Effects of Dandelion on Oxygen Free Radical Generating and Scavenging System of Brain in Streptozotocin-Induced Diabetic Rats (서양민들레가 Streptozotocin으로 유발한 당뇨 횐쥐의 뇌조직 중 유해 활성산소 생성 및 제거 효소계에 미치는 영향)

  • Many studies have shown that hyperglycemia leads to an increase of lipid peroxidation in diabetic patients and animals, reflecting a rise reactive oxygen species production. It is increasingly recognized that brain is another site of diabetic organ damage. Accordingly, this study was to investigate the effect of dandelion on oxygen free radical generating and scavenging system of brain in streptozotocin (STZ)-induced diabetic rats. Male Wistar rats were divided into diabetic (control) and diabetic-dandelion supplemented groups. Dandelion was supplemented for 4 weeks with dandelion leaf and root powder (DLP, DRP) or dandelion leaf and root water extract (DLW, DRW) based on 11.4 g of raw dandelion/kg diet. Diabetes was induced by single injection STZ (55 mg/kg B.W., i.p.)in a citrate buffer. Oxygen free radical generating enzymes, cytochrome P-450, amino-pyrine N-demethylase, aniline hydroxylase and xanthine oxidase, were lowered in dandelion supplemented-groups compared to the control group. Superoxide dismutase, catalase and gluthathione peroxidase activities of brain were also lower in dandelion leaf and root supplemented-group than in the control group, whereas glutathione S-transferase activity and gluthathione content were increased in dandelion supplemented-groups compared to the control group. With regard to the lipid peroxidation products, the malondialdehyde content of brain was lower in dandelion supplemented groups. Therefore, it could be suggested that powder and water extract of dandelion leaf or root are beneficial in preventing diabetic complication from lipid peroxidation and free radical in brain of diabetic rat brain.

Effect of Dietary Monascus Pigment on the Liver Damage Induced with $CCl_4$in Rats

  • In the biological world, there are a number of ecological fights for survival between each organism such as plants, animals and microorganism In such events, an organism can use its natural bioactive products as defence agent against other organisms. Furthermore, natural bioactive products can be utilized for medicine or functional food. Recently, we investigate the effect of Monascus pigment extracted from a fungus, Monascus anke, on the alcohol metabolism and blood lipid profile. In the present study, it is observed that Monascus pigment supplemented dietary may have a hepatoprotective effect on rat's liver damage induced with $CCl_4$ . By treatment with $CCl_4$(3 times, I.P), liver damage was reduced more in the rats fed 2% Monascus pigment extract supplemented diet than those fed standard diet, based on the serum levels of alanine aminotransferase, microsomal glucose-6-phosphatse activity and hepaic malondialdehyde content. On the other hand, oxygen free radical generating enzymes, hepatic P-450 dependent aniline hydroxylase, xanthine oxidase, and oxygen free radical scavenging enzymes, hepatic glutathione S-transferase, catalase, superoxide dismutase activities were generally higher both in $CCl_4$ treated group and control fed 2% Monascus pigment extract supplemented diet than those fed standard diet. In conclusion, the rats fed 2% Monascus pigment extract supplemented diet showed more reduced liver damage than those fed standard diet, which may be due to the acceleration of oxygen free radical metabolism.

A study on Histologically Change of the Skin and Liver in Skin Burn (피부화상에 의한 피부 및 간의 조직학적 변화에 관한 연구)

  • In order to investigate a pathogenesis of liver damage induced by skin burn, thermal injury was induced by scald burn on entirely dorsal surface in rats (total body surface area 30%) except for inhalated injury. At 5 and 24 h after scald bum, biochemical assay and morphological changes in skin tissue, serum and liver tissue were examined. The effects of bum injury on the levels of glutathione, lipid peroxide and on the activities of oxygen free radical generating and scavenging enzymes have been determined in association with observing of histologic and ultrastructural changes, measuring the protein concentration in plasma, and counting the number of intravascular polymorphonuclear leukocytes. The activity of xanthine oxidase, an enzyme of oxygen free radical generating system. was elevated (p<0.01) in serum, but not in skin and in liver tissue. Futhermore, thermal injury decreased not only the protein concentration in plasma but also the number of leukocytes, that indicates induction of edema formation with protein exudation and inflammation by neutrophil infiltration into the internal organs. These data suggest that acute dermal scald burn injury leads to liver damage, that is related to elevation of xanthine oxidase activity in serum. Xanthine oxidase may be a key role in the pathogenesis of liver damage induced by skin burn.

The effects of burn injury on the levels of glutathione, lipid peroxide and on the activities of oxygen of histologic and ultrasound changes, measuring. the protein concentration in plasma, and counting the number of intravascular polymorphonuclear leukocytes. Post burn 24 h, the content of glutathione was decreased (47.50%), whereas that of lipid peroxide was increased (37.01%), and the activity of superoxide dismutase was diminished (p<0.001). Thus decreasing the capacity of oxygen free radical scavenging enzymes led to oxidative injury in skin tissue. In liver tissue, at 24 h after scald burn, both the content of glutathione and the activity of catalase were markedly decreased (p<0.01, p<0.05), thus the imbalance between free radical generating and scavenging capacities has been induced.

A Generator of Gaseous Singlet Oxygen

  • In order to prevent side reactions due to free radical formation occurring in the reactions of singlet oxygen generated in solution phse, it was required that singlet oxygen is generated in gas phase from an apparatus of a solid-gas system. We have accomplished to construct an apparatus generating singlet oxygen in solid-gas system, which is composed of a flexible optical fibre tube connected by a dye-sensitizer probe containing rose bengal dye on polymer or inorganic material. Through the optical fibre tube visible light from a laser and an oxygen stream are passed into the sensitizer probe where singlet oxygen is generated. The determination of singlet oxygen was carried out by two methods. One involves the detection of the luminescence of singlet oxygen at 1268 mn and the other involves the chemiluminescence reaction of a dihydroisobenzofuran with singlet oxygen emitting luminescence at 456 nm.

Effects of Cordyceps Sinensis and Cordyceps Militaris on Antioxidation in liver of $CCl_4$-treated rats ($CCl_4$ 유도 간 독성에 대한 동충하초의 항산화 효과)

  • Objectives : This study was performed to investigate the effects of Cordyceps Sinensis (CS) and Cordyceps Militaris (CM) on anti oxidation in the livers of ${CCl_4}-treated$ rats. Methods : Hepatotoxicity in rats was induced by carbon tetracWoride. $CCl_4-induced$ rats were administered with the extract of CS and CM. Results : In vitro, CS and CM didn't affect levels of lipid peroxide and the activities and type conversion ratio of xanthine oxidase. However, hydroxyl radicals and DPPHL radicals were decreased. In vivo, in the ${CCl_4}-treated$ rats, lipid peroxide, the activities and type conversion ratio of xanthine oxidase and superoxide radicals were increased but superoxide dismutase was decreased. After CS and CM were administered to ${CCl_4}-treated$ rats, levels of lipid peroxide, the activities and type conversion ratio of xanthine oxidase and superoxide radicals were decreased but superoxide dismutase was increased. Conclusions : These results suggest that CS and CM decrease the activities of free-radical-generating enzymes which form lipid peroxide and increase the activities of oxygen free radical scavenging enzymes.

Effects of Aralia elata Water Extracts on Activities of Hepatic Oxygen Free Radical Generating and Scavenging Enzymes in Streptozotocin-Induced Diabetic Rats (두릅열수추출물이 당뇨유발 흰쥐의 간조직 중 유해 활성산소 대사효소계 활성에 미치는 영향)

  • Oxidative stress is currently suggested as a mechanism underyling diabetes. Accordingly, the present study was designed to evaluate the effect of Aralia elate water extracts (AEW) on activities of hepatic oxygen free radical generating and scavenging enzymes in streptozotocin (STZ)-induced diabetic rats. Male Wistar rats divided into nondiabetic group, diabetic group, and diabetic-AEW supplemented group. The extract was supplemented in 1.14% of raw Aralia elata/kg diet for 7 weeks. Diabetes was induced by injecting STZ (55 mg/kg BW, ip) once 2 weeks before sacrifying. The hepatic cytochrome P-450 content, xanthine oxidase and aminopyrine N-demethylase activities were significantly lowered in the diabetic group compared to the nondiabetic group. Whereas, the activities of aniline hydroxylase and oxygen free radical scavenging enzymes, superoxide dismutase, catalase, glutathione peroxidase, glutathione reductase, glucose-6-phosphate dehydrogenase and glutathione S-transferase, were significantly higher in the diabetic group than in the nondiabetic group. However, the supplementation of AEW normalized these enzyme activities in STZ-induced diabetic rats. When the AEW was supplemented with the diabetic rats, hepatic glutathione content was markedly elevated as well as lipid peroxide level was significantly lowered compared to those of the diabetic group. Thus, these results suggested that AEW supplement enhanced the activities of oxygen species metabolizing enzymes in STZ-induced diabetic rats.

Effect of Korean Traditional Tea Materials Water Extract on Hepatic Oxygen Free Radical Generating and Scavenging Enzyme Activities in Lead Administered Rats (한국전통차 재료의 열수추출물이 납투여 흰주의 간조직 중 유해 활성산소 생성과 제거효소 활성에 미치는 영향)

  • This study was designed to test the effect of Korean traditional tea materials on oxygen-free radical metabolism in lead (Pb) -administered rats. Male rats were divided into normal, Pb-control (Pb-Con) and Pb-water extract of green tea (Camellia sinensis; GT) , persimmon leaf (Diospyros kaki; PL) , safflower seed (Carhamus tinctorius: SS) , Du-Zhong (Eucommia ulmoides; EU) groups, respectively. Pb intoxication was induced by administration of lead acetate (25 mg/kg. B.W., oral) weekly. The extract was administered based on 1.26 g of raw material/kg B.W./day for 4 weeks. When the GT, PL, SS and EU were supplemented to the Pb-administered rats, hepatic lipid peroxide levels were significantly lower compared to the Pb-Con group. Hepatic cytochrom P-450 content and aminopyrine N-demethylase activity was lower in the Pb-Con group than in the normal group, whereas xanthine oxidase activity was significantly elevated in Pb-administered rats. The water extract of GT, PL, SS and EU supplementation attenuated changes in enzyme activities generating reactive oxygen species in the liver. Hepatic superoxide dismutase, catalase and glucose 6-phosphate dehydrogenase activities were significantly higher in the Pb-Con group than in the normal group, while monoamine oxidase activity also tended to increase in the Pb-administered rats. However, glutathione peroxidase and glutathione S-transferase activities, and glutathione content significantly decreased through Pb intoxication. The supplementation of GT, PL, SS and EU induced alleviation changes of hepatic antioxidant enzyme activity.

Antioxidative and Cytoprotective Effects of Isoflavones Isolated from Pueraria thunbergiana Flowers (갈화에서 분리한 이소플라본의 항산화 및 세포보호효과)

  • Antioxidative and cytoprotective effects of tectorigenin and glycitein isolated from the pueraria thunbergiana and its derivative, genistein, were determined. Among these three compounds, tectorigenin and glycitein bearing 6-methoxyl groups in both isoflavones showed significant free radical scavenging activities on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and xanthine/xanthine oxidase (XOD) generating superoxide anion radical. Tectorigenin only showed a slight inhibitory effect on XOD. We further studied the inhibitory effects of these isoflavones on the lipid peroxidation in rat liver microsomes induced by enzymatic and non-enzymatic methods. Each of them exhibited inhibitory effect on both ascorbic $acid/Fe^{2+}-{\;}and{\;}ADP/NADPH/Fe^{+3}-induced$ lipid peroxidation. Moreover, tectorigenin exhibited the highest protection of hydrogen peroxide damage on HepG2 and Vero cells among the three isoflavones, in the cytoprotective assay. It was suggested that the pattern of antioxidative and cytoprotective effect of isoflavones could be crucially by the aromatic substitution of oxygen-containing groups.

Effect of Toluene Treatment on the Xanthine Oxidase and Superoxide Dismutase Activities in Leukocyte of Bacterial Infected Rats (흰쥐에 toluene 투여가 백혈구 xanthine oxidase 및 superoxide dismutase 활성에 미치는 영향)

  • This study was designed to observe the effect of tohiene pretreatment on leukocyte variation in whole blood and the oxygen free radical generating, scavenging enzyme activities in neutrophil of bacteria infected rats. Toluene was administered 7 times intraperitoneally at levels of 9.45 mM/kg body weight to the rats and then infected with S. aureus $2\times10^7$ cfu/ml. The toluene treated-rats showed the significantly decreased numbers of lymphocyte and monocytes, but the similiar numbers of neutrophils with the control. Furthermore the increased neutrophils in blood of bacteria infected rats were reduced by the toluene pretreatment. Concomitantly the increased activities of xanthine oxidase and superoxide dismutase in neutrophil of bacteria infected rats were also decreased by the toluene pretreatment. On the other hand, injection of benzaldehyde to rats also led to similiar results in the count of leukocytes, xanthine oxidase and superoxide dismutase activities of neutrophil with those of toluene treated rats. These data suggest that toluene and its intermediate metabolite, benzaldehyde influence on the phagocytosis and defence mechanism of neutrophil.
