A Study on Changes in Antibacterial Activity of Pepsin-hydrolyzed Bovine Apo-lactoferrin at Various Method for Pasteurizations and pH Values (살균방법 및 pH 조건에 따른 Pepsin-hydrolyzed Bovine Apo-lactoferrin의 항균성 변화에 관한 연구)

  • This study was carried out to examine that pepsin-hydrolyzed bovine lactoferrin has applicabilities which are market milk and dairy products. The stability of pepsin-hydrolyzed bovine apo-lactoferrin and the change of its antibacterial character has been studied under various method for pasteurization (LTLT; 65$^{\circ}C$ / 30min., HTST ; 75$^{\circ}C$ / 15sec., UHT ; 135$^{\circ}C$ / 3sec.) and pH Values (pH 2.0, pH 4.0, pH 6.8). The ehated samples were assayed for minimal bacteriocidal concentrations (MBCs) and bacteriocidal effect against E. coli. The results obtained were summarized as follows: After fractionation of pepsin-hydrolyzed bovine lactofeerin by gel filtration. several peptide fractions were found that had strong antibacterial activity. SDS-PAGE showed that the one of these fractions with strong antibacterial activity, which had a molecular mass a range of 30∼33KDa. The MBCs for pepsin-hydrolyzed bovine lactoferrin fraction No. 2 against E. coli required to cause complete inhibition of growth varied within the range of 200∼400 $\mu\textrm{g}$/ml, depending on heat treatments and pH conditions. The peptide fraction No. 2 showed strong bacteriocidal activity against E. coli at LTLT and HTST treatments under acidic pH conditions. and was reduced activity at UHT treatment under pH 6.8 condition.

Effects of Ionizable Groups on the Redox Potentials of Cytochrome c₃from D. vulgaris Miyazaki F

  • The p2H dependence of the NMR chemical shifts of the proton signals of heme methyl groups and ionizable groups in the vicinity of the heme were investigated. The p2H titration of heme methyl signals in four macroscopic oxidation states by saturation tranfer method was performed in the range between p2H 5.2 and 9.0. While the p2H dependence of the heme methyl resonance in fully oxidized state was small, most resonances in the intermediate oxidation states showed certain shifts. Particularly, methyl resonances of heme 1 (sequential heme numbering) exhibited sharp p2H dependence in acidic range. β-CH2 of the propionate of hemes 1 and 4 were titrated in the range of p2H 4.5-9.0. Only the 6-propionate group of heme 1 was protonated in this p2H range and its titration curve was similar to those of methyl resonances of heme 1 in intermediate oxidation states. Analysis of the microscopic redox potentials showed that they change depending on p2H. The ionizable groups responsible for the p2H dependence of these potentials are 6-propionate of heme 1 in acidic range and His 67 in basic range.

Estimation of Chinese Cabbage Growth by RapidEye Imagery and Field Investigation Data

  • Chinese cabbage is one of the most important vegetables in Korea and a target crop for market stabilization as well. Remote sensing has long been used as a tool to extract plant growth, cultivated area and yield information for many crops, but little research has been conducted on Chinese cabbage. This study refers to the derivation of simple Chinese cabbage growth prediction equation by using RapidEye derived vegetation index. Daesan-myeon area in Gochang-gun, Jeollabuk-do, Korea is one of main producing district of Chinese cabbage. RapidEye multi-spectral imagery was taken on the Daesan-myeon five times from early September to late October during the Chinese cabbage growing season. Meanwhile, field reflectance spectra and five plant growth parameters, including plant height (P.H.), plant diameter (P.D.), leaf height (L.H.), leaf length (L.L.) and leaf number (L.N.), were measured for about 20 plants (ten plants per plot) for each ground survey. The normalized difference vegetation index (NDVI) for each of the 20 plants was measured using an active plant growth sensor (Crop $Circle^{TM}$) at the same time. The results of correlation analysis between the vegetation indices and Chinese cabbage growth data showed that NDVI was the most suited for monitoring the L.H. (r=0.958~0.978), L.L. (r=0.950~0.971), P.H. (r=0.887~0.982), P.D. (r=0.855~0.932) and L.N. (r=0.718~0.968). Retrieval equations were developed for estimating Chinese cabbage growth parameters using NDVI. These results obtained using the NDVI is effective provided a basis for establishing retrieval algorithm for the biophysical properties of Chinese cabbage. These results will also be useful in determining the RapidEye multi-spectral imagery necessary to estimate parameters of Chinese cabbage.

Changes of multi-purpose solutions for soft contact lens depending on using period or keeping temperature (사용기간 및 온도에 따른 소프트콘택트렌즈 다목적용액의 변화)

  • To investigate changes of multi-purpose solutions for soft contact lens(MPS) depending on using period or keeping temperature, we evaluate four brands of MPS. No significant difference was seen in protein deposit removing efficacy after samples had used for 24 weeks and kept at $4^{\circ}C$, $20^{\circ}C$ or $30^{\circ}C$. The pH values of the samples of 4 brands measured weekly over the 24 week testing period. The initial average pH value of samples were 7.0, 7.5, 7.6 or 8.2. One brand of MPS was in the range of the threshold for ocular awareness, which is outside the zone of 6.6 ~ 7.8. During the testing period, the pH value were decreased in using period-dependent manner. At the 24th week, the average pH values of samples turned to 6.6, 7.2, 7.2 or 7.7. However, the difference of keeping temperature was not associated with decreased levels of pH values. After 24 weeks, one of total 36 samples was contaminated by bacteria. Furthermore, the change of components was shown after 24 weeks in the analysis using thin layer chromatography and the analysis of UV absorption pattern. The results of our study provides that the keeping temperature of MPS is not the important factor of changes of MPS, but the using period of MPS can cause contact lens wearers discomfort.

The Binding of Food Dyes with Human Serum Albumin

  • The binding interactions between human serum albumin (HSA) and the edible food dyes amaranth, tartrazine and sunset yellow have been studied. Intrinsic association constants and the free energy changes associated with dye-protein binding at physiological pH for amaranth and tartrazine, and at two different pH values for sunset yellow have been calculated from ultrafiltration data. The temperature dependence $(20-40^{\circ}C)$ of the intrinsic association constants at pH 7.4 for amaranth-HSA and tartrazine-HSA mixtures have been measured, from which a plot of the van't Hoff isochore exhibits a marked change in slope around $30^{\circ}C$ indicating a possible change in protein conformation. The number of dye binding sites on HSA is reported for all the above conditions. HSA-ligand binding enthalpies have been used in conjunction with the N-B transitional binding enthalpy for HSA, to calculate the enthalpy for the N-B transition when ligands are bound with the protein.

Characteristics of the Turbidity Change of Clay Particulate Matter according to Its Surface Electrokinetic Behavior (점토성 광물입자의 표면 전기적 거동에 따른 탁도 변화 특성)

  • Montmorillonite is one of representative inorganic clay particles. As the characteristics of clay particulate matter in aqueous environment determine the efficiencies of wastewater treatment and some industrial operations, it is essential to understand its aquatic behavior in relation with turbidity. The change of electrokinetic potential of montmorillonite suspension shows that it tends to negatively increase as the pH of suspension increases. In addition, it is observed that its potential is around 0mV when the solution pH is ca. 5. The turbidity of suspension is shown to be very low when pH is lower than its isoelectric point. However, the turbidity gradually enhances according to beyond isoelectric point. These results reveal that the correlation between electrokinetic potential and turbidity for clay mineral suspension is peculiar which should be fundamentally considered for systematic treatment of wastewater.

  • A modified polyol process is developed to enhance Pt loading during the preparation of Pt/C catalysts. With the help of the zeta potential, the effect of pH on the electrostatic forces between the support and the Pt colloid is investigated. It is shown experimentally that the surface charge on the carbon support becomes more electropositive when the solution pH is changed from alkaline to acidic. However, this change does not affect the electronegative surface charge of Pt colloids already attained and stabilized by glycolate anions. This new behavior caused by the change in the solution pH accounts for the enhanced yield of the process and does not affect the Pt particle size. All our experimental results reveal that this simple modification is a cost effective method for the synthesis of highly Pt loaded Pt/C catalysts for fuel cells.

Radiation Sensitivity of Some Food Decay Fungi (몇몇 음식 부식 균류의 방사선 감수성에 대한 연구)

  • Five species of food decay fungl, Aspergillus flavus, Asp. uiger, Penicillum sp., Botrytis cinerea, and Rhizopus stolonifer, were examined for their radio sensitivity in several suspension media. Asp. flavus, Asp. niger and Penicillum sp. have almost the same sensitivity toward gamma rays. with D value in the range of 30 to 35 K rad, whereas Botrytis cinerea has a D value of approximately 55K rad and Rhizopus stolonifer, the most re4sistant fungus studied, has a crease in their radioresistnace when compared with spores irradiated in water. Asp. flavus and penicillium sp. spores irradiated in citrate buffer at pH3-7 showed almost no change in their radisensitiity with pH, but Botryis cinerea spores showed a distinct decrease in their radioresistnace at pH 6 and 7. Penicillum sp. spores irradiated in sucrose solutions showed no sinificant change in their radioresistance. Botrytis cinerea spores displayed a higher radioresistance when they were irradiated in sucrose solution than in water.

Variations of SCN Space in Metabolic Acidosis and Alkalosis in Rabbits (대사성 산증 및 알칼리증에 있어서 SCN 공간의 변화)

  • Thiocyanate space was determined in 23 bilaterally nephrectomized rabbits in acute metabolic acidosis and alkalosis. Acid-base disturbances were induced by the infusion of 0.3 N HCI or 0.3 N NaOH solution intravenously with the rate of 1 ml/min for 40 to 60 minutes. The blood pressure was monitored throughout the experiment and no changes in blood pressure was confirmed. The following results were obtained. 1. In the saline infused control rabbits, PH was 7.385 with negligible change in pH after the infusion, SCN space was 23.6% of body weight. 2. In the metabolic acidosis group, pH dropped from 7.417 to 7 130 and SCN space was 22.8% of body weight and suggested a negligible change in the extracellular space volume. 3. In the metabolic alkalosis group, pH increased from 7.393 to 7.478 and SCN space was 25.7% of body weight which confirmed a significant increase in the extracellular space volume.

Analysis of Pipe Wall-thinning Caused by Water Chemistry Change in Secondary System of Nuclear Power Plant (원전 2차계통의 수화학 변화가 배관감육에 미치는 영향 분석)

  • Pipe wall-thinning by flow-accelerated corrosion (FAC) is a significant and costly damage of secondary system piping in nuclear power plants (NPPs). All NPPs have their management programs to ensure pipe integrity from wall-thinning. This study analyzed the pipe wall-thinning caused by changing the amine, which is used for adjusting the water chemistry in the secondary system of NPPs. The pH change was analyzed according to the addition of amine. Then, the wear rate calculated in two different amines was compared at the steam cycle in NPPs. As a result, increasing the pH at operating temperature (Hot pH) can reduce the rate of FAC damage significantly. Wall-thinning is affected by amine characteristics depending on temperature and quality of water.
