Effect of some antibiotics on control of bacterial brown blotch of oyster mushroom caused by Pseudomonas tolaasii (몇가지 항생제의 느타리버섯 Pseudomonas tolaasii에 의한 세균성갈색무늬병 방제 효과)

  • Antibacterial activities of 33 antibiotics against Pseudomonas tolaasii causing the brown blotch disease on the edible mushroom Pleurotus ostreatus, were tested in vitro for the control of the disease. Tetracyclin, kanamycin, kasugamycin, and streptomycin showed strong antibacterial activity against P. tolaasii, having the minimal inhibitory concentration of 10, 10, 100 and 200ppm, respectively. These antibiotics showed similar control value of 72.9, 71.2, 68.1 and 62.7%, respectively when applied on the artificially infected mushroom beds. Mushroom yields in the tetracycline treated boxes were increased about 31.8% comparing to the control ones. Mycelial growth of P. ostreatus on the PDA supplemented with streptomycin and kanamycin were not affected, but were inhibited 10~20% and 40% with tetracyclin and kasugamycin treatment, respectively.

Characterization of a new commercial strain 'Mongdol' by intra-specific hyphal anastomosis in Pleurotus ostreatus (계통간 교잡에 의한 느타리 신품종 '몽돌'의 육성 및 그 특성)

  • A new commercial strain "Mongdol" of oyster mushroom was developed by hyphal anastomosis. It was improved with hybridization between monokaryotic strain derived from Pleurotus ostreatus ASI 0627 and dikaryotic strain derived from P. ostreatus ASI 2929. The optimum temperature of mycelial growth and fruiting body development were $25{\sim}30^{\circ}C$ and $12{\sim}18^{\circ}C$, respectively. When two different media including PDA (potato dextrose agar medium) and MCM (mushroom complete medium) were compared, the mycelial growth of this mushroom was faster in MCM than in PDA. Similar result was observed with the control strain P. ostreatus ASI 2504. Analysis of the genetic characteristics of the new cultivar "Mongdol" showed a different DNA profile as that of the control strain ASI 2504, when RAPD (Random Amplified Polymorphic DNA) primer URP3 and URP6 were used. Fruiting body production per bottle was about 106 g using demonstration farms. The color of pileus was blackish gray and the stipe was long. Therefore, we expect that this new strain "Mongdol" will satisfy the consumer's demand for high quality mushrooms.

An Investigation of the Major Dipteran Pests on the Oyster Mushroom(Pleurotus ostreatus) in Korea (한국 느타리버섯 재배지에 발생하는 주요 파리류 해충에 관한 연구)

  • This study was carried out to investigate the seasonal and regional occurrence and development of major pest flies on the oyster mushroom. The survey was performed in the oyster mushroom houses located at 16 places, selected randomly throughout South Korea from July to August and from October to November in 1997. The six species of flies were found including three pest species from Sciaridae, Cecidomyiidae and Scatopsidae flies. The pattern of occurrence was somewhat dissimilar depending on the survey periods and regions. Among these, the most serious and abundant pest, Lycoriella mali (Diptera: Sciaridae), occurred year around at the regions surveyed. In this study, Coboldia fuscipes (Diptera: Scatopsidae) was confinned as a mushroom pest. Its larvae damaged the mycelium, and occurred usually in summer from July to August. The paedogenetic Mycophila sp. (Diptera: Cecidomyiidae) was found abundantly on the oyster mushroom bed, and caused a severe damage in fall from October to November.

Two new species of Trichoderma isolated from commercially grown oyster mushroom, Pleurotus ostreatus (oral)

  • We describe two new Trichoderma species associated with oyster mushroom in Korea. Trichoderma green mould has been one of the most serious diseases of oyster mushroom in Korea. Of these the predominant species are two unrecorded species. We designed as Trichoderma sp. Korean type 1 (Th K1) and Trichoderma sp. Korean type 2 (Th K2), respectively. Th K1 and Th K2 can be distinguished from previously reported Trichoderma species as well as each other in morphological characteristics including growth rate at 35$^{\circ}C$, colony morphology, conidia shape and branch pattern of phialides. Sequence of the ITS region of rDNA, the protein coding translation elongation factor gene(EF-1${\alpha}$), and RNA polymeraseII (RPB2) not only clearly separated Trichoderma sp. Korean types from their closely related T. harzianum biotype but also distinguished them from each other. Analyses of the EF-1${\alpha}$ and RPB2 sequences were found to be more useful for establishing systematic relationships among Trichoderma isolates than those of the ITS sequence.   • The combination ratio of oyster mushroom cultivation supplements have used poplar sawdust(PS) 50% : beet pulp(BP) 30% : cotton seeds meal(CS) 20%. we substituted cotton seeds meal and beet pulp for food by-product dried wastes(DW) 10, 20. 30% respectively. In case of mixing with DW 10% except from BP, mycelial growth was inhibited a little, but mycelial density was accelerated. The more DW contents increase, the more mycelial growth was inhibited, but mycelial growth was accelerated a little, respectively. In case of mixing with DW except from CS, mycelial growth was similar to control almost. In case of mixing with DW except from CS and BP, mycelial growth was inhibited remarkably. CHUNCHU variety developed resistance to developed than SUHAN variety.

Comparison of amino acid and free amino acid contents between cap and stipe in Flammulina velutipes and Pleurotus ostreatus (팽이와 느타리버섯 자실체의 갓과 대에서 아미노산 및 유리아미노산 함량 비교)

  • The composition and content of amino acid and free amino acid were separately compared in cap and stipe of fruitbody obtained from different color strains in winter mushroom and oyster mushroom strains. The result showed different kinds and amount of amino acids according to the mushroom parts, strains and kinds. Tryptophane was not detected in any mushrooms. Alanine was also not detected in all oyster mushroom strains. The content of glutamic acid was the highest and that of methionine, phenylalanine and cysteine were very low in all mushroom strains. When compared by parts, the ratio of amono acids showed similar tendency in composition. Generally the content of amono acids showed higher in caps than in stipes. The profile and content of free amino acid showed a lot of variation between cap and strip in the intra and inter mushrooms.

  • The production scale of mushroom cultivation in Korea is approximately 600 billion won, which is 1.6% of the Korean gross agricultural output. Annually, ca. 190,000 tons of mushrooms are harvested in Korea. Although the numbers of mushroom farms and cultivators are constantly decreasing, the total mushroom yields are increasing due to the large-scale cultivation facilities and automation. The recent expansion of the well-being trend causes increase in mushroom consumption in Korea: annual per capita consumption of mushroom was 3.9kg ('13) that is a little higher than European's average. Thus the exports of mushrooms, mainly Flammulina velutipes and Pleurotus ostreatus, have been increased since the middle of 2000s. Recently, however, it is slightly reduced. However, Vietnam, Hong Kong, the United States, the Netherlands and continued to export, and the country has increased recently been exported to Australia, Canada, Southeast Asia and so on. Canned foods of Agaricus bisporus was the first exports of the Korean mushroom industry. This business has reached the peak of the sale in 1977-1978. As Korea initiated trade with China in 1980, the international prices of mushrooms were sharply fall that led to shrink the domestic markets. According to the high demand to develop new items to substitute for A. bisporus, oyster mushroom (Pleurotus ostreatus) was received the attention since it seems to suit the taste of Korean consumers. Although log cultivation technique was developed in the early 1970s for oyster mushroom, this method requires a great deal of labor. Thus we developed shelf cultivation technique which is easier to manage and allows the mass production. In this technique, the growing shelf is manly made from fermented rice straw, that is the unique P. ostreatus medium in the world, was used only in South Korea. After then, the use of cotton wastes as an additional material of medium, the productivity. Currently it is developing a standard cultivation techniques and environmental control system that can stably produce mushrooms throughout the year. The increase of oyster mushroom production may activate the domestic market and contribute to the industrial development. In addition, oyster mushroom production technology has a role in forming the basis of the development of bottle cultivation. Developed mushroom cultivation technology using bottles made possible the mass production. In particular, bottle cultivation method using a liquid spawn can be an opportunity to export the F.velutipes and P.eryngii. In addition, the white varieties of F.velutipes were second developed in the world after Japan. We also developed the new A.bisporus cultivar "Sae-ah" that is easy to grown in Korea. To lead the mushroom industry, we will continue to develop the cultivars with an international competitive power and to improve the cultivation techniques. Mushroom research in Korea nowadays focuses on analysis of mushroom genetics in combination with development of new mushroom varieties, mushroom physiology and cultivation. Further studied are environmental factors for cultivation, disease control, development and utilization of mushroom substrate resources, post-harvest management and improvement of marketable traits. Finally, the RDA manages the collection, classification, identification and preservation of mushroom resources. To keep up with the increasing application of biotechnology in agricultural research the genome project of various mushrooms and the draft of the genetic map has just been completed. A broad range of future studies based on this project is anticipated. The mushroom industry in Korea continually grows and its productivity rapidly increases through the development of new mushrooms cultivars and automated plastic bottle cultivation. Consumption of medicinal mushrooms like Ganoderma lucidum and Phellinus linteus is also increasing strongly. Recently, business of edible and medicinal mushrooms was suffering under over-production and problems in distribution. Fortunately, expansion of the mushroom export helped ease the negative effects for the mushroom industry.

Fruitbody Development of Pleurotus ostreatus via Bottle Cultivation Using Recycled Substrate

  • This study was carried out to determine the possibility of bottle cultivation utilizing recycled oyster mushroom culture waste as a cultivating substrate for P. ostreatus. Total nitrogen percentage was 0.76%, 1.13%, 1.16%, 1.36%, and 1.38% in the 1-, 2-, 3-, 4-, and 5-time mixed substrate, respectively; 0.95%, 1.04%, 1.34%, 1.36%, and 1.25% in the 1-, 2-, 3-, 4-, and 5-time postharvest substrate, respectively; and 0.72% and 0.68% in the 2- and 3-time nonadditive substrate, respectively. Weight of the fresh fruiting body harvest was 115 g, 120 g, 117 g, 118 g, and 114 g on 1-, 2-, 3-, 4-, and 5-time mixed substrate, respectively; and 105 g and 45 g on 2- and 3-time nonadditive substrate, respectively. The first mixed substrate (fresh) and recycled substrates generated no significant difference in the weight of fresh fruiting bodies harvested.

Development of new substrate using redginseng marc for bottle culture of oyster mushroom (Pleurotus ostreatus) (홍삼박을 이용한 병재배 느타리버섯의 첨가배지 개발)

  • This study was carried out to investigated mixing ratio of redginseng marc using organic waste resource for production cost-reduction of oyster mushroom. Redginseng marc were examined as substitute of cottonseed meal which was primary nutritive material of mushroom growing substrate. Total nitrogen and carbon source of redginseng marc was 45% and 2.7%, respectively and C/N ratio was 16.7. Total nitrogen source and pH of substrate mixed with redginseng marc was 4.6~4.9 and 2.5~2.8, respectively. The contents of $P_2O_5$, $K_2O$ and MgO were decreased by increasing redginseng marc, but increased at the 20% redginseng marc. The contents of CaO was increased by increasing redginseng marc, but there was no significant difference in $Na_2O$ content. The more mixing ratio of redginseng marc was increased in column test, the more mycelial growth growed slowly. Yields of fruiting body was no significant difference compared with control group by adding of 10~50% redsingeng marc instead of cottonseed meal, but decreased sharply at only 20% redsingeng marc without cottonseed meal. The L value of pileus by increasing redginseng marc decreased during mushroom harvest, but there was no significant difference in the a-value and the b-value.

Characterization and breeding of a new cultivar Pleurotus ostreatus 'Heuksol' (느타리버섯 신품종 '흑솔'의 육성 및 특성)

  • Oyster mushroom is a type of mushroom that is commonly cultivated and consumed in Korea. P. ostreatus 'Suhan' is a preferred cultivar for many mushroom farmers because it has a dark pileus and thick stipe. However, as it is very sensitive to environmental conditions, farmers consistently demand an alternative cultivar. To develop a new cultivar, the parental strains KMCC01680 ('Suhan') and KMCC00478 ('Gosol') were selected from various collected P. ostreatus strains by cultivating genetic resources. P. ostreatus 'Heuksol' was developed by the method of Mon-Mon crossing between monokaryotic strains derived from 'Suhan' and 'Gosol'. Thirty strains of 174 crossed strains were initially selected by cultivation experiments. After bulk cultivation tests, 'Heuksol' was selected. The nuclear DNA profile of 'Heuksol' was similar to those of the parental strains, 'Suhan' and 'Gosol', when RAPD (random amplified polymorphic DNA) primers and UPF (Universal PCR Fingerprinting) 2, 3, and 4 were used. The optimum temperature for mycelial growth was $30^{\circ}C$ for 'Heuksol', but medium-high temperatures were also appropriate, especially $13-20^{\circ}C$. The fruiting body production per bottle (1,100 mL) was approximately 140.8 g. When compared to the control strain 'Suhan', the thickness of the stipe of 'Heuksol' was greater than that of 'Suhan' (13.5 mm vs 9.4 mm). The pileus diameter of 'Heuksol' was similar to that of 'Suhan' and the pileus thickness of 'Heuksol' and 'Suhan' was 19.7 mm and 21.8 mm, respectively. 'Heuksol' had more a productive stipe number than 'Suhan' and the pileus of 'Heuksol' was dark gray, even at high temperatures. Therefore, it was suggested that this new cultivar, 'Heuksol', could provide an alternative to 'Suhan' and contribute to the profit of oyster mushroom farms.
