Yersinia enterocolitica 의 병원성 검정에 관한 연구 (Studies on the Pathogenic Test of Yersinia enterocolitica)

  • 국내외에서 분양 받고 국내식육에서 분리된 총 100균주의 Y. enterocolitica에 대한 병원성여부를 HEp-2세포 침투성 시험법을 기준으로 하고 여러 가지 병원성 확인 방법을 비교하였다. 혈청형만으로 병원성을 판단하기에는 충분하지 않았고 이외의 방법 즉, esculin과 salicin시험, pyrazinamidase 시험, biotype 등의 생화학적 특성에 바탕을 둔 실험 단독으로 병원성을 판단하기에는 완전하지 않았다. D-xylose 발효실험, CRMOX agar 시험과 자동응고반응(Autoagglutination)들은 병원성확인에 보조적인 역할만을 할 수 있었다. HEp-2 세포 침투성 시험법은 시간과 노력이 많이 소요되므로 이 방법을 대치할 수 있는 간단하고 신속, 정확하게 Y. enterocolitica 병원성을 확인할 수 있는 방법은 esculin시험에서 음성반응을 보이는 것만 PCR시험을 행하여 판별하는 것이라고 사려된다.

  • In order to investigate the pathogenic genes and genetic relationships of Y. pseudotuberculosis, we isolated 9 strains of Y. pseudotuberculosis from about 380 spring water sites in Seoul and carried out antibiotic susceptibility test, biological test and molecular typing. All isolated strains were distributed throughout the northeast area in Seoul (Mt. Bookhan, Mt. Soorak, Mt. Boolam and etc...).Antibiotic susceptibility test revealed that all the strains were susceptible to chloramphenicol, gentamicin, neomycin and amoxicillin/clavulanic acid, but were resistant to novobiocin and vancomycin.   • A fluorescent microsphere-based immunochromatographic strip test (FICT) was developed for the rapid, sensitive, and quantitative detection of porcine reproductive and respiratory syndrome virus (PRRSV) antibodies at the pen-side. The assay was based on the formation of a sandwich immune-complex (anti-pig IgG-PRRSV antibodies-NSP7/N), which was validated by a comparison with IDEXX-ELISA using 3325 clinical specimens. The diagnostic specificity, sensitivity, and accuracy of FICT were 97.28, 93.41, and 94.95%, respectively. FICT showed a good correlation with the virus neutralization assay. Overall, a promising pen-side diagnostic tool was developed for the rapid and quantitative detection of PRRSV antibodies within 15 min.

경북지역 가축에서 분리된 Salmonella typhimurium과 S enteritidis의 병원성 시험 (Pathogenicity of Salmonella typhimurium and S enteritidis isolated from domestic animals in Gyeongbuk province)

  • The result of studying the pathogenicity of Salmonella typhimurium and S enteritidis isolated from domestic animals in Gyeongbuk province were summarized as follows. In Congo-red binding test, S typhimurium had much more rough types than S enteritidis. In colicin production test, 4 strains of S typhimurium were positive but all of S enteritidis were negative. In hemolysin production test, all of S typhimurium and S enteritidis were negative. In Guinea pig serum resistant test, all of S typhimurium and S enteritidis were positive. As a result of pathogenicity test to mice, 54.4% of mice were died. Therefore, S typhimurium and S enteritidis were considered as highly pathogenic. S typhimurium DT104 and S enteritidis PT4 were more pathogenic to mice than other phage types of same serovar. S typhimurium and S enteritidis were considered not so pathogenic for 6-day-old chickens. The recovery rates of Salmonella stains from mice and chickens inoculated were 96.8%, and 54%, respectively. In chickens, proportional to the time from 2 weeks after challenge inoculation, the recovery rates were noticeably decreased.

  • Bacillus polyfermenticus SCD which is commonly called as Bisroot (equation omitted) has been appropriately used for the treatment of long-term intestinal disorder's. This strain strongly inhibited against methicillin resistance Staphylococcus aureus (MRSA) and various pathogenic microorganisms. Effects of B. polyfermentius SCD administration on death rates and egg-laying rates in two groups of hens were investigated. This strain decreased the death rates of two groups by 16.26% and 11.72%, respectively. Also this strain increased the egg-laying rates of those 2.74% and 2.66%, respectively Clinical tests of B. polyfermenticus SCD administration to healthy adults showed not adverse effects but decreased glyceride concentration from 154.52 mg/dl to 135.41 mg/dl after two week administrations.

Characterization of Phage-Resistant Strains Derived from Pseudomonas tolaasii 6264, which Causes Brown Blotch Disease

  • Pseudomonas tolaasii 6264 is a representative strain that causes bacterial blotch disease on the cultivated oyster mushroom, Pleurotus ostreatus. Bacteriophages are able to sterilize the pathogenic P. tolaasii strains, and therefore, they can be applied in creating disease-free mushroom cultivation farms, through a method known as "phage therapy". For successful phage therapy, the characterization of phage-resistant strains is necessary, since they are frequently induced from the original pathogenic bacteria in the presence of phages. When 10 different phages were incubated with P. tolaasii 6264, their corresponding phage-resistant strains were obtained. In this study, changes in pathogenic, genetic, and biochemical characteristics as well as the acquired phage resistance of these strains were investigated. In the phylogenetic analyses, all phage-resistant strains were identical to the original parent strain based on the sequence comparison of 16S rRNA genes. When various phage-resistant strains were examined by three different methods, pitting test, white line test, and hemolytic activity, they were divided into three groups: strains showing all positive results in three tests, two positive in the first two tests, and all negative. Nevertheless, all phage-resistant strains showed that their pathogenic activities were reduced or completely lost.

청대설진의안외감병여내상병적설상특정연구 (The tongue demonstration characteristic study between diseases caused by exogenous pathogenic factor and disorder of internal organs of the glossoscopy medical records in Qing Dynasty)

  • Objective To learn if there were different characteristics of tongue demonstration between diseases caused by exogenous pathogenic factor and disorder of internal organs. Methods Choosing 3059 cases from 51 medical records of Qing Dynasty, in which 1397 cases were diagnosed as diseases caused by exogenous pathogenic factor and 1110 were disorder of internal organs. Contrasting the constituent ratio of tongue demonstration using chi-square test. Results There were significant deviation between diseases caused by exogenous pathogenic factor and disorder of internal organs in color of tongue, color of fur and quality of fur (p<0.01). Conclusions the characteristics of tongue demonstration in diseases caused by exogenous pathogenic factor were high frequency of bright red tongue, yellow fur, blank fur and dry fur; and which in disorder of internal organs were high frequency of white tongue, thin fur, greasy fur and exfoliative fur.

Pear Skin Stain Caused by Mycosphaerella graminicola on Niitaka Pear (Pyrus pyrifolia Nakai)

  • Pear skin stains on 'Niitaka' pears, which occur from the growing stage to the cold storage stage, reportedly negatively influence the marketing of pears. These stains on fruit skin are likely due to a pathogenic fungus that resides on the skin and is characterized by dark stains; however, the mycelium of this fungus does not penetrate into the sarcocarp and is only present on the cuticle layer of fruit skin. A pathogenic fungus was isolated from the skin lesions of infected fruits, and its pathogenicity was subsequently tested. According to the pathogenicity test, Mycosphaerella sp. was strongly pathogenic, while Penicillium spp. and Alternaria spp. showed modest pathogenicity. In this present study, we isolated the pathogenic fungus responsible for the symptoms of pears (i.e., dark brown-colored specks) and identified it as Mycosphaerella graminicola based on its morphological characteristics and the nucleotide sequence of the beta-tubulin gene. M. graminicola was pathogenic to the skin of 'Niitaka' pears, which are one of the most widely growing varieties of pears in South Korea.

방선균이 생산하는 Cephalosporin 내성 병원성 Pseudomonas에 유효한 항생물질 (An Antibiotic from Actinomycetes Becoming Effective for Cephalosporin Resistant Pathogenic Pesudomonas sp.)

  • We isolated activnmycetes LAM-98-80 as strain producing an effective antibiotic for cephalosporin re-sistant pathogenic PSeudomonas sp. and identified as Streptomyces sp. LAM-98-80 from cultural and phyisological characteristics. We investigated the optimal culture conditions for producation of an anti-biotic becoming effective for cephalsporin-resistant pathogenic Pseudomonas sp. It was found that 1.5% soluble starch and 1.0% yeast extract were good as carbon and nitrogen source respectively. The pro-duction of antibiotic was also activated by 0.04% Mn2+ as 80% degree. The optimum initial pH on pro-ductio of antibiotic was pH 7.0. The culture condition for the maximal productivity of the antibiotic was at 3$0^{\circ}C$ for 5 days. The cephalosporin-resistant pathogenic Pseudomonas sp. as test bacteria was rev-ealed to resist antibiotic of cepha families but revealed to not resist those of $\beta$-lactam families ampicil-lin and amoxicillin. Parital purified antibiotic was stable for the pH from 3 to 9 and was also stable when treated at 70 $^{\circ}C$ for 1 hour, This antbiotic was effective against all gram positive and negative bac-teria but was not effective against molds and yeasts.

Effects of Compost Tea Making from Differently Treated Compost on Plant Disease Control

  • Antifungal activity of compost teas was evaluated in vitro and in vivo experiments. In vitro test, fourteen compost teas were produced using five different composts and through six different methods for compost tea production. Eleven pathogenic fungi were used as indicators of antifungal activity of compost teas. In vivo test, one of the compost teas used in vitro test was sprayed to cucumber leaves which were infected with powdery mildew in order to evaluate antifungal activity of compost tea. From the results of the tests, it was found that all compost teas used this study showed antifungal activity and the proper application of compost teas against pathogenic fungi for obtaining optimal effects was important to know.
