해조류의 Angiotensin-I 전환효소 저해작용 (Angiotensin-I Converting Enzyme Inhibitory Activity of Algae)

  • 해조자원의 기능 특성을 밝혀 그 이용도를 증진시키기 위한 연구의 일환으로 일반 해조류의 단백질 함량 및 물추출물과 해조 가수분해물의 ACE 저해효과를 조사한 결과는 다음과 같다. 1 단백질 함량은 김이 $39.6\%$로 가장 높았으며, 파래 $22.1\%$, 미역 $21.1\%$, 우뭇가사리 $18.3\%$, 청각 $15.7\%$, 톳 $12.4\%$, 다시마 $8.2\%$의 순으로 나타났다. 2. 해조 물추출물의 ACE 저해효과는 $50^{\circ}C,\;70^{\circ}C,\;98^{\circ}C$의 세 조건에서 각각 추출한 결과 $70^{\circ}C$에서 추출시 시료 모두에서 가장 높게 나타났으며, ACE 저해율이 가장 높은 $70^{\circ}C$를 기준으로 비교했을 때 우뭇가사리 $10.9\%$, 김 $9.3\%$, 파래 $8.9\%$, 미역 $8.2\%$, 톳 $7.5\%$, 다시마 $7.1\%$, 청각 $7.0\%$로 나타났다. 3. maxazyme과 papain에 의한 해조 가수분해물의 ACE 저해효과는 김이 파래, 미역, 우뭇가사리에 비해 월등히 높았고 pep-tide-nitrogen함량 역시 매우 높았다. 김의 경우 가수분해 8시간에서 ACE 저해효과가 maxazyme $31.3\%$, papain $27.9\%$로 가장 높았고 peptide-nitrogen함량도 이 시간에서 가장 높게 나타났다. 파래의 ACE 저해효과 역시 8시간에서 maxazyme $9.6\%$, papain $8.5\%$로 다른 시간대에 비해 대체로 높았으며 peptide-nitrogen함량은 16시간까지 완만히 증가하였다.

Studies on Skin Permeation with Polymer Micelles and the Cell Penetrating Peptide of Pyrus Serotina Var Stem Extracts

  • The stem extract from Pyrus serotina var has natural antioxidant ability, but the extraction method does not result in a soluble compound in cosmetic formulations. This study investigated the cosmetic efficacy of the Pyrus serotina var stem extract and its epidermis permeation ability when combined with polymer micelles and a cell penetrating peptide. The total concentration of polyphenol compounds was determined to be 103.1644 ± 1.38 mg/g in the ethanol extract and 78.97 ± 1.45 mg/g in the hydrothermal extract. The 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging effects were 55.94 ± 0.22% in the ethanol extract at 1,000 mg/L. Superoxide dismutase (SOD) activity rates were 104.05 ± 3.28% in the ethanol extract at 62.5 mg/L. The elastase inhibition rate was 67.21 ± 2.72% in the ethanol extract at 1,000 mg/L. An antimicrobial effect was observed in the Propionibacterium acnes strain. In the epidermal permeability experiment, it was confirmed that formulation of the polymer micelle containing the Pyrus serotina var stem extract and cell penetrating peptide (R6, hexa-D-arginine) showed small particle size and much better skin permeability. The cumulative amount of total Pyrus serotina var stem extract that penetrated to the skin over time increased over 24 hours in three formulations. The three formulations showed 51.61 ㎍/㎠ (Formulation 0), 75.97 ㎍/㎠ (Formulation 1) and 95.23 ㎍/㎠ (Formulation 2) skin penetration, respectively. Therefore, it was confirmed that the ethanol extracts of Pyrus serotina var stem showed good cosmetic efficacy and excellent epidermis permeation ability when combined with a polymer micelle and cell penetrating peptide. Thus, this extract has the potential to be used as a safe and natural cosmetic material in the future.

사습탕(瀉濕湯) 전탕액(煎湯液)이 백서(白鼠)의 신장기능(腎臟機能)에 미치는 영향(影響) (Effects of Saseuptang Water Extracts on the Renal Function in Rats)

  • The aim of this experiment was to elucidate the effects of Saseuptang water extract on the renal function plasma renin activity and plasma levels of atrial natriuretic peptide and aldosterone in rat The results were as follows; 1. Water balance decreased significantly after the administration of Saseuptang water extract, 0.4 and 0.8ml/kg. 2. Urine volume increased significantly after the administration of Saseuptang water extract, $0.4\;m{\ell}/kg$. 3. Urinary excretion of chloride increased significantly after the administration of Saseupthang water extract, $0.8\;m{\ell}/kg$. 4. Free water clearance increased significantly after the administration of Saseuptang water extract, $0.8\;m{\ell}/kg$. 5. Urinary excretion of creatinine increased significantly after the administration of Saseuptang water extract, $0.8\;m{\ell}/kg$. 6. Plasma levels of atrial natriuretic peptide (ANP) decreased significantly after administration of Saseupthang water extract, $0.8\;m{\ell}/kg$. These results suggest that the changes of urine volume after the administration of Saseuptang water extracts are related to the increments of glomerular filtration rate and free water clearance, and it is suggested that the changes of renal function by which Saseuotang may related to the renin-angiotensin and atrial natriuretic peptide system.

생육시기 및 부위별 고추의 항산화력 및 항암 Lunasin peptide의 동정 (Antioxidant Activity and Identification of Lunasin Peptide as an Anticancer Peptide on Growing Period and Parts in Pepper)

  • 부위간의 DPPH free radical scavenging 활성은 과육은 종자 보다 높았고 생육시기간에는 후기가 초기에 비하여 높았고, Xanthine oxidase 억제활성은 종자와 과육에 비하여 매우 높았고 동정된 항 산화물은 대부분이 지방산 및 페놀성 화합물로, 과육 및 종자가 성숙 될수록 1-eicosanol, palmitic acid, linoleic acid, linolenic acid 등의 지방산 및 benzonitrile의 함량이 증가되었다. 시료 채취 시기간의 POD활성은 과육은 성숙중기에 높았으나, 종자는 성숙이 경과될수록 낮았다. SOD 활성은 과육은 생육이 경과할수록 증가하였으나, 종자는 성숙할수록 낮아졌다. 고추종자 단백질은 western blot에 의하여 lunasin이 동정되었고, 개화 후 5주부터 lunasin 생합성 되었다. 고추 종자내의 100nM lunasin 펩타이드는 종양유발 유전자가 전이된 $2\~12$ 세포의 콜로니 형성을 억제시켰으며, 결과적으로 고추 종자내에는 암 예방적 특성을 갖는 lunasin peptide가 존재한다.

관절염 모델에서 백굴채전탕액이 척수와 척수신경절의 Calcitonin Gene-Related Peptide와 Substance P 면역반응에 미치는 영향 (The Effects of Herba Chelidonii Extracts on Calcitonin Gene-Related Peptide and Substance P Immunoreactive Response in Spinal Cord and Ganglia of Adjuvant-Induced Arthritis)

  • To investigate the anti-inflammatory and analgesic effects of Herba Chelidoniie, the extracts of Herba Chelidoniie treated in arthritic rat model. Complete Freund,s Adjuvant(CFA) were injected in the subcutaneous tissue of left foot paw of rats to induce arthritis. Herba Chelidonii extracts(HC) was administered immediately into the peritoneal cavity after CFA injection for 12 days. The immunohistochemical stainings for calcitonin gene-related peptide(CGRP) and substance P in the L4, L5 and L6 spinal dorsal horn and ganglia were done, and the paw swelling was measured with a micrometer and the blood leukocytes were counted. The results were as follows : The paw swelling of HC treated group was significantly decreased in 12th day after CFA injection compare to control group. The change of differential leukocytes counts of HC treated group increased the ratio of lymphocytes, and decreased the ratio of neutrophils compare to control group. The extent of CGRP immunoreactive nerve fiber of dorsal horn of HC treated group was weakly stained compare to control group. The number of CGRP immunoreactive neurons of L6 spinal cord of HC treated group was significantly decreased compare to control group. The extent of substance P immunoreactive nerve fiber of dorsal horn of He treated group was weakly stained compare to control group. The number of substance P immunoreactive neurons of L4, L5 and L6 spinal cord of HC treated group was significantly decreased compare to control group. These experimental results suggest that Herba Chelidonii extracts reduce the number of CGRP and substance P immunoreactive neurons and nerve fibers of spinal dorsal horns and ganglia, and decrease paw swelling in arthritic rat model, which may be closely related to analgesic and antiinflammatory effects of Herba Chelidonii.

양식장 넙치 폐사어를 이용한 단백질 소재의 개발에 관한 연구(1) -항산화 기능특성- (A Study on Development of Protein Materials using Dead Flatfish from Fish Farms(1) -Antioxidant Functional Properties-)

  • To develop the protein materials by the reutilization of dead flatfish from fish farms in Jeju island, the physicochemical characteristics and the functional activities of collagen peptide extracts were investigated. Flatfish skin collagen peptide (FSCP) and flatfish protein hydrolysate (FPH) were manufactured from dead flatfish. The differences of pH, moisture and fat contents between FSCP and FPH were not significant, fat contents were analyzed less than 0.3%, and trans-fat, saturated fat and cholesterol were not detected in both samples. Protein contents of FSCP and FPH showed about 92% and 95%, respectively. In the analysis of amino acids, glycine and hydroxy proline content in FSCP was 24.22% and 6.15%, respectively, showed a typical characteristics of the collagen protein, but essential amino acids contents such as threonine, valine, methionine, isoleusine, leusine and phenylalanine were relatively higher than those of FPH. Average molecular weight of FSCP was measured as 1,102 which was almost equal value with that of tuna collagen peptide. The antioxidant activities and functional properties showed high but did not show significant difference between two samples.

Effects of Cordyceps ophioglossoides extracts on the neuronal death and memory dificits

  • We investigated whether the mushroom extracts can protect neuronal death and ameliorate memory deficits in Alzheimer"s disease induced by $\beta$-amyloid peptide[A$\beta$(25-35)]. Cellular model of Alzheimer"s disease was produced by using SK-N-SH human neuronal cells treated with $A\beta$. Treatment with 40uM $A\beta$ for 48hours caused a 46% loss of cell viability. First, we examined the effects of 22 mushroom extracts on neuronal death using MTT assay. We found that 3 mushroom extracts increased viability of the cells from 46% to 87%. (omitted)

Assessment of the Inhibitory Activity of Peptide Extracts from Hanwoo Musculus Longissimus on Angiotensin I-Converting Enzyme

  • This study was performed to measure the angiotensin I-converting enzyme (ACE) inhibitory activity of peptide extracts derived from the enzymatic proteolysis of Hanwoo Musculus longissimus (M. longissimus) during cold storage. Thermolysin (80 ppm, w/w) and protease type XIII (100 ppm, w/w) were injected separately or in combination for the enzymatic proteolysis of sarcoplasmic and myofibrillar proteins prior to storage at $5^{\circ}C$ (T1) or at $-1^{\circ}C$ (T2) in a chilling room for 9 days. Beef injected with thermolysin (E2) and thermolysin+protease type XIII (E3) showed a significantly higher degree of hydrolysis at both storage temperatures (p<0.05). During the storage period, T1E2 at day 6 and T1E3 at day 9 showed the strongest ACE inhibitory activity with sarcoplasmic and myofibrillar protein proteolysates. Macromolecules greater than 10,000 Da were removed by ultra filtration, and the filtrates were separated into fractions using gel filtration. Five and three major fractions were collected from S-T1E2-6 and M-T1E3-9 extracts, respectively, and the $4^{th}$ fraction of the S-T1E2-6 extracts showed the highest ACE inhibitory rate of $61.96{\pm}7.41%$.

홍어의 항고혈압 활성물질 (ACE Inhibitory Materials from Raja kenojei)

  • 홍어를 발효하여 발효기간에 따른 항고혈압 효과를 조사하고 그에 따른 항고혈압성 물질을 분리하기 위해 GPC system을 사용하여 항고혈압 물질을 조 분리하였다. 항고혈압성 물질을 농축하기 위해 홍어 내장 및 가식부 열수 추출물을 대량 포집 하였다. 그리고 그에 따른 ACE 억제효과를 검색하였다. 즉, 홍어 가식 부의 ACE 저해 작용은 2% 첨가시 29%로, 다소 낮으나 상시 섭취될 수 있는 식품이란 측면에서 볼 때 그 유용성이 기대된다고 할 수 있으며 홍어 내장 열수 추출물의 ACE 저해 효과는 시료 2% 첨가의 경우 발효 0일째에 71.0%로 가장 높게 나타났다. 발효가 진행되면서 ACE 저해 효과는 감소하는 것으로 나타났다. 이러한 ACE 억제효과를 나타내는 성분을 추정하기 위하여 홍어 내장 열수 추출물의 일반성분과 원소분석을 실시한 결과 일반성분 중 순단백질이 58.7%로 가장 높게 나타났으며, 원소분석 결과 C, H, O, N의 성분비가 당류라기 보다는 peptide계인 것으로 나타났다. 또한, 질소화합물의 분석결과 ACE를 억제하는 기능성 peptide의 성분인 Tyr, Phe, Val, His 등이 가식부 보다 많아서 ACE를 억제하는 peptide를 함유하리라 예상되었다. ACE억제 물질을 조 분리하기 위하여 Sephadex G-25 column chromatography에 의해 분자량별로 분획한 결과, 분획물 들의 ACE 저해 효과는 분획물 B(111-160)의 농도 0.2%에서 67.8%로 가장 높은 ACE저해 효과를 나타내었다. 이상의 결과로 미루어 보아 홍어 내장 열수 추출물의 ACE 저해인자는 가열에 대하여 안정한 비교적 저분자의 peptide와 같은 물질이라고 추정할 수 있었다.

가열 유무에 따른 참담치(Mytilus coruscus) 추출물 내의 항균 펩타이드 변화 (Variation of Antimicrobial Peptide in the Extract of the Hard-shelled Mussel Mytilus coruscus Depending on Boiling)

  • This study was performed to confirm the optimal extraction method for antimicrobial peptides from the Hard-shelled mussel. Extractions were performed with two processes including 1% HAc/boiling and 1% HAc/non-boiling methods and used extracts for the comparison of the antimicrobial activity, protease stability, action mechanism, AU-PAGE (acid-urea PAGE), and HPLC chromatograms. 1% HAc/boiling extract showed potent antibacterial activities both against Gram-positive and negative bacterium but 1% HAc/non-boiling extract showed antibacterial activity only against Gram-positive bacteria. Treatment of 1% HAc/boiling extract with proteases retained almost antibacterial activity against B. subtilis, but abolished significant antibacterial activity against E. coli D31. Only 1% HAc/boiling extract showed two discrete clearing antibacterial zones including slow migrating and rapid migrating zones. Both extracts showed strong DNA-binding ability but did not show bacterial membrane permeabilizing ability. In comparison of the chromatogram obtained from C18 or cation-exchange HPLC, the eluted peaks from 1% HAc/boiling extract showed high hydrophobic property or absorbance compared to 1% HAc/non-boiling extract, respectively. The concentration of the purified antimicrobial peptide was also higher in 1% HAc/boiling extract than in 1% HAc/non-boiling extract. Our results suggest that the effective extraction condition for antimicrobial peptides from marine invertebrate is boiling process in a weak acetic acid solution (1%).
