  • Thirty-one medicinal plants in Korea are screened for anticholinergic and oxytocic activity The extracts of Cyperus rotundus L. and Scutellaria baicalensis showed anticholinergic action and other seven kinds of extracts showed weak action. The oxytocic action is observed with Tarraxacum platycarpium and Albizzia julibrissin $D_{URZ}$.

  • Subject : The application of eight extra Meridians theory to birth physiology of Korean Medicine. Objective : This study suggests physiological theories in child birth in Korean Medicine. Method : First, the application of eight extra Meridians and extraordinary organs theories to physiology, pathology, pregnancy, postnatal period of women were examined based on major publications and theories of some historical characters. This work provided clues for application of eight extra Meridians theory to birth physiology. Second, the progressing birth physiology of modern medicine based on the importance of the brain function was examined, with which the application of eight extra Meridians and extraordinary organs theories to the birth process showed close relationship. Conclusion : The following conclusions could be drawn. In traditional publications of Korean Medicine, the contents explaining birth physiology with eight extra Meridians theory were scarce, mainly due to the doctor's perception of birth as part of the natural female physiology along with menstruation and pregnancy. However, the contents explaining menstruation, pregnancy, postnatal physiology and pathology through eight extra Meridians theory could be found in classical texts such as Neijing, Jebyeong-wonhuron, Imjeunginameuian. Therefore, it is reasonable to apply the eight extra Meridians theory to the birth process as well. Modern medicine's understanding that pituitary and hypothalamus hormones are in play on womb in the birth process, such as Oxytocin, is in continuous progress. Moreover, the environmental condition that has been traditionally known ideal to the birth process in both the East and West-dark, warm, quiet and secluded-is being confirmed in modern research. The application of eight extra Meridians theory to birth physiology can help to explain these modern information, and thus make them more acceptable.

Effect of Water Extract of Maydis Stigma on Motility of Isolated Rabbit Uterus (옥수수 수염의 물 추출물이 토끼 적출 자궁운동에 미치는 영향)

  • Zea mays Linne belonging to Graminae family has been ordinarily made use of foods and fodder. Maydis Stigma, the stigma of maize, has been discarded without any special application with the exception of being applied as diuretic in the sphere of Chinese medicine, moreover, the pharmacological research toward Maydis Stigma as a medical herb has not been regretfully undertaken at all. The authors hereby paid attention to this point of view and made experiment to find out whether the water extract of Maydis Stigma might possess some pharmacological effect on the motility of the isolated rabbit uterus. The experiment was performed making the use of several agents related to the uterine motility such as acetylcholine, pilocarpine, epinephrine, atropine, barium chloride, quinine sulfate and oxytocin. The motility of the isolated rabbit uterus in Tyrode' solution was recorded with the electric kymograph according to Magnus method. The results of the experiment are as follows. 1. The motility of the isolated pregnant and nonpregnant rabbit uterus represents the tendency of gradual contractility in proportion to the concentration of ZW $10^{-4}$, $5{\times}10^{-4}$ and $10^{-3}$. 2. The uterine contractile effect of ZW $10^{-3}$ probably seems to antagonize the effect of epinephrine $10^{-6}$. 3. ZW $10^{-3}$ does not seem to have the significance with barium chloride, quinine sulfate and oxytocin respectively.

  • Since the blastocyst is broken and spreads out on a flat plastic culture dish (two dimensional culture) during in vitro development, it has been difficult to study the implantation process. It also has been difficult to analyse the interactions between endometrial epithelial and stromal cells because of the lack of a long-term in vitro model which can stimulate in vivo characteristics, as these cells eventually fail to proliferate or cease to express differentiated functions. Recently nontransformed cell lines, CUE-P and CUS-V2, derived from rat endometrial epithelium and stroma were reported. In this study, morphology of CUE-P and CUS-V2 was examined and oxytocin gene expression by CUE-P cells was demonstrated by RT-PCR. The CUE-P cells have a cuboidal morphology and CUS-V2 cells resemble fibroblast and exhibit a spindle-like morphology. In RT-PCR, same size of PCR products of oxytocin gene at hypothalamus, uterus and CUE-P cells were demonstrated. These results showed three dimensional culture system could be made by using the new cell lines.

Effects of $Ca^{2+}$ and $K^+$ on the Spike Action Potentials in Oxytocin-induced Uterine Contractions (칼슘 및 칼륨이온이 흰쥐 자궁근(子宮筋) 활동전압(活動電壓)에 미치는 영향(影響))

  • The influences of extracellular $Ca^{2+}\;and\;K^+$ upon the spike action potentials were studied in isolated uterine strips of rat. Regular, rhythmic uterine contractions were induced by the administration of oxytocin$(0.2{\sim}0.5\;I.U.)$, and recorded with force transducer. Spike action potentials were extracellularly measured by use of suction electrode, and compared with those recorded intracellularly by glass microelectrode. The results obtained were as follows : 1) The frequency and duration of spike bursts, and the number of spikes in a burst could be analyzed by use of both methods. But the absolute values of membrane potential were not measurable with the suction electrode. 2) The duration of contraction$(CD_{90};\;the\;duration\;of\;90%\;relaxation)$ was lengthened from the control 17.0 sec to 20.6 sec, in parallel with the increase of spike number from the control 21 to 26, as the increase in $Ca^{2+}$ concentration from 2 to 4 mM. 3) The amplitude and frequency of contractions were gradually decreased, simultaneously with the decrease in the number of spikes in a burst, when the $Ca^{2+}-antagonist$, verapamil was administered cumulatively. 4) The number of spikes was changed from the control 15 to 7, in cabs of the administration of ver)'low dose of verapamil$(10^{-6}\;g/l)$. 5) Increase in the numbers of spike bursts was well matched to the increase in frequency of contractions when extracellular $K^+$ was increased.

Effects of Different Music Genres on the Stress Levels of Jeju Crossbred Horses (마방에서 음악장르에 따른 Jeju crossbred의 스트레스 변화 연구)

  • This study investigated the effects of three music genres (classical, new age, and rock) on the stress levels of six Jeju crossbred horses (Jeju horse × Thoroughbred) in a horse stable. The horses were exposed to the three genres for seven days, and their stress levels were measured by analyzing physiological markers, including neurotransmitter (cortisol, β-endorphin, dopamine, serotonin, and oxytocin) plasma levels and creatine phosphokinase (CPK) and aldolase serum levels. The neurotransmitter analysis showed significant differences in cortisol levels between classical and new age music exposure. Dopamine levels decreased significantly only with new age exposure. Although there were no significant differences in β-endorphin levels between the three genres, β-endorphin levels decreased with increasing classical and new age music playback times and increased with increasing rock music playback times. There were no significant differences in serotonin levels between the three genres. Oxytocin levels decreased significantly with exposure to classical and rock music. The CPK and aldolase analyses showed that CPK levels decreased significantly only with exposure to new age music and increased after playback ended, while aldolase levels decreased significantly with classical and new age music exposure and increased after playback ended. These findings suggest that classical music and new age are the optimal music genres for the psychological stability of Jeju crossbred horses. Playing back an appropriate music genre could be used to improve breeding and promote the welfare of Jeju crossbred horses.

Studies on the Effects of Chokyungsan and Chunkeumchokyungtang (조경산(調經散)과 천금조경탕(千金調經湯)의 효능(效能)에 대(對)한 실험적(實驗的) 연구(硏究))

  • To elucidate the effects of Chokyungsan and Chunkeunchokyungtang, after oral administration of Chokyungsan and Chunkeunchokyungtang water extract in mice and rats, acute toxicity, analgesic, sedative, esoogenic actions, action on isolated uterine muscle were measured. The rlesults obtained were as follows: 1. The yield of water extract of Chokyungsan and Chunkeunchokyungtang was 24.5%, 32.2%, minimum lethal dose was 4,000mg/kg, which rarely had the acute toxicity in mice and rats. 2. The analgesic effects of Chokyungsan and Chunkeunchokyungtang by acetic acid induced writhing syndrome in mice were not remarkaely observed. 3. The relaxant action of Chokyungsan on on oxytocin induced contracted uterine muscle in estrogenized rats were not remarkably observed, but Chunkeunchokyungtang were remarked. 4. The sedative effects of Chokyungsan and Chunkeunchokyungtang by hexobabital sodium induced sleeping time in mice. 5. administration of Chokyungsan and Chunkeunchokyungtang caused remarkable increase in weight of rat's uterus.

  • General pharmacological properties of ι-muscone, the major component of musk, were investigatedin mice, rats and guinea pig. The administration of ι-muscone (1, 10, 100 mg/kg, p.o.) in mice had no effects in general behaviors, and no influences on analgesic actions and normal body temperature. Muscle relaxant action, intestinal propulsion and gastric secretion were not observed even at the high dose of 100 mg/kg. ι-Muscone (1, 10, 100 mg/kg, p.o.) given to conscious rats showed no effect on mean blood pressure and heart rate. It showed no direct effect at 2.4$\times$10.3 mg/ml and 2.4$\times$10-2 mg/ml in isolated uterus of rats and ileum of guinea pig, and also had no inhibition of contraction induced by oxytocin and histamine.
