Influence of Media and Hormones on the In Vitro Development of Porcine Follicular Oocytes (배지 및 첨가호르몬이 돼지난포란의 체외발생능에 미치는 영향)

  • This study was conducted to investigate the influence of media and hormones on in vitro maturation and development of porcine follicular oocytes. Basic media were used to TCM-199, Waymouth MB751/1 and BMOC-II, and hormones were used to hCG and FSH in each medium. The results obtained were summarized as follows : 1. The maturation rates of oocytes cultured in TCM-199 medium containing hCG, FSH and hCG+FSH were 78.05, 72.50 and 67.50%, respectively. The maturation rates of oocytes with hormones were significantly (P<0.05) higher than those of oocytes cultured without hormone. However, the cleavage rate(hCG 46.88%, FSH 31.04%. hCG+FSH 37.04%) of embryo cultured in TCM-199 containing hormone was significantly(P<0.05) lower than that(89.47%) of oocytes cultured without hormone. 2. The maturation rates of oocytes cultured in Waymouth MB751/1 medium containing hCG. FSH and hCG+FSH were 69.77, 71.43 and 80.00%, respectively. The maturation rates of oocytes with hormones were significantly(P<0.05) higher than those of oocytes cultured without hormone. However. the cleavage rate(hCG 46.67%. FSH 36.00%, hCG+FSH 35.71%) of embryo cultured in Waymouth MB751/1 containing hormone was significantly(P<0.05) lower than that(60.00%) of oocytes cultured without hormone. 3. The maturation rates of oocytes cultured in BMOC-II medium containing hormone were 66.67(control). 66.67(hCG). 91.89(FSH) and 81.82(hCG+FSH)%. respectively. showing the highest rate in FSH treatment. And, the cleavage rates of oocytes cultured in BMOC-II medium containing hormone were 81.82 (control, 79.17(hCG), 50.00(FSH) and 66.67(hCG+FSH)%, respectively.

THE RELATIONSHIP BETWEEN BUFFER INDEX CURVE AND FERMENTATION QUALITY OF SILAGE

  • The purpose of this study was to investigate the possibility of using the buffer index curve as a criterion of silage quality evaluation. The buffer index (or buffer capacity, $\beta$) value is the amount of 0.1N NaOH consumed in titrating from the beginning to the end of the chosen pH step. This value equals the slope of the titration curve at the chosen pH range (${\beta}={\Delta}NaOH/{\Delta}pH$). There were two patterns of buffer index curve. Poorly preserved silages had a peak at pH 5.0, whereas with well preserved silages the peaks tended to rise to the pH 3.75 or 4.00. Well preserved, wilted silages with high pH also appeared to have peak values at pH 3.75 or 4.00. There was a high positive correlationship between the concentration of lactic acid and peak value of good quality silage (r = 0.994, p < 0.01). Also a high positive correlationship (r = 0.899, p < 0.001) was found between the sums of concentration of acetic and butyric acids and the peak values of poor quality silages. The results of these experiments verified the value of the buffer index method as a criterion of silage quality evaluation.

Increased Plasma $PGE_2$ Levels after Administration of Radionuclides Used in Nuclear Medicine (핵의학 이용 방사핵종의 투여후 혈중 $PGE_2$의 변동)

  • After administration of $^{99m}TC,\;^{67}Ga,\;^{131}I,\;^{32}P\;and\;^{238}U(UO_2(NO_2)_3{\cdot}6H_2O)$, in male rats, toxic effects were examined by determining the biological materials in blood. Dosages of radio-nuclides injected are based on the amounts routinely administered to patients, and concentrations of BUN, creatinine, SGOT, SGPT and $PGE_2$ in plasma are determinated as indices to the biochemical response. No increase of creatinine was observed after injection of $^{99m}TC$. Concentrations of BUN, SGOT and $PGE_2$ were not significantly increased in comparison with before-administration. Administration of $^{67}Ga,\;^{131}I\;and\;^{32}P$, did not significantly change BUN, SGPT and SGOT, but largely increased $PGE_2$ than control levels. Besides, $^{238}U$ Showed the most severe toxicity. From the above results, we suggest that the determination of $PGE_2$ in plasma can be used as an index in case of elvaluating the effects of radiation toxicity by nuclides used in nuclear medicine.

Tumor Location Causes Different Recurrence Patterns in Remnant Gastric Cancer

  • Purpose: Tumor recurrence is the principal cause of poor outcomes in remnant gastric cancer (RGC) after resection. We sought to elucidate the recurrent patterns according to tumor locations in RGC. Materials and Methods: Data were collected from the Shanghai Cancer Center between January 2006 and December 2020. A total of 129 patients with RGC were included in this study, of whom 62 had carcinomas at the anastomotic site (group A) and 67 at the non-anastomotic site (group N). The clinicopathological characteristics, surgical results, recurrent diseases, and survival were investigated according to tumor location. Results: The time interval from the previous gastrectomy to the current diagnosis was 32.0±13.0 and 21.0±13.4 years in groups A and N, respectively. The previous disease was benign in 51/62 cases (82.3%) in group A and 37/67 cases (55.2%) in group N (P=0.002). Thirty-three patients had documented sites of tumor recurrence through imaging or pathological examinations. The median time to recurrence was 11.0 months (range, 1.0-35.1 months). Peritoneal recurrence occurred in 11.3% (7/62) of the patients in group A versus 1.5% (1/67) of the patients in group N (P=0.006). Hepatic recurrence occurred in 3.2% (2/62) of the patients in group A versus 13.4% (9/67) of the patients in group N (P=0.038). Patients in group A had significantly better overall survival than those in group N (P=0.046). Conclusions: The tumor location of RGC is an essential factor for predicting recurrence patterns and overall survival. When selecting an optimal postoperative follow-up program for RGC, physicians should consider recurrent features according to the tumor location.

A Study of the effects of Health Contract on the performance Level for Activity of Daily Living in the Hemiplegic patients (건강계약이 편마비 환자의 일상생활동작 수행정도에 미치는 영향)

  • The purpose of this study was to test the effects of health contract on the performance level for activity of daily living(ADL) in the hemiplegic patients. A quasi - experimental research for health contract, which was approved as an effective method of nursing intervention, was attempted to increase the performance of the hemiplegic patients' ADL. As a purpose sample, 69 hemiplegic patients hospitalized at Oriental medicine hospital of K university were taken and devided into the experimental group and the control group by means of random assignment. After Contracting with the experimental group, they were reinforced everyday for 20 minutes. The experimental group and the control group were observed and interviewed for five times at the in terval of 3~4 days by the trained nurses for this research. The data Collected through above mentioned methods were compute analyzed by t-test and ANOVA according to the purpose of this study. Sub Hypothesis 1,2,4,5 : “The hypothesis that the experimental group with health contract will demonstrate increased the performance level for eating(t=2.29, df=42.70, p= .027), personal hygiene and grooming (t=4.04, df=43.10, p= .000), dressing(t=3.32, df=67, p= .001) and undressing (t=3.47, df=48.44, p= .001) than the control group” was supported. Sub Hypothesis 3.6 : “The hypothesis that the experimental group with health contract will demonstrate increased the performance level for toiletting(t= .19, df=67, p= .849) and mobilization (t= .30, df=67, p= .765) than the control group” was not supported. The conclusion can be that the positive relationship between the nurse and the patient results in the desired performance level for ADL in the hemiplegic patients.

Effect of Dietary Copper Sources (Cupric Sulfate and Cupric Methionate) and Concentrations on Performance and Fecal Characteristics in Growing Pigs

  • This study was conducted to assess the effects of organic and inorganic copper on performance in growing pigs. A total of 100 pigs, average age 63 d and initial body weight 21.46${\pm}$1.13 kg, were assigned to five treatment groups. Dietary treatments included i) CON (basal diet, 0 ppm Cu), ii) T1 (basal diet with 67 ppm Cu as cupric sulfate, $CuSO_4$), iii) T2 (basal diet with 134 ppm Cu as $CuSO_4$), iv) T3 (basal diet with 67 ppm Cu as cupric methionate, CuMet) and v) T4 (basal diet with 134 ppm Cu as CuMet). Throughout the entire experimental period, ADG (average daily gain), ADFI (average daily feed intake) and G/F (gain: feed) ratios showed no significant differences. The dry matter digestibility was improved in the T1, T2, T3, and T4 treatments (p<0.05), as compared with CON. Nitrogen digestibility was improved in the T3 treatment group as compared with CON (p<0.05). As compared with the T1 treatment group, fecal pH values were improved in the CON, T3, and T4 treatment groups (p<0.05). Fecal Cu concentrations were significantly lower in the CON, T3, and T4 treatment groups than in T1 and T2 (p<0.05). The incidence of diarrhea was reduced when the pigs were fed on the T2, T3, and T4 diets as compared with CON. In conclusion, diets supplemented with 67 or 134 ppm Cu as CuMet may prove effective in improving nutrient digestibility and fecal pH value in growing pigs, and fecal Cu concentrations may be reduced by CuMet supplementation.
