Effect of Prophylactic Supplementation of Vitamin E and Se on Antioxidant Enzymes during Endotoxic Shock in Buffalo Calves

  • This study was conducted to determine the effects of prophylactic supplementation of vitamin E and Se on oxidative damage and antioxidant status. Fifteen healthy male buffalo (Bubalus bubalis) calves between the age of 6 to12 months were divided into three groups of five animals each: Group I-control, group II-endotoxic shock group infused with lyophilized E coli endotoxin @ 5 ${\mu}g$/kg body wt, and group III-supplemented with vitamin E @ 250 mg and Se @ 7.5 mg, one month prior to induction of endotoxic shock. All the animals in group II and group III exhibited signs of endotoxic shock. When the endotoxic shock was induced, there was significant (p<0.05) increase in the circulating levels of malonyl dialdehyde MDA (an indicator of lipid peroxidation). In the supplemented group III the magnitude of formation of MDA was also less as compared to group II at every stage of study. There was significant (p<0.05) decrease in circulating levels of SOD, GSH-Px, Catalase and G-6-PD activity from the normal (0 h) value with passage of time. As a result of endotoxic shock, these values reached a lowest value, and then showed a tendency towards the 0 h value. Prophylactic supplementation with vitamin E and Se was successful in reducing the quantum of oxidative damage due to formation of free radicals because of endotoxic shock.

  • This research sought to compare chlorine, chlorine dioxide and ozone as chemical disinfectants of drinking water, with inactivation of total coliform as the indicator. The inactivation of total coliform was tested against several variables, including the dose of disinfectant, contact time, pH, temperature and DOC. A series of batch processes were performed on water samples taken from the outlet of a settling basin in a conventional surface water treatment system that is provided with the raw water drawn from the mid-stream of the Han River. Injection of 1 mg/L of chlorine, chlorine dioxide and ozone resulted in nearly 2.4, 3.0 and 3.9 log inactivation, respectively, of total coliform at 5 min. To achieve 99.9 % the inactivation, the disinfectants were required in concentrations of 1.70, 1.00 and 0.60 mg/L for chlorine, chlorine dioxide and ozone, respectively. Bactericidal effects generally decreased as pH increased in the range of pH 6 to 9. The influence of pH change on the killing effect of chlorine dioxide was not strong, but that on ozone and free chlorine was sensitive. The activation energies of chlorine, chlorine dioxide and ozone were 36,053, 29,822 and 24,906 J/mol for coliforms with inactivation effects being shown in the lowest orders of these.

Sensitivity of Color Indicators to Fermentation Products of Kimchi at Various Temperatures (김치 발효산물에 대한 발색지시계의 온도별 민감성)

  • Application of the color indicators to kimchi packages was investigated in order to monitor the ripeness of commercial kimchi products during storage and distribution. Kimchi was packed in polypropylene (PP) tray and nylon/cast polypropylene (Ny/CPP) lid where the indicating sachet consisting of $CO_2$ absorbent and chemical dye (bromocresol purple and methyl red) was attached. The ripeness of kimchi during storage at $0{\sim}20^{\circ}C$ was measured in terms of pH and titratable acidity (TA), which were compared with Hunter color values of the indicators. The color of bromocresol purple dye turned from light blue to purple, while that of methyl red turned from light yellow to red. Regardless of the storage temperatures, Hunter b values of bromocresol purple type and Hunter a values of methyl red type appeared to be proportional to both the pH and TA values of kimchi. These results suggest that the color indicators be employed as one of the effective techniques for sensing the ripeness of packaged kimchi products without destructing the package.

  • The response properties of continuous automated system using an enzyme reactor for determination of urea were investigated. The enzyme reactor was constructed to packed-bed form which filled with nylon-6 beads (42∼48 mesh), which immobilized urease with glutaraldehyde, in teflon tube (2 mm I.D., 20 cm length). The system was composed of the enzyme reactor, gas dialyzer, and tublar PVC-nonactin membrane ammonium ion-selective electrode as an indicator electrode in serial order. The response characteristics of this system were as follows. That is, the concentration range of linear response, slope of linear response, detection limit, and conversion percentage were $5.5{\times}10^{-6}$$2.4{\times}10^{-3}M$, 57.8 mV/decade, $1.5{\times}10^{-6}$, and 80.8%, respectively. The optimum buffer and life time of urease reactor were 0.01M Tris-HCl buffer solution (pH 7.0∼7.8) and 0.01M phosphate buffer solution (pH 6.9∼7.5) and about 150 days, respectively. And the urease reactor had no interferences of the other physiological materials.

Evaluation of Corrosion Effects on Permanent Ground Anchors (영구 지반앵커에 대한 부식의 영향 평가)

  • The corrosion rate measurement procedure for the permanent ground anchors using polarization resistance measurements and electrochemical impedance spectroscopy is presented in this paper. The polarization resistance measurements were used to determine the correlation between corrosion rate in the steel and soil characteristics. The electrochemical impedance spectroscopy was used to predict the time dependent corrosion reaction and evaluate the different type of coating systems and the effect of cement grouting on the corrosion attack under various conditions. The results indicate that a low pH soil is a good indicator of a corrosive soil. The low pH soil condition (<5) in both clay and sand has a significant effect on the corrosion reaction of steel members in permanent found anchors. In the case of neutral and alkaline conditions beyond pH 6 in clay and sand, no consistent acceleration of corrosion was measured and the corrosion rate was constant regardless of variations of soil pH levels. Laboratory test data for porcelain clay indicate that the change of soil pH level has a small influence on the corrosion reaction in the steel member. The use of cement footing in the bonded length is sufficient to decrease the corrosion rate to a level close to 0.003∼0.01mm/y at the end of the given period. With epoxy and fusion bonded epoxy coating, the steel specimens remained unaffected and retained the original condition. It is suggested that epoxy and fusion bonded epoxy coating can provide effective protection against corrosion for a long time even in aggressive environment.

The Study of Donor-Acceptor Chromophores and Diketopyrrolopyrrole(DPP) Analogues (Donor-Acceptor 발색단과 디케토피롤로피롤(DPP) 유도체에 관한 연구)

  • The diketopyrrolopyrrole (DPP) pigment is a bicyclic 8-π-electron system containing two lactam units. Typical DPP derivative pigments have melting points of over 350°C and very low solubility in most solvents, and show absorption in the visible region with a molar extinction coefficient of 33,000 dm2mol−1 and strong photoluminescence with maxima in the range 500–600 nm. X-ray structure analyses of DPP show that the whole molecule is almost in one plane. The phenyl rings are twisted out of the heterocyclic plane and the intermolecular hydrogen bonding between neighboring lactam NH and carbonyl units influences the structure of the DPP pigment in the solid state. In this study, mono-N-alkylation and mono-N-arylation were undertaken for Pigment Red 264 or Pigment Orange 73 with alkyl halide and aryl halide, respectively, in the presence of sodium tert-butoxide as a base catalyst to improve the solubility of DPP pigments and their application as CO2 indicators. The synthetic yield was in the range 11–88%. The indicator dyes are highly soluble in organic solvents and shows pH-dependent absorption (λmax 501 and 572 nm for the protonated and deprotonated forms, respectively) and emission (λmax 524 and 605 nm for the protonated and deprotonated forms, respectively) spectra. The mono-N-alkylated and mono-N-arylated DPP pigment was identified by 1H-NMR (1H-Nuclear Magnetic Resonance Spectrometer), FT-IR (Fourier Transform Infrared Spectroscopy), and MS (Mass Spectrometry). According to the results of color and hue properties obtained by a color matching analyzer, the synthesized DPP pigment material can be used as a CO2 indicator.

Effect of Nutrient Solution Strength on pH of Drainage Solution and Root Activity of Strawberry 'Sulhyang' in Hydroponics (배양액의 농도가 배액의 pH와 딸기 '설향' 뿌리의 활성에 미치는 영향)

  • Experiments were conducted to investigate the optimum concentration of the nutrient solution in strawberry 'Sulhyang' with hydroponics in relationship between root activity and nutrient concentrations. Nutrient solutions for strawberry, made by Yamazaki, were supplied EC 0.5, 1.0, 2.0 $dS{\cdot}m^{-1}$ during experiment period. Growth of shoot and root of strawberries grown in visible plastic pot was observed during experiment. Petiole length was longest in plants grown in EC 1.0 $dS{\cdot}m^{-1}$, followed by 2.0 and 0.5 $dS{\cdot}m^{-1}$. Leaf width was longest in plants grown in EC 1.0 $dS{\cdot}m^{-1}$, followed by 0.5 and 2.0 $dS{\cdot}m^{-1}$. Fruit length, fruit diameter, fruit weight and yield were higher in EC 0.5 and 1.0 $dS{\cdot}m^{-1}$ than 2.0 $dS{\cdot}m^{-1}$ treatment but, soluble solids of the fruit did not show statistical differences among treatments. Shoot dry weight was heaviest in EC 1.0 $dS{\cdot}m^{-1}$, followed by 0.5 and 2.0 $dS{\cdot}m^{-1}$. Root dry weight was heavier in EC 0.5 and 1.0 $dS{\cdot}m^{-1}$ but significantly light in 2.0 $dS{\cdot}m^{-1}$. pH of the drainage solution was elevated in low nutrient concentration and lowered in high concentration. Also root activity was high in low nutrient concentration and low in high concentration. As a result, the optimum EC for strawberry 'Sulhyang' was EC 1.0 $dS{\cdot}m^{-1}$ in this experiment. It was confirmed that there was high relationship between root activity and pH of drainage solution. This result will be utilized as an indicator for strawberry hydroponics.

Cultural Conditions of Lactobacillus sp. GM7311 for the Production of Bacteriocin (Lactobacillus sp. GM7311에 의한 박테리오신의 생산 조건)

  • A lactic acid bacteria which showed antimicrobial activity was isolated from dairy products and identified as Lactobacillus sp. according to the morphological, physiological and biochemical properties, which was named Lactobacillus sp. SH 7311. The bacteriocin of Lactobacillus sp. GM 7311 showed a broad range of inhibitory spectrum against some gram positive and negative bacteria. Especially, Proteus mirabilis was highly sensitive to bacteriocin and used as indicator strain for further investigation. The optimal condition for the production of bacteriocin was showed on MRS broth at $37^{\circ}C$ and pM 6.0. Bacteriocin production of this strain cultured under optimal condition was increased late logarithmic phase to early stationary phase. This bacteriocin was fully active at the pH range $2.0\~5.0$, also was stable at $100^{\circ}C$ for 60 min. at pH 5.0, But about $40\%$ of bacteriocin activity was diminished by the treatment of acetone, ethanol, iso-butanol and ethyl ether during 2 hours at $4^{\circ}C$.

Characterization of Bacteriocin produced Lactobacillus bulgaricus acting on bovine mastitis pathogens

  • The antimicrobial substance produced by Lactobacillus bulgaricus was inactivated by pretense, which confirmed it as a bacteriocin and referred to 'bulgaricin HJ'. The bulgaricin HJ showed the inhibitory activity against mastitis pathogens, gram-positive and gram-negative bacteria. The optimal conditions for the production of bulgaricin HJ were at the temperature of $30^{\circ}C$ and 10 h after cultivation of L. bulgaricus. Staph. and Strep. agalactiae, common bovine mastitis pathogens, were treated with bulgaricin HJ by the agar well diffusion method and showed antimirobial activities to the bovine mastitis pathogens. The activity of the bulgaricin HJ was maintained at pH 6-7 and $100^{\circ}C$ for 60 min against the mastitis pathogens. The bulgaricin HJ was determined as class IV bacteriocin by various enzyme treatments. Colony forming units analysis with indicator strains by the treatments of bulgaricin HJ indicates that the mode of bacteriocin action was bactericidal rather than bacteriostatic.

Determination of Salicylate by Selective Poly(vinylchloride) Membrane Electrode Based on N,N'-1,4-Butylene Bis(3-methyl salicylidene iminato) Copper(II)

  • A new salicylate-selective electrode based on the complex N,N'-1,4-butylene bis(3-methyl salicylidene iminato) copper(II) as the membrane carrier was developed. The electrode exhibits a good Nernstian slope of 59.1 ${\pm}$ 1.0 mV/decade and a linear range of $1.0\;{\times}\;10^{-6}$-1.0 M for salicylate. The limit of detection was $5.0\;{\times}\;10^{-7}$ M. It has a fast response time 10 s and can be used for more than three months. The selective coefficients were determined by the fixed interference method (FIM) and could be used in the pH range 4.5-10.5. It was employed as an indicator electrode for direct determination of salicylate in pharmaceutical samples.
