활성슬러지 혼합미생물과 Nocardia asteroides에 의한 페놀화합물 분해시 양성자이온의 영향 (Proton Effect on the Degradation of Phenolic Compound by Activated Sludge and Nocardia asteroides)

  • This study was investigated to evaluate the effect of the sodium ion and pH on toxicity of dinitrophenol at high concentrations (0.41 to 0.54 mM), over a sodium concentration range of 0.1 mM to 107 mM and over a pH range of 5 to 9. The concentration of sodium ions in the activated sludge mixed liquor seemed to have very little effect on dinitrophenol toxicity. However, lack of sodium in the growth media resulted in a reduction of the dinitrophenol degradation rate by bacterial isolate from the activated sludge culture, which has been identified as Nocardia asteroides. Dinitrophenol inhibition was found to be strongly dependent on mixed liquor pH. The dinitrophenol degradation rate was highest in the pH range of 6.95 to 7.84; at pH 5.94 degradation of 75 mg/L dinitrophenol was significantly inhibited; at pH < 5.77, dinitrophenol degradation was completely inhibited after approximately 30% of the dinitrophenol was degraded.

  • Equilibrium swelling and pH-sensitivity of a polyelectrolyte copolymer hydrogel were controlled by employing copolymers with different hydrophilic-hydrophobic balances. Model pH-sensitive hydrogels, e.g., poly(methacrylic acid), poly(methacrylic acid-co-acrylamide), poly(methacrylic acid-co-2-hydroxyethylmethacrylate), poly(methacrylic acid-co-styrene) were synthesized at various monomer compositions. As hydrophobicity of the copolymer hydrogels increased, the equilibrium swelling decreased while the pH-sensitivity increased. In the case of poly(methacrylic acid-co-acrylamide), polymer-polymer interaction significantly affected the equilibrium swelling and provided a wide range control of pH-sensitivity.

  • Phenylvinylsulfone의 가수분해 속도상수를 자외선분광기를 사용하여 구하였으며 아울러 넓은 범위에서 잘맞는 반응속도식도 얻었다. 이 식에 의하면 넓은 pH범위, 특히 종전에 잘 규명된 바 없는 산성용매 속에서의 반응 및 수산화이온의 촉매역할등도 정량적으로 잘 설명할 수 있음을 알았다. 즉 pH7 이하에서는 물의 첨가로 부터 반응이 시작됨을 알 수 있었으며 pH9 이상에서는 수산화이온만이 반응에 참여함을 알았다.

pH와 산화환원전위에 따른 상등수-퇴적물에서의 인 형태 변화 (Speciation of Phosphorus Dependent upon pH and Oxidation Reduction Potential in Overlying Water and Sediment)

  • 퇴적물로부터 오염물질 용출은 상등수 수질에 많은 영향을 미친다. 본 연구에서는 pH와 산화환원전위에 따른 상등수 및 퇴적물의 인 존재형태의 변화를 관찰하였다. 상등수가 순환되어 호기성 상태를 유지하는 경우, 성층현상으로 인하여 혐기성을 띠는 경우, 퇴적물 상부에 모래 캡핑이 포설된 상태에서 혐기성을 띠는 경우 등 3가지 경우를 가정하여 상등수의 인 형태별 농도를 측정하였다. 퇴적물에서 인 용출은 상등수가 산성일 경우 증가하였으며, 캡핑은 인 용출량을 저감시키고, $PO_4-P$의 용출을 지연시키는 효과가 있는 것으로 나타났다. 상등수와 퇴적물이 혐기성일 때 상등수의 pe/pH가 인이 apatite 상태로 안정하게 존해하는 범위에서 상등수의 인농도는 감소하여 일정한 농도를 나타냈다. 세개의 칼럼에서 최종 평균 인 농도는 순환칼럼에서 0.223 mg/L, 비순환칼럼에서 0.342 mg/L, 캡핑칼럼에서 0.184 mg/L로 캡핑된 칼럼에서 가장 낮게 나타났다. 성층현상에 의해 수질이 악화될 수 있는 호소에 캡핑을 활용할 경우 퇴적물에 의해서 상등수의 인 농도를 감소할 수 있을 것으로 기대된다.

Synthesis and pH-Dependent Micellization of a Novel Block Copolymer Containing s- Triazine Linkage

  • Novel pH-sensitive moieties containing an s-triazine ring were synthesized with sulfonamide and secondary amino groups. The synthesized pH-sensitive moieties were used for the synthesis of a pH-sensitive amphiphilic ABA triblock copolymer. The pH-sensitive triblock copolymer was composed of diblock copolymers, methoxy poly(ethylene glycol)-poly ($\varepsilon$-caprolactone-co-D,L-lactide) (MPEG-PCLA), and pH-sensitive moiety. These copolymers could be dissolved molecularly in both acidic and basic aqueous media at room temperature due to secondary amino and sulfonamide groups. The synthesized s-triazine rings containing pH-sensitive compounds were characterized by ${^1}H-NMR,\;{^13}C-NMR$, and LC/MSD spectral data. The synthesized diblock and triblock copolymers were also characterized by ${^1}H-NMR$ and GPC analyses. The critical micelle concentrations at various pH conditions were determined by fluorescence technique using pyrene as a probe. Furthermore, the micellization and demicellization study of the triblock copolymer was done with pH-sensitive groups. The sensitivity towards pH change was further established by acid-base titration.

Detection of Wound-inducible Trans-Cinnamic Acid-4-Hydroxylase in Avocado, Persea americana, Roots

  • Trans-cinnamic acid-4-hydroxylase(tC4H) is the first cytochrome P450-dependent monooxygenase of the phenylpropanoid pathway. The roots of avocado seedlings were wounded and examined to determine whether the tC4H would be activated in response to wounding and/or whether tC4H activity be modulated by the application of exogenous p-coumarate. At the specified length of times, the wounded and treated roots were either frozen in liquid nitrogen or used immediately to extract microsomal proteins. The microsomal proteins were subjected to immunoblot analysis using polyclonal antibodies against CYP73 of tC4H gene. In this study, tC4H was induced in wounded roots sealed in bags within 6 hours, and in low level({TEX}$10^{-8}${/TEX}M) of p-coumarate solution within 24 hours, whereas the olution without p-coumarate and high levels of p-coumarate solution repressed tC4H induction in wounded roots. These results indicate that tC4H is induced by wounding in the root of avocado, and is inhibited by the application of exogenous p-coumarate.

Effect of low intensity pulsed ultrasound in activating the mitogen-activated protein kinase signaling pathway and inhibition inflammation cytokine synthesis in chondrocytes

  • Objective: Low intensity pulsed ultrasound (LIPUS) has been shown to accelerate cell proliferation and tissue healing in both animal models and clinical trials. However, details of the clinical effects of LIPUS have not been well characterized. The aim of this study was to investigate the effect of LIPUS on mitogen-activated protein kinase (MAPK) activation in rat articular chondrocytes. Design: Cross-sectional study. Methods: Chondrocyte were cultured in six well cell culture plates for 72 hours at $37^{\circ}C$ with 5% $CO_2$, and then exposed to LIPUS at 1.5 MHz frequency and $30-mW/cm^2$ power. Changes in chondrocyte activities were evaluated in response to oxydative stress in dose-dependent (0 and 300 uM) and time-dependent (0-24 hr) manner. The cell viability were analyzed using MTT [3-(4.5-dimethylthiazol-2-yl)-2.5 diphenyltetrazolium bromide]. The expression of p38 MAPK was measured using western blotting. Results: Oxidative stress was induced in rat chondrocytes using hydrogen peroxide ($H_2O_2$). The cell viability was decreased in chondrocytes after the $H_2O_2$ dose and time-dependent treatment. The p38 MAPK phosphorylation occurred at a significantly increased rate after $H_2O_2$ treated (p<0.05). Expression of p38 MAPK was decreased in the p38 inhibitor groups compared with the oxidative stress-induced chondrocyte damage via the p38 MAPK signaling pathways (p<0.05). Conclusions: It could be concluded that LIPUS can inhibit oxidative stress-induced chondrocyte damage via the p38 MAPK signaling pathways.

유드라짓과 알긴산 나트륨 매트릭스를 이용한 4-Aminopyridine의 서방성 제제설계 (Formulation Design of Sustained-Release Matrix Tablets Containing 4-Aminopyridine)

  • 4-Aminopyridine (AP) is a potassium channel blocker used in the treatment of neurological disorders such as multiple sclerosis and Alzheimer disease. AP‘s window of therapeutic effect appears to correlate with its plasma halflife (3.5 hours). It demonstrates pH-dependent solubility because of a weakly basic drug. In addition, the resulting release from conventional matrix tablets decreases with increasing pH-milieu of the gastrointestinal tract. The aim of this study is to design sustained release matrix tablet containing AP, overcoming this problem. $Eudragit^{\circledR}$ L 100 (EuL) and sodium alginate were used in an effort to achieve pH independent drug release. The effect of sodium alginate and EuL on drug release from matrix tablet was investigated. The drug release behavior from the different tablets was analyzed by $t_{20%},\;t_{40%},\;t_{60%}$, The exponential diffusion coefficient n, kinetic constant K were calculated according to the Korsmeyer-Peppas equation. The drug release from matrix tablets prepared with sodium alginate was decreased with increasing the content of sodium alginate in pH 7.4 while there is no significant difference in pH 1.2. The exponent n values were determined to be approximately 0.5 and 0.8 respectively, in both pH 1.2 and 7.4. These values indicate diffusion-based anomalous mechanism and erosion-based anomalous mechanism, respectively. The drug release from sodium alginate matrix tablets prepared with solid dispersion of EuL containing drug showed a slow drug release in an acidic medium and a more fast drug release in phosphate medium, compared with sodium alginate matrix tablets prepared with physical mixture. These results may be attributed to the gel forming ability of sodium alginate and pH dependent solubility of EuL. Therefore, sustained-release AP matrix tablets using sodium alginate and EuL were successfully prepared.

Structure-Activity Relationships of Dimethylsphingosine (DMS) Derivatives and their Effects on Intracellular pH and $Ca^{2+}$ in the U937 Monocyte Cell Line

  • We recently reported that dimethylsphingosine (DMS), a metabolite of sphingolipids, increased intracellular pH and $Ca^{2+}$ concentration in U937 human monocytes. In the present study, we found that dimethylphytosphingosine (DMPH) induced the above responses more robustly than DMS. However, phytosphingosine, monomethylphytosphingosine or trimethylsphingosine showed little or no activity. Synthetic C3 deoxy analogues of sphingosine did show similar activities, with the C16 analogue more so than C18. The following structure-activity relationships were observed between DMS derivatives and the intracellular pH and $Ca^{2+}$ concentrations in U937 monocytes; 1) dimethyl modification is important for the DMS-induced increase of intracellular pH and $Ca^{2+}$, 2) the addition of an OH group on C4 enhances both activities, 3) the deletion of the OH group on C3 has a negligible effect on the activities, and 4) C16 appears to be more effective than C18. We also found that W-7, a calmodulin inhibitor, blocked the DMS-induced pH increase, whereas, KN-62, ML9, and MMPX, specific inhibitors for calmodulin-dependent kinase II, myosin light chain kinase, and $Ca^{2+}$-calmodulin-dependent phosphodiesterase, respectively, did not affect DMS-induced increases of pH in the U937 monocytes.

천연염료자원 탐색 및 염색특성(I) - 거북꼬리(Boehmeria tricuspis Makino)추출물의 pH에 따른 염색특성 - (The Search and Dyeing Properties of Natural Dyes Resources(I) - The Dyeing Properties of Boehmeria tricuspis Makino Extracts by pH -)

  • 미이용 천연염료자원에 대한 탐색연구의 일환으로 거북꼬리(Boehmeria tricuspis Makino)를 선발하고 열수 및 알칼리 조건에서 추출한 추출물을 사용하여 pH 조건에 따른 염착량 및 색상, 명도, 채도의 변화에 대하여 조사하였다. 면, 한지, 실크에 대한 열수 및 알칼리추출물 처리 염색물의 최대흡수파장 (${\lambda}\;max$)은 400 nm였으며, pH 조건에 따른 염착량(K/S)은 실크의 알칼리추출물 처리 염색물을 제외하고 전체적으로 산성조건으로 갈수록 증가하는 경향을 보였으나 pH 7 전후에서 큰 변화를 나타내지는 않았다. Munsell의 색상은 주로 알칼리추출물 처리 염색물은 YR 계열의 색상을, 열수추출물 처리 염색물은 R 계열의 색상을 나타냈다. 명도는 알칼리 조건으로 갈수록 증가하고, 채도는 감소하는 경향을 보였으나 pH 7을 전후하여 현저한 변화를 나타내지는 않았다. 따라서 거북꼬리 추출물로 염색할 경우 염색액의 최적 pH 조건은 pH 7로 조정하여 사용하는 것이 바람직할 것으로 사료되었다.
