Cryptotanshinone (CPT) is a natural compound isolated from traditional Chinese medicine Salvia miltiorrhiza Bunge. In the present study, the regulatory effect and potential mechanisms of CPT on tumor necrosis factor alpha ($TNF-{\alpha}$) induced lectin-like receptor for oxidized low density lipoprotein (LOX-1) were investigated. Human umbilical vein endothelial cells (HUVECs) were cultured and the effect of $TNF-{\alpha}$ on LOX-1 expression at mRNA and protein levels was determined by Real-time PCR and Western blotting respectively. The formation of intracellular ROS was determined with fluorescence probe $CM-DCFH_2-DA$. The endothelial ox-LDL uptake was evaluated with DiI-ox-LDL. The effect of CPT on LOX-1 expression was also evaluated with SD rats. $TNF-{\alpha}$ induced LOX-1 expression in a dose- and time- dependent manner in endothelial cells. $TNF-{\alpha}$ induced ROS formation, phosphorylation of $NF-{\kappa}B$ p65 and ERK, and LOX-1 expression, which were suppressed by rotenone, DPI, NAC, and CPT. $NF-{\kappa}B$ inhibitor BAY11-7082 and ERK inhibitor PD98059 inhibited $TNF-{\alpha}-induced$ LOX-1 expression. CPT and NAC suppressed $TNF-{\alpha}-induced$ LOX-1 expression and phosphorylation of $NF-{\kappa}B$ p65 and ERK in rat aorta. These data suggested that $TNF-{\alpha}$ induced LOX-1 expression via ROS activated $NF-{\kappa}B/ERK$ pathway, which could be inhibited by CPT. This study provides new insights for the anti-atherosclerotic effect of CPT.
Recently, there are a lot of diseases all around the world. Out of them, Atherosclerosis (AS) is the most common cause of stroke, cardiovascular mortality, and myocardial infarction. The macrophage-derived foam cell, which is formed by oxidized low-density lipoprotein (oxLDL), is the crucial marker for AS. In this study, we report a label-free capacitance imaging technique with multi-electrode array (MEA). The lipid-rich aorta arch lesions, which are derived from an apolipoprotein-E receptor-deficient (apoE-/-) mouse, exhibit higher capacitance than the lipid-free aorta arch, allowing the capacitance imaging of lipid region in atherosclerosis. To improve the contacts between MEA and tissue, polypyrrole(PPy)-coated multi walled carbon nanotubes (MWNTs) multi electrode array (PPy-MWNTs-MEA) was fabricated. Compared to TiN-MEA, PPy-MWNTs-MEA yielded lower contact impedance and better capacitance images. In addition, we have also developed a flexible MEA using single walled carbon nanotubes on a PET substrate. The lipid region could be discriminated in the capacitance images of the lipid-rich aorta arch lesions measured using flexible MEA, demonstrating a feasibility of in vivo applications.
Lysophosphatidylcholine (LPC) is a major phospholipid component of oxidized low-density lipoprotein (ox-LDL) and is implicated in its atherogenic activity. This study investigated the effects of LPC on cell viability, intracellular calcium homeostasis, and the protective mechanisms of chlorogenic acid (CGA) in human umbilical vein endothelial cells (HUVECs). LPC increased intracellular calcium ($[Ca^{2+}]_i$) by releasing $Ca^{2+}$ from intracellular stores and via $Ca^{2+}$ influx through store-operated channels (SOCs). LPC also increased the generation of reactive oxygen species (ROS) and decreased cell viability. The mRNA expression of Transient receptor potential canonical (TRPC) channel 1 was increased significantly by LPC treatment and suppressed by CGA. CGA inhibited LPC-induced $Ca^{2+}$ influx and ROS generation, and restored cell viability. These results suggested that CGA inhibits SOC-mediated $Ca^{2+}$ influx and ROS generation by attenuating TRPC1 expression in LPC-treated HUVECs. Therefore, CGA might protect endothelial cells against LPC injury, thereby inhibiting atherosclerosis.
이 연구는 고지혈증에 대한 생리 반응의 조절과 관련하여 Dendropanax morbifera 잎의 효과를 비교하기 위한 것이다. 추출물은 표본 연령, 수확시기, 추출 방법에 따라 달랐다. 이 추출물이 처리된 RAW 264.7 세포는 산화 저밀도 지단백질에 의해 자극되었고, 세포 지질 축적은 Oil Red O (ORO) 염색법을 사용하여 정량화되었다. HepG2 세포에서 에탄올을 사용하여 독성을 유도한 다음, 상기와 같이 지질 축적을 정량화하였다. 추출물의 3-hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) reductase 억제 활성을 측정하였다. RAW 264.7 세포에서 가장 지질 축적을 억제한 시료는 8년생 11월 채취 주정 60% 추출물로 처리된 세포에서 관찰되었다. 에탄올로 유도한 HepG2 세포에서 가장 큰 보호효과를 나타낸 시료는 5년생 5월 채취 주정 30% 추출물이었다. 가장 강력한 HMG-CoA reductase 억제 작용을 한 시료는 8년생 5월 채취 열수 100% 추출물이다. 전체적으로 봤을 때 지질 축적 억제는 주정 추출물에서, 콜레스테롤 합성 효소 저해 효과는 열수추출물에서 좋은 효과를 보여 이들 원료를 기반으로 지질 대사 개선 기능성 원료를 개발할 수 있을 것이라 기대한다.
포유류에 존재하는 Collectin-Placenta 1 (CL-P1)을 포함한 여러 종류의 scavenger 수용체는 주로 내피 세포, 대식 세포 및 평활근 세포 표면에 발현되는 분자이다. 이들 분자는 산화 된 저밀도 지질 단백질 (oxLDL)에 결합하여 처리 할 수 있는 세포 표면 당 단백질이다. 이들 분자 중 케르세틴이 CL-P1 활성화에 어떤 영향을 미치는가를 확인하였다. 케르세틴은 산화 반응을 담당하는 자유 라디칼의 제거제 역할을 하여 산화를 중지시키는 항산화제로 알려져 있다. 본 논문에서는 마우스 CL-P1 유전자 promoter 부분의 전사 시작 점부터 -500 번째 염기까지의 단편을 DNA 중합효소를 이용하여 클로닝 하였다. 그 후에 대식세포 계열인 RAW264.7 및 섬유아세포계열의 NIH3T3 세포에 도입하여 케르세틴이 CL-P1 유전자 발현에 어떠한 영향을 미치는지에 대한 연구를 하였다. 이 부위에는 세포주기 조절 인자인 E2F 결합부위를 비롯해서 여러 종류의 전사 인자가 결합하는 염기서열이 다수 위치하고 있다. 이러한 500염기 단편을 pGL4.10 기본 벡터 및 프로모터에 연결시킨 후 세포에 도입시켰다. 그리고 배양 중에 케르세틴을 처리하여 유전자 발현양을 형광 색소 발현기법으로 측정하였다. 그 결과 유전자 발현이 시작되는 앞쪽 부분의 -250에서 -350사이의 염기들이 CL-P1 단백질을 만드는데 중요하다는 것을 확인하였다. 그 중에서도 E2F결합 부위가 결정적인 것 이라는 것을 DNA 돌연변이 실험을 통해 확인 하였다. 또한 부착 세포인 RAW264.7 배양액에 케르세틴을 첨가 한 결과, 배양용기 표면에서 탈락하는 현상을 확인하였다. 즉 발현된 CL-P1단백질이 케르세틴에 의해 세표 표면의 부착 분자에도 영향을 주는 것을 확인하였다.
본 연구는 성숙도가 다른 떫은감 분말의 이상지질혈증 개선 기능을 확인하기 위하여 Wistar계 흰쥐를 사용하여 시험하였다. 이상지질혈증 유도를 위해 7주간 고지방/고콜레스테롤 식이를 급여하였으며, 미성숙감과 성숙감 건분은 7%, 양성대조군으로 탄닌은 1%의 수준으로 식이에 함께 넣어 같은 기간 동안 제공하였다. 체중의 증가, 혈장과 간 조직의 지질 및 콜레스테롤 수준을 바탕으로 이상지질혈증 모델이 형성되었음을 확인하였다. 탄닌, 성숙감, 미성숙감을 섭취한 경우에는 체중, 혈장 Apo B 및 ox-LDL 수준이 유의적으로 감소하였으며, 특별히 미성숙감은 LDL-C이 감소하는 경향을 나타냈다. 탄닌, 성숙감, 미성숙감의 섭취는 간에서 병리조직학적 손상과 총지질 및 중성지방의 수준을 감소시키는데 유사한 효과를 나타냈다. 그러나 유전자 발현은 서로 다른 양상으로, 탄닌군은 콜레스테롤 합성과 관련된 HMGCR 유전자 발현을 감소시키고, 성숙감은 간에서 담즙산 생성을 통한 콜레스테롤 배출과 관련된 CYP7A1 유전자 발현을 증가시키고, 미성숙감은 지방산 합성과 관련된 FAS 유전자 발현을 억제하는 것으로 나타났다. 이상의 결과로 감은 탄닌 성분을 매개로 콜레스테롤 합성을 억제하고, 성숙도에 따라 미성숙감은 중성지방합성을 억제하고 성숙감은 담즙산 생성을 통한 콜레스테롤 배출을 증가하는 기전으로 콜레스테롤과 중성지질대사 개선을 목적으로 한 기능성 소재로 개발될 가능성이 있는 것으로 판단된다. 향후 변에서 답즙산 분석 등을 통한 기전 확인이 필요할 것이며, 혈중 oxLDL 수준과 같은 항산화 기능과 지질대사 간 상호 영향을 추가적으로 연구해야 할 것으로 사료된다.
