전남산 젓갈의 지방산 조성 (Fatty Acid Composition of Salt-Fermented Seafoods in Chonnam Area)

  • 김치의 부재료로 쓰이는 멸치젓과 새우젓의 수분함량은 각각 31.14~39.01% 및 30.33~36.59%이고 밑반찬용으로 쓰이는 꼴뚜기젓, 모치젓 및 석화젓 등은 50%정도였다. 지방함량은 멸치젓이 5.51~5.85%로 가장 높았고 모치젓, 석화젓, 꼴뚜기젓 및 새우젓의 순이었다. 젓갈의 지방산 조성비는 시료에 따라 달랐지만 주요 지방산은 $C_{l6:0}$, $C_{20:5}$, $C_{l6:1}$, $C_{22:6}$, 및 $C_{l8:1}$ 등이었고, n-3 고도불포화지방산 비율이 높은 시료로는 꼴뚜기젓이 39.11%로 가장 높았고, 새우젓, 석화젓, 멸치젓 및 모치젓의 순이었다. n-3 고도불포화지방산 중에서 EPA는 비교적 석화젓(17.71%)과 꼴뚜기젓(16.38%)이 높았으며 DHA는 꼴뚜기젓(22.22%)과 새우젓 (15.13~21.50%)이 높았으나 특히 모치젓은 가장 낮은 함량을 보였다.보였다.

  • Reproductive condition of the tropical blacklip pearl oyster, Pinctada margaritifera collected during the period July through September 2003 from Chuuk Lagoon, Federated State of Micronesia was investigated using histology. The level of gonad development for each pearl oyster was determined with an average score of five microscopic fields, and the average score was used as the maturity index (ML). All wild pearl oysters collected in July did exhibit fully ripe eggs in their ovaries ($45{\sim}50{\mu}M$ in diameter), indicating that they were ready for spawning. In mid August most wild pearl oysters were in spawning and M1 dropped dramatically from mid- to late September, suggesting that the wild pearl oyster completed spawning during this period. In contrast, the cultivated pearl oysters collected in mid-September held ripe eggs in the ovaries and only a few of them spawned, indicating that gonad maturation of the cultivated pearl oyster was somewhat slower than that of the wild pearl oyster in Chuuk Lagoon during the summer period. Histological analysis also indicated that spawning of the pearl oyster is rather incomplete and they may spawn continuously during summer.

Evaluation of the Bacteriological Quality of a Shellfish-growing Area in Kamak Bay, Korea

  • The sanitary quality of a shellfish-growing area in Kamak Bay, Korea, was assessed through a bacteriological examination of seawater and oysters from January 2004 to December 2006. From seawater samples collected at 28 stations, the range of geometric mean (GM) and the estimated 90th percentile for most-probable-number (MPN) values of fecal coliforms were <1.8-2.4 MPN/100 mL and 1.8-6.0 MPN/100 mL, respectively. The observed fecal coliform GM and the estimated 90th percentile did not exceed the fecal coliform water quality standards of 14 MPN/100 mL and 43 MPN/100 mL. Therefore, the bacteriological quality of seawater at this shellfish-growing area met the Korean Shellfish Sanitation Program (KSSP) criteria for a growing area used for export. The range of the fecal coliform GM and the estimated 90th percentile MPN values of oyster samples were 19.2-160 MPN/100 g and 20.2-166.9 MPN/100 g, respectively, and the range of the viable cell count was 30-1900 CFU/g. Thus, the fecal coliform value for the oysters and the viable cell count were less than the current shellfish quality standards of 230 MPN/100 g and 50,000 CFU/g, respectively. The bacteriological quality of the oysters complied with the criteria for domestic use and export of shellfish.

통영시 연안의 양식굴(Crassostrea gigas)에서 검출된 노로바이러스의 정량분석 (Norovirus Quantification in Oysters Crassostrea gigas Collected from Tongyeoung, Korea)

  • Norovirus (NoV) is a major cause of food poisoning outbreaks in Korea. Most NoV outbreaks originate from environmental contamination, but bivalves such as oysters are also important vectors. Oyster Crassostrea gigas contamination by NoV has been reported in Korea, but no quantitative analyses of NoV have been performed. We investigated the NoV concentration in 21 oyster samples from a Korean commercial oyster-growing area with confirmed fecal contamination from January to December 2012, using real-time reverse transcription-polymerase chain reaction. Additionally, we assessed the NoV concentration after heating to investigate the effects of heat treatment on NoV-infected oysters. In NoV-positive samples, the cycle threshold (Ct) values were 37.43-39.41 and 36.77-39.30, while viral concentrations were $8.97{\times}10^2-2.24{\times}10^2$ and $3.05{\times}10^2-7.47{\times}10^1$ copies/g for genogroups I and II, respectively. After heat treatment, NoV genogroup I decreased by 83.4%, 88.0%, 89.4% and 100% at $60^{\circ}C$, $68^{\circ}C$, $70^{\circ}C$, and $100^{\circ}C$, respectively, for 15 min, while genogroup II respectively decreased by 67.3%, 76.3%, 80.1%, and 89.8% under the same conditions.

Cellular Biomarker of Membrane Stability and Hydrolytic Enzyme Activity in the Hemocytes of Benzo(a)pyrene-exposed Pacific oyster, Crassostrea gigas

  • The Pacific oysters, Crassostrea gigas, were stressed with different concentrations of benzo(a) pyrene and depurated to determine the hemocyte lysosomal membrane stability and hydrolytic enzymatic activity as a biomarker candidate to the chemical, using NRR (neutral red retention) and API ZYM System, respectively. The membrane damage measured as NRR decrease was significant with the increase of chemical concentration and exposure time (P<0.05), providing a possible tool for biomarker. Interestingly, the control showed intrinsic stress probably due to captive life in the laboratory, and a recovering trend was also found during the depuration. The benzo(a)pyrene-exposed oysters showed increased enzyme activities in alkaline phosphatase, esterase (C4), acid phosphatase, naphthol-AS-BI-phospho­hydrolase, $\beta$-galactosidase, $\beta$-glucuronidase, and N-acetyl- $\beta$-glucosaminidase. Of them, only two enzymes, acid phosphatase and alkaline phosphatase, showed some potential available for the generation of enzymatic biomarker in the oyster. The results are suggestive of the potential availability of the cellular and enzymatic properties as a biomarker. However, considering that a robust biomarker should be insensitive to natural stress coming from normal physiological variation, but sensitive to pollutants, a concept of intrinsic stress the animal possesses should be taken into consideration. This reflects the necessity of further research on the intrinsic stress affecting the cellular and enzymatic properties of the chemical­stressed oysters prior to using the data as a biomarker.

Genetic Relationships of Four Korean Oysters Based on RAPD and Nuclear rDNA ITS Sequence Analyses

  • Random amplified polymorphic DNA (RAPD) marker and sequence analyses of the internal transcribed spacer (ITS) region of ribosomal DNA were used to assess phylogenetic relationships of four Korean oyster species. The average number of species-specific markers identified from five universal rice primers (URPs) by RAPD-PCR was 1.8 for Crassostrea gigas, 3.2 for C. nippona, 3.6 for C. ariakensis, and 4.6 for Ostrea denselamellosa. The length of the ITS (ITS1-5.8S-ITS2) region ranged from 1,001 to 1,206 bp (ITS1, 426-518 bp; 5.8S, 157 bp; and ITS2, 418-536 bp), while the GC content ranged from 55.5-61.1% (ITS1, 56.8-61.8%; 5.8S, 56-57.3%; and ITS2, 54.1-62.2%). A phylogenetic analysis of the oysters based on our RAPD, ITS1, and ITS2 sequence data revealed a close relationship between C. gigas and C. nippona and a distant relationship between the genera Crassostrea and Ostrea. Our results indicated that RAPD and ITS sequence analysis was a useful tool for the elucidation of phylogenetic relationships and for the selection of species-specific markers in Korean oysters.

자연정화에 의한 양식굴(Crassostrea gigas) 중 노로바이러스 저감화 (Defecation of Norovirus from the Oyster Crassostrea gigas by Depuration Following Translocation of the Growing Area)

  • The efficacy of depuration following growing area translocation for the defecation of norovirus was evaluated under experimental conditions using oysters Crassostrea gigas previously subjected to bioaccumulation of this virus at a waste treatment plant discharge site. Three trials were assayed in an open experimental system with a commercial oyster farm located in a shellfish growing area approved by the Korean Shellfish Sanitation Program. Real-time reverse-transcription polymerase chain reaction (RT-PCR) was used to quantify viruses in the digestive glands of oysters. The final viral loads in oysters after 12 days remained under the detection limit (10 copies/g digestive gland) of the real-time RT-PCR. This reduction trend showed two-phase removal kinetics, with an initial slow reduction or slight increase in viruses during the first 2 days of depuration and subsequent stabilization with 0.12 to 2.64 log unit norovirus copies/g digestive gland per 2 days of depuration for the remaining time.

수온과 염분농도가 Vibrio vulnificus 증식에 미치는 영향 (Effect of temparature and salinity on the multiplication of Vibrio vulnificus)

  • V. vulnificus 오염으로부터 패류의 안전성을 조사하기 위해 굴을 V. vulnificus에 인위감염시키고 4℃와 25℃에서 패각 유무에 따른 V. vulnificus 균수 변화를 조사하였다. V. vulnificus가 있는 인공해수에 침지시킨 굴에서 분리된 V. vulnificus 균수는 인공해수 속의 균수보다 많았다. 굴을 4℃에 보관하는 경우는 실험기간 동안 V. vulnificus 균수의 차이가 관찰되지 않았지만 25℃에 보관하는 경우는 빠른 균수의 증가가 관찰되어 보관온도가 균수 변화에 많은 영향을 주었다. 여러 염분농도, 5, 15 및 35 ‰의 인공해수에서 V. vulnificus 균수는 25℃와 4℃에서 모두 감소하였다. 그러나 4℃에서 균수의 감소율은 25℃에서 보다 빨랐고 4℃에서 염분도 V. vulnificus 균수 감소율에 영향을 주어 35 ‰의 인공해수에서 가장 높은 감소율을 보였다.

Detection of Ostreid Herpesvirus 1 from adult Pacific Oysters Crassostrea gigas Cultured in Korea

  • The presence of ostreid herpesvirus 1 (OsHV-1) and the percentage of viral DNA detected in Pacific oyster Crassostrea gigas adults were investigated monthly between May and November 2012 at three locations along the southern coast of Korea. Among 210 oysters examined by polymerase chain reaction (PCR) analysis, OsHV-1 DNA was detected in only one oyster collected in August. The low detection rate of OsHV-1 DNA was consistent with the lack of reported OsHV-1-associated disease in C. gigas cultured in Korea. The sequence of the present PCR product amplified with the C2/C6 primer pair was identical to that of OsHV-1 ${\mu}Var$ except for one nucleotide, and the sequence amplified with Del36-37F2/Del36-37R showed a 605-bp deletion as in OsHV-1 ${\mu}Var$. Although these sequence data are insufficient to determine genotype, the results suggest that the herpesvirus detected was similar to OsHV-1 ${\mu}Var$. This is the first report on the presence of OsHV-1 in adult Pacific oysters cultured in Korea.

Infection Status of Estuarine Fish and Oysters with Intestinal Fluke Metacercariae in Muan-gun, Jeollanam-do, Korea

  • The source of human infection with intestinal flukes was surveyed in estuarine fishes, including the dotted gizzard shad, common sea bass, common blackish goby, redlip mullet, black sea bream, and oyster collected from Muan-gun, Jeollanam-do, Korea during August and September 2007. Collected fishes and oysters were artificially digested in pepsin-HCl solution and examined under a stereomicroscope. In 36 shads (Konosirus punctatus) and 20 basses (Lateolabrax japonicus) examined, Heterophyopsis continua metacercariae were found in 58.3% and 100%, and their average numbers were 12.0 and 6.3 per infected fish, respectively. In 34 gobies (Acanthogobius flavimanus) examined, metacercariae of H. continua were detected in 79.4%, Stictodora lari in 97.1%, and Acanthotrema felis in 92.1%, and their average numbers were 45.8, 189.3, and 235.3 per infected fish, respectively. In 37 redlip mullets (Chelon haematocheilus), Heterophyes nocens metacercariae were found in 56.8%, Pygidiopsis summa in 94.6%, and Stictodora fuscata in 45.9%, and the average metacercarial densities were 17.4, 31.3, and 35.1 per infected fish, respectively. In 30 black sea breams (Acanthopagrus schlegeli) and 45 oysters (Crassostrea gigas) examined, no metacercariae were detected. From the above results, it has been confirmed that the dotted gizzard shad, common sea bass, common blackish goby, and redlip mullet from Muan-gun, Jeollanam-do, Korea are infected with the metacercariae of heterophyid flukes.
