  • Optimum conditions for the enzymatic hydrolysis of isolated sesame meal protein were investigated. Optimum conditions by papain were $60^{\circ}C$, pH 6.0, 3% enzyme concentration to substrate and 1.5% substrate concentration, respectively. The optimum operating conditions using pepsin were $55^{\circ}C$, pH 9.0, 3% enzyme concentration to substrate and 1% substrate concentration. The optimum operating conditions using trypsin were $60^{\circ}C$, pH 9.0, 1% enzyme concentration to substrate and 1% substrate concentration.

The Effect of Exercise Training (EXE) on Myocardium Glucose Metabolic Phenotypic Proteins and HSP-60 Protein Expression after Ischemia/Reperfusion Injury in STZ-induced Rats (지구성 운동이 STZ-당뇨 유발 쥐의 허혈/재 관류 손상 후 심근의 당대사 관련 표현형 단백질과 HSP-60 단백질 발현에 미치는 영향)

  • The objective of this study was to identify EXE (1 hr a day at 21 m/min for 5 day/wk, at 0 % grade for 6 wk) on myocardium glucose metabolic phenotypic proteins (AMPK-PGC-1${\alpha}$-GLUT-4) and HSP-60 protein expression after ischemia/reperfusion injury (IRI) in STZ-induced rats. EXE was performed using STZ-induced diabetic rats on a rodent treadmill (28 m/min, 1 hr/day, 5 day/wk for 6 wk). The results of this study suggest that i) serum insulin level was not changed among groups (p>l0.05). ii) the LVDP level increased significantly in the STZ-EXE-IRI group compared to the STZ-IRI group at 60 min (p<0.01), 70 min (p<0.05) and 80 min (p<0.05) after reperfusion, respectively, and iii) AMPK phosphorylation (p<0.01), PGC-1${\alpha}$ protein (p<0.001), GLUT-4 protein (p<0.001) and HSP-60 protein expressions (p<0.05) increased significantly in the STZ-EXE-IRI group compared to the STZ-IRI group. In conclusion, the findings of the present study reveal that EXE may provide therapeutic value to insulin dependent diabetic patients with peripheral insulin resistance and myocardium injury by improving glucose metabolic proteins (AMPK-PGC-1${\alpha}$-GLUT-4) and heat shock protein-60 (HSP-60), along with increasing LVDP levels and decreasing glucose levels. Therefore, EXE protects the STZ-induced diabetic myocardium injury against ischemia/ reperfusion injury.

Production of extracellular protein from Bacillus sp. WY-60 (Bacillus sp. WY-60에 의한 균체외 단백질의 분비조건)

  • A bacteria strain producing extracellular protein was isolated and identified from soil samples, and the optimum conditions of producing protein were investigated. Eleven strains of bacteria were isolated from soil samples. Among which WY-60 strain showed a very strong capability of producing protein and identified as a Bacillus sp. The optimum composition of nutrient medium for the production of the protein by WY-60 was fructose 4.0%, polypeptone 1.0%, $NH_4NO_3$ 0.1%, $K_2HPO_4$ 0.1%, $MgSO_4{\cdot}7H_2O$ 0.005%, $CaCO_3$ 1.0% and the optimum pH and temperature were 8.0 and $30^{\circ}C$, respectively. The penicillin G and lincomycin added to the above medium were effective for the protein production of the WY-60, but other antibiotics were non-effective. The maximum production of protein was obtained after 5 days culture.

Effects of the Dietary Protein and Energy Levels on Growth in Fat Cod (Hexagrammos otakii Jordan et Starks) (사료의 단백질 및 에너지 함량이 쥐노래미 성장에 미치는 영향)

  • Two-month feeding experiment was conducted to investigate the optimum dietary protein level and energy to protein ratio in fat cod (Hexagrammos otakii Jordan et STARKS). The fish averaging 29 g were fed with one of the isocaloric diets containing 30, 40, 50 or $60\%$ of protein, or with one of the isoproteic diets containing 9, 10, 11 or 12 of available energy/protein (E/P) ratio. Weight gain and feed efficiency increased significantly with dietary protein level up to $50\%$, then decreased with $60\%$ protein diet (P<0.05). Daily protein intake increased significantly with dietary protein level, whereas protein efficiency ratio decreased with dietary protein level (P<0.05). Second order polynomial regression analyses of percent weight gain and daily protein intake may indicate that the adequate dietary protein level is $45\%$ and daily protein requirement per 100g fish is 1.5g for maximal growth. Weight gain, feed efficiency and protein efficiency from fish led the diet containing 12 of E/P ratio were significantly higher than those from fish fed the other diets (P<0.05). Daily feed or protein intake from fish fed the diet containing 12 of E/P ratio was significantly lower than those from fish fed the other diets (P<0.05). Daily lipid intake increased significantly with dietary E/P ratio (P<0.05).

  • To extract insoluble proteins and improve functional properties of sesame meal proteins was treated with phytase and protease from Aspergillus sp. It was found that the optimum pH, optimum temperature, optimum treatment time and optimum unit of enzyme for extraction of protein were pH $10{\sim}12$ (alkaline), $60^{\circ}C$, 11 hr. and 900 units of phytase and 60 units of protease, respectively. The foaming capacity, foaming stability, oil absorption and water absorption of sesame meal protein after treatment with phytase and protease were increased as compared to the control.

  • We have isolated a large cylindrical protein complex from hyperthermophile archaeon Thermococcus profundus. Structural analysis by image processing of electron micrographs suggests that the complex is composed of two stacked rings of eight subunits each; the ring enclose a central channel. The purified protein was shown to be a homomultimer of 60 kDa subunit (P60 complex). It exhibits an extremely thermostable ATPase activity with a temperature optimum of $80^{\circ}C$. This protein complex may play an important role in the adaptation of thermophile archaeon to life at high temperature.

  • The present study has been performed to evaluate Porphyromonas gingivalis (P.gingivalis) heat shock protein(HSP)60 as a candidate vaccine to inhibit multiple bacteria-induced alveolar bone loss. Rats were immunized with P.gingivalis HSP60 and experimental alveolar bone loss was induced by infection with multiple periodonto -pathogenic bacteria. Post-immune rat anti-P.gingivalis HSP IgG levels were significantly elevated and have demonstrated highly significant inverse relationship with the amount of alveolar bone loss induced by multiple bacteria. Results from PCR detection of subgingival bacterial plaque indicated that the vaccine successfully eradicated the multiple pathogenic species. We concluded that P.gingivalis HSP60 could potentially be developed as a vaccine to inhibit periodontal disease induced by multiple pathogenic bacteria.

  • Since periodontal infections are suggested as risk factors for the development of cardiovascular diseases, the present study was performed to evaluate the T cell immune responses specific to Pophylomonas gingivalis(P. gingivalis) heat shock protein(hsp) 60 and T-cell and B-cell epitope specificities for P. gingivalis hsp60 in atherosclerosis. Anti-P, gingivalis IgG antibody titers were elevated in all patients. We could establish P. gingivalis hsp-specific T cell lines from the atheroma lesions, a mixture of $CD4^+$ and $CD8^+$ cells producing the cytokines characteristic of both Th1 and Th2 subsets. of 108 overlapping synthetic peptides spanning whole P. gingivalis hsp60 molecule, ten peptides with common epitopes specificities for both T-cell and B-cell were identified. it was concluded that P. gingivalis hsp60 might K involved in the immunoregulatory process of atherosclerotic diseases with epitope specificities.

Factors in effecting the activities of the protein remover (단백질 제거제의 작용에 영앙을 미치는 요인)

  • We investigated the question whether protein removing activities of enzyme cleaner - protein remover for soft contact lens - are associated with the material of soft contact lens as well as action time, temperature and pH of enzyme solution. We used a subtilisin cleaner as protein remover and estimated the protein amount remained on soft contact lens after using the subtilisin cleaner under the different conditions. The remained protein in soft contact lens was greatly decreased until treatment for 60min, but no significant differences were found from 60min to 24hr. The cleaning effect of the enzymatic treatment in the range of $15{\sim}30^{\circ}C$ was constant. however, there was a significant decline of the protein removing effect at $10^{\circ}C$ and less. The pH of the solution was also important for the efficacy of the enzymatic treatment. The activity of the enzyme cleaner was highest in pH 8.0 and significantly decreased a pH below 7. The pH dependence was found to be related to the conformational change of subtilisin. Furthermore, significant differences in the protein deposit removing efficacy of the subtilisin cleaner were found among the soft contact lens materials.

Effects of Dietary Supplementation of Lysine Cell Mass (LCM) in Juvenile Israeli Carp, Cyprinus carpio (치어기 잉어에 있어 사료내 Lysine 부산물의 첨가효과)

  • This experiment was conducted to evaluate the lysine cell mass (LCM) as a dietary fish meal (EM) protein replacer in juvenile Israeli carp, Cyprinus carpio. Fishmeal, a major animal protein source in the control diet, was replaced by tCM on the protein equivalent base, Fish averaging 1,7 $\pm$ 0.1 g (Mean $\pm$ SD) fed one of nine diets containing isonitrogenous and isocaloric basis of $38\%$ crude protein and 15.2 kJ available energy/g diet: control, $100\%$ $FM; LCM_20$, $80\%$ $FM+20\%$ $LCM; LCM_40$, $60\%$ $FM+40\%$ $LCM; LCM_60$, $40\%$ $FM+60\%$ $LCM; LCM_100$, $100\%$ $LCM; LCM_20$l, $80\%$ $FM+20\%$ $LCM+0.07\%$ $Lysine; LCM_40$l, $60\%$ $FM+40\%$ $LCM+0.14\%$ $Lysine; LCM_60$l $40\%$ $FM+60\%$ $LCM+0.22\%$ Lysine; LCM_100l, $100\%$ LCM+$0.35\% Lysine. After 6 weeks of feeding trial there was no significant difference in weight gain (WG), feed efficiency (FE), protein efficiency ratio (PER) and specific growth rate (SGR) among fish fed control and $LCM_20$ (P>0.05), while fish fed $LCM_40,\;LCM_60,\;LCM_100,\;LCM_40l,\;LCM_60l\;and\;LCM_100l$ diets had a significantly lower WG, FE, PER and SGR than did fish fed control diet (P<0.05). There was no significant difference in WG, PER and SGR among fish fed control and $LCM_20$l diets (P>0.05), while fish fed $LCM_20$l S had a significantly lower FE than did fish fed control diet (P<0.05). No significant difference was observed in hematocrit and condition facto, among fish fed nine diets (P>0.05). Therefore, these results indicated that LCM could replace FM up to $20\%$ and dietary synthetic lysine supplementation did not show any positive growth effects in juvenile Israeli carp.
