The buffalo is an important livestock resource in several countries of South Asia and the Mediterranean regions. However, reproductive efficiency is compromised due to known problems of biological and management origins, such as lack of animal selection and poor nutrition. Under optimal conditions puberty is attained at 15 to 18 months in river buffalo, 21 to 24 months in swamp buffalo and is influenced by genotype, nutrition, management and climate. However, under field conditions these values deteriorate up to a significant extant. To improve reproductive efficiency, several protocols of oestrus and ovulation synchronization have been adopted from their use in commercial cattle production. These protocols yield encouraging pregnancy rates of (30% to 50%), which are comparable to those achieved in buffaloes bred at natural oestrus. The use of sexed semen in buffalo heifers also showed promising pregnancy rates (50%) when compared with conventional non-sexed semen. Assisted reproductive technologies have been transferred and adapted to buffalo but the efficiency of these technologies are low. However, these latest technologies offer the opportunity to accelerate the genetic gain in the buffalo industry after improving the technology and reducing its cost. Most buffaloes are kept under the small holder farming system in developing countries. Hence, future research should focus on simple, adoptable and impact-oriented approaches which identify the factors determining low fertility and oestrus behaviour in this species. Furthermore, role of kisspeptin needs to be explored in buffalo.
This experiment was carried out to investigate the effects of long-term applications of organic matters on the chemical properties and on the application levels of nitrogen in Fluvio-Alluvial plain of Jeonbug series. The amounts of application of rice straw and compost, the sources of organic matters, were 500 and 1,000 kg/10a in combined with the different nitrogen levels of 0.15 and 20 kg/10a, respectively. The results obtained from the 9 year's experiment during 1979 to 1987 were summarized as follows: I. In long-term application of Organic matter the soil pH showed the lowest value in the 3rd-4th year at rice straw and 5th-6th year at compost but it varied less in control plot for 9 years. 2. Organic matter content in the soils was gradually increased by yearly application of organic matter, while it was higher in rice straw than in compost since the 5th year. 3. The contents of available silica and available phosphate in soil were rapidly increased by long-term application of organic matter and it was especially higher in rice straw than compost, but it was gradually decreased in control plot. 4. The contents of exchangable cations (Ca, Mg and K) and the total nitrogen were increased by long-term application of organic matter they were in rice straw than in compost. But they showed decreasing tendency in control plot. 5. The soil Eh was lower in order of rice straw, compost and control plot however application of orgnic matter resulted in increasing soil Eh due to the rapid reduction of soil from panicle formation to heading stage in rice cultivation. 6. The number of panicles per $m^2$ and the number of granis per panicle were increased by increment of nitrogen levels in all treatments and especially largest in rice straw application. But 1,000 grain weight increased in low nitrogen level with long-term applications of organic matter. 7. It was estimated that the application levels of nitrogen by long-term application of organic matter were 21, 24 and 20 kg/10a for 1st-3rd, 4th-6th and 7th-9th year in rice straw application, and 16 and 19 kg/10a for 1st-3rd and 4th-9th year in compost application, respectively.
The characteristics of the heavy metal contamination in the soils affected by the tailings of the Palbong mine have been studied. The soils in the studied area consist mostly of loam by the particle size analysis, but a little of it, located near the stream, consist of loamy sand to sandy loam, finally to loam downward. The organic contents of soils are significantly low aoom 2 percent and the pH is in acidic ranging 6.0 $\pm$ 0.1. The samples of the parent rocks, the normal soils, the tailings and the channel deposits from the studied area were chemically analysed. From the result, the heavy metal concenlration of the soils is a little Jow compared with that of the parent rocks, shows the hydrologic process of the surface and the groundwater. The contamination of the tailings from the ore mining are high in lead, copper and arsenic. In the channel deposits the concenlrations of lead and copper are abnormally high but that of arsenic is uniquely low. And most of heavy metal contamination are decreased with the distance from the mine. It is caused by the properties of the surface and the ground water during the process of the heavy metal migration. The correlation-coefficient between sand and silt contents and the concentrations of Cd, Cu and Pb are significant but the amounts of As and Hg are increased with the clay contents. The dispersion of the heavy metals with the distance shows that the concentrations of them in the soils sampled at distance of 100 m to 200 m along the stream started near the Palbong mine are extremely high compared with those from other distances. These discrepancies are significant in Cd, Cu, Pb and Hg, but low in As. All the samples contain below detection limit of Cr+6 In the present stream water the concentrations of the heavy metals are not detected. So, it is interpreted that the concentrations in the soils are caused by the activities of the mining during the operation and have been continued by the dispersion from the tailings since after the closure of the mining, especially by the surface and ground water. The concentrations are diminished with the distance from the mining site, but in the interval of 800-2000 m increases abruptly. In the soil samples counted on the dispersion direction by wind, the lowering of the concentration is relatively uniform with the distance from the mining site. So, the rapid increase of the heavy metal concentrations is presumed to have been caused by the ground-water movement. In the migration of the heavy metals, the groundwater conditions, such as pH, Eh, the contents of colloidal particles, and Mn and Fe oxides are closely involveo. Integrating with these factors, it is interpreted that the groundwater conditions which have caused the heavy metal contamination of the studied area are those that the pH is about 3 in oxidized conditions, the contents of the colloidal particles are low, and Mn and Fe oxides are not involved in the migration of the heavy metals. Meanwhile, the vegetables growing on the soils in the studied area are not affected by the contamination of the heavy metals.
To find out the oxidation process of potential acid sulfate soil(PASS) along with time. the PASS were treated with lime and ammonia water to adjust soil pH in laboratory column condition. pH range of PASS showed 6.5 to 7.5. however, complete oxidized PASS by $H_2O_2$ showed 2.1 to 2.5. After pilling the PASS under the natural condition. oxidation occured slowly from surface of the pilled soil. The oxidation of PASS proceeded slowly when the soil was in submerged condition. but quickly in dried condition. The content of sulfide-sulfur in PASS sharply decreased after exposing to the air and the decreasing rate was greater in dried than in submerged condition. The content of sulfate-sulfur continuously decreased in submerged condition. but increased in dried condition. Contents of $Fe^{+{+}}$ and $Al^{+{+}}$ in PASS were generally increased with time and the increasing rate was greater in submerged than in dried condition. Liming to PASS was slowly acting to pH change and ammonia water caused fast pH change within a short period of time. The contents of sulfate-sulfur and exchangeable aluminum in drainage water decreased with time and the contents of sulfide-sulfur and ferrous iron were increased.
This study was conducted to evaluate the effects of red earth addition in sandy loam paddies. Clay contents of the red earths were 19% (loam) and 35% (clay loam). The treatments were 3 levels of red earth and 2 levels of fertilizer. The results were summerized as follows: 1. Water decrease rate was lowered with the increase of clay by the red earth application in sandy loam paddies. 2. Red earth addition increased CEC and available $SiO_2$ in sandy loam, and integrated soil improvement showed higher pH and lower Eh than standard fertilizer application. 3. Rice yields in the red earth addition of loam were decreased by 2-3% at standard fertilizer application and increased by 2-4% at integrated soil improvement. Otherwise, rice yields in the red earth application of clay loam were increased by 1-4% at standard fertilizer application and 3-8% at integrated soil improvement.
As-rich alluvial groundwaters occurring in the agricultural area of Mankyeong River watershed were geochemically studied. 15 out of 29 investigated wells (52%) showed As levels exceeding the WHO drinking water standard ($10{\mu}g/L$). Their chemistry is characterized by low Eh levels, low $NO_3$ and $SO_4$ concentrations, and high pH, alkalinity, Fe, $NH_4$, and $PO_4$ levels. This suggests that arsenic is enriched by the reductive dissolution of As-bearing Fe-/Mn-(hydro)oxides, the commonest process in Bangladesh and West Bengal of India, of which groundwaters are severely contaminated by As. It was also revealed that As concentrations in the area are strongly regulated by the presence of agrochemicals such as $NO_3$ and $SO_4$.
In this study, an electrical resistivity survey and a drilling investigation were conducted at an animal carcass disposal site. Chemical analysis of leachate collected from the site was also performed (sampling times: May 2011 and June 2012). Five lines of dipole-dipole electrical resistivity surveys were carried out, along with drilling investigations at 3 points within the disposal areas and 11 points near the disposal site. Two-dimensional inverse modeling of the collected resistivity data was performed to evaluate the properties (size, depth, and form) of the disposal site. Leachate analysis showed that pH of leachate decreased from 7.4 to 6.7, while Eh changed from -358 mV to -48 mV over time. In addition, dissolved ions increased due to the progression of carcass decomposition. Results of the electrical resistivity survey indicated that low resistivity zones (minimum value, $0.64{\Omega}m$) existed at a depth of 8 m from the surface. Considering the bedrock location and carcass disposal depth, there was no evidence of bedrock contamination by leachate. The results of the electrical resistivity survey are consistent with those of the drilling investigation, which indicates that electrical resistivity effectively depicted the properties of the disposal site. This study demonstrates that electrical resistivity survey is a suitable technique for investigation of animal carcass disposal sites.
The objective of this research was to examine the effect of ammonium thiosulfate(ATS) on urease activity and on biological and chemical properties of flooded paddy soil especially having high organic matter content by comparing with the effect of sodium thiosulfate(STS). The results obtained are summarized as follows. 1. The hydrolysis of urea was inhibited at 3 and 5 days after treatment of thiosulfate(ATS and STS) +glucose and thiosulfate only, respectively. The inhibitory effect of ATS on urea hydrolysis was slightly lower than that of STS in glusoce-added soils, but when the glucose was not added, the effects of ATS and STS were not different significantly. 2. The soil pH and Eh was lowered by 0.3~0.5 units and 30~120 mV, respectively, when incubated flooded soil with ATS and glucose at $25^{\circ}C$. 3. Soil respiration rate in/flooded soil was increased by 10~70% with the treatment of ATS during the 20 day experimental period. 4. The contents of acetic and butyric acid in thiosulfate treatment soil was below $10{\mu}g/g$, which was lower than that($220.3{\mu}g/g$) of critical growth inhibition of rice.
To investigate quality characteristics of kimchi prepared for the winter around Chonnam area, home made kimchi samples collected from 22 area, and they were stored at -1${\pm}$1$^{\circ}C$. The results were as follows : The pH and acidity of kimchi samples were 4.75 and 0.84%. respectively. Salt concentration was 3.50% and in Redox potential measurement, Eh value was -134.08mV. Ascorbic acid and reducing sugar contents were 10.l8mg% and 13.25mg%, respectively, In color measurement, L value was 52.29 and a and b values were 19.68 and 27.69, respectively. Total viable count was 5.5${\times}$10$\^$6/ and lactic acid bacteria count and yeast were 4.6${\times}$10$\^$5/ and 8.8${\times}$10$\^$5/. respectively. Properties of hardness of kimchi measured instrumentally was 9.26kgf. Alcohol insoluble solids(AIS) content was 5.53% and hot water soluble pectin(HWSP) content and sodium hexametaphosphate soluble pectin(NaSP) content were 17.35% and 29.65%, respectively, also hydrochloric acid soluble pectin(HClSP) content was 53.0%.
A gene for cellulolytic xylanase of Bacillus circulnns ATCC21365 was cloned on pUC 19 in Eschwichia coli. The recombinant plasniid pXLI80 contained an 1.8 id, inselt composed of0.5 kb and 1.3 kb PslI fragments derived from B, circulans. The 0.5 kh fragment in the upstream region of 1.3 kb one was confirmed lo be indispensable for not only expression but also hyperexpression of the cloned gene. The transformant overproduced the xylanase 135 times greater than that produced by the orlginal B circulnns. The optimum pH and temperature of the cloned enzyme we]-e pH 5.2 and $60^{\circ}C$, respectively. The SDS-PAG electrophoretic mobility and zyiiogram of this enzyme derived from whole cell extracts or c~~lture supematants or E. coli(pXL180) indicated a molecular weight of 45,000 and nonprocessing of the enzyme in the peilplasln of E. coli.
