Integration of microbial antagonists with fungicides was tried to control the gray mold caused by Botrytis cinerea on pepper in greenhouse conditions and to reduce fungicide uses. All of the selected bacterial antagonists, Bacillus amyloliquefaciens BL3, Paenibacillus polymyxa BL4, and Pseudomonas putida Cha94, completely inhibited the conidial germination of B. cinerea until 30 days after treatment. However, bacterial colonization of pepper phylloplane was poor in BL4, while the other bacterial isolates and the fungal antagonist Trichoderma harzianum TM colonized well on the phylloplane, maintaining the population density of 104-105 cfu/g until 15 days after microbial treatments. Out of 13 kinds of selected fungicides used for gray mold diseases, polyoxin B and BKF 1995 showed the most discriminatory activity on the fungal growth between B. cinerea and TM. TM grew readily on the media containing those fungicides, while B. cinerea showed poor or no mycelial growth on them. The selected fungicides and antagonists alone reduced incidence of gray mold on pepper, showing disease indices of about 2.4 to 3.0, while its was increased up to 4.2 in the untreated control. Alternate treatments with the antagonists and 2-fold diluted fungicides inhibited the disease incidence as much as the antagonists or fungicides alone, and reduced the secondary inoculum more than the single treatments. This suggests that integration of antagonists and fungicides may be an efficient way to reduce fungicide sprays with reliable control efficacy of the disease. However, there was not much difference in the early and mid-term disease progress among the treatments and the untreated control, probably due to extremely favorable environmental conditions for the disease development in this experiment.
Fungi produce various secondary metabolites that have beneficial and harmful effects on other organisms. Those bioactive metabolites have been explored as potential medicinal and antimicrobial resources. However, the activities of the culture filtrate (CF) and metabolites of whiterot fungus (Schizophyllum commune) have been underexplored. In this study, we assayed the antimicrobial activities of CF obtained from white-rot fungus against various plant pathogens and evaluated its efficacy for controlling anthracnose and gray mold in pepper plants. The CF inhibited the mycelial growth of various fungal plant pathogens, but not of bacterial pathogens. Diluted concentrations of CF significantly suppressed the severity of anthracnose and gray mold in pepper fruits. Furthermore, the incidence of anthracnose in field conditions was reduced by treatment with a 12.5% dilution of CF. The active compound responsible for the antifungal and disease control activity was identified and verified as schizostatin. Our results indicate that the CF of white-rot fungus can be used as an eco-friendly natural product against fungal plant pathogens. Moreover, the compound, schizostatin could be used as a biochemical resource or precursor for development as a pesticide. To the best of our knowledge, this is the first report on the control of plant diseases using CF and active compound from white-rot fungus. We discussed the controversial antagonistic activity of schizostatin and believe that the CF of white-rot fungus or its active compound, schizostatin, could be used as a biochemical pesticide against fungal diseases such as anthracnose and gray mold in many vegetables.

We determined threshold environmental factros to initiate infection of pepper plants by Botrytis cinerea, a fungal pathogen of pepper gray mold, in two greenhouse conditions. A new efficient spore-trapping method was developed to estimate population density of airborne conidia in the greenhouses, and spore release was measured using a Kerssies' selective medium. At a given day, spores were released greater during daytime (mostly from 7:30 am to 10:30 am and at 4:30 pm) than nighttime. Diurnal and nocturnal temperatures in the greenhouse-1 were about $25^{\circ}$ and $17^{\circ}$,and relative humidity was 100% for prolonged 24 h due to rain on December 17, 1997. Population density of air-borne conidia was 3.0$\times$103 conidia/ $0.5\textrm{m}^3$ after two days, and the initial infection occurred in ten days. During the same period of time in the greenhouse-2, diurnal temperature was about $25^{\circ}$ and nocturnal temperature was below $15^{\circ}$, and population density of air-borne conidia was 104 conidia/ $0.5\textrm{m}^3$. Under these conditions, the initial infection started in three days. This indicates that the early infection occurs under which diurnal temperature is approximately $25^{\circ}$, nocturnal temperature is maintained below $15^{\circ}$, and population density of air-borne conidia is 104 conidia/ $0.5\textrm{m}^3$ at saturated relative humidity condition.
Bacillus licheniformis N1, previously developed as a biofungicide formulation N1E to control gray mold disease of plants, was investigated to study the bacterial traits that may be involved in its biological control activity. Two N1E based formulations, bacterial cell based formulation PN1E and culture supernatant based formulation SN1E, were evaluated for disease control activity against gray mold disease of tomato and strawberry plants. Neither PN1E nor SN1E was as effective as the original formulation N1E. Fractionation of antifungal compounds from the bacterial culture supernatant of B. licheniformis N1 indicated that two different cyclic lipopeptides were responsible for the antimicrobial activity of the N1 strain. These two purified compounds were identified as iturin A and surfactin by HPLC and LCMS. The purified lipopeptides were evaluated for plant disease control activity against seven plant diseases. Crude extracts and purified compounds applied at 500 ${\mu}g/ml$ concentration controlled tomato gray mold, tomato late blight and pepper anthracnose effectively with over 70% disease control value. While iturin showed broad spectrum activity against all tested plant diseases, the control activity by surfactin was limited to tomato gray mold, tomato late blight, and pepper anthracnose. Although antifungal compounds from B. licheniformis N1 exhibited disease control activity, our results suggested that bacterial cells present in the N1E formulation also contribute to the disease control activity together with the antifungal compounds.

In commercial greenhouses, senescent flower petals or flowers of vegetables such as tomato, strawberry, hot pepper and zucchini squash were blighted to be removed from fruits within five days after spraying of Trichoderma harzianum YC459 (TORY), a biocontrol agent for the gray mold rot of vegetables caused by B. cinerea The mechanism for selective colonization of senescent floral tissues by T. harzianum YC459 was elucidated using fresh and senescent (Hays and 14days after flowering, respectively) floral tissues of zucchini squash (Cucurbita moschata Duchesne). The spores of T. hrzianum YC459 were produced more on agar and liquid culture media supplemented with 5% dry powder of senescent floral tissues than fresh tissues during 15days. Mycelial growth was also much better in the media with senescent tissues than with fresh tissues. Enzyme activities of amylase, polygalacturonase and cellulase in the liquid media which might be involved in the colonization of tissues by T. harzianum YC459 were compared. The activities of three enzymes were much higher in the media with senescent floral tissues than with fresh floral tissues reaching to the maximum during 9 to 12days of incubation. Based on the results, the removal of senescent floral tissues, a possible inoculum source of the pathogen, may be another mechanism for biocontrol of gray mold rot of vegetables by T. harzianum YC459.

Biological control of important fungal diseases such as Phytophthora blight of red pepper, gary mold rot of vegetables, and powdery mildew of many crops was attempted using an antagonistic bacterium, Bacillus sp. AC-1 in greenhouses and fields. The antagonistic bacterium isolated from the rhizosphere soils of healthy red pepper plant was very effective in the inhibition of mycelial growth of plant pathogenic fungi in vitro including Phytophthora capsici, Rhizoctonia solani, Pyricularia oryzae, Botrytis cinerea, Valsa mali, Fusarium oxysporum, Pythium ultimum, Alternari mali, Helminthosporium oryzae, and Colletotrichum gloeosporioides. Culture filtrate of antagonistic Bacillus sp. AC-1 applied to pot soils infested with Phytophthora capsici suppressed the disease occurrence better than metalaxyl application did until 37 days after treatment in greenhouse tests. Treatments of the bacterial suspension on red pepper plants also reduced the incidence of Phytophthora blight in greenhouse tests. In farmers' commercial production fields, however, the controlling efficacy of the antagonistic bacteria was variable depending on field locations. Gray mold rot of chinese chives and lettuce caused by Botrytis cinerea was also controlled effectively in field tests by the application of Bacillus sp. AC-1 with control values of 79.7% and 72.8%, respectively. Spraying of the bacterial suspension inhibited development of powdery mildew of many crops such as cucumber, tobacco, melon, and rose effectively in greenhouse and field tests. The control efficacy of the bacterial suspension was almost same as that of Fenarimol used as a chemical standard. Further experiments for developing a commercial product from the antagonistic bacteria and for elucidating antagonistic mechanism against plant pathogenic fungi are in progress.
A Pseudomonas strain SG3 producing biosurfactant and showing antifungal and insecticidal activities was isolated from agricultural soil severely contaminated with machine oils. The antagonistic bacterium inhibited mycelial growth of all of the tested fungal pathogens. The fermentation broth of SG3 also effectively suppressed the development of various plant diseases including rice blast, tomato gray mold, tomato late blight, wheat leaf rust, barley powdery mildew and red pepper anthracnose. An antifungal substance was isolated from the fermentation broth of SG3 by ethyl acetate partitioning, silica gel column chromatography and preparative HPLC under the guide of bioassay. The chemical structure of the antifungal substance was determined to be rhamnolipid B by mass and NMR spectral analyses. The antifungal biosurfactant showed a potent in vivo antifungal activity against gray mold and late blight on tomato plants. In addition, rhamnolipid B inhibited mycelial growth of B. cinerea causing tomato gray mold and zoospore germination and mycelial growth of P. infestans causing tomato late blight. Pseudomonas sp. SG3 producing rhamnolipid B could be used as a new biocontrol agent for the control of plant diseases occurring on tomato plants.
Plants are the promising reservoirs for natural products with their diverse secondary metabolites. Many invasive plants have been introduced in Korea, which adversely affect on the native ecosystem but holds difficulty removing them due to their proliferation. In this study, we evaluated disease control efficacy of methanol extracts from four invasive plant species against 7 representative crop pathogens. Methanol extract of Phytolacca americana effectively suppressed rice blast, tomato gray mold, and tomato late blight in a dose dependent manner. The methanol extract of Amorpha fruticosa also exhibited potent antifungal activity against pepper anthracnose in a concentration dependent way. These data suggest that the extracts of P. americana and A. fruticosa can be developed as plant disease protection agents against rice blast, tomato gray mold, tomato late blight, and pepper anthracnose. Furthermore, more extensive research will be required to identify and isolate active compounds from problematic invasive plant species to develop valuable agrochemicals.
Electron beam (EB) irradiation was tested to determine the dose required to eradicate plant pathogens, such as Botrytis cinerea and Agrobacterium rhizogenes, from the infected seeds without affecting the germination rate of the irradiated vegetable seeds, including crown daisy, cucumber, hot pepper, green onion, leaf lettuce, and radish seeds. EB irradiation of 1.5 kGy and 2 kGy was sufficient to kill 100% of hairy root disease bacteria and gray mold conidia, respectively. EB irradiation showed no effect or minimal effect on the germination rate of the crown daisy, cucumber, green onion, and radish seeds. However, the germination rate of the hot pepper and leaf lettuce seeds was significantly reduced by using 2 kGy of EB irradiation. Difference in susceptibility to EB irradiation appears not to be related to the weight of each seed, but to the intrinsic characteristic of each plant. Conclusively, EB irradiation might be a useful way to decontaminate crown daisy, cucumber, green onion, and radish seeds.

Since 1993, a total of 50 problematic plant diseases unrecorded in Korea were surveyed in Gyeongnam province. Totally 34 new host plants to corresponding pathogens investigated in this study were 5 fruit trees, 9 vegetables, 12 ornamental plants, 3 industrial crops, and 5 medicinal plants. Among the newly recorded fruit tree diseases, fruit rot of pomegranate caused by Coniella granati and Rhizopus soft rot of peach caused by Rhizopus nigricans damaged severely showing 65.5% and 82.4% infection rate. Among the vegetable diseases, corynespora leaf spot of pepper caused by Corynespora cassiicola and the crown gall of pepper caused by Agrobacterium tumefaciens, powdery mildew of tomato caused by Oidiopsis taurica were the most severe revealing 47.6%, 84.7%, and 54.5% infection rate in heavily infected fields, respectively. In ornamental plants, collar rot of lily caused by Sclerotium rolfsii, gray mold of primula caused by Botrytis cinerea, soot leaf blight of dendrobium caused by Pseudocercospora dendrobium, sclerotinia rot of obedient plant caused by Sclerotinia sclerotiorum showed 32.7 to 64.8% disease incidence. On three industrial plants such as sword bean, broad bean, and cowpea, eight diseases were firstly found in this study. Among the diseases occurring on broad bean, rust caused by Uromyces viciae-fabae and red spot caused by Botrytis fabae were the major limiting factor for the cultivation of the plant showing over 64% infection rate in fields. In medicinal plants, anthracnose of safflower caused by Collectotrichum acutatum was considered the most severe disease on the plant and followed by collar rot caused by Sclerotium rolfsii.(중략)
