Pluronic F-68 and oxygen vectors were applied to increase the cell growth of Angelica gigas Nakai in aqueous two-phase system (ATPS). ATPS was composed of 3.6% (w/v) polyethylene glycol (PEG) 20,000 and 2.8% (w/v) crude dextran. n-Hexadecane, n-dodecane and FC-40 were used as oxygen vectors to enhance the oxygen transfer in ATPS. With 2$\sim$10 g/L of Pluronic F-68, addition of of n-hexadecane and FC-40 significantly enhanced the oxygen transfer rate as well as the maximum cell mass in a medium with ATPS. However, n-dodecane reduced the cell growth in all treatments. Maximum cell densities were increased up to 27.5% with 10 g/L of Pluronic F-68 and up to 40.2% with 8% (v/v) n-hexadecane compared to those of the controls without Pluronic F-68 and oxygen vectors. It was confirmed that the cell growth could be increased in ATPS using n-hexadecane.
Effects of supplementing various 0.1-1.0%(w/v) surfactants and 0.1-1.0%(v/v) oxygen vectors on postthaw growth following cryopreservation of transgenic Nicotiana tabacum(GUS) in suspension culture were investigated. The postthaw cell growth was increased 20.68% and 23.66% compared to control when Pluronic F-68 and polyethylene glycol(PEG) were added in recovery medium respectively. Addition of n-hexadecane and FC-40 also enhanced the recovered growth by 14.89% and 20.68%. These results demonstrate that the marked Protection could be achieved by using surfactants and oxygen vectors for plant cells recovered from cryostorage.
It has been commonly known that cell growth is inhibited by the lack of dissolved oxygen and mass transfer inhibition of nutrients at stationary phase in fed-batch culture. In this study, Pluronic F-68 and oxygen vectors were added in Angelica gigas Nakai suspension culture in order to enhance cell growth in fed-batch culture. It was observed that the addition of 6%(w/v) Pluronic F-68 promoted cell growth up to 6.1% compared to control and that the use of 4%(v/v) n-hexadecane markedly enhanced cell growth up to 11.4%.
The effects of aliphatic hydrocarbons (n-hexadecane and n-dodecane) on the volumetric oxygen mass transfer coefficient $(k_L\;a)$ were studied in flat alveolar airlift reactor and continuous stirred tank reactors (CSTRs). In the flat alveolar airlift reactor, high aeration rates (>2vvm) were required in order to obtain efficient organic-aqueous phase dispersion and reliable $k_La$ measurements. Addition of 1% (v/v) n-hexadecane or n-dodecane increased the $k_La$ 1.55- and 1.33-fold, respectively, compared to the control (superficial velocity: $25.8{\times}10^{-3}m/s$, sparger orifice diameter: 0.5 mm). Analysis of the gas-liquid interfacial area a and the liquid film mass transfer coefficient $k_L$ suggests that the observed $k_La$ increase was a function of the media's liquid film mass transfer. Addition of 1% (v/v) n-hexadecane or n-dodecane to analogous setups using CSTRs led to a $k_La$ increase by a factor of 1.68 and 1.36, respectively (superficial velocity: $2.1{\times}10^{-3}m/s$, stirring rate: 250 rpm). These results propose that low-concentration addition of oxygen-vectors to aerobic microbial cultures has additional benefit relative to incubation in purely aqueous media.
Dissolved oxygen level of cell culture media has a critical effect on cellular metabolism, which governs specific productivity of recombinant proteins and mammalian cell growth However, in the cores of cell aggregates or cell-immobilized beads, oxygen level frequently goes below a critical level. Mammalian cells have a number of genes induced in the lower level of oxygen, and the genes contain a common cis-acting element (-RCGTG-), hypoxia response element (HRE). By binding of hypoxia inducible factor-l (HIF-I) to the HRE, promoters of hypoxia inducible genes are activated, which is a survival mechanism. In this work, to develop a CHO cell capable of producing recombinant proteins in immobilization and high density cell culture efficiently, mammalian expression vectors containing human tissue-type plasminogen activator (t-PA) gene controlled by HRE were constructed and stably transfected into the CHO cells. In $Ba^{2+}$ -alginate immobilization culture, CHO/pCl/dhfr/2HRE-t-PA cells produced 2 folds higher recombinant t-PA activity than CHO/pCl/dhfrlt-PA cells without $CoCl_2$ treatment. Furthermore, in repeated fed batch culture, productivity of t-PA in immobilized CHO/pCI/dhfr/2HRE-t-PA cells was 121 ng/ml/day, total production of 0.968 mg/day at 11 days culture while CHO/pCIIdhfrlt-PA cells was 22.8 ng/ml/day. All these results indicate that HRE is very useful for the enhancement of protein productivity in mammalian cell cultures.

Plasmid DNA (pDNA) is a product of interest for many biopharmaceutical companies and research laboratories, because of increase in the number of gene therapy protocols that use nonviral vectors. This work was undertaken to study the effect of antibiotic and dissolved oxygen concentration (DOC) on the production of a ColE 1-type plasmid (pVAX1-LacZ) hosted in Escherichia coli $DH5\alpha$ and cultured in a batch fermentor with 0.751 of Terrific Broth. A decrease in the DOC from $60\%\;to\;5\%$ was shown to increase the specific pDNA concentration approximately 1.5-fold, due to the downregulation of growth. Additionally, this increase in the pDNA concentration led to a 2.2-fold increase in the purity of cell lysates obtained after cell lysis. However, the use of higher DOC led to 2.8-fold higher volumetric productivity as a consequence of a faster growth rate, reducing the fermentation time from 24 to 8 h. Interestingly, the specific pDNA concentration, and pDNA productivity and purity were always higher $(10-15\%)$ in the absence of antibiotic. Overall, the data indicate that nonselective conditions can be used without compromising yield, productivity, and purity of pDNA.
Hydrogen is seen as one of the important energy vectors of the next century. Hydrogen as a renewable energy source, provides the potential for a sustainable development particularly in the transportation sector. Hydrogen driven vehicles reduce both local as well as global emissions. The laboratory of transporttechnology (University of Gent) converted a GM/Crusader V-8 engine for hydrogen use. Once the engine is optimised, it will be built in a low-floor midsize hydrogen city bus for public demonstration. For a complete control of the combustion process and to increase the resistance to backfire (explosion of the air-fuel mixture in the inlet manifold), a sequential timed multipoint injection of hydrogen and an electronic management system is chosen. The results as a function of the engine parameters (ignition timing. injection timing and duration, injection pressure) we given. Special focus is given to topics related to the use of hydrogen as a fuel: ignition characteristics (importance of electrode distance), quality of the lubricating oil (crankcase gases with high contents of hydrogen), oxygen sensors (very lean operating conditions), noise reduction (configuration and length of inlet pipes). The advantages and disadvantages of a power regulation only by the air to fuel ratio (as for diesel engines) against a throttle regulation (normal gasoline or gas regulation) are examined. Finally the goals of the development of the engine are reached: power output of 90 kW, torque of 300 Nm, extremely low emission levels and backfire-safe operation.
Plant viruses are responsible for worldwide production losses of numerous economically important crops. The most common plant RNA viruses are positivesense single-stranded RNA viruses [(+)ss RNA viruses]. These viruses have small genomes that encode a limited number of proteins. The viruses depend on their host's machinery for the replication of their RNA genome, assembly, movement, and attraction to the vectors for dispersal. Recently researchers have reported that chloroplast proteins are crucial for replicating (+)ss plant RNA viruses. Some chloroplast proteins, including translation initiation factor [eIF(iso)4E] and 75 DEAD-box RNA helicase RH8, help viruses fulfill their infection cycle in plants. In contrast, other chloroplast proteins such as PAP2.1, PSaC, and ATPsyn-α play active roles in plant defense against viruses. This is also consistent with the idea that reactive oxygen species, salicylic acid, jasmonic acid, and abscisic acid are produced in chloroplast. However, knowledge of molecular mechanisms and functions underlying these chloroplast host factors during the virus infection is still scarce and remains largely unknown. Our review briefly summarizes the latest knowledge regarding the possible role of chloroplast in plant virus replication, emphasizing chloroplast-related proteins. We have highlighted current advances regarding chloroplast-related proteins' role in replicating plant (+)ss RNA viruses.
The aim of this study was to examine the metabolic adjustment of lactate dehydrogenase (EC 1.1.1.27, LDH) isozymes to a change in dissolved oxygen (DO) in bluegill (Lepomis macrochirus). After bluegills were adapted to a constant environment in an aquarium, the DO was changed to investigate the activity of LDH isozyme and the relative ratio of subunits A, B, and C for each tissue. When the DO was decreased from 18 ppm to 6 ppm, LDH in skeletal muscle, heart, and brain tissues recovered to the level of control activity within 12, 12, and 6 hr, respectively. LDH activity changed in accordance with a change in DO. The compensation was performed rapidly and is thought to be an important function of LDH in enabling bluegills to adapt to their environment. In bluegill heart, eye, and brain tissues, the relative ratio of subunit A increased and showed a tendency to recover similarly to the subunit ratio of control groups up to 12 hr. It is thought that the anaerobic metabolism using subunit A was increased in the initial stage when DO was changed. In addition, the results revealed that subunit C was more similar to subunit A than subunit B. In bluegills, subunits A and C of LDH seem to be evolutionarily similar. LDH isozymes, mainly containing subunits A and C, are likely responsible for the function of pyruvate reductase, which plays a role in making the bluegill adapt to a hypoxic environment through anaerobic metabolism.
The dissolved oxygen level of any cell culture environment has a critical effect on cellular metabolism. Specifically, hypoxia condition decreases cell viability and recombinant protein productivity. In this work, to develop CHO cells producing recombinant protein with high productivity, mammalian expression vectors containing a human tissue-type plasminogen activator (t-PA) gene with hypoxia response element (HRE) were constructed and stably transfected into CHO cells. CHO/2HRE-t-PA cells produced 2-folds higher recombinant t-PA production than CHO/t-PA cells in a $Ba^{2+}-alginate$ immobilized culture, and 16.8-folds in a repeated batch culture. In a non-aerated batch culture of suspension-adapted cells, t-PA productivity of CHO/2HRE/t-PA cells was 4.2-folds higher than that of CHO/t-PA cells. Our results indicate that HRE is a useful tool for the enhancement of protein productivity in mammalian cell cultures.
