Cloning and Expression of an $\alpha$-Amylase Gene from Bacillus circulans in B. subtilis and B. megaterium (Bacillus circulans $\alpha$-amylase 유전자의 Basillus subtilis와 Bacillus megaterium에서의 클로닝 및 발현)

  • A Baczllus circdans KCTC3004 $\alpha$-amylase gene contained in a recombinant plasmid pAL850 was transferred into a new shuttle vector plasmid pALSIlI by ligating linearlzed DNAs of pUC19 and pUB110. B. subtilis RM125 and B. megatenurn ATCC14945 transfonned with pALS111 produced the $\alpha$-amylase substantially Most of the enzyme was produced during the exponential growth period. The maxiinurn activities of the $\alpha$-amylase produced by the Bucillus transformants were compared with that of the B. circulans gene donor strain. The B. subtilis RM125(pALS111) enzyme showed the actlvicy 95 times higher than that of the gene donor cells, followed by the B, nzegaterium ATCC14945(pALSlll) enzyme with activity 34 limes higher than that of the gene donor cells. While E coli secreted about 10% of the produced enzyme, B. subtilis excreted the enzyme inlo the medium wholly and B. megaterirun about 98% ofthe total product. The plasmid pALSI11 was quite stable inB. nzegaterium (92%), inoderately stable in B. subtilis (76%), but was unstable in E. coli (38%). The SDS-PAGE and zymogram of this enzyme produced in E. coli(pALS111), B. subtilis( pALS111) or B. megateril~m (pALS111) indicated a molecular weight of 55,000. The enzymes overproduced in three different host cells hydrolyzed starch to produce mainly maltoaiose and mallooligosaccharides.

Effects of Artificial Stimulations on the activation of oocyte and the expression of cyclin B1 protein in mouse oocytes (쥐 난자의 활성화 처리가 난자의 활성화 및 Cyclin B1 단백질발현에 미치는 영향)

  • This study was performed to investigate the effects of artificial stimulation on the increase of the oocyte activation, to evaluate the expression of cyclin B1 protein levels in enucleated mouse oocytes, and to investigate correlation between the oocyte activation and the cyclin B1 protein levels. The oocyte activation was induced by 7% ethanol (EtOH) or 10μg/ml Ca-ionophore with or without 10μg/ml cycloheximide (CH). The activation rate was significantly higher in both single (p<0.05) and combined (p<0.01) stimulated groups compared to control group. The cyclin B1 protein level was significantly reduced in both stimulated groups (p<0.05), except for EtOH+CH treatment group. The expression of cyclin B1 protein showed a higher negative correlation with activation rate in EtOH+CH (r=0.61, p<0.05) and Ca+CH (r=0.86, p<0.01) stimulation groups, but not in a both single stimulation groups. Taken together, it can be suggested that single (EtOH and Ca- ionophore) and combined (EtOH+CH and Ca+CH) stimulation increases the oocyte activation, especially combined stimulation, because it induces the degradation of cyclin B1 protein after artificial stimulation treatments in mouse oocytes.

  • This study was undertaken to determine thermodynamic characteristics of B. subtilis p-4 and B. licheniformis p-5 proteases isolated from fermented anchovy paste. $K_m$ values of two proteases for casein as a substrate were 0.38mM in p-4 protease and 0.18mM in p-5 protease, respectively. Denaturation constants($K_D$) of p-4 and p-5 proteases were $12.2{\times}10^{-5}/sec\;and\;19.0{\times}10^{-5}/sec\;at\;40^{\circ}C,\;and\;35.7{\times}10^{-5}/sec\;and\;46.3{\times}10^{-5}/sec\;at\;50^{\circ}C$, respectively. Activation energies($E_a$) of p-4 and p-S pmteases were 19.6 Kcal/mole and 15.2kcal/mole, respectively. Free energy of activation(${\Delta}G^*$), activation enthalpy(${\Delta}H^*$) and activation entropy(${\Delta}S^*$) at $40^{\circ}C$ were 23.21Kcal/mole, 18.98Kcal/mole and -13.50 eu, respectively for p-4 protease and 22.93Kcal/mo1e, 14.58Kcal/mole and -26.68 eu, respectively for p-5 protease. The major amino acids in p-4 protease(151 residues of amino acid) were Gly, Glu, Pro, Asp, Ser, Ala, Lys and Leu, while those in p-5 protease(247 residues of amino acid) were Gly, Glu, Asp, Ala and Leu. It may be concluded that heat denaturation of two proteases showed liner regression curve and p-5 protease was more sensitive to heat than p-4 protease.

  • The caregiving experiences of 100 family menbers of outpatients with schizophrenia and schizoaffective disorders were investigated for the presence of positive(positive family-patient relationship. patient' contribution to the family) and negative caregiving experience(objective and subjective burden) and their predictors. This study attempts to make the analysis of caregiving experience more useful by expanding the focus to incoporate these positive aspects of the experience of family caregiver. Objective burden consists of two elements: 'disruption of family life', 'care'(amount of caregiving related to activity of daily living). Subjective burden is defined as emotional reactions to the care giving and it comprised of 6 emotional subdimensions such as 'stigma', 'grief'. 'worry', 'pity', 'fear', 'despair'. Also we investigate the severity of patients' disturbing behaviors into two categories, positive and negative disturbing behaviors and patient' contribution to the family as a predictors of positive and negative caregiving experiences. This study use Pearson's correlation coefficient, Hierardhical regressions in the SAS Program. The results are as follows: 1. Respondents reported moderate level of objective burden 'disruption of family life' (mean = 2.48, range = 1-4), and 'care' (mean=2.54, range = 1-4), and slightly high level of total subjective burden(mean = 2.19, range = 1-4). Mean scores for the measure of the severity of behavioral disturbance indicated that the caregiver experienced negative disturbing behaviors around almost 'somtimes'(mean=2.28, range = 1-4), and positive disturbing behaviors 'almost not frequent'(mean=2.78. range=1-4). So they reported that they perceived patient's negative disturbing behaviors more than positive disturbing behaviors. Mean scores for the measure of the patient' contributions (mean = 1.99. range = 1-4) indicated that caregivers experienced these contributions a little. It means that there should be a positive aspect of possibilities of patient' family roles that can be developed in the daily life. Mean scores for the measure of the positive family-patient relationship indicated that caregivers experienced moderate level of positive family-patient relationship(mean=2.52, range = 1-4). 2. Hierardhical regression analysis 1) Hierardhical regression of 'disruption of family life' showed that the interaction between positive disturbing behaviors and patient' contributions (B = .20. p = .022) and caregiver's educational level(B=.06. p=.000) were 'significant and Hierardhical regression of 'care' showed that 'negative disturbing behaviors'(B= .35. p= .007). 'patient' contributions'(B= .28, p= .019). 'family income'(B=-.l1. p=.096) were significant. 2) Hierardhical regression of 'total subjective burden', 'stigma', 'grief', 'worry', 'pity'. 'fear', 'dispair' showed that 'positive disturbing behaviors'(B=.51. p=.000). 'negative disturbing behaviors' (B = .17, p = .026), 'caregiver's educational level'(B = .03. p=.036), 'family income'(B=.08. p=.041) were significant predictors of 'total subjective burden': 'positive disturbing behaviors'(B=.32. p=.066). 'negative disturbing behaviors'(B=.24, p=.096) 'durations of illness'(B= .03. p= .079) were significant predictors of 'stigma' 'negative disturbing behaviors'(B=.28. p=.005). 'patient sex'(B=-.32. p=.022). 'positive disturbing behaviors'(B=.28. p=.020), 'patient age'(B=.02. p=.010), 'caregiver age'(B=-01, p= .002) were significant predictors of 'grief' 'negative disturbing behaviors'(B= .28, p= .005). 'patient sex'(B= -.32. p=.039), 'caregiver age'(B=-.02, p= .023). 'caregiver's educational level'(B= .04, p = .044) were significant predictors of 'worry' 'patient sex'(B=-.46. p=.005). 'negative disturbing behaviors'(B= .28. p=.018), 'caregiver age'(B=-.01, p=.037) were significant predictors of 'pity' 'positive disturbing behaviors'(B=.83. p=.000). Self-Esteem, Body Image and Factors Influencing on Depression of Infertile Women (난임여성의 자아존중감, 신체상과 우울에 미치는 영향요인)

  • This purpose of this study was to investigate self-esteem, body image and depression in infertile women and to identify factors influencing depression. There were statistically significant difference in self-esteem on variables such as age, education, occupation, pressure of treatment cost. There were statistically significant difference in body image on variables such as period of marriage, education, pressure of treatment cost. There were statistically significant difference in depression on variables such as age, education, family structure, pressure of treatment cost. Self-esteem was positively correlated with body image(r=.688, p=.000). Depression was negatively correlated with self-esteem(r=-.710, p=.000) and body image(r=-.526, p=.000). Factors influencing depression were self-esteem(b=-0.66, p=.000), Living with parents-in-law(b=-7.92, p=.001), and age over 40 years(b=-1.72, p=.033), which explained 63.5%.

  • The aim of this study is to present the basic reference data of age and specipic gait parameters for comparisons of the gait characteristics depended on Unilateral Trans-Femoral or Trans-Tibial Prostheses. The basic gait parameters were extracted from 10 Adult, 10 above knee(A/K) patients and 10 below knee(B/K) patients, 50 to 60 years of age using VICON 512 Motion Analyzer. The results were as follows; 1) The mean Cadence of the above knee(A/K) patients and below knee(B/K) patients were $87.77{\pm}8.64$ steps/min, to $99.84{\pm}11.14$ steps/min.(p<0.05) 2) The mean Walking Speed of the above knee(A/K) patients and below knee(B/K) patients were $0.84{\pm}0.15$ m/s, to $0.96{\pm}0.25$ m/s.(p>0.05) 3) The mean Stride Length of the above knee(A/K) patients and below knee(B/K) patients were $1.14{\pm}0.14$ m, to $1.14{\pm}0.22$m.(p>0.05) 4) The mean maximal angles of joint on the hip flexion motion for different above knee(A/K) patients and below knee(B/K) patients were $34.75{\pm}10.18_{\circ}$, to $32.32{\pm}6.34_{\circ}$ .(p>0.05) 5) The mean maximal angles of joint on the knee flexion motion for different above knee(A/K) patients and below knee(B/K) patients were $66.97{\pm}15.08_{\circ}$, to $52.65{\pm}9.21_{\circ}$ .(p<0.05) 6) The mean maximal angles of joint on the ankle dorsiflexion motion for different above knee(A/K) patients and below knee(B/K) patients were $14.41{\pm}4.82_{\circ}$, to $10.04{\pm}3.49_{\circ}$ .(p>0.05) 7) The mean maximal angles of joint on the ankle plantarflexion motion for different above knee(A/K) patients and below knee(B/K) patients were $5.77{\pm}3.17_{\circ}$, to $2.75{\pm}4.49_{\circ}$ .(p>0.05)

  • We say operators A, B on Hilbert space satisfy Fuglede-Putnam theorem if AX = X B for some X implies $A^*X=XB^*$. We show that if either (1) A is p-hyponormal and $B^*$ is a class y operator or (2) A is a class y operator and $B^*$ is p-hyponormal, then A, B satisfy Fuglede-Putnam theorem.

Burnout, Secondary Traumatic Stress, and Compassion Satisfaction of Military Officers Responding to COVID-19 (COVID-19 대응 군 간부의 소진, 이차적 외상성 스트레스 및 공감만족)

  • Purpose: The purpose of this study was to identify individual and occupational factors influencing burnout, secondary traumatic stress, and compassion satisfaction in military officers who experienced supporting civilians in responding to COVID-19. Methods: This descriptive study was conducted on 140 special forces' military officers who experienced responding to disasters within 3 months. Data were collected through an online survey from September to November 2021. Results: The mean scores for burnout, secondary traumatic stress, and compassion satisfaction were 21.67±6.03, 20.54±8.21, and 39.72±8.12 out of 50, respectively. The significant factors that influenced burnout and secondary traumatic stress were 'higher passive stress copying styles' (B=0.17, p<.001; B=0.31, p=<.001, respectively) and 'lower social support (B=-0.11, p=.031; B=-0.10, p=.001, respectively). The compassion satisfaction was more significantly associated with 'higher self-efficacy' (B=0.37, p=<.001), 'higher active stress copying styles' (B=0.19, p=.006), and 'education responding to disasters' (B=2.04, p=.029). Conclusion: The results suggest that the strategies to increase social support, self-efficacy, and active stress coping styles should be considered in developing educational programs for military officers responding to disasters to minimize burnout, secondary traumatic stress and to improve compassion satisfaction.

The Vegetational History of korea During the Holocene Period (홀로세 중 한국의 식생사)

  • The vegetational and environmental history of Korea during the Holocene period has reconstructed by the use of folral, palaeoclimatogical, archaeological data and written records. From 10,000 to 6,700 yeat B.P. in mid-eastern Korea, deciduous broadieaved trees gradually increased trees, notably at c.8,000 years B.P. From 6,700 to 4,500 years B.P., the vegetation remained dominated by Quercus and pinus(Diploxylon). Vegetational informations suggest the postglacial hypsithermal from c.8,000 to 4,500 years b.p. After 4,500 until 1,400 years B.P., the admixture of cryophilous trees in midland and thermophilous trees in south seems to imply the latitudinal divisions of vegetation and climate. The presence of cultivated rice pollen as well as non-arboreal pollen and spores indicates an increased rate of deforestation. From 1,400 years B.P., there was a sharp decrease in the representation of temperate trees, but the presence of the cryophilous genera Abies and pinus (Haploxylon) is noticea-ble and might be related to the occurrence of Little Ice Age. Fossil pollen diagram from western Korea and eastern Korea since 6,250 and 10,000 yeats B.P., respectively have enabled to compare vegetational dis-tribution patterns and changes in the Holocene period. In western Korea, Alnus- dominated vegetation continued from 6,250 to 1,500 years B.P., but pinus took over Alnus from 1,500 years B.P. In eastern Korea, however, Quercus and pinus dominated from 10,000 to 2,000 years B.P., but pinus be-came predominant from 2,000 years B.P. On a time- spatial basis, different vegetational distribution pattern can be recognised between western and east-ern Korea. The use of seven historical records which is unique to Korea also enabled to reconstruct the distributional pattern and temporal change of vegetation from 1425 to 1928.
