Nectarine beverage treated with cellulase and pectinase enzymes was measured for mineral contents, total flavonoids, and free amino acids and DPPH radical scavenging effect, nitrite scavenging effect. Total flavonoid contents of the no treatment, treated with pectinase, with cellulase, and with both measured 0.146 mg/mL, 0.167 mg/mL, 0.148 mg/mL and 0.171 mg/mL, respectively. DPPH was measured as 13.42% with no treatment and more than 28.98% with enzyme treatments. Nitrite scavenging effect with no treatment was 79% at pH 1.2. Whereas, it was measured above 90% while treated with enzymes at pH 1.2. And also, the nitrite scavenging effect was slightly higher at pH 3.0, pH 4.0 and pH 6.0 than no treatment. Results of free amino acids analysis revealed that, aspartic acid, serine, alanine, $\gamma$-aminobutylic acid, and glutamic acid were present with the amount ranging from 86.71% to 94.14% from total detected free amino acids. Ornithine and taurine were also observed from the beverages. The mineral contents, nitrogen element (T-N) of enzyme treatment of nectarine beverages were measured slightly higher than T-N of no treatment, however, the $P_2O_5$ was similar. Moreover, CaO, MgO and $K_2O$ in the beverages were measured above 45 mg/L, 85 mg/L and 2,133 mg/L, respectively.

Prunus mume Siebold & Zucc., a member of the Rosaceae family (called Maesil in Korea), has been widely distributed in East Asia, e.g. Korea, Japan and China, and its fruit has been used as a traditional drug and health food. In this study, we evaluated physicochemical properties and physiological activities of condensed Prunus mume juice treated with pectinase (PJ). The values of total acidity, pH, sugar contents, turbidity moisture content of the PJ were 35.81%, 2.73, $54.36^{\circ}Brix$, 2.75 and 51.32%, respectively. The PJ had effective DPPH radical scavenging activity, reducing power effect, $H_2O_2$ scavenging activity and ${\beta}$-carotene bleaching effect. DPPH radical scavenging activities of PJ was 46.31%; their reducing power ($OD_{700}$) was 1.80; $H_2O_2$ scavenging activity of PJ was 91.62%; and ${\beta}$-carotene bleaching effect of PJ was 73.02%. Also, PJ showed effective levels of ${\alpha}$-glucosidase inhibition activity. The cell viability was measured by SRB assay. The PJ significantly decreased the cell viability of mouse melanoma cells (B16) and human melanoma cells (SK-MEL-2 and SK-MEL-28) in a dose-dependent manner, however, there was no effect on human keratinocyte HaCaT. In morphological study, PJ-treated SK-MEL-2 cells showed distorted and shrunken cell masses. Total polyphenol contents and total flavonoid contents of PJ were 588.31 mg% (gallic acid equivalent) and 860.45 mg% (rutin equivalent). The antiproliferative effect of PJ seems to be associated with the antioxidant activity of its flavonoid and polyphenol contents. In conclusion, PJ may be beneficial in development of a functional food material.

효소는 주로 면직물의 전처리 및 염색가공에 사용되고 있는데, 공업적으로 적용되고 있는 예로는 아밀라제에 의한 호발, 셀룰라제에 의한 감량 및 유연가공을 들 수 있고, 최근 미국과 일본을 중심으로 연구되고 있는 펙티나제에 의한 정련은 공업화 가능성이 매우 높은 것으로 보고되고 있으며, 글루코스 옥시다제에 의한 표백은 아직연구 초기 단계에 있다. 효소정련이란 기존의 수산화나트륨 대신에 효소를 사용하여 행하는 정련을 말하며, 장점으로는 50-6$0^{\circ}C$ 정도의 온도에서 행하기 때문에 기존 정련법과 비교시 에너지와 용수를 절약할 수 있는 것, 폐수 중에 방출되는 오염물질의 양을 현격하게 줄일수 있을 것, 우수한 생분해성으로 인한 수질오염을 감소시킬 수 있을 것, 부드럽고 스무스하여 자연스런 태를 부여하는 것 및 재현성이 높은 것 등이다. 쥬스제조시 청징공정에 있어서 UF의 응용은 현재까지 그리 주목받아 오지 못했다. 그러나 UF는 기존의 여과방법보다 더 간단하고 저렴한 기술이며 더 좋은 품질의 제품을 생산할 수 있다. 일반적인 사과쥬스 제조에 있어서 기존의 청징방법은 보통 2단계를 필요로 하는데 먼저 Pectinase와 같은 효소처리된 원료쥬스를 Decanter라는 회전진콩 여과기로 여과한 다음 가압여과기로 여과를 하게 된다. 이 두단계에 있어서 많은 양의 규조토가 필요한데 UF를 사용하여 청징여과를 하게 되면 규조토의 사용을 안하므로 생산비 절감 뿐만아니라 폐기물처리 문제까지도 해결해 준다. 도한 제품의 질, 특히 덫징도가 증가하며 수율도 높일 수 있고 노동비를 절감할 수 있으며, 효소는 막을 통과하지 않기 때문에 농축액에 축적되어 효소의 재사용에 따른 효소사용량 절감을 가져올 수 있다.
Antagonistic fluorescent pseudomonads isolated from rhizospheric soil of rice were characterized by 16S rRNA amplicon and fatty acid methyl ester (FAME) analyses. Antagonistic isolates were grown in the fermentation media, and production of antibiotics was confirmed by thin-layer chromatography (TLC) and high-performance liquid chromatography (HPLC). Production of fungal cell-wall-degrading enzymes such as protease, cellulase, pectinase, and chitinase was determined. Dendrogram based on the major and differentiating fatty acids resulted into 5 clusters, viz., cluster I (P. pseudoalcaligenes group), cluster II (P. plecoglossicida group), cluster III (P. fluorescens group), cluster IV (P. aeruginosa group), and cluster V (P. putida group). Characteristic presence of high relative proportions of cyclopropane (17:0 CYCLO w7c) was observed in antagonistic bacteria. Data revealed biodiversity among antagonistic fluorescent pseudomonads associated with the rice rhizosphere. Results presented in this study will help to identify the antagonistic isolates and to determine their mechanisms that mediate antagonism against fungal pathogens of rice.
Soft-rot bacterial pathogen, Erwinia sp., was isolated from chinese cabbage tissue showing soft-rot symptom. This bacterial strain caused soft-rot to chinese cabbage and potato, and it was identified as Erwinia carotovora subsp. carotovora LY34 (E. c. subsp. carotovora LY34). Erwinia carotovora subsp. carotovora LY34 did not have hemicellulase but extracellular cellulase, pectinase, polygalacturonase, protease activity. The results of the microscopy showed that chinese cabbage tissue and potato tissue were macerated by infection of E. c. subsp. carotovora LY34. In analysis of the cellulases activity of the isolated cellulose-degradation enzymes from E. c. subsp. carotovora LY34 total protein, three cellulase activity bands were detected by non-denaturation gel electrophoresis method and five cellulase activity bands were detected by CMC-SDS-PAGE direct stain method.

Purification and characterization of pectate lyase from Bacillus sp HSA-925. Bacillus sp. HSA-925 isolated from soil produced constitutively an extracellular pectate lyase when cultivated in LB broth. The pectate lyase(EC 4.2.2.2) was purified from the cuylture broth by preciptation with ammonium sulfate, followed by column chromatography on CM-cellulose C-50 and repeated gel filtration on Sephadex G-75G. The enzyme had a molecular weight of 32-33 kDa. The activity was mazimum at pH 9.5 AND 45$\CIRC $C. The enzume activity was stable at 55$\circ $C for 15 min and between pH7-12. The activation energy, Km and V$_{max}$ for the pectate lyase were 5.8779 kcal/mol, 6.33$\times $10$^{-2}$ mol/ml and 2.09$\times $10$^{2}$ $\mu $mol/min respectively. The enzyme was activated by Ca$^{2+}$, Cu$^{2+}$ and inhibited by Li$^{+}$, Hg$^{2+}$, EDTA.

Aspergillus kawachii extracellular pectinases were screened in liquid cultures with different carbon sources. The fungus grown on citrus pectin or lemon pomace produced at least one of these inducible pectinases: acidic polygalacturonase, pectin lyase, pectin methylesterase, $\alpha$-L-arabinofuranosidase, $\alpha$-1,5-endoarabinase, $\beta$-D-galactosidase/exogalactanase, and $\beta$-1,4-endogalactanase. The lemon-pomace filtrates also contained significant $\alpha$-L-rhamnosidase and $\beta$-D-fucosidase activities. Most of the screened pectinases were active at pH 2.0-2.5, indicating that the A. kawachii enzymes were acidophilic. Under the culture conditions employed we could not detect enzymatic degradation of soybean rhamnogalacturonan. The A. kawachii pectinase-production-related regulatory phenomena of induction-repression resemble those described for other Aspergillus sp.
Rice callus was treated with 0.56M sucrose solution mixed with 5% pectinase and 10% cellulase, and the protoplasts isolated were transferred to 0.25 M sodium nitrate to induce protoplasmic fusion. Callus tissues were macerated well and degradation of cell walls also proceeded satisfactorily. When the protoplasts were transferred to sodium nitrate solution, many giant roundish protoplasts and some multilobed complex protoplasmic bodies were observed. Most of the fusions took place immediately after the protoplasts were transferred to sodium nitrate. Some multilobed protoplasts which failed to fuse in the initial stage took longer time, about two hours, to get completely fused and rounded-off. Multilobed protoplasmic bodies were invariably multinucleate, while giant round protoplasts had either several nuclei or had one nucleus of large size. Nuclear fusion, also, seemed to occur immediately after the protoplasts were transferred to sodium nitrate.
The optimal conditions for the protoplast isolation from the leaves of pea (Pisum sativum L. cv. Sparkle) and barley (Hordeum vulgare L. cv. Baecdong) were determined in order to achieve a somatic hybridization between two species. It was revealed that the use of 0.5M sorbitol as an osmoticum was appropriate for pea. The yield of intact protoplasts was the highest (40%) when pea leaves were incubated in the enzyme solution for 4 hours. In case of barley, the optimal concentrations of cellulase, pectinase and mannitol as the enzyme solution were 2%, 1% and 0.35M, respectively. And the yield of barley protoplasts was the highest(87%) when leaves were incubated in this enzyme solution for 3.5 hours. A fusion of protoplasts from pea and barley was induced by PEG treatment enriched with calcium salts within 60 minutes.
