Two-Dimensional Electrophoresis Analysis of Proteins between Bacillus licheniformis DM3 and Its Antifungal Activity Deficient Mutant (이차원전기영동법을 이용한 길항세균 Bacillus licheniformis DM3와 이의 항진균 활성 결여 돌연변이균주간 단백질 비교 분석)

  • In the course of screening for antifungal agents, a bacterial strain, DM3 was isolated from a mud sample collected at Daechon in Chungnam province and identified as Bacillus licheniformis based on API 50CHB test. It has antifungal activity against 12 plant pathogenic fungi in paper disc assay. At the 95% lethal dose of gamma radiation ($^{60}Co$, 10 kGy, $D_{10}=2.32\;kGy$), the antifungal activity deficient mutant (mDM3) against Colletotrichum gloeosporioides was induced From 2-D electrophoresis analysis, serine hydroxymethyltransferase (45.0 kDa), hypothetical protein(40.7 kDa), NifU protein homolog(15.4 kDa), and resolvase(12.5 kDa) homologous proteins were detected only in B. licheniformis DM3. Lysozyme(18.1 kDa) and alkyl hydroperoxide reductase(15.6 kDa) homologous proteins were expressed uniquely in B. licheniformis mDM3. Further studies are needed to reveal that these proteins from B. licheniformis DM3 could be closely related to the antifungal activity against plant pathogenic fungi.

  • The purpose of this study was to determine the thermal inactivation of Salmonella enteritidis, Salmonella typhimurium and E. coli O111 in liquid cultures treated with microwave energy. Furthermore, this study was to introduce new methodologies for studying nonthermal microwave effects on microorganisms, using controlled microwave energy and specially designed apparatuses. For the automatic temperature control during microwave heating, the real time data acquisition and computation system is designed with BASIC routine. The automatic temperature control system used in the experiments perform relatively stable control at the experiment temperature of 45, 50, 55 60$^{\circ}C$ and 65$^{\circ}C$ for 30 minutes. The effects of microwave heating on liquid cultures was compared with that of conventional heating, still reduces effectively the number of pathogenic bacteria in liquid cultures. While no particular differences between microwave heating and conventional heating was observed in the activation of E. coli at 45$^{\circ}C$ test, the activation of Sal. enteritidis and Sal. typhimurium was slightly reduced during the microwave treatments.

Alterations of Protein Expression in Macrophages in Response to Candida albicans Infection

  • Although macrophages are an important first line of cellular defense, they are unable to effectively kill phagocytosed C. albicans. To determine the physiological basis of this inability, we investigated the alterations of macrophage proteins caused by C. albicans infection. Since the formation of C. albicans hyphae caused cell death, proteins were prepared 3 h after infection and examined by two-dimensional gel electrophoresis (2-DE). The most prominent changes were in glycolytic enzymes, which could have caused energy depletion of the infected cells. Also changed were proteins involved in maintenance of cellular integrity and NO production. Treatment of the macrophages with either cytochalasin D or taxol did not alter their inability to kill C. albicans. Our results indicate that multiple factors contribute to cell death as the pathogenic form of C. albicans becomes fully active inside macrophage cells.

Endophytic Trichoderma gamsii YIM PH30019: a promising biocontrol agent with hyperosmolar, mycoparasitism, and antagonistic activities of induced volatile organic compounds on root-rot pathogenic fungi of Panax notoginseng

  • Background: Biocontrol agents are regarded as promising and environmental friendly approaches as agrochemicals for phytodiseases that cause serious environmental and health problems. Trichoderma species have been widely used in suppression of soil-borne pathogens. In this study, an endophytic fungus, Trichoderma gamsii YIM PH30019, from healthy Panax notoginseng root was investigated for its biocontrol potential. Methods: In vitro detached healthy roots, and pot and field experiments were used to investigate the pathogenicity and biocontrol efficacy of T. gamsii YIM PH30019 to the host plant. The antagonistic mechanisms against test phytopathogens were analyzed using dual culture, scanning electron microscopy, and volatile organic compounds (VOCs). Tolerance to chemical fertilizers was also tested in a series of concentrations. Results: The results indicated that T. gamsii YIM PH30019 was nonpathogenic to the host, presented appreciable biocontrol efficacy, and could tolerate chemical fertilizer concentrations of up to 20%. T. gamsii YIM PH30019 displayed antagonistic activities against the pathogenic fungi of P. notoginseng via production of VOCs. On the basis of gas chromatography-mass spectrometry, VOCs were identified as dimethyl disulfide, dibenzofuran, methanethiol, ketones, etc., which are effective ingredients for antagonistic activity. T. gamsii YIM PH30019 was able to improve the seedlings' emergence and protect P. notoginseng plants from soil-borne disease in the continuous cropping field tests. Conclusion: The results suggest that the endophytic fungus T. gamsii YIM PH30019 may have a good potential as a biological control agent against notoginseng phytodiseases and can provide a clue to further illuminate the interactions between Trichoderma and phytopathogens.

One-step phyto-mediated fabrication of silver nanoparticles and its anti-microbial properties

  • This manuscript describes the one-step eco-friendly green fabrication of silver nanoparticles (AgNPs) through the in-situ bio-reduction of an aqueous solution of silver nitrate using Syzygium aromaticum leaf extract. UV-vis spectroscopy shows a characteristic SPR peak around 442 nm. FTIR spectroscopy showed that the AgNPs were capped with bioactive phyto-molecules. TEM images revealed oval and spherical particles with a mean diameter of ~12.6 nm. XRD analysis revealed crystalline and face-cantered cubic AgNPs. The phytosynthesized AgNPs showed broad-spectrum anti-microbial activity against two foodborne pathogenic bacteria, Listeria monocytogenes and Staphylococcus aureus. The AgNPs showed a prominent ability to inhibit biofilms formed by L. monocytogenes and S. aureus in laboratory conditions through a crystal violet assay. The results suggest that the AgNPs could be a novel nanotool to develop effective antimicrobial and anti-biofilm agents in food preservation.

  • The purpose of this article was to compare 'The Discourse on the Constitutional Symptoms and Disease' of ${\ulcorner}$Dongyi Soose Bowon${\lrcorner}$ written in 1894(Old Edition(舊本)) with that of ${\ulcorner}$Dongyi Soose Bowon${\lrcorner}$ published in 1901(In Edition(印本)), and to find the idea of pathologic mechanism and classification of 'the Exterior and Interior disease'. the conclusions were as follows. 1. The classification of constitutional symptoms and disease of Soeumin and Soyangin in 'Old Edition(舊本)' was almost equal to that in 'In Edition(印本)' 2. In pathological mechanism of constitutional symptoms and disease of Soeumin and Soyangin, 'The Exterior Disease' could be explained as the disease resulted from fight between 'Yang-chi(陽氣)(Hot-chi(熱氣))'of 'Thoracic vertebrae' and 'Yin-chi(陰氣)(Cold-chi(寒氣))' of 'Bladder' and 'The Interior Disease' between 'Hot-chi(熱氣)(Stomach-chi(胄氣))' of 'Stomach' and 'Cold-chi(寒氣)' of 'Large intestine'. 3. 'The Exterior Symptoms and Disease of the Exterior and the Interior Disease(表裏之表病)' could be explained as the disease occurring at the Branch portion(large portion)(標) by overcoming of Pathogenic factors but Vital energy still sufficient, and 'The Interior Symptoms and Disease of the Exterior and the Interior Disease(表裏之裏病)' occurring at Root portion(small portion)(裏) by invasion of Pathogenic factors and Vital energy almost exhausted. 4. In the classification of constitutional symptoms and disease of Taeumin, 'The Exterior Symptoms and Disease of the Exterior and the Interior Disease(表裏之表病)' in 'Old Edition(舊本)' were rearranged to 'The Exterior Disease' in 'In Edition(印本)', 'The Interior Symptoms and Disease of the Exterior and the Interior Disease(表裏之裏病)' to 'The Interior Disease'. 5. It was assumed that 'The Exterior and the Interior Disease' of Taeumin could be explained in relation between the exterior and e interior, based on the Healthy energy(保命之主) and e concept of the Branch and the Root portion

A Putative Early Response of Antifungal Bacillus lentimorbus WJ5 Against the Plant Pathogenic Fungus, Colletotrichum gloeosporioides, Analyzed by a DNA Microarray

  • The global RNA transcription profiles of Bacillus lentimorbus WJ5 under an in vitro co-culture with Colletotrichum gloeosporioides were analyzed in order to study the antagonistic bacteria-fungi interactions. Using a filter membrane system, B. lentimorhus WJ5 was exposed to the spores of C. gloeosporioides at the late exponential stage. The transcription profiles of the B. lentimorhus WJ5, both with and without a challenge from C. gloeosporioides, were analyzed using custom DNA chips containing 2,000 genome fragments. A total of 337 genes were expressed, with 87 and 47 up- and down-regulated, respectively. Of these, 12 genes, which were involved in central carbon metabolisms, and 7 from minor catabolism were relatively highly up-regulated (> 10 fold) and down-regulated (< 0.2 fold), respectively. Nine genes, which were thought to be related to the antifungal activity, were also up-regulated, but their levels were not so high (2.0 - 9.7 folds). From the results, during the early stage of the co-culture of B. lentimorbus WJ5 and C. gloeosporioides, nutrient competition seemed to occur; therefore, the genes from central carbon metabolisms could be up-regulated, while those from minor catabolism could be down-regulated.

  • In order to evaluate the biological activities of some fractions from the fungal(Pleurotus sajorcaju) fermentation products of sugar cane bagasses, the antimutagenicity, the glyceollin elicitor activity on soybean and the effect on the stem elongation in pea were observed. The alcohol extract fraction and DMSO soluble fraction had excellent antimutagenicity even though it is weaker than that of the extracts from the fruiting bodies. All of the extracts had the ability to elicit glyceollins in soybean cotyledons and these extracts could be helpful for plants to protect themselves from pathogenic contaminations. IAA and the extracts had shown synergistic effects on pea stem elongation in all experimental groups positively determined.

  • Objectives : The aim of this study is to review allergic rhinitis and to help make full use of oriental medical therapy in this disease. Methods : We investigated the published papers with the key word 'allergic rhinitis' and refered to several important old records Results : 1. Allegic rhinitis in Oriental Medicine is known as bigu, bunche and the main symptoms is rhinorrhea, sneezing, nasal obstruction. 2. The cause of allergic rhinitis in Oriental Medicine is dysfunctions of the spleen, lung and kidney systems bring weakness to the vital energy(正氣) making wind and cold(風寒邪氣) energy easy to intrude. 3. Determination of the patient's constitution and understanding of the weakness of spleen, lung and kidney systems helps improve the treatment rate, it is used to treat allergic rhinits with herbal medicines, acupunctures and so on. 4. In acupuncture therapy, the mainly used acupuncture points are 迎香(younghyang), 印堂(yindang), 鼻通(bitong), 上星(sangsung), 合谷(habgouk). Conclusions : The Oriental Medicine treatment principal of rhinitis is strengthening the body resistance and eliminating the pathogenic factors according to the individual characters of each patients and the symptoms.

  • In the literatual studies on the causes and treatments of the melasma, the results were as follows. 1. Melasma is the blackish patch on the face, is belong to the cartegory of the 'Myunjin(面塵)', 'Myunganzung(면간증)', 'Jakban(雀斑)' etc. in Oriental medicine 2. Melasma is deeply associated with Yangming channel in Meridian, with the spleen, stomach, heart and kidney in Viscera. 3. The pathogenic factors of Melasma is divided five parts. One is insufficient of Yangming's energy in Neijing(內經). Two is incoordination between vital energy and blood caused by wind-evil and phlegm-retention syndrome. Three is anxiety impairing the spleen. Four is kidney-asthenia and fire-hyperactivity. Five is heat-evil. 4. In the treatments of Melasma, Sthenia-syndrome was used cooling blood and activating blood circulation, or dispelling wind-evil and promoting meridian, or expelling fire-evil and removing toxic material etc. Asthenia-syndrome was used invigorating the liver and kidney, or nourishing yin and keeping fire downwards etc. 5. Melasma is concerned with sun-light, is mostly seen in female. 6. In the prescription of Melasma, it was used Jujesamultang(酒製四物湯加減), Okyong -san(玉容散), Chunghwasungitang(沖和順氣湯), Okyongseosisan(玉容西施散), Yukmiji-hwanghwan(六味地黃丸) etc.
