This experiment was conducted to elucidate the effects of fruit bagging on the amounts of pesticide residues on/in pears with two pesticides, chlorpyrifos 25% WP, and penconazole 5% WP, and two pear cultivars, Niitaka and Hwangeum-bae. Residues of chlorpyrifos and penconazole in bagged pears were only $0.4{\sim}27%$ of those in non-bagged one. Residues of both pesticides in bagged and non-bagged pears were steeply reduced in the field but slowly reduced during storage. Residues of chlorpyrifos were more in the peel than in the flesh, while penconazole in bagged pear was evenly distributed in the peel and flesh. Chlorpyrifos was evenly distributed in outer bag and inner bag irrespective of bag materials, while most of penconazole was found in outer bag rather than in inner bag. To produce safer pear from pesticide residues, removal of bag before storage is recommended.

To develop scab-resistant pear (Pyrus spp.) varieties with fruits that are as crisp and juicy as Asian pears, a cross was made between 'Whangkeumbae' and 'Bartlett' varieties (P. pyrifolia ${\times}$ P. communis) at the Pear Research Institute of the National Institute of Horticultural & Herbal Science, Rural Development Administration, in 1994. Among the 285 seedlings, 'Greensis' was first selected in 2006 for its good eating quality and named in 2012 after regional adaptation tests in nine regions and ten experimental plots from 2007 to 2012. The tree showed a vigorous growth habit and semi-spreading characteristics, like 'Whangkeumbae'. The optimum fruit harvest date was also around Sept. 26 and fruit was round in shape and green in skin color at maturity. Average fruit weight was 470g, and the soluble solids content was $12.4^{\circ}Brix$. The flesh was very crisp and juicy, and had good eating quality. Its' leaf size was similar with 'Bartlett' and smaller than 'Whangkeumbae'. The average of full bloom date of 'Greensis' was determined as Apr. 26, which was six days later than 'Whangkeumbae' and similar with 'Bartlett'. S genotypes of 'Greensis' were identified as $S_4S_e$ by S-allele PCR product sequencing analysis. It seems that the $S_4$ allele was inherited from 'Whangkeumbae' and the Se allele from 'Bartlett'. 'Greensis' displayed strong resistance to scab disease caused by Venturia nashicola, similar to European pear cultivars like 'Beurre Hardy' and, 'Conference'. 'Greensis' was also highly resistant to black leaf spot (Alternaria kikuchiana) in the field
Physical properties and compositions of dietary fiber sources (DFS) from peels of Asian pear fruit, Hosui, Niitaka and Chuwhangbae, were examined at different growth stages, young, unripe, and ripe fruits, to investigate their use as functional ingredients. Total dietary fiber (TDF) contents of DFS prepared from peels of three cultivars at three stages were high, 74.00-88.38%, with most being insoluble dietary fiber. Total phenolic compound contents of DFS were 1.64-4.46mg/g, with those of Niitaka and Chuwhangbae increasing with growth stages. Density of DFS from young fruit was significantly lower than those from unripe and ripe fruits (p<0.05), because particle size of DFS increased with growth stages within cultivars. Water-holding capacity (WHC) and oil absorption were 3.11-6.03g water/g solid and 1.98-2.57g oil/g sample, respectively. Young fruits, which showed lowest WHC value, had highest oil absorption value. Particle size of DFS had significant correlation with density, WHC, and oil absorption (p<0.05). Physical properties of DFS were mainly influenced by cellulose and uronic acid.
This was carried out to develop a chromosome-doubled (12x) persimmon that will be used as a crossing parent to select seedless persimmon cultivars with the change of the consumption trend recently. To obtain a chromosome-doubled (12x) persimmon, colchicine was applied at the meristem of seedlings in vitro derived from cross among hexaploid persimmon (Diopyros kaki Thunb.). These were treated with 0.03%, 0.05% and 0.1% colchicine respectively for doubling chromosome, and it was most effective at the concentration of 0.05% colchicine. After colchicine treatment, we conducted tests to elucidate conditions for inducing shoot and root development. As the result, the shoots grew best when cultivated at 1/2MS media plus 10 and $30{\mu}M$ zeatin respectively, and the roots grew best when cultivated at 1/2MS media after dipping for 5 seconds at 10 mM NAA+5% DMSO. We also compared seedlings that have chromosome (6x) do not doubled and crossing parents (6x) and chromosome-doubled seedlings (12x). As the result, these chromosome-doubled seedlings (12x) showed lower stomatal density and larger stomatal size.

Precise, fast, and cost-effective identification of crop cultivars is essential for plant breeder's rights. Traditional methods for identification of persimmon cultivars are based on the evaluation of sets of morphological characteristics. However, it is difficult to distinguish closely related cultivars using only morphological traits. This study was conducted to develop DNA markers for identification of the 32 persimmon cultivars in Korea and Japan. A total of 309 randomly amplified polymorphic DNA (RAPD) markers were identified using 40 different random primers. Various number of polymorphic bands ranged from 4 (OPP-08) to 14 (UBC159) were detected with an average of 7.7. Resulting 57 RAPD fragments were selected, and their sequences were determined for developing sequence characterized amplified region (SCAR) markers. As a result, 15 of 57 RAPD fragments were successfully converted to SCAR markers. Single polymorphic bands of the same size as or smaller than the RAPD fragments were amplified depending on SCAR markers. Among these markers, a combination of eight SCAR markers (PS225_200, PSN05_420, PSF13_523, PSN11_540, PS372_567, PS485_569, PSP08_635, and PS631_735) provided sufficient polymorphisms to identify 32 persimmon cultivars. These newly developed markers will be a fast and reliable tool to identify persimmon cultivars.
This study was conducted to elucidate the cause of micro-cracking in the exocarp of 'Jinmi' peach (Prunus persica). through the microscopic observation of fruit skin development in 4 varieties such as 'Jinmi', 'Kanoiwa Hakuto', 'Kawanakajima Hakuto', and 'Yumyeong'. Micro-cracking was noted in 59.1% of 'Jinmi', 30.6% of 'Kanoiwa Hakuto', 21.5% 'Kawanakajima Hakuto' and 6.4% of 'Yumyeong', respectively. The development of intercellular spaces, which increased rapidly with the fruit development, was easily observed at 69 days after full bloom. Histological studies revealed that the number of outer epiderm cell layers of 'Jinmi' was smaller than that of the other three cultivars, and thinner than the 'Kawanakajima Hakuto' and 'Yumyeong'. Moreover, 'Jinmi' exhibited smaller and flatter shapes in the sub-epidermal cell layer than those of the 'Kawanakajima Hakuto' and 'Yumyeong' at harvest season. Therefore, these results suggest that micro-cracking of 'Jinmi' fruit skin was due to poor-developed outer epidermis and well-developed intercellular spaces just under exocarp as compared with other varieties.
Root-knot nematodes (Meloidogyne spp.) are the most destructive group of plant-parasitic nematodes. Plants infected by Meloidogyne spp. develop above-ground symptoms, stunting, yellowing, nutrient deficiencies, and gall formations with typical hook-shaped root tips. Infected plants experience yield losses. During 2018-2019 survey, leaf chlorosis rice plants were found in 206 fields of 67 counties in Guangxi, China, around 30 days after transplanting. Galls and hooked tips on the roots and pear-shaped females were observed. About 32.04% of fields were infested with the nematode. The nematodes were identified as Meloidogyne graminicola base on morphological and molecular analysis. To the best of our knowledge, this is the first report of M. graminicola on rice plants in Guangxi, China. The results of this study urge the discovery of resistant cultivars and the development of management strategies.
This study was performed to confirm physiological activities according to cultivars and parts of Ulsan pear (wonhwang, pungsu, whangkeum, whasan and shingo). Total contents of phenolic compounds from peel, pulpy substance and core were 699.3-800.6, 51.5-112.5 and 254.0-401.5 mg/100 g as tannic acid equivalent, respectively. There were difference contents by cultivars, and peel and core of shingo and pulpy substance of wonhwang showed high contents, respectively. Total flavonoid contents of peel, pulpy substance and core were 125.2-164.2, 25.9-35.9 and 45.1-60.0 mg/100g, respectively and those of shingo cultivar showed comparatively high. Electron donating ability was in the order of peel (66.1-90.7%), core (48.5-82.8%) and pulpy substance (24.9-58.2%), and whasan cultivar showed comparatively low. Nitrite scavenging activity was in the order of peel (58.2-100.8%), core (59.5-86.2%), pulpy substance (39.9-82.5%). There were little difference by cultivars of core but peel and pulpy substance of shingo cultivar showed comparatively low nitrite scavenging activity. And as the concentration of each extract increased, nitrite scavenging activity increased. Xanthine oxidase inhibition rate was in the order of peel (14.1-75.4%), core (5.3-71.8%), pulpy substance (2.2-67.5%). And there little difference by cultivars and which was increased as the concentration of each extract increased.

Most deciduous trees in temperate zone are dormant during the winter to overcome cold and dry environment. Dormancy of deciduous fruit trees is usually separated into a period of rest by physiological conditions and a period of quiescence by unfavorable environmental conditions. Inconsistent and fewer budburst in pear orchards has been reported recently in South Korea and Japan and the insufficient chilling due to warmer winters is suspected to play a role. An accurate prediction of the flowering time under the climate change scenarios may be critical to the planning of adaptation strategy for the pear industry in the future. However, existing methods for the prediction of budburst depend on the spring temperature, neglecting potential effects of warmer winters on the rest release and subsequent budburst. We adapted a dormancy clock model which uses daily temperature data to calculate the thermal time for simulating winter phenology of deciduous trees and tested the feasibility of this model in predicting budburst and flowering of Niitaka pear, one of the favorite cultivars in Korea. In order to derive the model parameter values suitable for Niitaka, the mean time for the rest release was estimated by observing budburst of field collected twigs in a controlled environment. The thermal time (in chill-days) was calculated and accumulated by a predefined temperature range from fall harvest until the chilling requirement (maximum accumulated chill-days in a negative number) is met. The chilling requirement is then offset by anti-chill days (in positive numbers) until the accumulated chill-days become null, which is assumed to be the budburst date. Calculations were repeated with arbitrary threshold temperatures from $4^{\circ}C$ to $10^{\circ}C$ (at an interval of 0.1), and a set of threshold temperature and chilling requirement was selected when the estimated budburst date coincides with the field observation. A heating requirement (in accumulation of anti-chill days since budburst) for flowering was also determined from an experiment based on historical observations. The dormancy clock model optimized with the selected parameter values was used to predict flowering of Niitaka pear grown in Suwon for the recent 9 years. The predicted dates for full bloom were within the range of the observed dates with 1.9 days of root mean square error.
We tested fruits quality and skin coloration of two pear cultivar 'Wonhwang' and 'Whasan' produced by bagging with different kinds of paper bags in the Cheonan area, Chungnam Province, Korea. The fruits were bagged at 35, 45, 55 or 65 days after full bloom (DAFB) with 3 kinds of double layer paper bag which consisted of grey color, yellow color and newspaper for outer layer. The great extent of light interruption was observed in grey colored outer paperbag among three kinds of paperbag which showed only 0.46% of lowest light transmittance when compared with 43.7% and 40.0% of yellow paper and newspaper, respectively. The development of skin redness ($a^*$) increased with the delay of bagging time in two pear cultivars. But the fruits treated at 65 DAFB showed uneven coloration and excessive development of redness which represented low fruit external appearance value. Uniform flesh firmness was attained at the fruits bagged with grey outer color paperbag regardless of bagging time, although the fruit quality indices including soluble solids and titratable acidity did not change significantly by using different kind of fruit bags and bagging time in two pear cultivar. Based on our results, it was explained that the grey-colored fruit bag had positive effect on the development of skin coloration without any detrimental effect on fruit quality factors in 'Wonhwang' and 'Whasan' pears.
