Distribution of Antimicrobial Resistant Genes in Acinetobacter calcoaceticus-baumannii Complex Isolated from Clinical Specimens in Chungcheong, Korea (충청지역의 임상검체로부터 분리된 Acinetobacter calcoaceticus-baumannii Complex를 대상으로 항균제 내성 유전자 비교분석)

  • Species that belong to the Acinetobacter calcoaceticus-baumannii (Acb) complex are major causes of hospital-acquired infections. They are important opportunistic pathogens. These species are usually multidrug resistant (MDR), and the therapeutic options to treat the infections caused by these species are limited. In the present study, we investigated fluoroquinolone resistance mechanisms in 53 ciprofloxacin resistant Acinetobacter species isolates in Chungcheong, Korea. Antimicrobial susceptibilities were determined using the disk-diffusion method. Detections of genes and identification of mutations associated with fluoroquinolone resistance were carried out using PCR and DNA sequencing. In our study, 47 out of 53 ciprofloxacin resistant Acinetobacter isolates harbored sense mutations at the 83rd residue (serine to leucine) in the gyrA gene as well as at the 80th residue (serine to leucine) in the parC gene. Among the 47 isolates harboring sense mutations in gyrA and parC gene, 44 isolates were A. baumannii and 3 isolates were A. pittii. Plasmid-mediated quinolone resistance (PMQR) determinants were detected in isolates in our study. Among the 46 ciprofloxacin resistant A. baumannii isolates, 41 showed type A, B, or F banding patterns on their REP-PCR profiles. This result suggests that clonal relation and horizontal spreading of the bacterial isolates have been around hospitals in Chungcheong area. To prevent colonization and disseminations of fluoroquinolone resistance Acb complex isolates, continuous investigation and monitoring of antimicrobial resistant determinants of MDR isolates are needed.

Changes in Soil Temperature, Moisture Content, Light Availability and Diameter Growth After Thinning in Korean Pine (Pinus koraiensis) Plantation (잣나무 임분 내 간벌 후 토양 온도, 수분, 광 유효도 및 직경생장 변화)

  • This study was conducted to investigate the changes in soil temperature, soil moisture content, light availability (photosynthetically active radiation, PAR) and diameter growth in 46-year-old Korean pine (Pinus koraiensis Siebold et Zuccarini) plantation located in Gwangneung experimental forest after 4 years from thinning. Three different thinning intensities [control, 45%(T45), and 60%(T60) basal area reduction] were applied in 2004. Mean soil temperature and soil water content were $8.9^{\circ}C$ and 14.3% for control, $10^{\circ}C$ and 16.1% for T45 and $10.2^{\circ}C$ and 16.1% for T60, respectively. Mean soil temperature and moisture content were significantly different among the treatment plots (p<0.05). Mean PAR in control, T45 and T60 was 44, 143 and178 ${\mu}mol/m^2$/sec, respectively and the differences were also statistically significant among the treatment plots (p<0.05). Mean annual diameter growth in control, T45 and T60 was 0.4, 1.5 and 1.2 mm, respectively. Diameter growth of Pinus koraiensis was steadily increased after thinning in comparison to control. Consequently, the results suggested that soil temperature, soil moisture content and light availability were likely to last beyond the 4 years after thinning in this Korean pine plantation.

  • Neutrophils and lymphocytes are essential inflammatory cells in the pathogenesis of allergy. In this study, we evaluated the role of house dust mite (HDM) in the interaction between allergic lymphocytes and neutrophils. The extract of Dermatophagoides pteronissinus (DP) showed a stronger anti-apoptotic impact on neutrophil apoptosis in the coculture of allergic neutrophils with allergic lymphocytes when compared with that in allergic neutrophils alone. DP increased IL-6, IL-8, MCP-1, and GM-CSF in allergic lymphocytes, and the increased cytokines were inhibited by rottlerin-an inhibitor of the protein kinase C (PKC) ${\delta}$, as well as by SB202190-a p38 MAPK inhibitor. DP activated p38 MAPK in a time-dependent manner. The activation of p38 MAPK was suppressed by PAR2i, which is a protease-activated receptor (PAR) 2 inhibitor, and rottlerin. Both aprotinin-a serine protease inhibitor-and E64-a cysteine protease inhibitor-were not effective on cytokine secretion of lymphocytes. These results, despite increased cytokines in allergic lymphocytes via DP, did not show any differences between asthma and allergic rhinitis. Molecules, including cytokines, released by DP in lymphocytes inhibited the migration of neutrophils. This finding may further elucidate the pathogenic mechanism of allergic diseases due to HDM.

Separation and Determination of Co(II) and Ni(II) Ion as their 4-(2-Pyridylazo) resorcinol Chelates by Reversed-Phase Capillary High-Performance Liquid Chromatography (역상 모세관-고성능 액체 크로마토그래피에 의한 코발트와 니켈 이온의 4-(2-피리딜아조)레조루신올 킬레이트로서의 분리 및 정량)

  • Separation and determinations of Co(II) and Ni(II) ions as their 4-(2-pyridylazo)resorcinol(PAR) chelates by reversed-phase capillary high-performance liquid chromatography(RP-CpHPLC) were performed. Among many capillary columns, Vydac C4 column was selected and acetonitrile solution was used as mobile phase. The effect of pH and MeCN concentration(%) on the retention factor, k and peak intensity was examined and discussed. As a results, it was found that 22.5% MeCN and pH 5.60 was adequate as mobile phase for the separation of the two metal ions and determination of Co(II) ion, but the mobile phase condition for Ni(II) ion determination was 22.5% MeCN of pH 7.20. Detection limit(D.L., S/N=3) of Co(II) and Ni(II) ions were $2.0{\times}10{-7}$ M(14.9 ppb) and $1.0{\times}10{-6}$ M(59.2 ppb), respectively.

Beaucoup parlent de musical, mais beaucoup de musicals sont $mont{\acute{e}}s$ sur la $sc{\grave{e}}ne$ sans des recherches $n{\acute{e}}cessaires$ suffisantes. C'est pour cette raison que la plupart des spectacles musicaux qui sont $cr{\acute{e}}es$ en $Cor{\acute{e}}e$ sont $destin{\acute{e}}s$ en faillite $d{\grave{e}}s$ la naissance. D'ailleurs les musicals $liscenci{\acute{e}}s$ par $l^{\prime}Am{\acute{e}}rique$, la France qui sont $mont{\acute{e}}s$ sur la $sc{\grave{e}}ne$ de $Cor{\acute{e}}e$ aujourd'hui sont assez pour nuire le $march{\acute{e}}$ $cor{\acute{e}}en$ $constitu{\acute{e}}$ autour de musical. Quels sont donc les $probl{\grave{e}}mes$ que posent les musicals $liscenci{\acute{e}}s$? $Premi{\grave{e}}rement$ ils encouragent la mime, pas la $cr{\acute{e}}ation$. Donc, $d{\grave{e}}s$ le $d{\acute{e}}but$, il n'y avait pas de $consid{\acute{e}}rations$ artistiques dans et sur le musical. Comme c'est par le commerce que le musical est $r{\acute{e}}pandu$, s'il y aura un $probl{\grave{e}}me$ quelconque, il est $tr{\grave{e}}s$ possible qu'on cherche ${\grave{a}}$ $r{\acute{e}}soudre$ le $probl{\grave{e}}me$ par la vue de commerce. En comprenant les $probl{\grave{e}}mes$ $mentionn{\acute{e}}s$ $l{\grave{a}}$-dessus, il faut $pr{\acute{e}}parer$ le changement de $march{\acute{e}}$ $cor{\acute{e}}enne$ de musical. Le moyen le plus $su{\hat{r}}$ de se $pr{\acute{e}}parer$ est de trouver la $r{\acute{e}}solution$ dans la culture. Effect of Panax ginseng on Latency of Passive Avoidance Response and Neuronal Damage of Hippocampus

  • The effects of crude saponin (SAP) and alkaloid (ALK) fractions of Panax ginseng C.A. Meyer on the detrimental effects of electroconvulsive shock (ECS) and scopolamine on passive avoidance response (PAR) were studied in male Sprague-Dawley rats, referring their effects on the neuronal injury and plasticity of hippocampus in response to electrolytic lesion of left entorhinal cortex (ECL). The detrimental ECS effect on PAR was attenuated by pre- and post-treatments with SAP and ALK, respectively, or by pretreatment with aminoguanidine (AG), an inhibitor of diamine oxidase and NO synthase. And the detrimental scopolamine effect on PAR was also inhibited by pre-treatment with ALK or AG, and by post- treatment with SAP or ALK, respectively. On the 7th day after ECL, the brain sections stained by cresyl violet and by acetylcholinesterase (AChE) histochemistry, respectively, showed the chromatolysis and numeral decrease of neurons and the reduction of AChE reactivity in the hippocampus CA1 area and to a lesser extent, in the dentate gyrus. The neuronal cell death of the CA1 area was significantly reduced by SAP, ALK, or AG, and the reduction of AChE reactivity was significantly attenuated by SAP or ALK and to a lesser extent by AG. These results suggests that the protective effect of ginseng SAP and ALK fractions on ECS- or scopolamine-induced impairment of PAR may be ascribed in part to preservation of hippocampal neurons, particularly cholinergic neurons.

Characteristics of Photosynthesis with Growing Stages by different Shading Materials in Panax Ginseng C. A. Meyer (차광재료에 따른 생육시기별 인삼의 광합성작용의 특성)

  • To investigate the influence of shading materials with growing stages in Panax Ginseng C. A. Meyer, the diurnal change of photosynthesis, stomatal conductance, transpiration and its any correlation were measured. The net photosynthetic rate and stomatal conductance of ginseng were higher in the morning than in the broad day. The net photosynthetic rate was increased as the PAR (Photosynthetically Action Radiation) was increased and it was reached the maximum at the $200\;{\mu}mol/m^2/s$ of PAR in overall leaves. Transpiration rate was increased in the afternoon compared to in the morning. The transpiration rate was higher in rain shelter shading plate than in polyethylene net. A linear equation was obtained between net photosynthetic rate and stomatal conductance in the morning. SPAD was higher in rain shelter shading plate than in polyethylene net through all growth stages. It may result from the decrease of growth progress. From investigating photosynthetic characteristics, we concluded that shading plate of rain shelter was more an efficient material to ginseng growth.

Changes of Chlorophyll Fluorescence and Photosynthesis under Different Shade Materials in Korean Ginseng(Panax ginseng C. A. Meyer) (해가림자재에 따른 인삼의 엽록소 형광 반응 및 광합성 변화)

  • This study was conducted to investigate the influence of shading material on the chlorophyll fluorescence, photosynthesis, transpiration, stomatal conductance and its any correlations in Panax ginseng C.A.Meyer. Fo was higher in polyethylene shade net than in silver-coated shading plate, but this treatment caused a lower Fm in comparison with silver-coated shading plate. Also, Fv/Fm and PhiPS2 showed higher in silver-coated shading plate than in polyethylene shade net. The relationship between net photosynthetic rate and transpiration, stomatal conductance were increased as the PAR (Photosynthetic active radiation) was increased and reached maximum at the $200-400\;{\mu}mol/m^{2}/s$ of PAR in all of leaves, and the higher in silver-coated shading plate than in polyethylene shade net. A linear equation was obtained between net photosynthetic rate and transpiration, net photosynthetic rate and stomatal conductance. SPAD was higher in silver-coated shading plate than in polyethylene shade net.

  • An ion-association chromatography was applied for the separation and determination of individual rare earth elements (REE) contained in mineral monazite. Prior to the determination, the group separation of REE was achieved by a cation exchange column of Dowex 5OW-X8 resin. The quantitative recovery of REE by the resin column, free from coexisting elements in monazite, was confirmed with radioactive tracers as well as with ICP-MS. Individual REE at ppm level was separated on reversed-phase column ($\mu$-Bondapak $C_{18}$) using gradient elution from 0.05 to 0.3 M $\alpha$-hydroxyisobutyric acid at pH 4.6. The individual REE was detected at 546 nm following post-column reaction with PAR (4-(2-pyridylazo)-resorcinol monosodium salt).

Characterization of Quinolone-Resistant Clinical Isolates of Escherichia coil in Korea

  • Twenty-eight clinical isolates of Escherichia coil, composed of thirteen norfloxacin resistant isolates (MIC of >16${\mu}$g/ml), one intermediately resistant isolate (MIC of 8${\mu}$g/ml), and fourteen susceptible isolates (MIC of <4${\mu}$g/ml), were randomly selected to study the norfloxacin resistance mechanism and phylogeny in clinical isolates in Korea. Eleven nofloxacin resistant isolates and one susceptible isolate were multi-drug resistant (MDR). Every norfloxacin resistant isolate with MIC higher than 32${\mu}$g/ml had the same three mutations: Ser83\longrightarrowLeu and Asp87\longrightarrowAsn or Tyr in GyrA and Ser80\longrightarrowIle in ParC. Whereas a resistant isolate with MIC of 16${\mu}$g/ml had three mutations but Asp87 in GyrA was replaced with Gly instead of Asn. The intermediately resistant isolate had the same two mutations in GyrA but a different mutation in ParC, Glu84\longrightarrowLys. Among the susceptible isolates, two isolates with MIC of 4${\mu}$g/ml had one mutation: Ser83\longrightarrowiLeu in GyrA, and no mutation was found in the susceptible isolates. Resistant isolates showed higher efflux activity than the susceptible ones, with random amplification of polymorphic DNA (RAPD), six susceptible isolates form a separate group from the rest of the isolates.
