  • It has recently been suggested that the release of "endocrine disrupters (EDs)" into the environment has resulted in adverse health effects on wild life populations and humans (Golden et al., 1998; Tyler et al., 1998; Kang et al., 2000). Human sperm counts have declined significantly throughout the world during the past fifty years, and which is a significant public health concern (Carlsen et al., 1992; Carlsen et al.. 1995). In addition, the EDs persisting in the environment are known to disrupt the normal endocrine systems of wildlife (Colborn, 1995; Crewet al., 1995; Folmer et at, 1996; Sumpter, 1995; Tyler, 1998). Some estrogenic chemicals bind to estrogen receptors (Bolger et al.. 1998), interfere with the binding of physiological ligands to steroid hormone-binding proteins (Danjo, 1997; Milligan et al., 1998). and show immunotoxicity (Sakae et al., 1998). 식물유래 천연항균제를 이용한 화장품에서의 방부시스템에 관한 연구 (The Study of Preservative System in Cosmetics using Botanical Antimicrobial)

  • 본 연구는 고삼추출물과 황금추출물의 항균활성을 스킨케어화장품의 기본제형인 화장수, 로션에 적용, 관찰하여 식물유래의 천연항균제로써의 화장품에서의 방부시스템을 연구하였다. Test group에 고삼추출물 1%와 황금 추출물 1%, 0.5%씩의 고삼추출물과 황금추출물을 함유시켜 parabens와 imidazolidinyl urea가 함유된 positive control을 Escherichia coil, Pseudomonas aeruginosa, Staphylococcus aures, Candida albicans의 4가지 균에 대하여 실험하였다. 그 결과 고삼추출물과 황금추출물이 함유된 group은 control과 비교하여 4가지 균에 대하여 우수한 항균활성을 보였다. 고삼추출물과 황금추출물을 함께 사용하였을 때의 항균활성은 positive control과 비슷하게 나타나므로써 탁월한 항균활성을 보였다.

MICROBIOLOGICAL QUALITY OF BRAZILIAN LIPSTICKS AFTER NORMAL USE BY CONSUMERS

  • Lipsticks are cosmetics which do not contain water and are usually preserved with parabens. When submitted to the Challenge Test, theses products did not reach the CTFA criteria, which means that microbiological contamination could occur before the end of its shelf life. The aim of this study was to evaluate the contamination level of 130 lipsticks after its use. Microorganisms were isolated from 14,6 % of the samples. However, only in two samples (1,5%) the contamination level exceeded the 100 CFU/g level, which means that, although the preservative system was not efficient to eliminate bacteria, the lack of free water was enough to prevent the microbial development. Total bacteria and fungi were determined by conventional methodology, according to CTFA Microbiological Guidelines. The microbes were then isolated and characterized as normal skin flora microorganisms. This suggest that products were contaminated by the constant application of lipsticks by consumers. This could lead to cross contamination when the same product is shared by several people. Extra care should be taken into account when this type of products are available to be tested by several consumers in demonstration displays.

Simultaneous HPLC determination of multiple compounds in a cosmetic lotion

  • 고속액체크로마토그래피 방법을 사용하여 동시에 arbutin, 메탄올에 녹인 methyl, ethyl, prpyl, butyl parben과 glablidien(유용성감초추출물)을 파장 254와 276 nm에서 Gradient methanol로 octdecyl culumn을 사용하여측정하였다. arbutin와 glablidien 농도 0.5-1.0 ug/ml 파라벤류는 0.1-2.0 ug/ml에서 검량선이 직선으로 작성되었다. 검량이 직선으로 나타나 정량분석을 할 수 있다는 것을 알 수 있었다. 샘플 전처리가 크로마토그래피로 측정하기에 좋은 방법이란 것을 판정하기 위하여 일반로션을 사용하여 판정하였다. 이 방법의 정확도는 모든 측정물질(arbutin, methylparaben, ethylparaben, propylparaben, butylparaben, glablidine)의 회수율 상대표준편차(RSD)가 (0.28-2.55%) 나타나 신뢰성 있는 결과를 보였다.A high-performance liquid chromatographic method for the simultaneous determination of arbutine, a mixture of methyl, ethyl, propyl, butyl parabens dissolved in methoanol and Glrablidine(Oil Soluble Licorice), was studied by using a ODS C18 column and a methanol gradient at 254 and 276 nm. Calibration curves were found to be linear in the 0.5-1.0 ug/ml range(compounds arbutine, glrablidin) and 0.1-20 ug/ml (compounds methylpaaben, ethylparaben, propylparaben, butylparaben). Linear regression analysis of the data demonstrates the efficacy of the method in terms of precision and accuracy. An extraction method is developed and validated in order to apply this chromatographic method to a cosmetic lotion. The presision of this method, calculated as the relativ standard deviation(RSD) of the recoveries(0.28-2.55%) was excellent for all compounds.

환경성 내분비계장애물질(EDCs)의 우선관리순위 결정법 개발 및 적용 (Development and Application of a Chemical Ranking and Scoring System for the Management of Endocrine Disrupting Chemicals)

  • Background: Exposure to endocrine disrupting chemicals (EDCs) has been considered one of the main causes of a range of endocrine diseases in modern society. An EDC priority list considering exposure, toxicity, and societal concern should be established for EDC management. Methods: The chemical ranking and scoring (CRS) system for EDCs was based on exposure, toxicity,and societal concern. The exposure score system was based on usage, circulation volume, bioaccumulation, and detection in consumer products. The toxicity score system was based on carcinogenicity and reproductive and developmental toxicity. The societal concern score system was based on domestic or international regulations and mass media reports. Results: A total of 165 EDCs were considered in the CRS system. The top-five priority EDCs were Bis(2-ethylhexyl) phthalate (DEHP), Benzene, Bisphenol A, Dibutyl phthalate (DBP) and Trichloroethylene. Phthalates, bisphenols and parabens were identified as priority chemical groups. Conclusion: We developed a CRS system for EDCs to identify priority EDCs for management. This will be a foundation to provide an EDC management plan based on scientific decision-making.

국내 화장품 원료성분에 대한 유해성 조사 (Hazard Investigation of Cosmetic Ingredients in Korea)

  • Objective: The purpose of this study was to investigate the hazards of cosmetic ingredients in Korea. Methods: An Excel database of cosmetic ingredients was developed on a website(Korea Cosmetic Ingredient Dictionary) and used for a hazard assessment based on Tox-free, a database containing toxic information such as on carcinogens, mutagens or reproductive toxicants(CMRs) and endocrine disrupting chemicals(EDCs), as well as other regulated chemicals in Korea. Results: A total of 16,605 chemicals were registered with the cosmetic ingredient dictionary and 308 of them were identified as either CMRs or EDCs. CMRs included formaldehyde and nickel gluconate, and EDCs included parabens, benzophenon, styrene, and toluene. Reproductive toxicants such as xylene, zinc chloride, toluene, and formaldehyde were regulated by the Occupational Safety and Health Act and the Act on the Registration and Evaluation, etc. of Chemical Substances(the Chemical Control Act). Conclusions: Regulations on cosmetics components should be strengthened, and the right to know about cosmetics containing hazardous chemicals should be guaranteed.

Synergistic Antimicrobial Effect of Lonicera japonica and Magnolia obovata Extracts and Potential as a Plant-Derived Natural Preservative

  • Most people use cosmetics to protect their skin. Preservatives are often used to prevent their contamination upon use. There has been a great demand for natural preservatives due to recent reports on the side effects of parabens. Therefore, we evaluated the antimicrobial activities of Lonicera japonica and Magnolia obovata extracts and determined their potential as natural preservatives. We found that the 50% ethanol extract from L. japonica had antibacterial activity only against S. aureus and P. aeruginosa, while the ethyl acetate fraction showed antimicrobial activity against all six microbial strains tested. On the other hand, the 70% ethanol extract and the ethyl acetate fraction from M. obovata showed antimicrobial activity against all six strains. A synergistic effect against S. aureus, B. subtilis, and C. albicans was confirmed when two ethyl acetate fractions having antimicrobial activity against all six strains were used in combination. Synergistic activity against B. subtilis was also confirmed through kill-time analysis. High-performance liquid chromatography was performed to identify the components of each extract. Based on the minimum inhibitory concentration and the results of a disc diffusion assay, we confirmed that caffeic acid and luteolin influenced the antimicrobial activity of L. japonica and that the antimicrobial activity of M. obovata was influenced by the interaction of magnolol and honokiol with other components. Therefore, this study suggests that the combination of L. japonica and M. obovata extracts may be used as a plant-derived natural preservative.

국민환경보건기초조사 1~3기의 연구성과 검토 (A Review of the Literature Using the Korean National Environmental Health Survey (cycle 1-3))

  • Objectives: The Korean National Environmental Health Survey provides representative biomonitoring data for environmental pollutants in South Korea. Over the last decade, there have been various studies published using this data. In this study, we aimed to provide information and implications by reviewing each study. Methods: We searched comprehensive electronic databases from PubMed, Google Scholar, and Naver Academic database using the key words 'Korean National Environmental Health Survey' and 'KoNEHS' through March 2021. A total of 57 studies were selected after reviewing the relevance of the data. Results: The most frequently studied pollutants were heavy metals (10), Cotinine (8), Bisphenol A (7), and Phthalates (6), in that order. In particular, Phthalates, Bisphenol A, and Parabens were often studied together (6). A decline in urinary cotinine and heavy metals in the body was shown over time among studies on exposure association. It was demonstrated that Phthalates and Bisphenol A were significantly related to obesity and diabetes from the studies of health impacts. Cross-section study design, spot urine, and insufficient health status information were mostly reported as limitations of the data. Conclusion: Since research has been focused on adults, further investigations of children and adolescents are required. In this regard, it is necessary to maintain the consistency of the data structure and provide integrated weights for all ages. In addition, it would allow the measurement of several environmental pollutants by considering subsample design. Lastly, integrated studies with multi-cycles and the health effects from co-exposure to multiple chemicals would be expected to provide important knowledge.

UPLC-MS/MS를 이용한 소변 시료 중 내분비계 교란물질 27종 동시분석법 확립 (Simultaneous Analysis Method for 27 Endocrine Disrupting Chemicals in Human Urine using UPLC-MS/MS)

  • 내분비 교란 화학물질(EDCs)은 생체 외부로부터 유입되어 인체의 내분비 기관 내에서 호르몬 작용을 교란시키는 화합물이다. 파라벤, 벤조페논, 비스페놀, 프탈레이트 등이 대표적이며, 현재 광범위한 분야에서 사용되고 있다. 하지만 이들에게 지속적으로 노출되면 혈당조절, 생식, 대사, 신경계 발달, 임신, 출산, 성장 등에 부정적인 영향을 끼칠 수 있다. 본 연구에서는 EDCs의 노출정도를 파악하기 위하여 인체시료(소변)를 liquid-liquid-extraction을 사용하여 전처리한 뒤 UPLC-MS/MS로 효과적이고 빠르게 분석하였다. 이와 같이 분석조건을 확립하고, 분석법의 유효성 검증을 통해 동시분석법의 신뢰도를 평가하였다. 결과는 정확도가 75.28~122.36%, 정밀도가 2.16~22.74%의 수치를 보였다. 본 연구에서 확립된 분석법은 추후 인체시료 중 EDCs의 노출을 평가하고 모니터링할 수 있는 연구의 방법론으로 이용될 수 있을 것이다.
