석회암지대 지하수 및 하천수 ${\delta}13CDIC$값의 변화 요인

  • 2004년 7월부터 2005년 2월까지 8개월 동안 강원도 정선지역 탄산염지대 지하수 및 하천수의 ${\delta}^{13}C$의 조사 결과, 탄산염 지대 지하수의 13C는 $-12.07{\sim}-8.63$ (평균 -10.34 ), 탄사염지대 하천수의 ${\delta}^{13}C$$-10.32{\sim}-6.80$ (평균 -7.944 ) 이었다. 하천수와 지하수 ${\delta}^{13}C$는 수온(T)과 음의 상관관계를 보이고 물의 전기전도도(EC)와 양의 상관관계를 보이며, 여름보다 겨울에 높은 값을 보이는 반면 산화환원 전위(Eh), 용존산소(DO), pH와는 상관관계가 미약함을 보이고 있다. 탄산염지대 지하수 및 하천수의 ${\delta}^{13}C$는 연구지역의 지하수와 하천수가 탄산염암의 용해, 대기 $CO_2$와 용존 $CO_2$의 교환, 유입된 대기 $CO_2$가 물 분자와 반응하여 ${HCO_3}^-$ 이온으로 전환될 때 있는 분별작용의 효과에 의해 주로 영향을 받았으며 수중생물의 신진대사에 의한 변화는 미미한 것으로 나타났다.

Discussion on Genesis of the Zhezkazgan Copper Deposit in Kazhkstan (카자흐스탄 제스카즈간 동광상의 성인 고찰)

  • Geology of the Zbezkazgan copper deposit in Kazhkstan is mainly composed of Permian and Carboniferous sedimenary rocks in which copper minerals are mainly contained in grey sandstone of Carboniferous age. There are 28 layers of copper ore bodies in Zbezkazgan suite. Thickness of the ore bodies ranges from one to 35 meters, grade of the crude ore ranges from 2 to 5 wt % Cu and the extension of the orebodies is 5 to 7 km. Microscopic study on specimens from the Zbezkazgan ore deposit has exposed clues to understand the origin of this deposit. Alternatively deposited grey sandstone and red sandstone are mainly composed of quartz and feldspar grains. A big difference between the grey sandstone and the red sandstone is in grain size, the former is larger than the latter. Chalcocites as main copper minerals have cemented through grain boundary. It is assumed that quartz, feldspar and copper were derived from granitoid in which copper mineralization had taken place before exposing to weathering. The chalcocites were precipitated by a sudden change of geochemical condition (Eh, pH, temperature, etc.) of fluid which had carried quartz, feldspars, copper ions and sulphate during formation of grey sandstones. The copper ions and sulphate were stable in fluid during sedimentation of oxidation environment, however, the copper ions were no more stable at the reduced environment and changed to stable forms to precipitate copper minerals by reaction of copper ions and hydrogen sulfides. This chemical precipitation of copper minerals in the sandstone attributes to the assumption of hydrothermal origin on this sedimentary origined deposit.

Effect of Phospho-gypsum on reduction of methane emission from rice paddy soil

  • Phospho-gypsum a primary waste by-product in phosphate fertilizer manufacturing industry and a potential source of electron acceptors, such as mainly of sulfate and a trace amount of iron and manganese oxides, was selected as soil amendment for reducing methane $(CH_4)$ emissions during rice cultivation. The selected amendment was added into potted soils at the rate of 0, 2, 10, and 20 Mg $ha^{-1}$ before rice transplanting. $CH_4$ flux from the potted soil with rice plant was measured along with soil Eh and floodwater pH during the rice cultivation period. $CH_4$ emission rates measured by closed chamber method decreased with increasing levels of phospho-gypsum application, but rice yield markedly increased up to 10 Mg $ha^{-1}$ of the amendment. At this amendment level, total $CH_4$ emissions were reduced by 24% along with 15% rice grain yield increment over the control. The decrease in total $CH_4$ emission may be attributed due to shifting of electron flow from methanogenesis to sulfate reduction under anaerobic soil conditions.

Characteristics of Occurrence and Distribution of Natural Radioactive Materials, Uranium and Radon in Groundwater of the Danyang Area (단양지역 지하수중 자연방사성물질 우라늄과 라돈의 산출과 분포특징)

  • Natural radionuclides in groundwater in the Danyang area were investigated to characterize the behaviors of uranium and radon with respect to lithology and physico-chemical components, which can aid our understanding of their occurrence, properties, and origins. To this end, a total of 100 groundwater samples were collected and analyzed, and radionuclide levels were used to construct detailed concentration maps. The water type of the groundwater, assessed using a Piper diagram, is mainly Ca-Na-$HCO_3$. The concentrations of uranium range from 0.02 to $251.0{\mu}g/L$ (average, $3.85{\mu}g/L$) and only 1% exceed USEPA's MCL (Maximum Contaminant Level). Uranium is enriched in groundwaters of Cretaceous granites and Precambrian metamorphic rocks, whereas it is depleted in groundwaters of sedimentary rocks. The concentrations of radon range from 13 to 28,470 pCi/L (average, 2397 pCi/L). Only 15% of the samples exceed AMCL (Alternative Maximum Contaminant Level) of 4000 pCi/L. The radon concentration is highest in groundwater of Cretaceous granites and lowest in groundwater of sedimentary rocks. In conclusion, the distribution and occurrence of radionuclides are intimately related to the basic geological characteristics of the rocks in which the radiogenic minerals are primarily contained. The behavior of uranium is only weakly related to that of radon (correlation coefficient = 0.15). There are also weak correlations between radionuclides and the main chemical components, pH, EC, Eh, and well depth. Of note, the correlation coefficient between radon and $SiO_2$ is 0.68, and that between radon and $HCO_3$ is -0.48. Factor analysis shows that radionuclides behave somewhat independently of each other because there are no significant factors that control the behavior of chemical components as well as radionuclides. The detailed concentration maps during this study will be used to establish useful database of radionuclide distribution and geological properties throughout Korea.

Effect of Irrigation Water Depth on Greenhouse Gas Emission in Paddy Field (논물 담수심이 온난화 가스 배출에 미치는 영향)

  • The increasing emission of greenhouse gases may change agricultural environment. The agronomic productivity will depend upon change of temperature, precipitation, solar radiation and fertilization. This study was conducted to investigate greenhouse gas emission with irrigation water depth in paddy field. Area of each experiment plot is $70m^2$, Three treatments with three replications were carried out in this experiment, which was laid out as randomized complete block design. The treatments of irrigation water were maximum field water capacity and 4 and 8 cm depth. The application rate of fresh rice straw was $8,000kg\;ha^{-1}$ in combination with chemical fertilizers ($110kg\;N\;ha^{-1}$, $45kg\;P_2O_5\;ha^{-1}$ and $57kg\;K_2O\;ha^{-1}$). The $CH_4$ emission was highest at 32 days after rice transplanting with rice straw treatment. The $CH_4$ emission in the plot of maximum field water capacity was lower compared with 4 and 8 cm of irrigation depth. $CH_4$ and $N_2O$ emission under different water depth in the paddy field were 30 and $1.52kg\;ha^{-1}$ at 8 cm depth, 281 and $1.71kg\;ha^{-1}$ at 4 cm depth, and 219 and $2.01kg\;ha^{-1}$ at water saturated condition. The total emission of greenhouse gases equivalent to $CO_2$ emission with rice straw application were $6,939kg\;CO_2\;ha^{-1}$ at 8 cm depth plot, $6,431kg\;CO_2\;ha^{-1}$ at 4 cm depth plot and $5,222kg\;CO_2\;ha^{-1}$ at water saturated condition. The GWPs without rice straw application were $4,449kg\;CO_2\;ha^{-1}$ at 8 cm depth plot, $3,702kg\;CO_2\;ha^{-1}$ at 4 cm depth plot and $4,579kg\;CO_2\;ha^{-1}$ at water saturated condition.

Expression of Cell Cycle Related Genes in HL60 Cells Undergoing Apoptosis by X-irradiation (HL60 세포주에서 방사선 조사에 의한 Apoptosis와 세포 주기 관련 유전자의 발현 변화)

  • Purpose : To evaluate changes in expression of cell cycle related genes during apoptosis induced in HL60 cells by X-irradiation to understand molecular biologic aspects in mechanism of radiation therapy. Material and Methods : HL-60 cell line (promyelocytic leukemia cell line) was grown in culture media and irradiated with 8 Gr by linear accelerator (6 MV X-ray). At various times after irradiation, ranging from 3 to 48 hours were analyzed apoptotic DNA fragmentation assay for apoptosis and by western blot analysis and semi-quantitative RT-PCR for expression of cell cycle related genes (cyclin A, cyclin B, cyclin C, cyclin Dl, cyclin E, cdc2, CDK2, CDK4, $p16^{INK4a}$, $p21^{WAF1}$, $p27^{KIP1}$, E2F, PCNA and Rb). Results : X-irradiation (8 Gy) induced apoptosis in HL-60 cell line. Cycline A protein increased after reaching its peak 48 h after radiation delivery and cyclin E, E2F, CDK2 and RB protein increased then decreased after radiation. Radiation induced up-regulation of the expression of E2F is due to mostly increase of Phosphorylated retinoblastoma proteins (ppRb). Cyclin Dl, PCNA, CDC2, CDK4 and $p16^{INK4a}$ protein underwent no significant change at any times after irradiation. There was not detected $p21^{WAF1}$ and $p27^{KIP1}$ protein. Cyclin A, B, C mRNA decreased immediately after radiation and then increased at 12 h after radiation. Cyclin Dl mRNA increased immediately and then decreased at 48 h after radiation. After radiation, cyclin E mRNA decreased with the lapse of time. CDK2 mRNA decreased at 3h and increased at eh after radiation. CDK4 mRNA rapidly increased at 6 to 12 h after radiation. There was no change of expression of $p16^{INK4a}$ and not detected in expressin of $p21^{WAF1}$ and $p27^{KIP1}$ mRNA. Conclusion : We suggest that entry into S phase may contribute to apoptosis of HL60 cells induced by irradiation. Increase of ppRb and decrease of pRb protein are related with radiation induced auoptosis of HL60 cells and tosis of HL60 cells induced by irradiation. Increase of ppRb and decrease of PRb protein are related with radiation induced apoptosis of HL60 cells and this may be associated with induction of E2F and cyclinE/CDK2. These results support that $p21^{WAF1}$ and $p27^{KIP1}$ are not related with radiation induced-apoptosis.

Characteristics of Vanadium Leaching from Basaltic Soils of Jeju Island, Korea (제주도 현무암 기원 토양의 바나듐 용출 특성)

  • To understand the characteristics of vanadium leaching from soils formed by the weathering of basalts, paleo soil at Gosan, Jeju Island, Korea, and several present-day soils from neighboring areas were collected. Leaching experiments were carried out by two approaches: 1) batch experiments under various geochemical conditions (redox potential (Eh) and pH) and 2) continuous leaching experiments under conditions similar to those of natural environments. From the batch experiments, leached vanadium concentrations were highest under alkaline (NaOH) conditions, with a maximum value of $2,870{\mu}g/L$, and were meaningful (maximum value, $114{\mu}g/L$) under oxidizing ($H_2O_2$) conditions, whereas concentrations under other conditions (acidic-HCl, $neutral-NaHCO_3$, and $reducing-Na_2S_2O_3$) were negligible. This indicated that the geochemical conditions, in which soil-water reactions occurred to form groundwater with high vanadium concentrations, were under alkaline-oxidizing conditions. From the continuous leaching experiments, the pH and leached vanadium concentrations of the solution were in the ranges of 5.45~5.58 and $6{\sim}9{\mu}g/L$, respectively, under $CO_2$ supersaturation conditions for the first 15 days, whereas values under $O_2$ aeration conditions after the next 15 days increased to 8.48~8.62 and $9.7{\sim}12.2{\mu}g/L$, respectively. Vanadium concentrations from the latter continuous leaching experiments were similar to the average concentration of groundwater in Jeju Island ($11.2{\mu}g/L$). Furthermore leached vanadium concentrations in continuous leaching experiments were highly correlated with pH and Al, Cr, Fe, Mn and Zn concentrations. The results of this study showed that 1) alkaline-oxidizing conditions of water-rock (soil) interactions were essential to form vanadium-rich groundwater and 2) volcanic soils can be a potential source of vanadium in Jeju Island groundwater.

Optimal Conditions of Mycelial Growth and Exopolysaccharide Production in Submerged Culture of Phellinus baumii (Phellinus baumii으로부터 세포외 다당체 생산의 최적화)

  • The polysaccharide isolated from Phellinus species has been known as a folk remedy, including antitumor and immune-stimulating activities. However, there are lacks of knowledge about mycelial growth and exopolysaccharide (EH) production in its submerged culture. We investigated the optimal conditions on mycelial growth and EPS production in Phellinus baumii. The optimal temperature and initial pH for mycelial growth and EPS production in shake flask culture of P. baumii were proved to be 3$0^{\circ}C$ and pH 5.0, respectively. In case of carbon source, cellobiose and maltose were highly efficient for mycelial growth and fructose and mannitol were also relatively favorable for EPS production. Yeast extract was the most suitable nitrogen source for mycelial growth and EPS production. The composition of optimal culture medium was determined to be fructose 20 g/L, yeast extract 20 g/L, and $CaCl_2$ 0.55 g/L, respectively. Under the optimal culture condition, the maximum mycelial biomass and EPS achieved in a 5-L stirred-tank fermenter were 17.43 g/L and 3.6 g/L, respectively. It was found that the EPS was a glycoprotein onsisted of mainly arginine (14.1%) and glycine (12.0 %) in protein moiety and mainly mannose (48.7%) and arabinose (38.4%) in carbohydrate moiety.

Geochemical Equilibria and Kinetics of the Formation of Brown-Colored Suspended/Precipitated Matter in Groundwater: Suggestion to Proper Pumping and Turbidity Treatment Methods (지하수내 갈색 부유/침전 물질의 생성 반응에 관한 평형 및 반응속도론적 연구: 적정 양수 기법 및 탁도 제거 방안에 대한 제안)

  • The formation of brown-colored precipitates is one of the serious problems frequently encountered in the development and supply of groundwater in Korea, because by it the water exceeds the drinking water standard in terms of color. taste. turbidity and dissolved iron concentration and of often results in scaling problem within the water supplying system. In groundwaters from the Pajoo area, brown precipitates are typically formed in a few hours after pumping-out. In this paper we examine the process of the brown precipitates' formation using the equilibrium thermodynamic and kinetic approaches, in order to understand the origin and geochemical pathway of the generation of turbidity in groundwater. The results of this study are used to suggest not only the proper pumping technique to minimize the formation of precipitates but also the optimal design of water treatment methods to improve the water quality. The bed-rock groundwater in the Pajoo area belongs to the Ca-$HCO_3$type that was evolved through water/rock (gneiss) interaction. Based on SEM-EDS and XRD analyses, the precipitates are identified as an amorphous, Fe-bearing oxides or hydroxides. By the use of multi-step filtration with pore sizes of 6, 4, 1, 0.45 and 0.2 $\mu\textrm{m}$, the precipitates mostly fall in the colloidal size (1 to 0.45 $\mu\textrm{m}$) but are concentrated (about 81%) in the range of 1 to 6 $\mu\textrm{m}$in teams of mass (weight) distribution. Large amounts of dissolved iron were possibly originated from dissolution of clinochlore in cataclasite which contains high amounts of Fe (up to 3 wt.%). The calculation of saturation index (using a computer code PHREEQC), as well as the examination of pH-Eh stability relations, also indicate that the final precipitates are Fe-oxy-hydroxide that is formed by the change of water chemistry (mainly, oxidation) due to the exposure to oxygen during the pumping-out of Fe(II)-bearing, reduced groundwater. After pumping-out, the groundwater shows the progressive decreases of pH, DO and alkalinity with elapsed time. However, turbidity increases and then decreases with time. The decrease of dissolved Fe concentration as a function of elapsed time after pumping-out is expressed as a regression equation Fe(II)=10.l exp(-0.0009t). The oxidation reaction due to the influx of free oxygen during the pumping and storage of groundwater results in the formation of brown precipitates, which is dependent on time, $Po_2$and pH. In order to obtain drinkable water quality, therefore, the precipitates should be removed by filtering after the stepwise storage and aeration in tanks with sufficient volume for sufficient time. Particle size distribution data also suggest that step-wise filtration would be cost-effective. To minimize the scaling within wells, the continued (if possible) pumping within the optimum pumping rate is recommended because this technique will be most effective for minimizing the mixing between deep Fe(II)-rich water and shallow $O_2$-rich water. The simultaneous pumping of shallow $O_2$-rich water in different wells is also recommended.

Evaluation of Manganese Removal from Acid Mine Drainage by Oxidation and Neutralization Method (산화법과 중화법을 이용한 산성광산배수 내 망간 제거 평가)

  • Two oxidizing agents (KMnO4, H2O2), and one neutralizing agent (NaOH) were applied to evaluate Mn removal in mine drainage. A Mn2+ solution and artificial mine drainage were prepared to identify the Fe2+ influence on Mn2+ removal. The initial concentrations of Mn2+ and Fe2+ were 0.1 mM and 1.0 mM, respectively. The injection amount of oxidizing and neutralizing agents were set to ratios of 0.1, 0.67, 1.0, and 2.0 with respect to the Mn2+ mole concentration. KMnO4 exhibited a higher removal efficiency of Mn2+ than did H2O2 and NaOH, where approximately 90% of Mn2+ was removed by KMnO4. A black MnO2 was precipitated that indicated the oxidation of Mn2+ to Mn4+ after an oxidizing agent was added. In addition, MnO2 (pyrolusite) is a stable precipitate under pH-Eh conditions in the solution. However, relatively low removal ratios (6%) of Mn2+ were observed in the artificial mine drainage that included 1.0 mM of Fe2+. The rapid oxidation tendency of Fe2+ as compared to that of Mn2+ was determined to be the main reason for the low removal ratios of Mn2+. The oxidation of Fe2+ showed a decrease of Fe concentration in solution after injection of the oxidizing and neutralizing agents. In addition, Mn7+ of KMnO4 was reduced to Mn2+ by Fe2+ oxidation. Thus, the concentrations of Mn increased in artificial mine drainage. These results revealed that the oxidation method is more effective than the neutralization method for Mn removal in solution. It should also be mentioned that to achieve the Mn removal in mine drainage, Fe2+ removal must be conducted prior to Mn2+ oxidation.
