  • Probiotics are very closely related to gut microbiome and recognized as beneficial microorganisms for our health. They have various biological effects such as inhibition of pathogenic bacteria, activation of beneficial bacteria, prevention of diarrhea and constipation, enhanced immune activity etc. Prebiotics, non-digestible carbohydrates such as galactooligosaccharide and fructooligosaccharide, are utilized by beneficial gut bacteria such as bifidobacteria and lactobacilli, resulting in production of short chain fatty acids which inhibit pathogenic bacteria in the gut and function for human health. Synbiotics are introduced for synergistic effects when probiotics are combined with prebiotics and now commercially available. At the moment many functional ingredients are developed and commercialized. Probiotics, prebiotics, and synbiotics might be hot items in the functional food market and the values will increase according to the results of human gut microbiome researches. To meet the situation, systematic and scientific studies as well as marketing effects should be accompanied.

  • This study was attempted to see more clear relationships among the enterobacteria, especially between the intestinal normal flora and pathogenic bacteria. It has been known that some intestinal normal flora produce the bactrial metabolites that are harmful to other enteric bacteria. One of the metabolites is known as colicin, the protein fraction, which possesses certain degree of inhibitory effect against other bacterial growth fraction, whih possesses certain degree of inhibitory effect against other bacterial growth. As a preliminary study for a colicin purification, the antagonistic effect of E, coli to groups of Salmonella and Shigella has been studied by means of in vitro quantitative culture method. 1. E.coli showed definite inhibitory effects aganist both Salmonella and Shigella groups in the mixture of two organisms. 2. The inhibitory effects of E.coli in the E.coli-Salmonella and the E.coli-Shigella mixture occurred from 4 hours incubation following the inoculation. 3. Even the complete inhibition of pathogenic enteric bacterial growth was noticed in the E.coli-Salmonella mixture at overnight incubation. 4. Among the diluted mixtures, 1:100, 1:1,000, and 1:10,000, survival rate of pathogenic enteric bacteria in the mixtures with E.coli showed least affected at the 1:1,000 dilution. 5. It was found that the antagonistic effect aganist groups of Salmonella-shigella was depending upon the groups of the genera.

Wnt5a attenuates the pathogenic effects of the Wnt/β-catenin pathway in human retinal pigment epithelial cells via down-regulating β-catenin and Snail

  • Activation of the Wnt/β-catenin pathway plays a pathogenic role in age-related macular degeneration (AMD) and is thus a potential target for the development of therapeutics for this disease. Here, we demonstrated that Wnt5a antagonized β-catenin response transcription (CRT) induced with Wnt3a by promoting β-catenin phosphorylation at Ser33/Ser37/Thr41 and its subsequent degradation in human retinal pigment epithelial (RPE) cells. Wnt5a decreased the levels of vascular endothelial growth factor (VEGF), tumor necrosis factor-α(TNF-α), and nuclear factor-κB (NF-κB), which was up-regulated by Wnt3a. Furthermore, Wnt5a increased E-cadherin expression and decreased cell migration by down-regulating Snail expression, thereby abrogating the Wnt3a-induced epithelial-mesenchymal transition (EMT) in human RPE cells. Our findings suggest that Wnt5a suppresses the pathogenic effects of canonical Wnt signaling in human RPE cells by promoting β-catenin phosphorylation and degradation. Protective Effects of a Novel Probiotic Strain of Lactobacillus plantarum JSA22 from Traditional Fermented Soybean Food Against Infection by Salmonella enterica Serovar Typhimurium

  • Lactobacillus species have been shown to enhance intestinal epithelial barrier function, modulate host immune responses, and suppress the growth of pathogenic bacteria, yeasts, molds, and viruses. Thus, lactobacilli have been used as probiotics for treating various diseases, including intestinal disorders, and as biological preservatives in the food and agricultural industries. However, the molecular mechanisms used by lactobacilli to suppress pathogenic bacterial infections have been poorly characterized. We previously isolated Lactobacillus plantarum JSA22 from buckwheat sokseongjang, a traditional Korean fermented soybean food, which possessed high enzymatic, fibrinolytic, and broad-spectrum antimicrobial activity against foodborne pathogens. In this study, we investigated the effects of L. plantarum JSA22 on the growth of S. Typhimurium and S. Typhimurium-induced cytotoxicity by stimulating the host immune response in intestinal epithelial cells. The results showed that coincubation of S. Typhimurium and L. plantarum JSA22 with intestinal epithelial cells suppressed S. Typhimurium infection, S. Typhimurium-induced NF-κB activation, and IL-8 production, and lowered the phosphorylation of both Akt and p38. These data indicated that L. plantarum JSA22 has probiotic properties, and can inhibit S. Typhimurium infection of intestinal epithelial cells. Our findings can be used to develop therapeutic and prophylactic agents against pathogenic bacteria.

인천 연안지역의 해양환경요인과 병원성 비브리오균의 분포와의 관련성 (Relationships of Pathogenic Vibrios and Environmental Factors Affecting their Occurrence in the Seawater of Incheon Coastal Area)

  • The aim of this study was to investigate the effects of environmental factors such as temperature, salinity, turbidity and pH on the growth of pathogenic Vibrios. In this study, we was obtained the samples from 2 different sites of the Incheon coastal area between January 2012 and December 2012. The water temperature in August and September was the high. the Incheon port changes the width of a small, wherease in the case of Hanjin harbor of changes of larger width. Salinity and turbidity showed significant differences, whereas temperature and hydrogen ion concentration was not significant. Pathogenic vibrios was determined using the real-time PCR method. Pathogenic vibrios in the Incheon port and Hanjin harbor were detected in 11 samples (91.67%) and 9 samples (75.0%) of Vibrio cholerae, 7 samples (58.3%) and 6 samples (50.0%) of V. vulnificus, 10 samples (83.3%) and 12 samples (100.0%) of V. parahaemolyticus, respectively. Pathogenic Vibrio bacteria were the highest at $26.8^{\circ}C$ of seawater in August. Quantitative results were the following: 102 $cell/m{\ell}$ in Vibrio cholerae, 7.876 $cell/m{\ell}$ in V. vulnificus, and 503.4 $cell/m{\ell}$ in V. parahaemolyticus, respectively. The enumeration of pathogenic vibrios showed a positive correlation with temperature and pH, but was negatively correlated with salinity and turbidity.

Antifungal Effects of Silver Nanoparticles (AgNPs) against Various Plant Pathogenic Fungi

  • This research is concerned with the fungicidal properties of nano-size silver colloidal solution used as an agent for antifungal treatment of various plant pathogens. We used WA-CV-WA13B, WA-AT-WB13R, and WA-PR-WB13R silver nanoparticles (AgNPs) at concentrations of 10, 25, 50, and 100 ppm. Eighteen different plant pathogenic fungi were treated with these AgNPs on potato dextrose agar (PDA), malt extract agar, and corn meal agar plates. We calculated fungal inhibition in order to evaluate the antifungal efficacy of silver nanoparticles against pathogens. The results indicated that AgNPs possess antifungal properties against these plant pathogens at various levels. Treatment with WA-CV-WB13R AgNPs resulted in maximum inhibition of most fungi. Results also showed that the most significant inhibition of plant pathogenic fungi was observed on PDA and 100 ppm of AgNPs.

병원성사상균이 Alfalfa 유식물의 생육에 미치는 영향 (Effect of Pathogenic Fungi on Growth of Alfalfa Seeding)

  • This study was conducted to investigate the effects of pathogenic hngi(Fusarium culmorurn, Fusarium oxyspomm fsp. lycopersici, Fusaiurn solani, Rhizoctonia solani), pregermination time(0, 6, 9, 12, 18, 24, 48 hrs.) and temperature($12^{\circ}C, 18^C{\circ}, $28^C{\circ}$) on growth of alfalfa seedling. The survival rate of alfalfa was decreased by pathogenic hngi and treatment of pregermination and survival rate of alfalfa resulted in higher than that of nontreatment of pregermination. Pathogenicity of fungi was increased as increased the temperature(P<0.05). The emergence rate of alfalfa was decreased when pathogenic fungi were inoculated. However, the emergence rate was increased when seeds were pregerminated(P<0.05)

Effects of Ribosomal Protein L39-L on the Drug Resistance Mechanisms of Lung Cancer A549 Cells

  • Background: Cancer is a major threat to the public health whether in developed or in developing countries. As the most common primary malignant tumor, the morbidity and mortality rate of lung cancer continues to rise in recent ten years worldwide. Chemotherapy is one of the main methods in the treatment of lung cancer, but this is hampered by chemotherapy drug resistance, especially MDR. As a component of the 60S large ribosomal subunit, ribosomal protein L39-L gene was reported to be expressed specifically in the human testis and human cancer samples of various tissue origins. Materials and Methods: Total RNA of cultured drug-resistant and susceptible A549 cells was isolated, and real time quantitative RT-PCR were used to indicate the transcribe difference between amycin resistant and susceptible strain of A549 cells. Viability assay were used to show the amycin resistance difference in RPL39-L transfected A549 cell line than control vector and null-transfected A549 cell line. Results: The ribosomal protein L39-L transcription level was 8.2 times higher in drug-resistant human lung cancer A549 cell line than in susceptible A549 cell line by quantitative RT-PCR analysis. The ribosomal protein L39-L transfected cells showed enhanced drug resistance compared to plasmid vector-transfected or null-transfected cells as determined by methyl tritiated thymidine (3H-TdR) incorporation. Conclusions and Implications for Practice: The ribosomal protein L39-L gene may have effects on the drug resistance mechanism of lung cancer A549 cells.

Effects of Compost Tea Making from Differently Treated Compost on Plant Disease Control

  • Antifungal activity of compost teas was evaluated in vitro and in vivo experiments. In vitro test, fourteen compost teas were produced using five different composts and through six different methods for compost tea production. Eleven pathogenic fungi were used as indicators of antifungal activity of compost teas. In vivo test, one of the compost teas used in vitro test was sprayed to cucumber leaves which were infected with powdery mildew in order to evaluate antifungal activity of compost tea. From the results of the tests, it was found that all compost teas used this study showed antifungal activity and the proper application of compost teas against pathogenic fungi for obtaining optimal effects was important to know.

  • Purpose: The purpose of this study was to develop standard syllabuses of 4 subjects (Structure and Function of Human Body, Pathogenic Microbiology, Pathophysiology, Mechanisms and Effects of Drug) of bionursing. Methods: We developed standard syllabuses of 4 subjects through 3 step discussions and analysis. At first each committee for 4 subjects made the syllabus, after then all researchers discussed together. Finally each committee concluded final one. Committee members consisted of professors who teach that subject at nursing college. Results: Standard syllabus for Biological Nursing I-1 or Structure & Function of Human Body I consisted of 3 parts and 9 themes and for Biological Nursing I-2 or Structure & Function of Human Body II was 3 parts and 9 themes. Standard syllabus for Biological Nursing II or Pathogenic Microbiology consisted of 8 themes and Biological Nursing III or Pathophysiology consisted of 2 parts and 19 themes. Standard syllabus for Biological Nursing IV or Mechanisms & Effects of Drug consisted of 2 parts and 13 themes. Conclusion: The standard syllabuses and titles of 4 subjects of bionursing need to be disseminated to all the nursing college by Academy of Korean Biological Nursing Science. Further researches to investigate curriculum for bionursing and to identify the effects of 4 standard syllabuses were recommended.
