 3.0-11.0, while that of P. crustosum was induced in the acidic pH range pH 3.0-5.0. Patulin production levels were dependent on the carbon sources in the media and maximal patulin production by P. griseofulvum and P. crustosum was observed in media containing glycerol and fructose, respectively.

Production of Mycelium and Expolysaccharides by Fed-batch Culture of Agaricus blazei (Agaricus blazei의 유가식 배양을 통한 균사체 및 세포외 다당체 생산)

  • DO-stat fed-batch cultures of Agaricus blazei were carried out using various feeding solutions, for the production of mycelial biomass and exopolysaccharides (EPS). It was observed to be more effective to use a feeding solution containing both carbon and nitrogen sources than that containing only carbon source. The best result was obtained when a feeding solution containing 450 g/l glucose, 60 g/l yeast extract, 30 g/l soytone peptone was used. The maxium mycelial biomass and EPS concentrations were 36.5 g/l and 10.9 g/l, respectively, at 100 hours of cultivation. The mycelial and EPS productivities were 0.37 g/l-h and 0.11 g/l-h, respectively. As compared with the batch culture, the mycelial biomass concentration and its productivity were 6.0- and 2.2-folds increased, respectively. The EPS concentration and its productivity were increased by 4.7 times and 1.8 times, respectively.

Rapid Detection of Salmonella spp. in Fresh-Cut Cabbage by Real-Time PCR (Real-Time PCR을 이용한 신선편이 양배추에서 Salmonella spp.의 신속검출)

  • This study was conducted to find out the minimal time needed for detection of Salmonella spp. which exist at very low concentration in foods by using real-time PCR. The sal-F and sal-R sequences were used as primers and sal-P was used as a probe. The detection limit of Salmonella spp. was $3.77{\times}10^2\;cfu/mL$ in buffered peptone water (BPW). Microbial growth was monitored after artificially inoculated Salmonella spp. into BPW. The obtained growth curve was well fitted with the equation, y=$0.0127x^2$+0.5927x-0.4317 ($R^2$=0.99), if assuming that 1 cell exists in 25 g sample (0.04 cfu/mL). The microbial concentration will be reduced to 10 fold by adding BPW during sample treatment, so actual initial concentration at the starting point of enrichment is 0.004 cfu/mL. At this condition, real-time PCR detection would be possible only when microbial concentration increase occurs to exceed the detection limit (377 cfu/mL). The time needed for microbial increase was calculated from the growth curve equation as 7 hours and 20 minutes. Therefore the total time required for detection was less than 10 hours including the PCR operating time.

Purification and Characterization of Phospholipase D from Actinomycetes KF923. (방선균 KE923이 생산하는 Phospholipase D의 정제 및 특성)

  • In order to screen microorganisms producing phopholipase D (PLD) had high transphosphatidylation activity, about 1,000 Actinomycetes strains were isolated from the 63 soil samples, collected over 6 local area in Korea. When the hydrolytic activity in the supernatant was determined, 131 strains produced PLD more than 0.3 U/ml. Among 131 culture broths tested, 23 ones had transphosphatidylation activity higher than 20% and finally one strain (Actinomycetes KF 923), which had highest hydrolytic and transphophadylation activity, was selected. Actinomycetes KF923 showed the highest hydrolytic activity (13 U/ml) and phosphatidylation activity (95%) after 48 h fermentation using the P medium (yeast extract 1%, peptone 1%, glucose 1.5%, glycerol 1%, $CaCo_3$ 0.4%, pH 7.2). PLD was purified from the culture broth of Actinomycetes KF923 and the specific activity of purified PLD was 567 U/mg. The molecular weight of PLD was about 55 kD and the optimum pH and temperature were 6.0 and $60^{\circ}C$, respectively. The stability of PLD toward pH and temperature were high around pH 8.0 and below $40^{\circ}C$. Special metal ions were not necessary to the PLD activity.
