인체 혈구암세포 U937에서 해양해면동물에서 추출된 Pectenotoxin-2에 의한 Apoptosis의 유발에 관한 연구 (Induction of Apoptosis by Pectenotoxin-2 Isolated from Marine Sponges in U937 Human Leukemic Cells)

  • 본 연구에서는 U937 인체 백혈병 세포의 증식에 미치는 PTX-2의 영향을 조사한 결과, PTX-2의 처리에 따라 U937 세포는 처리 농도 및 처리 시간 의존적으로 심한 형태적 변형과 함께 증식이 억제되었다. 이러한 PTX-2 처리에 의한 U937 세포의 증식억제는 apoptosis 유발과 관련이 있었으며, 이를 DAPI staining에 의한 apoptotic body 형성, flow cytometry를 이용한 sub-G1 세포 빈도의 정량적 분석을 통하여 확인하였다. 이러한 PTX-2 처리에 의한 U937 세포의 apoptosis 유발은 Bcl-2 family에 속하는 anti-apoptotic 인자인 Bcl-$X_L$의 발현 감소 및 IAPs family에 속하는 유전자들의 선택적 발현 감소와 연관성이 있음을 알 수 있었다. 이상의 결과들은 인체 암세포에서 PTX-2의 항암작용을 이해하는데 중요한 자료가 될 것이고 나아가 PTX-2을 포함한 그와 유사한 항암제 후보물질들의 연구에 있어서 기초 자료로서 사용될 수 있을 것으로 생각된다.

치과위생사의 감염관리 실천도 측정도구의 개발과 타당화 (Construction and Validation of Infection Control Practice Scale for Dental Hygienist)

  • 치과위생사의 감염관리 실천도를 측정하는 척도를 개발하고 타당화하기 위해 254명의 치과위생사를 대상으로 설문조사를 실시하였다. 예비척도는 리커트 4점 척도로서 21개의 문항으로 구성되었으며, 신뢰도 분석, 탐색요인분석, 확인요인분석을 통해 척도의 차원과 항목을 축소하고 신뢰도와 타당도를 검증하였다. 1. 탐색요인분석과 신뢰도 분석을 통해 4개 요인과 12개의 항목으로 척도를 수정하였다. 수정된 척도의 요인은 '예방접종 및 정기검진'(2항목), '기구 소독 및 멸균'(3항목), '손위생 관리'(4항목), '개인 보호'(3항목)가 추출되었으며, 전체분산 중 58.296%를 설명하고 있다. 예비척도 중 '환경 관리' 요인이 제외되었다. 2. 확인요인분석을 통해 한 문항을 제외하였다. 최종적인 측정모형은 4개 요인과 11개 항목으로 구성되었으며, 모형의 적합도는 $x^2$= 79.593(df = 38, 0 = 0.000), RMR= 0.045, GFI = 0.940, CFI = 0.904, AGFI = 0.896, NFI = 0.837, TLI = 0.861, RMSEA = 0.67로 나타나 대체로 기준을 충족하였다. 모든 측정변수의 요인부하량은 유의하였으나(p < 0.001), 세 변수만이 0.7 이상으로 나타났다. 평균분산추출값과 구성개념신뢰도는 대체로 기준을 충족하지 못하였으나, 모든 요인의 평균분산추출값은 각 요인간 상관관계제곱값 보다 크게 나타났다. 3. 요인분석을 통해 추출한 네 개의 요인이 치과위생사의 '감염관리 실천률 인식'에 미치는 영향을 알아보기 위해 네 요인을 독립변수로 선정하고 실천률 인식을 종속변수로 선정하여 회귀분석을 실시하였다. $R^2$는 0.304, 수정된 $R^2$은 0.431, F = 25.813, p = 0.000이며, '손위생 관리'를 제외한 세 변수의 회귀계수는 통계적으로 유의하게 나타났다.

Unstable Pathologic Vertebral Fractures in Multiple Myeloma : Propensity Score Matched Cohort Study between Reconstructive Surgery with Adjuvant Radiotherapy and Radiotherapy Alone

  • Objective : Although radiotherapy (RT) is recommended for multiple myeloma (MM) involving spine, the treatment of choice between reconstructive surgery with RT and RT alone for pathologic vertebral fractures (PVFs) associated with structural instability or neurologic compromises remains controversial. The purpose of this study was to evaluate the clinical efficacies of reconstructive surgery with adjuvant RT for treatment of MM with PVFs by comparing with matched cohorts treated with RT alone. Methods : Twenty-eight patients underwent reconstructive surgery followed by RT between 2008 and 2015 in a single institution, for management of PVFs associated with structural instability of the spine and/or neurologic compromises (group I). Twenty-eight patients were treated with RT alone (group II) after propensity score matching in a 1-to-1 format based on instability of the spine, as well as age and performance. Clinical outcomes including the overall survival rates, duration of independent ambulation, neurological status, and numeric rating scale (NRS) for back pain were compared. Results : Clinical and radiological features before treatment were similar in both groups. The median survival period was similar between the two groups. However, the mean duration of independent ambulation was significantly longer in group I (88.8 months; 95% confidence interval [CI], 66.0-111.5) than in group II (39.4 months; 95% CI, 25.2-53.6) (log rank test; p=0.022). Deterioration of Frankel grade (21.4% vs. 60.7%, p=0.024) and NRS for back pain (2.7±2.2 vs. 5.0±2.7, p=0.000) at the last follow-up were higher in the group II. Treatment-related complications were similar in both groups. Conclusion : In patients with unstable PVFs due to MM, reconstructive surgery may yield superior clinical outcomes compared with RT alone in maintaining independent ambulation and neurological status, as well as pain control despite similar median survival and complications.

인체폐암세포에서 Bcl-2 발현저하 및 caspase 활성을 통한 청조구폐탕의 apoptosis 유발에 관한 연구 (Apoptosis of Human Lung Carcinoma Cells through the Inhibition of Bcl-2 Expression and Activation of Caspase by Chungjogupae-tang)

  • We previously reported the anti-proliferative effect of Chungjogupae-tang (CJGPT) in human lung carcinoma A549 cells, which was associated with the induction of cyclin-dependent kinase inhibitor p21 in a tumor suppressor p53-independent manner. CJGPT treatment also resulted in the inhibition of prostaglandin E2 release A549 cells by the down-regulation of cyclooxygenase-2. In the present study, we investigated the pathway of the induction of apoptotic cell death by CJGPT in A549 cells. It was found that CJGPT could inhibit the cell viability and induce the apoptotic cell death of A549 cells in a dose-dependent manner as measured by hemocytometer counts, flow cytometry analysis and agarose gel electrophoresis. Apoptosis of A549 cells by CJGPT was associated with a down-regulation of anti-apoptotic Bcl-2 and inhibitor of apoptosis proteins (IAPs) expression. Additionally, DNA fragmentation by CJGPT was connected with the activation of inhibitor of caspase-activated DNase/DNA fragmentation factor 45 (ICAD/DFF45) protein expression.

대퇴사두근에 대한 탄력테이핑 적용이 보행주기에 미치는 효과 (The Effect of Elastic Taping on the Quadriceps Femoris to the Gait Cycle)

  • Background: The purpose of present study was to investigate the effect of quadriceps femoris taping in normal gait using 3D motion capture technique. Method: Twenty healthy volunteers, have no musculoskeletal problems, were recruited as subjects for this study. In experimental group, 10 males (mean age : 23.6${\pm}$.40) and 10 females (mean age : 21.3${\pm}$.36) were included. Methods of therapeutic taping that taping of patellar inferior and medial gliding and quadriceps. To obtain the dynamic data, we captured the motion of subject attached markers without taping during repeated gaits five times or more in 7 m Capture volume of gait analysis center. The result was obtained as a mean value in three times. After taping on quadriceps femoris, the same procedure was carried out. Statistical analyses were performed using statistical software packagess SPSS WIN 12.0(SPSS, Chicago, IL, USA). Differences were tested for statistical significance using paired t-test, independent t-test, chi-squared test for comparisons between the male and female. Results: The date of 20 subjects who carried out the whole experimental course were statistically analyzed. 1. gait velocity was showed that female group had more significantly increased than male group(p<.05). 2. step length was showed that female group had more significantly increased than male group(p<.05). 3. step width was showed that female group had more significantly increased than male group(p<.05). Conclusion: Elastic taping on quadriceps femoris promoted cadence, gait velocity, step length and step width in normal subject group.

인체유방암세포에서 Cyclooxygenase-2 활성 및 Prostaglandin E2 생성에 미치는 어성초 추출물의 영향 (Antiproliferative effect of Houttuynia cordata Thunb was Associated with the Inhibiton of Cyclooxygenase-2 Expression in Human Breast Carcinoma Cells)

  • Houttuynia cordata Thunb, well known as 'E-Sung-Cho' in Korea, is a traditional medicinal plant generally used in Oriental medicine therapy. In the present study, we investigated the effect of water extract of H. cordafa (WEHC) on the growth of human breast carcinoma MCF-7 cells. Exposure of MCF-7 cells to WEHC resulted in growth inhibition and apoptosis in a dose-dependent manner as measured by MTT assay and fluorescent microscope. The anti-proliferative effect of WEHC was associated with a dose-dependent up-regulation of cyclin-dependent kinase inhibitor p21 in a p53-independent fashion. We found WEHC decreased the levels of cyclooxygenase (COX)-2 and inducible nitric oxide synthease (iNOS) expression without significant changes in the expression of COX-1 , which was correlated with a decrease in prostaglandin $E_2{\;}(PGE_2)$ synthesis. Taken together, these findings provide important new insights into the possible molecular mechanisms of the anti-cancer activity of H. cordata.

섬수 추출물에 의한 T24 인체 방광암세포의 증식억제에 관한 연구 (Growth Arrest by Bufonis Venenum is Associated with Inhibition of Cdc2 and Cdc25C, and Induction of p21WAF1/CIP1 in T24 Human Bladder Carcinoma Cells)

  • Bufonis venenum (dried toad venom; Chinese name, Chan su) is a traditional Chinese medicine obtained from the skin venom gland of the toad. It has long been used in treating arrhythmia and other heart diseases in China and other Asian countries. Additionally, Bufonis venenum has been reported to selectively inhibit the growth of various lines of human cancer cells. In the present study, it was examined the effects of extract of Bufonis venenum (EBV) on the growth of human bladder carcinoma cell line T24 in order to investigate the anti-proliferative mechanism and induction of apoptosis by EBV. Treatment of T24 cells to EBV resulted in the growth inhibition, morphological change and induction of apoptotic cell death in a dose-dependent manner. Flow cytometric analysis revealed that EBV treatment caused G2/M phase arrest of the cell cycle and down-regulation of cyclin A, cyclin B1 and Cdc2, which was associated with a marked up-regulation of cyclin-dependent kinases (Cdks) inhibitor p21 (WAF1/CIP1) in a p53-independent manner. The Cdc25C expression was also significantly inhibited by EBV treatment, however Wee1 kinase expression was not affected. The induction of apoptotic cell death by EBV was connected with down-regulation of anti-apoptotic Bcl-XS/L expression without alteration pro-apoptotic Bax expression. Taken together, these findings suggest that EBV may be a potential chemotherapeutic agent for the control of human bladder carcinorma cells and further studies will be needed to identify the active compounds that confer the anti-cancer activity of EBV.

청소년에서 우울 및 불안 증상과 수면 곤란의 성별에 따른 차별적 관련성 (Differential Relations of Depression, Anxiety and Sleep Disturbances by Gender in Young Adolescents)

  • 연구목적 청소년에서 성별에 따른 불안, 우울과 수면 장애의 유병율을 조사하고 우울, 불안과 수면 곤란의 관련성을 성별에 따라 분석하고자 한다. 방 법 서울시 마포구 내 7개 중학교의 1학년 학생 1,422명을 대상으로 자가 보고 질문지인 Children's Depression Inventory(CDI), Beck Anxiety Inventory(BAI), Pittsburgh Sleep Quality Index(PSQI)를 실시하였다. CDI, BAI, 그리고 PSQI는 각각 다음과 같이 분류되었다 : $\leq$21, 22-25, 26-28, 그리고 $\geq$29 ; $\leq$21, 22-26, 27-31, 그리고 $\geq$32 ; $\geq$5, <5. 각각의 cut-off 점수는 CDI 22, BAI 22 그리고 PSQI는 5점 이었다. 두 군간 연속 변수 비교에는 독립 t-검정, 비 연속변수 비교에는 $\chi^2$-검정, 불안과 우울 증상이 수면 곤란에 미치는 영향은 로지스틱 회귀분석을 이용하였다. 결 과 여학생에서 평균 CDI 점수($12.52{\pm}8.32$ vs. $10.39{\pm}7.52$ ; p=0.003), BAI($7.77{\pm}7.9$ vs. $9.84{\pm}9.044$ ; p<0.001) 그리고 PSQI($4.57{\pm}2.67$ vs. $3.64{\pm}2.30$ ; p=0.013)는 남학생보다 유의하게 높았다. 그러나 성별, 과거 정신과 치료력을 통제한 후, 매우 높은 수준의 우울과 불안 상태에서 남학생이 수면 곤란을 경험할 위험도가 여학생에 비해 현저히 높았다(CDI aOR, 14.66 ; 95% CI, 4.17-51.53, BAI aOR, 32.99 ; 95% CI, 4.26-255.39). 결 론 이 연구 결과는 여학생에 비해 남학생에서 우울과 불안의 정도가 심각할 때 수면 곤란을 경험할 위험이 더욱 높다는 것을 시사한다.

Paclitaxel에 의한 관절연골 세포의 capase-비의존적 mitotic catastrophe 유도 (Paclitaxel Induced Caspase-Independent Mitotic Catastrophe in Rabbit Articular Chondrocyte)

  • Paclitaxel은 미세소관의 탈중합을 억제하는 시약으로 알려져 있다. Paclitaxel은 다양한 세포에서 세포 내 방추체를 안정화시킴으로써 유사분열 억제 및 세포사멸을 유도한다. 본 실험에서는 토끼 관절 연골세포에서 paclitaxel이 연골세포의 증식과 사멸에 미치는 효과에 대한 연구를 수행하였다. MTT assay를 수행한 결과 paclitaxel은 연골세포에서 농도 의존적으로 세포 증식을 억제한다는 것을 확인 할 수 있었으며, FACS analysis와 Western blot analysis를 수행한 결과, paclitaxel이 G2/M 정지를 유도하는 것을 확인하였다. 또한, paclitaxel이 비정상적인 세포 분열유도와 핵 단편분절 유도없이 일어나는 mitotic catastrophe 즉, caspase-3 비의존적인 세포사멸을 유도하였다. Paclitaxel을 처리한 세포에서 일어나는 이러한 mitotic catastrophe에 의한 세포 죽음은 G1/S기의 진행을 억제하는 시약인 thymidine을 처리하는 것에 의해 억제되는 것을 확인할 수 있었다. 이러한 결과를 종합해 볼 때, paclitaxel에 의한 토끼 관절 연골 세포에서의 세포 죽음은 caspase-3 비의존적인 mitotic catastrophe에 의해 일어나는 것으로 사료되어진다.

  • Objective : The effect of water extract of Chungjogupae-tang (CJGPT) was investigated on the growth of human lung carcinoma A549 cells. Methods : MTT assay and fluorescent microscope performed to compare and examine the efficacy of CJGPT treatment on the cytostaticity of lung cancer cells in proportion to time and doses, and DAPI staining and Western blot analysis were used to examine their effect on apoptosis. In addition the quantitative RT-PCR was used to examine to lung cancer cells growth and Progtaglandin E2 and Telomerase activity were measured Results : Exposure of A549 cells to CJGPT resulted in the growth inhibition and apoptosis in a dose-dependent manner as measured by MTT assay and fluorescent microscope. The antiuoliferative effect by CJGPT treatment in A549 cells was associated with morphological changes such as membrane shrinking and cell rounding up. CJGPT treatment resulted in an up-regulation of cyclin-dependent kinase inhibitor p21(WAF1/CIPl) in a p53-independent fashion. We found that CJGPT treatment decreased the levels of cyclooxygenase (COX)-2 and inducible nitric oxide synthease (iNOS) expression without significant changes in the expression of COX-1, which was correlated with a decrease in protaglandin E2 (PGE2) synthesis. CJGPT treatment also inhibited the levels of human telomerase reverse transcriptase (hTERT) and telomerase-associated protein (TEP)-1 mRNA expression, however the activity of telomerase was slightly increased by CJGPT treatment. Conclusion : These findings suggested that CJGPT-induced inhibition of human lung carcinoma A549 cell growth was connected with the induction of apoptotic cell death and the results provided important new insights into the possible molecular mechanisms of the anti-cancer activity of CJGPT.
