Regulation of alkaline-resistant laccase from Perenniporia tephropora KU-Alk4 was proved to be controlled by several factors. One important factor was the initial pH, which drove the fungus to produce different kinds of ligninolytic enzymes. P. tephropora KU-Alk4 could grow at pH 4.5, 7.0, and 8.0. The fungus produced laccase and MnP at pH 7.0, but only laccase at pH 8.0. The specific activity of laccase in the pH 8.0 culture was higher than that in the pH 7.0 culture. At pH 8.0, glucose was the best carbon source for laccase production but growth was better with lactose. Low concentrations of glucose at 0.1% to 1.0% enhanced laccase production, while concentrations over 1% gave contradictory results. Veratryl alcohol induced the production of laccase. A trace concentration of copper ions was required for laccase production. Biomass increased with an increasing rate of aeration of shaking flasks from 100 to 140 rpm; however, shaking at over 120 rpm decreased laccase quantity. Highest amount of laccase produced by KU-Alk4, 360 U/mL, was at pH 8.0 with 1% glucose and 0.2 mM copper sulfate, unshaken for the first 3 days, followed by addition of 0.85 mM veratryl alcohol and shaking at 120 rpm. The crude enzyme was significantly stable in alkaline pH 8.0~10.0 for 24 hr. After treating the pulp mill effluent with the KU-Alk4 system for 3 days, pH decreased from 9.6 to 6.8, with reduction of color and chemical oxygen demand at 83.2% and 81%, respectively. Laccase was detectable during the biotreatment process.
The Biochemical Methane Potential (BMP) of fresh leachate and domestic wastewaters codigestion was determined by laboratory Bach Tests at $35^{\circ}C$ over a period of 90 d using a wide range of leachates volumetric ratios from 0% to 100%. To simulate wastewaters plant treatment step, all the ratios were first air stripped for 48 h before anaerobic incubation. The kinetic of biogas production was assessed using modified Gompertz model and exponential equation. The results obtained showed that cumulative biogas production was insignificant in the case of wastewaters monodigestion while the codigestion significantly improves the BMP. Air stripping pretreatment had positive effect on both ammonium concentration and volatiles fatty acids with reduction up to 75% and 42%, respectively. According to the Modified Gompertz model, the optimal anaerobic co-digestion conditions both in terms of maximal biogas potential, start-up period and maximum daily biogas production rate, could be achieved within large leachate volumetric ratios from 25% to 75% with a maximum BMP value of 438.42 mL/g volatile solid at 50% leachate ratio. The positive effect of codigestion was attributed to a dilution effect of chemical oxygen demand and volatile fatty acid concentrations to optimal range that was between 11.7 to $32.3gO_2/L$ and 2.1 to 7.4 g/L, respectively. These results suggested that the treatment of fresh leachate by their dilution and co digestion at wastewaters treatment plants could be a promising alternative for both energetic and treatment purposes.

At a time like this I was studied the effect on nutrients removal through lighting period in aerobic high rate pond and we know that waste stabilization pond method is the most economical and energy saving wastewater treatment technology than others. At the results which was studied through operating the reactor-l artifically main-tained at a temperature, $25^{\circ}C$, a light intensity, 3000lux, and a lighting period, 24hrs and the reactor-2 artifically maintained at a tern야rature, $25^{\circ}C$ and a light intensity 3000lux, and a lighting period period, 12hrs, It has appeared for 24hrs.-lighting period -reactor-1 to be prior to the reactor-2. The attained results are that 1. reactor-1 is prior to reactor-2 on oxygen-generation 2. reactor-1 is prior to reactor-2 on algal production 3. COD removal efficiency, 90.76%, T-N removal efficiency, 80%, T-P removal e efficiency, 74.47 % in reactor-2, in reactor-1 COD removal efficiency, 94.85 %, T-N removal efficiency, 98.07%, T-P removal efficiency, 72.13% are, so the treatment efficiency of reactor-1 is more excellent than things of reactor-2 4. it appeared that the detention time is 8, 9days.
There is a significant demand to make various dissolved gases in water. However, the conventional aeration method shows low gas mass transfer rate and gas utilization efficiency. In this study, a novel rotating wheel entraining gas method was developed for making high dissolved $O_2$ and $O_3$ in water. It produced higher concentration and higher transfer rate of dissolved $O_2$ and $O_3$ than conventional bubble aeration method, especially almost 100% of gas transfer efficiency was achieved for $O_3$ in enclosed reactor. For application of rotating wheel entraining gas method, aerobic bio-reactor and membrane bio-reactor (MBR) were successfully used for treatment of domestic and pharmaceutical wastewater, respectively; and vacuum ultraviolet $(VUV)/UV+O_3/O_2$ reactors were well used for sterilization in air/water, removal of dust particles and toxic gases in air, and degradation of pesticide residue and sterilization on fruits and vegetables.
Purpose: Performance of slurry composting and biofiltration with methane production (SCB-M) using swine manure and sawdust was evaluated. The suitable specific liquid input (SLI) was determined at lab-scale SCB. Method: In lab-scale SCB, the SLI test was performed at liquid input rate of 0.04, 0.09, $0.17cc/cm^3$ with constant sawdust volume. In pilot-scale SCB-M, the swine manure was fed to methane digester at organic loading rate (OLR) of 0.25-0.5 g VS/L/d. The effluent from methane digester was filtered using SCB. Results: The SLI at $0.04cc/cm^3$ showed good performance in terms of retention time. In pilot-scale SCB, the removal of $NH_3$-N and total nitrogen (T-N) was found to be around 59% and 28%, respectively. Similarly, volatile fatty acid (VFA) and total chemical oxygen demand (TCOD) removal was found be 56% and 43%, respectively. Conclusions: For SCB-M process, the SLI of $0.04cc/cm^3$ is recommended. The performance of swine manure treatment was improved more by using SCB-M system than using methane digester only.

Background: Those classified as Tae-Eum (TE)-type people in Sasang constitutional medicine (SCM) are prone to obesity. Although extensive clinical observations have confirmed this tendency, the underlying physiological mechanisms are unknown. Here, we propose a novel hypothesis using integrative physiology to explain this phenomenon. Methods: Hypoactive lung function in the TE type indicates that respiration is attenuated at the cellular level - specifically, mitochondrial oxygen consumption. Because a functional reduction in cellular energy metabolism is suggestive of intrinsic hypoactivity in the consumption (or production) of metabolic energy, we reasoned that this tendency can readily cause weight gain via an increase in anabolism. Thus, this relationship can be derived from the graph of cellular metabolic power plotted against body weight. We analyzed the clinical data of 548 individuals to test this hypothesis. Results: The statistical analysis revealed that the cellular metabolic rate was lower in TEtype individuals and that their percentage of obesity (body mass index >25) was significantly higher compared to other constitutional groups. Conclusion: Lower cellular metabolic power can be an explanation for the obesity trend in TE type people.
Objective: The objective of this study was to examine the effect of using a tunnel ventilation system within the dairy barn environment upon the productivity of dairy cows during the winter season. Methods: The study was performed at the University Farm, Faculty of Agriculture, Utsunomiya University. Twenty-one Holstein dairy cows (5 heifers and 16 multiparous) were enclosed in a stall barn. Unventilated (UV) and tunnel-ventilated (TV) was operated by turns every other week, and a number of key parameters were measured in the barn, including tunnel ventilation output, temperature, relative humidity, gas concentrations (oxygen [$O_2$], carbon dioxide [$CO_2$], and ammonia [$NH_3$]). Also, skin and rectal temperature, respiratory rate, blood gas concentrations, and bacterial count were measured from nipple attachments on ten cows. The amount of fodder left uneaten, and general components and somatic cell count of the milk were measured. Results: As for our dairy barn environment, air temperature dropped significantly with the passage of time with TV. Humidity was significantly higher with TV at 0600 h compared to UV, while $CO_2$ and $NH_3$ concentrations with UV were significantly higher than with TV at 0000 h and 0600 h. Skin temperature was significantly lower with TV compared to UV at 0000 h and 0600 h. Respiratory rate was also significantly lower at 0600 h with TV than with UV. Bacterial count for the nipple attachments was significantly lower with TV than with UV at 0600 h. The amount of leftover fodder was significantly less with TV in comparison with UV. Conclusion: Our results suggest that a TV system in the winter barn results in environmental improvements, such as reductions in unfavorable gas concentrations and bacterial growth. Consequently, it is expected that barns utilizing a TV system will be beneficial for both animal health and production.

Anaerobic digestion (AD) refers to the biological process which can convert organic substrates to biogas in the absence of oxygen. The aim of this study was to determine the capability of feedstock to produce biogas and to quantify the biogas yield from different feedstocks. A co-digestion approach was carried out in a continuous stirred tank reactor operated under mesophilic conditions and at a constant organic loading rate of 0.0756 g COD/ L.day, with a hydraulic retention time of 25 days. For comparison, mono-digestion was also included in the experimental work. 2 L working volumes were used throughout the experimental work. The seed culture was obtained from composting as substrate digestion. When the feedstock was added to seeding, the biogas started to emit after three days of retention time. The highest volume of biogas was observed when the seeding volume used for 1000mL. However, the lowest volume of biogas yield was obtained from both co-digestion reactors, with a value of 340 mL. For methane yield, the highest methane production rate was 0.16 L CH4/mg. The COD with yield was at 8.6% and the lowest was at 0.5%. The highest quantity of methane was obtained from a reactor of Euphorbiaceae peel with added seeding, while the lowest methane yield came from a reactor of Euphorbiaceae stems with added seeding. In this study, sodium bicarbonate (NaHCO3) was used as a buffering solution to correct the pH in the reactor if the reactor condition was found to be in a souring or acidic condition.
Objective: We increased the nuclear maturation rate of antral follicle derived oocytes by using a pre-in vitro maturation (IVM) culture system and improved the developmental potential of these porcine pathenotes by supplementing with melatonin. Furthermore, we investigated the expression patterns of genes involved in cumulus expansion (HAS2, PTGS2, TNFAIP6, and PTX3) derived from small and medium antral follicles before and after oocyte maturation. Methods: Only the cumulus oocyte-complexes (COCs) derived from small antral follicles were induced with [Pre-SF(+)hCG] or without [Pre-SF(-)hCG] the addition of human chorionic gonadotropin (hCG) during the last 7 h of the pre-IVM period before undergoing the regular culture system. The mature oocytes were investigated on embryonic development after parthenogenetic activation (PA). Melatonin (10-7 M) was supplemented during in vitro culture (IVC) to improve the developmental potential of these porcine pathenotes. Results: A pre-IVM culture system with hCG added during the last 7 h of the pre-IVM period [Pre-SF(+)hCG] effectively supported small antral follicle-derived oocytes and increased their nuclear maturation rate. The oocytes derived from medium antral follicles exhibited the highest nuclear maturation rate in a regular culture system. Compared with oocytes cultured in a regular culture system, those cultured in the pre-IVM culture system exhibited considerable overexpression of HAS2, PTGS2, and TNFAIP6. Porcine embryos treated with melatonin during IVC exhibited markedly improved quality and developmental competence after PA. Notably, melatonin supplementation during the IVM period can reduce and increase the levels of intracellular reactive oxygen species (ROS) and glutathione (GSH), respectively. Conclusion: Our findings indicate that the Pre-SF(+)hCG culture system increases the nuclear maturation rate of small antral follicle-derived oocytes and the expression of genes involved in cumulus expansion. Melatonin supplementation during IVC may improve the quality and increase the blastocyst formation rate of porcine embryos. In addition, it can reduce and increase the levels of ROS and GSH, respectively, in mature oocytes, thus affecting subsequent embryos.

We performed the experiment to evaluate the effect of different DO concentrations (0.5, 4.5 and 9.0 $mgO_2L^{-1}$) and water depths (20, 50 and 80 cm) on the filtering rate, mortality, and pseudifeces production of Unio douglasiae against the cyanobacterial bloom (mainly Microcystis aeruginosa). A solitary-living bivalve U. douglasiae was collected in the upstream region of the North Han River (Korea). The harvested mussels were carefully transferred to the laboratory artificial management system, which was controlled temperature $(18{\pm}2^{\circ}C)$, flow rate (10L $h^{-1}$), food $(Chlorella^{TM})$, sediment (pebble and clay), light intensity (ca. $20{\mu}mol$ photons), and photocycle (12 L : 12 D). In the field observation, the mussel mortality was significantly correlated with water temperature, pH and DO concentration (P<0.05). The mortality was decreased with water depth; 65, 90, 80% of mortality at 20, 50, 80 cm water-depth, respectively. Filtering rate (FR) showed the highest value at 50 cm water depth, and thereby the concentration of chlorophyll-${\alpha}$ decreased continuously by 94% of the control at the end of the experiment. In contrast, FR decreased by 34% of the initial concentration at 20 cm water depth. Over the given water-depth range, the mussel FR ranged from $0.15{\sim}0.20L\;gAFDW^{-1}hr^{-1}$ during the 18hrs of experiment, and thereafter, they appeared to be approximately 0.11, 0.26 and 0.30 L $gAFDW^{-1}hr^{-1}$ at 20, 50 and 80cm water depth, respectively. FR was highest with the value of 0.46L $gAFDW^{-1}hr^{-1}\;at\;0.5mgO_2 L^{-1}$ at the early stage of the experiment, while it increased with DO concentration. Maximum pseudofaeces production was 11.2 mg $gAFDW^{-1}hr^{-1}\;at\;9.0mgO_2L^{-1}$. Our results conclude that U. douglasiae has a potential to enhance water quality in eutrophic lake by removing dominant cyanobacteria, but their effects vary with environmental parameters and the water depth at which they are located.
