Cloning and Base Sequence Determination of Replication Initiation Gene (rep) Isolated from Staphylococcus aureus DH1 R-plasmid pSBK203 (Staphylococcus aureus DH1에서 분리된 R-plasmid pSBK203의 복제 개시 유전자(rep) 분리 및 염기서열 결정)

  • A replication initiation gene was identified and its nucleotide sequence has been determined from a 3.8 kb, chloramphenicol acethyltransferase conferring R-plasmid pSBK203 of Staphylococcus aures. Location of the replication related region of pSBK 203 was determined by interuption with pUC 119 at XBaI and MspI sites which resulted in inactivation of replication in Bacilius subtilis. Base sequence of this region revealed on open reading frame of 942 base pairs, which encoded a 314 amino acid protein. Base sequence homology with other rep of pT181 family plasmids such as pT181, pC221, pC223, pS194, pU112, and pCW7 was ranged from 78% to 97% and the predicted amino acid sequence homology was from 72% to 95%.

Changes of Body Composition and Aging-related Hormones by Rhythmic Exercise Program in Elderly Women (율동적 운동에 따른 여성 노인의 체구성 및 노화관련 호르몬 변화)

  • The purpose of this study was to examine the changes of body composition and aging-related hormones by rhythmic exercise in elderly women. The subjects in this study were 20 olders living in a rural community, >60 years of age and were performed rhythmic exercise program during 12 weeks. The measurement of the body composition, body mass (t=3.125, p=.006), body mass index (t=3.225, p=0.004), body fat mass (t=3.782, p=0.001), waist-to-hip ratio (t=3.867, p=0.001) were significant difference after exercise program, but had no effect on lean body mass. Also, growth hormone (t=-2.221, p=0.039) and somatomedin-C (t=-9.186, p=0.000) were significantly increased after rhythmic exercise, however, rhythmic exercise had no effect on DHEA-S. These results suggest that rhythmic exercise appeared to effect of control on body composition and some aging-related hormones in elderly women.

Novel Compound Heterozygote Mutations of the SLC25A13 Gene in an Infant with Neonatal-onset Type II Citrullinemia Detected by Newborn Mass Screening

  • Citrin deficiency caused by the SLC25A13 gene mutations is associated with both neonatal-onset type II citrullinemia (CTLN2), also known as neonatal intrahepatic cholestasis caused by citrin deficiency and adult-onset CTLN2. Neonatal-onset CTLN2 is an autosomal recessive disorder characterized by poor growth, intrahepatic cholestasis, and increased serum citrulline. A 16-days old infant with hyperammonemia was referred for evaluation of increased plasma citrulline diagnosed using tandem mass spectrometry. Blood amino acid analysis showed significant elevation of citrulline. Mild elevation in serum galactose levels had been found. DNA analysis of the SLC25A13 gene in this patient showed two novel compound heterozygous mutations, c.221C>T in exon4 and c.1645C in exon16 (p.[Ser74Phe]+[Gln549X]). We suggest that infants with a high serum citrulline level on a tandem mass screening test are candidates for gene analysis and blood amino acid analysis for neonatal-onset CTLN2.

  • Cultural conditions for $\alpha$-D-Glucosidase Inhibitor production from Streptomyces sp. YS-221-B isolated from soil arid identified as Streptomyces flauovirens or a subspecies of it were investigated. When the strain was cultured in a flask containing 2% glucose, 0.3% asparagine, 0.0002% riboflavin, 0.05% $K_2$HPO$_4$, 0.1% MgSO$_4$.7$H_2O$, 0.05% NaCl, pH 8.0 at 3$0^{\circ}C$, maximum production of the inhibitor was obtained after 8-9 days of cultivation. Sugar alcohols such as mannitol, i-inositol, erythritol as carbon sources, asparagine and beef extract as nitrogen sources were favorable for inhibitor production. Among vitamins, riboflavin, p-aminobenzoic acid, pyridoxamine and folic acid promoted the production of inhibitor, but depressed by the addition of hesperidine, and also depressed by cobalt, lithium and ferrous salts.

A Cold-Adapted Carbohydrate Esterase from the Oil-Degrading Marine Bacterium Microbulbifer thermotolerans DAU221: Gene Cloning, Purification, and Characterization

  • A cold-adapted carbohydrate esterase, CEST, belonging to the carbohydrate esterase family 6, was cloned from Microbulbifer thermotolerans DAU221. CEST was composed of 307 amino acids with the first 22 serving as a secretion signal peptide. The calculated molecular mass and isoelectric point of the mature enzyme were 31,244 Da and pH 5.89, respectively. The catalytic triad consisted of residues Ser37, Glu192, and His281 in the conserved regions: GQSNMXG, QGEX(D/N), and DXXH. The three-dimensional structure of CEST revealed that CEST belongs to the ${\alpha}/{\beta}$-class of protein consisted of a central six-stranded ${\beta}$-sheet flanked by eight ${\alpha}$-helices. The recombinant CEST was purified by His-tag affinity chromatography and the characterization showed its optimal temperature and pH were $15^{\circ}C$ and 8.0, respectively. Specifically, CEST maintained up to 70% of its enzyme activity when preincubated at $50^{\circ}C$ or $60^{\circ}C$ for 6 h, and 89% of its enzyme activity when preincubated at $70^{\circ}C$ for 1 h. The results suggest CEST belongs to group 3 of the cold-adapted enzymes. The enzyme activity was increased by $Na^+$ and $Mg^{2+}$ ions but was strongly inhibited by $Cu^+$ and $Hg^{2+}$ ions, at all ion concentrations. Using p-nitrophenyl acetate as a substrate, the enzyme had a $K_m$ of 0.278 mM and a $k_{cat}$ of $1.9s^{-1}$. Site-directed mutagenesis indicated that the catalytic triad (Ser37, Glu192, and His281) and Asp278 were essential for the enzyme activity.

Optimization of Environmental Parameters for Extracellular Chitinase Production by Trichoderma harzianum SJG-99721 in Bioreactor (Trichoderma harzianum SJG-99721의 체외 분비 chitinase 생산에 미치는 생물 반응기에서의 반응 최적화 연구)

  • A self-directing optimization procedure was applied to determine the best environmental factors in operating the bioreactor. The self-directing optimization process was employed to determine the best conditional combination of multi parameters, pH, temperature, aeration rate and mixing rate toy maximal production of chitinase by Trichoderma harzianum SJG-99721 in batch mode fermentation. Among these factors, the parameters of pH and aeration rate were found to be particularly important on mycellial growth and chitinase activity. pH 4.89, an aeration rate of 3.22 ι per minute and an agitation rate of 225 rpm was found to be the best combination. By the optimization, chitinase activity was dramatically increased from an initial value of 4.221 U under basic conditions to n final value of 16.825 U.

Biochemical Characterization of Protease Produced by Cordyceps nutans

  • The fruiting body of Cordyceps is derived from the pupa or larva of insects infected by the entomopathogenic fungi Cordyceps. The fruiting body has been used as an anti-cancer and anti-inflammatory ingredient in traditional Chinese medicine. The biochemical characteristics of protease isolated from Cordyceps nutans were investigated in this study. The culturing period for production of protease by C. nutans was 10days. The acidity was pH 7.0, and the temperature was $25^{\circ}C$. The carbon and nitrogen sources for the production of the protease were glucose and yeast extract, respectively. The ratio of C/N was 2% glucose and 0.6% yeast extract. 0.06% $CuSO_4$ was used as the inorganic salt. The investigation into the acidity of the protease produced by C. nutans revealed that the optimal pH and temperature were pH 7.0 and $30^{\circ}C$. The stability of the protease was shown as pH 6.0~9.0 and $30{\sim}50^{\circ}C$. The investigation into the influence of the metal ions on the enzyme activation of C. nutans revealed that it was inhibited in $ZnSO_4$ and activated in $FeSO_4$.

  • The relative retrogradation of Jeungpyun investigated with different millimg methods of rice flour. The relative retrogradation of Jeungpyun was reduced in order that of W-C, W-P, D-M, D-J in DSC(Differential Scanning Calorimetry) method while W-C, D-M, D-J, W-P in Diastase method. In wet milling method, the relative retrogradation of Jeungpyun by Pin mill(W-P) was lower than Jeungpyun by colloid mill(W-C). In dry milling method, the relative retrogration of Jeungpyun by jet mill(D-J) was lower than Jeungpyun by micro mill(D-M). The relative retrogradation of Jeungpyun by DSC method was similar to the Diastase method. The relative retrogradation of Jeungpyun was decreased with decreasing particle size and setback value for amylogram and increasing damaged starch.

Quality Changes in Hot Sauce with Red Pepper Powder and/or Kochujang during Storage (고춧가루 및 고추장을 이용한 핫소스의 저장 중 품질 특성 변화)

  • To develop Korean-style hot sauce using red pepper powder and/or Kochujang, its physiochemical and microbiological characteristics were investigated during storage at 20 and $30^{\circ}C$ for 56 days. The capsanthin contents and the value of surface color decreased during storage, but pH and titratable acidity were not changed. The sensory quality showed a decreased tendency during storage. Among the microorganisms, viable cell count decreased, but yeast, lactic acid bacteria and E. coli were not detected during storage. From the correlationship between sensory color and capsanthin content, the shelf-lives of hot sauce were predicted to be 221.7 days at $20^{\circ}C$, and 85.3 days at $30^{\circ}C$. While they were 230.0 days at $20^{\circ}C$ and 70.0 days at $30^{\circ}C$ in case of using Kochujang, hot sauce using both materials showed shelf-lives of 204.0 days at $20^{\circ}C$, 67.3 days at $30^{\circ}C$, respectively.
