Genetic Variation and Relationships of Korean Native Chickens and Foreign Breeds Using 15 Microsatellite Markers

  • The purpose of this study was to assess the genetic variation and establish the relationship amongst breeds and strains using 15 chicken specific microsatellite markers. A total of 285 unrelated DNA samples from four Korean native chicken strains (Black strain of Korean native chicken; KL, Red Brown strain of Korean native chicken; KR, Ogol strain of Korean native chicken; KS and Yellow Brown strain of Korean native chicken; KY) and three introduced chicken breeds (F strain of White Leghorn; LF, K strain of White Leghorn; LK, Rhode Island Red; RC and Cornish; CN) were genotyped to estimate within and between breed genetic diversity indices. All the loci analyzed in 15 microsatellite markers showed a polymorphic pattern and the number of alleles ranged from 5 to 14. The polymorphism information content (PIC) of UMA1019 was the highest (0.872) and that of ADL0234 was the lowest (0.562). The expected total heterozygosity (He) within breed and mean number of observed alleles ranged from 0.540 (LF) to 0.689 (KY), and from 3.47 (LK) to 6.07 (KR), respectively. The genetic variation of KR and KY were the highest and the lowest within Korean native strains, respectively. The genetic distance results showed that Korean native chicken strains were separated with the three introduced chicken breeds clustered into another group. The lowest distance (0.149) was observed between the KR and KL breeds and the highest distance (0.855) between the KR and LK breeds. The microsatellite polymorphism data were shown to be useful for assessing the genetic relationship between Korean native strains and other foreign breeds.

  • 1. We have observed the frequency , geographi variation and microgeographic variation of the elytral pattern types and the elytral edge of the population of Lady-beetle, Harmonia axyridis PALLACE, collected in Jinju and other 6 localitites in South Korea. 2. the number of specimen collected were 6,625 in which Succinea was 90.32% : Axyridis ; 0.74% : Spectabilis , 3.82% ; and Conspicua 5.07%. 3. We have found that 92.26% of all specimens have elytral edge. 4. There were some differences in the compositional of the elytral pattern types and elytral edge to the population among the Western seaside , inland and southern seaside of South Korea , whereas as great difference was in the compositional proportion tot he population of Spectabilis between the western and southern seaside . There was also a gret difference of the elytral edge between the inland and southern seaside. 5. A micro-geographic variation was observed in Jinju , located on southern area of South Korea. In the samples collected from the pine trees at Jinju area the Succinea occupied 73.25%, and 69.61 % were found to be provided with elytral edge, while in the general population 92.46% of specimen were Succinea and 98.35% were found to have elytral edge.

댕댕이나무(Lonicera caerulea var. edulis) 국내 잔존 집단의 유전적 다양성 (Genetic Diversity of Lonicera caerulea var. edulis in South Korea)

  • Lonicera caerulea var. edulis is a rare species found in some alpine region of Korea. Genetic variation in L. caerulea var. edulis has been investigated by examining 161 individuals from six natural populations: Mt. Seorak 1, Mt. Seorak 2, Mt. Jeombong, Mt. Bangtae, Mt. Gyebang, Mt. Halla. The mean genetic diversity for all the six populations was 0.25 (S.I.). The highest genetic diversity was found in Mt. Seorak (S.I.=0.3158) and the lowest was in Mt. Gyebang (S.I.=0.1047). Comparatively low level of genetic diversity was observed (Ae=1.25, P= 64.6%, S.I.=0.25), which is a typical pattern for rare tree species. AMOVA showed exceptionally large proportion of genetic variations both for among populations (34.69%) and within populations (65.31%). Excluding Mt. Gyebang, the genetic variation among and within population was 18.71% and 81.29% respectively. The UPGMA dendrogram based on genetic distance is not suitable for geographic relationship. Genetic distance of Mt. Gyebang was most distant from the other populations. Excluding Mt. Gyebang, the genetic identities among the five populations were 0.95 to 0.97, which is very high similarity level of genetic identity. This low level of genetic variations and the lack of site in nature indicates that Lonicera caerulea var. edulis demanded a serious conservation.

한국산 바위솔속(돌나물과) 5종에 대한 유전적 변이 (Genetic variation in five species of Korean Orostachys (Crassulaceae))

  • 한국산 돌나불과 바위솔속 식물의 동위효소 변이와 분류군의 실체를 알아보기 위해 5종 24 개 집단을 대상으로 전분전기영동을 실시하였다. 동위효소 분석 결과로 나타난 전형질도에 의하면 5종의 한국산 바위솔속 식물은 2개의 주요 군으로 나누어 졌다. 그리고 이들 두 군은 기존에 분자 형질과 형태 형질을 기초로 한 결과에서 바위솔속을 Appendiculatae아절과 Orostachys아절로 나누는 처리와 일치하였다. 한국산 바위솔속 종간의 낮은 유전적 동질성은 본 속의 종들이 지리적 종분화 과정을 통해 점진적으로 분화되었음을 시사한다. 유사한 생활사와 생식 양상을 갖고 있는 종들의 유전적 자료와 비교해 보면, 넓게 분포하는 바위솔이 좁게 분포하는 다른 종들에 비해 상대적으로 높은 유전자 변이를 나타내고 있으나, 바위솔속 식물들은 매우 낯은 유전적 변이를 보여주고 있으며, 이는 격리된 서식처, 집단 내 적은 수의 개체, 서식처의 파괴, 근친교배, 무성생식 등의 결과일 가능성이 크다. 정동진에서 채집된 둥근바위솔 집단(POP 21) 은 다른 둥근바위솔과는 유전적으로는 이질적인 균으로 기존의 화분학적, 형태학적 연구 결과와도 일치하고 있다. 본 동위효소 자료는 최근 신분류군으로 발표된 울릉연화바위솔과 진주바위송의 분류학적 처리를 지지하고 있다.

Assicuation between Genetic Variation of the Insulin Receptor Gene and Essential Hypertension in the Korean Population

  • Essential hypertension is a multifactorial disease, and has been shown to be associated with insulin resistance. The relationship between the genetic variation of the insulin receptor (INSR) gene and essential hypertension In Korean population was investigated by the Nsi 1 restriction fragment length polymorphism (RFLP) pattern of this gene. The observed genotype frequencies of INSR gene were not deviated from those expected for the Hardy-Weinberg equilibrium (HWE), but a significant association was observed between essential hypertension and N1 allele of Nsi 1 RFLP at the INSR gene ($X^2$-test; P<0.05). Moreover, the frequency of N1 allele was significantly different between normotensives and essential hypertensives in subgroups that were not obese ($X^2$-test; P<0.05). These data suggest that the Nsil RFLP of INSR gene may be a useful genetic marker for essential hypertension in Korean population.

  • Genetic and morphological variations of Rhinogobius giurinus were surveyed. Populations of R.giurinus were clearly divided into two forms (Form-A and Form-B). Starch gel variation of this species. Three loci (Aco, Mpi, aGpd) out of 27 showed fixed allelic differences between Form-A and Form-B and they are well differentiated from each other genetically (Rogers' S=0.871). These two forms, moreover, are found to be allopatric in distribution and morphologi cally different in body length and caudal fin color pattern. Therefore, they are considered as two distinct taxa of subspecific rank. In addition, the genetic relationships among 5 taxa within 4 species of the genus Rhinogobius were investigated. Three species of the Rhinogobius brunneus complex (R.sp. OR, R. sp. CB and R. sp. CO) are well differentiated from each other genetically and two taxa of R. giurinus are genetically divergent from three species of the Rhinosotius brunneus complex (average Nei's D=0.603, average Rogers'=0.534).

Comparison of Population Genetic Structure of Two Seashore-Dwelling Animal Species, Periwinkle Littorina brevicula and Acorn Barnacle Fistulobalanus albicostatus from Korea

  • The genetic structure of marine animals that inhabit the seashore is affected by numerous factors. Of these, gene flow and natural selection during recruitment have strong influences on the genetic structure of seashore-dwelling species that have larval periods. Relative contributions of these two factors to the genetic structure of marine species would be determined mainly by the duration of larval stage. The relationship between larval period and genetic structure of population has been rarely studied in Korea. In this study, genetic variations of cytochrome oxidase subunit I (COI) were analyzed in two dominant species on rocky shore habitats in the Korean peninsula: periwinkle Littorina brevicula and acorn barnacle Fistulobalanus albicostatus. Both species are not strongly structured and may have experienced recent population expansion. Unlike periwinkle, however, barnacle populations have considerable genetic variation, and show a bimodal pattern of mismatch distribution. These results suggest that barnacle populations are more affected by local adaptation rather than gene flow via larval migration. The bimodal patterns of barnacle populations observed in mismatch distribution plots imply that they may have experienced secondary contact. Further studies on seashore-dwelling species are expected to be useful in understanding the evolution of the coastal ecosystem around Korean waters.

특정변화패턴 식별을 위한 염기서열 집단간의 다형성 분석 및 시각화 도구 (A Polymorphism Analysis and Visualization Tool for Specific Variation Pattern Identification in Groups of Nucleotide Sequences)

  • 유전체는 생명체가 가지고 있는 모든 유전적 정보를 담고 있다. 특정 종 내에서는 개체별로 고유의 특성이 나타나며, 이 특성은 유전체의 염기서열 분석을 통해 확인할 수 있다. 종내 개체들 사이에 조금씩 다른 염기에 대해 유전적 연관성을 규명 짓고, 더 나아가 질병과의 연관성을 찾는 전장유전체 연관분석 연구가 많이 진행되고 있다. 종 내의 조금씩 발생하는 염기변이를 파악하는 것은 개체의 다형성을 파악하기 위해 중요하다. 이 논문에서는 종 내 여러 개체의 염기서열에서 대립형질 빈도의 특정변화패턴을 쉽게 파악할 수 있는 분석 및 시각화 도구를 제안한다. 그리고 수두 대상포진 바이러스의 계대 배양한 pOka strain 염기서열 데이터를 이용해 실험하여 분석과 시각화의 실용성을 보인다. 본 제안도구를 통해 종 내의 대립형질 빈도의 변화를 탐색하고 유전적 요인을 찾는 연구효율의 증진을 기대할 수 있다.

EST-SSR Based Genetic Diversity and Population Structure among Korean Landraces of Foxtail Millet (Setaria italica L.)

  • Understanding the genetic variation among landrace collections is important for crop improvement and utilization of valuable genetic resources. The present study was carried out to analyse the genetic diversity and associated population structure of 621 foxtail millet accessions of Korean landraces using 22 EST-SSR markers. A total of 121 alleles were detected from all accessions with an average of 5.5 alleles per microsatellite locus. The average values of gene diversity, polymorphism information content, and expected heterozygosity were 0.518, 0.594, and 0.034, respectively. Following the unweighted neighbor-joining method with arithmetic mean based clustering using binary data of polymorphic markers, the genotypes were grouped into 3 clusters, and population structure analysis also separated into 3 populations. Principal coordinate analysis (PCoA) explained a variation of 13.88% and 10.99% by first and second coordinates, respectively. However, in PCoA analysis, clear population-level clusters could not be found. This pattern of distribution might be the result of gene flow via germplasm exchanges in nearby regions. The results indicate that these Korean landraces of foxtail millet exhibit a moderate level of diversity. This study demonstrated that molecular marker strategies could contribute to a better understanding of the genetic structure in foxtail millet germplasm, and provides potentially useful information for developing conservation and breeding strategies.

RAPD 표지인자를 이용한 흑오미자의 자웅동주 및 자웅이주 식물의 동정 (Identification of Monoecious and Dioecious Plants of. Schisandra nigra Using the RAPD Markers)

  • 본 연구는 국내의 경우 제주도의 일부지역에서만 자생하는 흑오미자(Schisandra nigra)를 RAPD법을 이용하여 자웅동주 및 자웅이주 식물의 특이적 Marker를 탐색하고저 실시 하였다. 10-mer로 구성된 80종류의 random primer를 사용하여 흑오미자를 분석한 결과 기존의 재배되고 있는 오미자(Schisandra chinensis) 또는 남오미자(Kadsura japonica)와는 다른 band pattern을 보여 주었다. 흑오미자의 자웅동주. 암그루, 숫그루의 3품종을 상기와 동일하게 80개의 primer를 사용하여 RAPD를 분석한 결과, 5종류의 random primer(OPA-17, OPA-19, OPB-3, OPB-9, OPB-16)에 대해서는 각 품종에 대한 특이적인 band가 검출되었다. 숫그루, 암그루 식물과는 다르게 자웅동주 식물은 3개체(1호 2호, 3호)간에도 서로 다른 band pattern을 보이는 특징을 갖고있다. 숫그루 특이적인 band pattern은 OPB-3 primer을 이용할 경우 750bp에서 검출되었고, 암그루는 OPA-19 primer에서 950bp, 1690bp와 OPB-3 primer의 경우 700bp에서 자웅동주 및 숫그루에서 나타나지 않는 band가 검출되었다. 이러한 결과는 흑오미자의 숫그루 및 암그루에서 나타난 특이적인 band가 유묘시기에 암ㆍ수 개체를 조기에 구별하는 genetic marker로 사용할 수 있으리라 기대된다.
