Changes in Physicochemical Properties of Baik-kimchi during Fermentation (백김치 숙성중 물리화학적 특성변화)

  • To elicit the effect of fermentation on food quality of the watery Chinese cabbage pickles without fish sauce and red pepper paste(Baik-kimchi), changes in physicochemical properties and microstructure of fiber components were studied. Better water holding capacity(WHC) was showed in Baik-kimchi fermented at $25^{\circ}C$ than that of Baik-kimchi fermented at 5$^{\circ}C$. WHC measured at pH 2 and 6 were ranged from 10.18 to 16.79g/g dried sample for Baik-kimchi fermented at $25^{\circ}C$ and 6.51~14.58g/g dried sample for sample for samples at 5$^{\circ}C$, respectively. The higher WHC was resulted in pH controlled freeze-dried sample to pH 6 than that measured in pH 2 sample. The settling volume(SV) and oil adsorption capacity(OAC) increased with fermentation period and kept the same value for a little while, but slightly decreased in the over ripening period. Baik-kimchi fermented at $25^{\circ}C$ exhibited more shrunk microstructure of parenchyma cell and xylem than those of Baik-kimchi fermented at 5$^{\circ}C$. The appearance of SDF of the both Baik-kimchi ripened at 5$^{\circ}C$ and $25^{\circ}C$ could give granular shape, whereas the overripened Baik-kimchi had smooth surface of SDF. On the other hand, the IDF retained the original shape during fermentation.

Anatomical Structures and Fiber Quality of Four Lesser-Used Wood Species Grown in Indonesia

  • This study aimed to investigate the anatomical structure and fiber quality of four lesser-used wood species namely Benuang (O. sumatrana), Duabanga (D. moluccana), Pisang Merah (H. hellwigii), and Terap (A. odoratissimus). This study evaluated its suitability for raw material in pulp and paper manufacturing. The anatomical structure was observed macro- and microscopically. Macroscopic structures were observed directly to the wood samples, while microscopic characteristics were observed through microtome specimens. Fiber dimension was measured through macerated specimens and fiber quality was analyzed following the Rachman and Siagian's method. Results showed that these four timber species have similarity in the indistinct growth ring, diffuse porous in a radial pattern, rounded solitary vessel outline, 1 to 3 cells of ray width, deposits within the rays, fiber length, and cell wall thickness. Differences were found on vessel diameter, vessel grouping, vessel frequency, tyloses existence, type of axial parenchyma, and ray height. Based on fiber length and its derived values, the wood fibers of all species studied are suitable for pulp and paper manufacturing. They belong to the II quality class. The produced pulp and paper would have good quality, especially in tensile, folding, and tear strength. To promote their utilization, silviculture aspect of these four species has to be well understood.

Effect of Clonorchis sinensis infection on hepatic oval cell proliferation in rats (랫트에서 Clonorchis sinensis 감염이 간 oval cell 의 증식에 미치는 영향)

  • The prevalence of primary liver carcinoma (PLC) is relatively high in Clonorchis sinensis (CS) endemic areas in Korea. PLC is a malignant tumor which can be subclassified into hepatocellular carcinoma and cholangiocarcinoma(CC). CC has been associated with clonorchiasis, but it is unclear whether clonorchiasis is associated with hepatocarcinogenesis. This experiment was designed to investigate relationships between clonorchiasis and early changes of hepatocarcinogenesis. Sixteen Sprague-Dawley rats weighing 150g were divided into two groups of 8 rats in each. All rats were fed choline-devoid(CD) diet for 4 weeks. Group 1 was given 0.015-0.020% diethylnitrosamine(DEN) as drinking water for 1 week. After one week, the rats were treated orally with 1% N-acetylaminofluorene(AAF) (5 times per week for 2 weeks). Group 2 was treated equally to group 1 except for CS infection during AAF treatment. Two rats in each group were sacrificed at 4th, 5th, 6th and 7th week of the experiment. Livers were stained with OV -6, proliferating cell nuclear antigen(PCNA) and GST-p. Results were as follows: Group 2 livers showed more oval cell proliferation in parenchyma and portal areas at the 4th, 5th, 6th and 7th weeks than did livers of group 1 (p<0.01). PCNA was mostly localized in oval cell populations, rather than hepatocytes and biliary cells. The ratio of oval cells to hepatocytes was much higher in group 2 than in group l(p<0.01 The ratio of hepatocytes to biliary cells is higher in group 2 than in group 1 (p<0.05), More group 2 acidophilic foci reacted to GST-p monoclonal antibody than in the noninfected group. It appeared that CS infection promoted potentially precancerous acidophilic foci and oval cell proliferation.

LJ-2698, an Adenosine A3 Receptor Antagonist, Alleviates Elastase-Induced Pulmonary Emphysema in Mice

  • Emphysema, a major component of chronic obstructive pulmonary disease (COPD), is a leading cause of human death worldwide. The progressive deterioration of lung function that occurs in the disease is caused by chronic inflammation of the airway and destruction of the lung parenchyma. Despite the main impact of inflammation on the pathogenesis of emphysema, current therapeutic regimens mainly offer symptomatic relief and preservation of lung function with little therapeutic impact. In the present study, we aimed to discover novel therapeutics that suppress the pathogenesis of emphysema. Here, we show that LJ-2698, a novel and highly selective antagonist of the adenosine A3 receptor, a G protein-coupled receptor involved in various inflammatory diseases, significantly reversed the elastase-induced destructive changes in murine lungs. We found that LJ-2698 significantly prevented elastase-induced airspace enlargement, resulting in restoration of pulmonary function without causing any obvious changes in body weight in mice. LJ-2698 was found to inhibit matrix metalloproteinase activity and pulmonary cell apoptosis in the murine lung. LJ-2698 treatment induced increases in anti-inflammatory cytokines in macrophages at doses that displayed no significant cytotoxicity in normal cell lines derived from various organs. Treatment with LJ-2698 significantly increased the number of anti-inflammatory M2 macrophages in the lungs. These results implicate the adenosine A3 receptor in the pathogenesis of emphysema. Our findings also demonstrate the potential of LJ-2698 as a novel therapeutic/preventive agent in suppressing disease development with limited toxicity.

Clinicopathological Significance of Osteopontin in Cholangiocarcinoma Cases

  • Cholangiocarcinoma (CCA) is generally a rare primary liver tumor of the bile duct with extremely poor clinical outcomes due to late diagnosis. Osteopontin (OPN) is the most abundant expressed gene in intrahepatic CCA and its involvement in tumor aggressiveness suggests it could be a useful prognostic biomarker. However, the prognostic significance of OPN expression in CCA is still controversial. We therefore immunohistochemically studied OPN expression in 354 resected CCAs and correlated the results with patient clinicopathological parameters. OPN expression was separately scored according to the percentage of cancer cells or degree of stromal tissue staining and classified as low (score 0-1) and high (score 2-3). OPN expression in CCA cells was found in 177 out of 354 patients (56.5%), whereas stroma was positive in 185 out of 354 patients (52.3%). Univariate analysis with several of the aforementioned parameters revealed that stromal but not cancer cell OPN expression was significantly associated with tumor size, tumor direct invasion into normal liver parenchyma, regional lymph node metastasis and higher staging. The combination of cancer cell and stromal OPN expression demonstrated a positive trend for linkage with lymph node metastasis. Multivariate analysis identified gender, the presence of lymphatic permeation and lymph node metastasis, but not OPN expression, as independent prognostic factors. This study confirms the presence of stromal OPN expression in tumor aggressiveness but not survival in CCA patients.

Morphological studies on the major salivary gland of the Korean native goat (한국재래산양의 큰침샘에 대한 형태학적 연구)

  • The morphological findings of the parotid, mandibualr and sublingual salivary glands of the Korean native goat have been investigated by the histological and histochemical observation using the light microscope. Tissues were fixed with 10% neutral buffered formalin and Bouin's solution, and embedded in paraffin. The tissue sections were stained with hematoxylin and eosin, Heidenhain's azocarmine-aniline blue, alcian blue, toluidine blue, periodic acid Schiff, aldehyde fuchsin, alcian blue-periodic acid Schiff and aldehyde fuchsin-alcian blue. Some sections were stained with the alcian blue after each teatment of diastase digestion, methylation, methylation-saponification, and neuraminidase digestion. The results were as follows ; 1. The major salivary glands were compound. tubuloacinar glands, and the parenchyma was composed of acini, intercalated ducts, striated ducts and excretory ducts. 2. The acini were composed of serous cells in the parotid gland, and mucous cells, serous cells and seous demilunes in the mandibular gland. The acini of the sublingual glands were composed of mucous cells and serous demilunes. 3. In histochemistry, the serous cells of the parotid gland contained neutral mucin and enzyme-liable silaic acid. 4. The serous cells and demilunes of the mandibular gland contained neutral mucin and enzyme-liable sialic acid, and the mueous cells contained sulfated mucin, enzyme liable sialic acid and neutral mucin 5. In the sublingual gland, the mucous cells contained sulfated mucin, enzyme-resistant sialic acid and neutral mucin, and the serous demilunes contained neutral mucin and enzyme-resistant sialic acid.

Anatomical Characteristics of Major Korean Ash Species (한국산 물푸레나무속(屬) 주요 수종의 해부학적 특성)

  • Anatomical characteristics of Mulpurenamu (Korean ash, Praxmus rhynchopbylla), Deulmaenamu (Manshurican ash, Fraxinus mandsburica) and Sheamulpure (Sieboldiana ash, Fraxinus sieboldiana) grown in Korea were examined by an optical microscopy and an image analysis. Some characteristics such as boundary of annual rings, shape and size of vessel elements, arrangement of axial parenchyma cells in cross section, and cell volumetric composition showed significant differences between the sample species. In radial variation of elements, fiber length and vessel size increased from the pith for about 10 to 15 years and then reached a more or less constant. The results of this study can be used for identification of wood and indices of wood quality in Fraxinus spp.

Poor Prognosis of Grade 2 Spread Through Air Spaces in Neuroendocrine Tumors

  • Background: Spread through air spaces (STAS) has recently emerged as a prognostic factor in lung adenocarcinoma, but little is known about the association of STAS and its grade with recurrence in neuroendocrine tumors (NETs). This study investigated the prognostic effect of STAS grade in NETs after curative resection. Methods: Seventy-seven patients were retrospectively reviewed, including 9 with typical carcinoid (TC), 6 with atypical carcinoid (AC), 26 with large cell neuroendocrine carcinoma (LCNEC), and 36 with small cell carcinoma (SCC). STAS was defined as the presence of floating tumor cells within air spaces in the lung parenchyma beyond the edge of the main tumor. STAS was classified as grade 1 or 2 depending on whether it was found within or beyond one ×10 objective lens field away from the main tumor margin, respectively. Results: Fifty-four patients (70%) had STAS, including 22% with TC, 50% with AC, 69% with LCNEC, and 86% with SCC. Patients with STAS had more nodal metastasis, lymphatic and vascular invasion, tumor necrosis, and tumor subtypes other than TC. Among STAS cases, grade 2 STAS was present in 33% of AC, 78% of LCNEC, and 87% of SCC. The 5-year recurrence-free survival (RFS) rate was 81%, 63%, and 35% in patients with no STAS, grade 1, and grade 2 STAS, respectively. Multivariate analysis found that grade 2 STAS was an independent negative prognostic factor for RFS. Conclusion: Although STAS itself was not associated with a poor prognosis, grade 2 STAS was an independent negative prognostic factor for RFS.

Ginsenoside Rg3-enriched Korean Red Ginseng extract attenuates Non-Alcoholic Fatty Liver Disease by way of suppressed VCAM-1 expression in liver sinusoidal endothelium

  • Background: The incidence and clinical importance of nonalcoholic fatty liver disease (NAFLD) has emerged. However, effective therapeutic strategies for NAFLD have yet to be found. Panax ginseng (P. ginseng) is a traditional herb in Eastern Asia with therapeutic effects in many chronic disorders. However, the precise effects of ginseng extract on NAFLD are currently unknown. In present study, the therapeutic effects of Rg3-enriched red ginseng extract (Rg3-RGE) on the progression of NAFLD were explored. Methods: Twelve-week-old C57BL/6 male mice were fed a chow or western diet supplemented with high sugar water solution with or without Rg3-RGE. Histopathology, immunohistochemistry, immunofluorescence, serum biochemistry, western blot analysis, and quantitative RT-PCR were used for in vivo experiment. Conditionally immortalized human glomerular endothelial cell (CiGEnC) and primary liver sinusoidal endothelial cells (LSECs) were used for in vitro experiments. Results: Eight weeks of Rg3-RGE treatment significantly attenuated the inflammatory lesions of NAFLD. Furthermore, Rg3-RGE inhibited the inflammatory infiltrate in liver parenchyma and the expression of adhesive molecules to LSECs. Moreover, the Rg3-RGE exhibited similar patterns on the in vitro assays. Conclusion: The results demonstrate that Rg3-RGE treatment ameliorates NAFLD progression by inhibiting chemotaxis activities in LSECs.

Ultrasonographic and Computed Tomographic Features of Dermoid Cyst of the Neck in a Dog: A Case Report

  • A 13-year-old castrated male mixed-breed dog presented for a health screening with a small nodule on the left hindlimb, which was revealed to be a mast cell tumor. A CT scan was performed to evaluate the mast cell tumor, and it incidentally revealed a well-marginated, fluid-attenuating, non-contrast enhanced mass in the deep ventral region of the left lower neck. Ultrasonography confirmed a well-capsulated mass with a cranio-ventrally protruding lesion extending into sternohyoid muscles. The outer layer of the mass had a hypoechoic thick wall and the internal parenchyma contained hyperechoic foci and multiple hyperechoic parallel lines. The mass was surgically resected. Histopathological examination confirmed a dermoid cyst characterized by cornifying squamous epithelial cells, keratinaceous debris, and hair shaft fragments. The dog showed no signs of recurrence or additional abnormalities three months post-surgery. This report highlights the importance of considering dermoid cysts in the differential diagnosis of neck masses on imaging examinations using CT or ultrasonography.
