Rapid and Efficient Detection of 16SrI Group Areca Palm Yellow Leaf Phytoplasma in China by Loop-Mediated Isothermal Amplification

  • Areca palm yellow leaf (AYL) disease caused by the 16SrI group phytoplasma is a serious threat to the development of the Areca palm industry in China. The 16S rRNA gene sequence was utilized to establish a rapid and efficient detection system efficient for the 16SrI-B subgroup AYL phytoplasma in China by loop-mediated isothermal amplification (LAMP). The results showed that two sets of LAMP detection primers, 16SrDNA-2 and 16SrDNA-3, were efficient for 16SrI-B subgroup AYL phytoplasma in China, with positive results appearing under reaction conditions of 64℃ for 40 min. The lowest detection limit for the two LAMP detection assays was the same at 200 ag/μl, namely approximately 53 copies/μl of the target fragments. Phytoplasma was detected in all AYL disease samples from Baoting, Tunchang, and Wanning counties in Hainan province using the two sets of LAMP primers 16SrDNA-2 and 16SrDNA-3, whereas no phytoplasma was detected in the negative control. The LAMP method established in this study with comparatively high sensitivity and stability, provides reliable results that could be visually detected, making it suitable for application and research in rapid diagnosis of AYL disease, detection of seedlings with the pathogen and breeding of disease-resistant Areca palm varieties.

A Study on Thermographic Change of DITI by Sugi Therapy. (수기요법(手氣療法)에 따른 체열변화(體熱變化)의 비교(比較) 연구(硏究))

  • Objective: I carried out this study for research on thermographic change of DITI by Sugi Therapy. Methods: This study was conducted on 22 normal cases (M : F =4:18) with no past history to observe the effect of Sugi Therapy. 22 cases were divided into 2 groups. One group is a control group treated by only Acupuncture Therapy. The other group is a treatment group treated by Acupuncture Therapy and Sugi Therapy. I measured temperature of abdomen and palm by DITI (Digital Infrared Thermographic Imaging) before and after treatment Result: 1. Overall the temperature difference of the treatment group was bigger than that of the control group. 2. On the palm, the average value of the treatment group was -0.56±0.70, which is lower 10 times than -0.06±0.91 of the control group. 3. On Chung-wan(CV12), the average value of the treatment group was 1.20±0.49, which is 2.15 times higher than 0.56±0.44 of the control group. 4. On Kwanwon(CV4), the average value of the treatment group was 0.77±0.73, which is 1.38 times higher than 0.55±0.39 of the control group. 5. To compare the temperature difference on the palm, Chung-wan(CV12) and Kwanwon(CV4) before and after treatment, the order is like this :Chung-wan(CV12) > Kwanwon(CV4) > palm of all treatment group and control group. Conclusion: I came to a conclusion that Sugi Theraphy is likely to work better with Acupuncture Therapy better than only Acupuncture Therapy, especially on Chung-wan(CV12).

Dietary Manipulation and Increase in Plasma Unsaturated Fatty Acids in Sheep

  • Forty three 7-month old, Barbados Black $Belly{\times}Malin$ crossbred sheep were used for the trial. They were allotted into three treatment groups fed varying levels of oil palm (Elaeis guineensis) frond pellets and commercial sheep pellets. Treatment diets were 80% commercial pellet+20 % (% w/w) oil palm frond pellet (CON group, n=15), 50% commercial pellet+50% oil palm frond pellet (% w/w) (HAF group, n=14) and 80% oil palm frond pellet+20% (% w/w) commercial pellet (OPF group, n=14). The plasma fatty acid profiles from these animals were compared before and after 14 weeks of feeding. Results showed that total unsaturated fatty acid content in the CON group had increased by 10% (p<0.01) from the pre-treatment values. All three treatment groups had significantly different plasma n-6 and n-3 polyunsaturated fatty acid contents at the end of the trial. In fact, the CON group had significantly (p<0.01) more n-6 polyunsaturated fatty acid content compared to its own initial values, and also the values from the HAF and OPF groups. However there was a significant (p<0.01) decline in plasma n-3 polyunsaturated fatty acids in all groups. The final total unsaturated to saturated fatty acid content ratio was significantly (p<0.01) highest in the CON group, demonstrating the high plasma unsaturated fatty acid content in these animals. This study shows the plasma unsaturated fatty acids in sheep can be increased by dietary manipulation.

  • The purpose of this study was to develop the FSR sensor suit that controls walking assist device for paraplegic patients. The FSR sensor suit was to detect user's intent and patterns for walking by measuring pressure on the palm and the sole of user's foot. It consisted of four modules: sensing pressure from palm, changing modes and detecting pressure on the palm/at the wrist, sensing pressure from the soles of user's foot, and host module that transmit FSR data obtained from sensing modules to PC. Sensing modules were connected to sensing pads which detect analog signals obtained from the palm or the sole of foot. These collect signals from the target regions, convert analog signals into digital signals, and transmit the final signals to host module via zigbee modules. Finally, host modules transmit the signals to host PC via zigbee modules. The study findings showed that forces measured at the palm when using a stick reflected user's intent to walk and forces at the sole of the user's foot revealed signals detecting walking state.

Stabilization Characteristics of Upgraded Coal Using Palm Oil Residues (팜 잔사유를 이용한 고품위화 석탄의 안정화 특성분석)

  • In this research, the stabilization characteristics of upgraded coal using palm oil residues were investigated. The Eco coal, which is the Indonesian low-rank coal, was used as a raw material. The low-rank coal was mixed with palm fatty acid distillate (PFAD), and then dried in a nitrogen atmosphere at $107^{\circ}C$. The trend of spontaneous combustion of upgraded coal was studied by measuring of crossing-point temperature (CPT), low temperature oxidation and moisture readsorption. The results of the CPT measuring and low temperature oxidation showed that the propensity of spontaneous combustion of the upgraded coal was improved compared to the dried coal. The moisture readsorption characteristics of the upgraded coal was also improved. The upgraded coal was stabilized through the surface coating with PFAD, and stability of upgraded coal was proportional to the content of PFAD.

  • In this study, we investigated the feasibility of torrefied OPF (oil palm fronds) as the fuel. The torrefaction was performed at 200, 250, 300 and $350^{\circ}C$ during 1 and 2 hours, respectively. As raising the torrefaction temperature and increasing the processing time, the GHV (gross heating value) of torrefied OPFs was increased. Moreover, we found that the torrefaction temperature is more important factor than the processing time. However, the proper torrefaction temperature was asked because the higher torrefaction temperature leaded to the lower torrefied OPF yield. TGA (thermo-gravimetric analysis) data released that the torrefaction at $250^{\circ}C$ could significantly decompose the hemicellulose and the almost cellulose was decomposed at $300^{\circ}C$. In addition, the grindability of biomass was improved after torrefaction, so that it can reduce energy consumption in comminution.

  • Recently, both the integration of product data during design and production and the effective management of information during full lifecycles have attracted attention from shipyards and ship owners as a result of recycling regulations and a desire for efficient operations. Generally, PLM (Product Lifecycle Management) supports a collaborative environment during the BOL (Beginning of Life) stage, while an ALM (Asset Lifecycle Management) system provides all of the information required to maintain, overhaul, and discard/recycle all or part of a vessel during the MOL (Middle of Life) and EOL (End of Life) stages. The main goal of this paper is to suggest the fundamental configuration of a PALM (Product Asset Lifecylce Management) system and a method that can be used to utilize a marine vessel's lifecycle information during the MOL, emphasizing the maintenance information during the middle of life. The authors also suggest a PALM system configuration in which lifecycle information can be collected by a PEID (Product Embedded Information Device) integrating a microcomputer, sensors, and wireless network communication. Through a prototype PALM system, the suggested features and PALM system configuration are implemented.

Ingredient Contents of Nipa Palm(Nypa fruticans Wurmb.) according to Different Extraction Methods (추출방법에 따른 니파팜의 성분 함량)

  • This study was conducted to confirm the change in the contents of the ingredients according to the extraction method of nipa palm. The contents were analyzed by extraction according to the ethanol ratio, extraction time, and extraction temperature. The contents of polyphenols and flavonoids according to the ratio of the extraction solvent were the highest at 36.91 and 27.62 mg/g, respectively, when extracted with 50% ethanol. Polyphenols and flavonoids according to extraction temperature and extraction time showed the highest content of 40.83 and 37.63 mg/g, respectively, when extracted for 6 hours at 60℃. The contents of the major component of nipa palm according to the ethanol ratio were 2.08 mg/g in 70% ethanol for 5-O-caffeoylshikimic acid, 0.10 mg/g for 4-hydroxybenzoic acid in 30% ethanol, and 0.12 mg/g for 3,4- hydroxybenzoic acid in 50% ethanol. It is expected that it can be used as basic research data when developing natural materials such as food and cosmetics through the change in the contents of the ingredients contained in nipa palm according to the extraction methods.

Thermooxidative Stability of Soybean Oil, Beef Tallow and Palm Oil during Frying of Steamed Noodles (증숙면 튀김 과정 중 대두유, 우지, 팜유의 가열 산화 안정성)

  • Thermooxidative stabilities of soybean oil, beef tallow and palm oil were studied during frying of steamed noodles. Steamed noodles were fried in $150^{\circ}C$ oils for 70 sec at the interval of 30 min. The frying oil was taken every 8 hrs for the analysis of peroxide value (PV) and free fatty acid (FFA) content, fatty acid composition, and tocopherol and tocotrienol content. A little change was shown in PV and FFA content in soybean oil during frying; on the other hand, rapid increase in beef tallow and palm oil was observed. Unsaturated fatty acid content was the highest in soybean oil, followed by palm oil and beef tallow. While fatty acid composition in soybean oil was not changed during frying, unsaturated fatty acid content decreased and saturated fatty acid increased in beef tallow and palm oil, which showed susceptibility to the oxidation. The ratio of linoleic acid to palmitic acid did not show difference with frying time in soybean oil: however, it decreased in other oils with a high correlation with frying time and higher decreasing rate in palm oil was observed. These suggested that soybean oil was the most stable to thermooxidation and the stability was followed by beef tallow and palm oil. Tocopherol was disappeared during frying and 87.5, 81.1, and 73.1% were remained in soybean oil after 8, 16 and 24 hour frying, respectively. Also the rate decreased in the order of ${\gamma}-,\;{\beta}-\;and\;{\alpha}-tocopherol$. However, 34.2 and 169.0 ppm tocopherol and tocotrienol which were present in control samples of beef tallow and palm oil were completely disappeared by 8 hr frying. Therefore, high thermooxidative stability of soybean oil resulted from higher residual amount of tocopherol during frying, and lower stability of palm oil than beef tallow was partly due to high degree of unsaturation.

Role of FT-IR in Assessing Lubricant Degradation - A Study on Palm Oil Methyl Ester Blended Lubricant

  • In this paper, studies were made on the palm oil methyl ester (POME) added lubricants using FT-IR for monitoring oil degradation. In order to assess the degradation characteristics of POME added lubricant by FT-IR, static oxidation test was conducted using three different blended lubricants (viz, zero percent POME, five percent POME and ten percent POME with mineral-based oil) for 280 hrs. The oxidation temperature was set at $140^{\circ}C$. FT-IR quantitative data indicate an increased in oxidation products which was formed from 10% POME added lubricants after 280 hrs of oxidation test. The 5% POME added lubricant and mineral-based lubricant (without POME) showed less oxidation product after the test. From the FT-IR spectrum analysis of the oxidized oils it could be concluded that 5% POME can improve the performance of mineral-based oil by forming protective films.
