A Study on the Actual Status of Heat Transfer oils in Industries for Process Safety Management (공정안전관리 사업장의 열매체유 사용실태에 관한 연구)

  • Heat transfer oils are used in applications such as chemical plant heating systems, refinery heat exchange systems, certain gas processes, injection molding systems, and pulp and paper processing. These oils are extremely stable and resistant to thermal and oxidative degradation. In the event of a spill or accidental release of heat transfer oils, it can be ignited easily when there is an ignition source. This study discusses the status of safety management through the actual status of the heat transfer oils to prevent fire and explosion accidents in industries for process safety management. The actual status of the heat transfer oils in process system of industries surveyed by a questionnaire developed. The results of this study can be used to help establishment of safety management to prevent fire and explosion accidents, such as the management of heat transfer oils, safe operation and maintenance in heat transfer oil processes.

Protective Effect of Hwansodan in Serum and Glucose Deprivation Induced-apoptotic Death of PC12 Cells Via Ho-1 Expression (영양혈청 결핍성 PC12 세포고사에서 HO-1의 발현 증가를 통한 환소단의 보호 효과)

  • The water extract of Hwansodan has been traditionally used for treatment of ischemic brain damage in oriental medicine. However, little is known about the mechanism by which the water extract of Hwansodan rescues cells from neurodegenerative disease. PC12 pheochromocytoma cells have been used extensively as a model for studying the cellular and molecular mechanisms of neuronal cell damages. Under deprivation of growth factor and ischemic injury, PC12 cells spontaneously undergoes apoptotic cell death. Serum and glucose deprivation markedly decreased the viability of PC12 cells, which was characterized with apparent apoptotic features such as membrane blebbing as well as fragmentation of genomic DNA and nuclei. However, the aqueous extract of Hwansodan significantly reduced serum and glucose deprivation-induced cell death and apoptotic characteristics through reduction of intracellular peroxide generation. Pretreatment of Hwansodan also ingibited the activation of caspase-3, in turn, degradation of ICAD/DFF45 was completely abolished in serum and glucose deprivated cells. Furthermore, pretreatment of Hwansodan obviously increased heme oxygenase 1 (HO-1) expression in PC12 cells. Taken together, the data suggest that the protective effects of Hwansodan against serum and glucose deprivation induced oxidative injuries may be achieved through the scavenging of reactive oxygene species accompanying with HO-1 induction.

Hepatic Gene Expression Analysis of Gadolinium Chloride Treated Mice

  • Gadolinium chloride ($GdCl_{3}$) was known to block Kupffer cells and generally its toxicity study based on blocking these cells. Therefore, $GdCl_{3}$ frequently used to study toxic mechanisms of hepatotoxicants inducing injury through Kupffer cells. We also tried to investigate the effect of $GdCl_{3}\;on\;CCl_{4}$ toxicity, typical hepatotoxicants. Administration of $GdCl_{3}$ to mice significantly suppressed AST (asparatate amino transferase), ALT (alanine amino transferase) levels which were increased by $CCl_{4}$ treatment. However, $GdCl_{3}$ didn't inhibit the phagocytotic activity of Kupffer cells. Malondialdehyde (MDA) is a good indicator of the degree of lipid peroxidation. In this study, MDA increased by $GdCl_{3}$ administration not by $CCl_{4}$. To understand the toxicity of $GdCl_{3}$, we analyzed global gene expression profile of mice liver after acute $GdCl_{3}$ injection. Four hundred fifty two genes were differentially expressed with more than 2-fold in at least one time point among 3 hr, 6 hr, and 24 hr. Several genes involved in fibrogenesis regulation. Several types of pro-collagens (Col1a2, Col5a2, Col6a3, and Col13a1) and tissue inhibitor of metal-loproteinase1 (TIMP1) were up regulated during all the time points. Genes related to growth factors, chemokines, and oxidative stress, which were known to control fibrogenesis, were significantly changed. In addition, $GdCl_{3}$ induced abnormal regulation between lipid synthesis and degradation related genes. These data will provide the information about influence of $GdCl_{3}$ to hepatotoxicity.

Effect of binder composition on the stability of lipid oxidation and antioxidants of nut-sesame yeotgangjeong (Korean traditional candy bar) (결착제 조성이 견과-깨 엿강정의 저장 중 지방질 산화와 산화방지제 안정성에 미치는 영향)

  • Lipid oxidation and antioxidant stabilities of nut-sesame yeotgangjeong (Korean traditional candy bar) with different binders were evaluated during storage in the dark. S1, S2, and S3 binders consisted of starch syrup, sugar, and water (58:34:7.6, w/w/w); starch syrup, jocheong, honey, trehalose, and water (43.5:11.6:2.9:34:7.6, w/w/w/w/w); and starch syrup, jocheong, honey, sugar, trehalose, and water (43.5:11.6:2.9:17:17:7.6, w/w/w/w/w/w); respectively. Lipid oxidation was determined by peroxide and p-anisidine values, and tocopherol, lignan, and polyphenol contents were determined spectrophotometrically. Peroxide values were significantly higher (p<0.05) in yeotgangjeong samples with S2 or S3 binder than in those with S1; however, the converse was true for tocopherol degradation. Lignan was the most stable antioxidant in yeotgangjeong. The results suggest that tocopherols have a priority over lignans to control the lipid oxidation of yeotgangjeong during storage; reduced lipid oxidative stability after substituting sugar with trehalose might be because it increases oxygen diffusivity and inhibits the antioxidant action of tocopherols on lipids.

EFFECT OF TOP END CONDITION OF FUEL BED CONTAINER ON DOWNWARD SMOLDER SPREAD

  • An experimental study was performed of natural-convection downward smolder spread across a sawdust bed peripherally enclosed with an insulating container, to examine the effect of the open- ing condition at the top end on downward smolder spread. Experiments were conducted by using relatively coarse sawdust and 25-cm-long cylindrical container The variations of temperature profiles along the bed axis with time were determined far different opening conditions and were com-pared with those in smolder spread from open top to open bottom. It was shown that the smolder zone initiated from open top toward closed bottom penetrates the bed with keeping high peak temperature like the case of open top to open bottom spread, although mean spread rate is smaller. This indicates that the downward smolder zone can be sustained stably if sufficient air or oxygen Is supplied from the back of it by natural convection even if upward draft entering from the bottom of the bed is absent. When the top end was partially closed by mounting a cover after stable smolder spread had begun from open top toward open bottom, the temperature at the peak decreased more than 200 K and the smolder zone became to spread with thickening residue. In this case, the shape of temperature profiles continuously changed or decayed until end-effect at the open bottom end enhanced the reaction. The temperature at the shrunk peak, free from the end-effect, was almost identical with the temperature at the exothermic oxidative-degradation zone in smolder spread from open top to open bottom. from these results, it can be inferred for natural-convection downward smolder spread that the oxidation reaction of the char is very sensitive to the oxygen supply by natural convection in the space above the smolder zone, and that the top end opening condition strongly alters the completeness of reactions, structure, and behavior of the smolder zone.

Bioconversion enhances anti-oxidant and anti-inflammation activities of different parts of the Mulberry Tree (Morus alba L.), especially the leaf (Mori Folium)

  • The mulberry tree (Morus alba L.) has been traditionally used in Chinese medicine to treat inflammatory diseases. We investigated the effects of bioconversion on different components of the mulberry tree, and determined changes in the physiological activities. Ethyl acetate-soluble fractions of five different segments (fruit, Mori Fructus; leaf, Mori Folium; twig, Mori Ramulus; root, Mori Cortex; and mistletoe, Loranthi Ramulus) of the mulberry tree show enhanced anti-oxidant effects in the 2,2-diphenyl-1-picrylhydrazyl, and 2,2'-azinobis-(3-ethylvenzothiazoline-6-sulfonic acid) assays, and enhanced anti-inflammatory effects of lipopolysaccharide (LPS)-stimulated nitric oxide (NO) production in RAW 264.7 macrophages, after being treated with a crude enzyme extract from Aspergillus kawachii, in the following order of activity: Mori Folium>Mori Cortex>Mori Ramulus>Mori Fructus>Loranthi Ramulus. Ethyl acetate- soluble fraction of mulberry leaves (Mori Folium) that underwent bioconversion was most effective, and was devoid of any cytotoxicity. The fraction was also effective against mRNA expression of LPS-induced pro-inflammatory cytokines, such as inducible nitric oxide synthase, cyclooxygenase-2, tumor necrosis $factor-{\alpha}$, $interleukin-1{\beta}$, and interleukin-6. In addition, the fraction was effective in LPS-induced phosphorylation of mitogen-activated protein kinases and IKK, and $I{\kappa}B$ degradation, followed by translocation of the nuclear $factor-{\kappa}B$ from the cytoplasm to the nucleus. Thus, bioconversion increased the anti-oxidative and anti-inflammatory activities of the mulberry leaf.

Effects of Bombycis Corpus on Male Osteoporosis (백강잠(白殭蠶)이 남성 골다공증에 미치는 영향)

  • To investigate the effect of Bombycis Corpus on male osteoporosis, we performed Dual Energy X-Ray Absorptiometry(DEXA) and histochemical methods. The animals were used ICR-based male mice of 8 weeks and 50 weeks, respectively. ICR male mice at 8 weeks were used in the control group, and ICR male mice at 50 weeks were used in aging group and Bombycis Corpus group(BC group). In the aging group, 0.5 ml of distilled water was administered once a day for 6 months. In BC group, Bombycis Corpus(0.78g/kg) was dissolved in distilled water for 6 months once a day. As a result, Bombycis Corpus decreased bone loss, increased bone density by reducing the loss of bone matrix in the femur due to aging, and increased osteoblast - induced osteopontin(OPN) and osteocalcin(OPC) positivite reaction. In addition, administration of Bombycis Corpus decreased Reaction of activation of nuclear factor kappa B ligand(RANKL) positive reaction, increased osteoprotegerin(OPG) positive reaction, and decreased matrix metalloproteinase-3(MMP-3) and 8-hydroxy-2'-deoxyguanosine(8-OHdG) positivite reaction. Taken together, Bombycis Corpus increases the activity of osteoblasts, inhibits osteoclast function, promotes osteoblast function, inhibits bone tissue degradation, and inhibits bone loss due to oxidative stress. It was observed that Bombycis Corpus reduced bone loss and increased bone density caused by aging to improve male osteoporosis. Therefore, Bombycis Corpus may be used as a preventive and therapeutic agent for male osteoporosis.

Syringaresinol derived from Panax ginseng berry attenuates oxidative stress-induced skin aging via autophagy

  • Background: In aged skin, reactive oxygen species (ROS) induces degradation of the extracellular matrix (ECM), leading to visible aging signs. Collagens in the ECM are cleaved by matrix metalloproteinases (MMPs). Syringaresinol (SYR), isolated from Panax ginseng berry, has various physiological activities, including anti-inflammatory action. However, the anti-aging effects of SYR via antioxidant and autophagy regulation have not been elucidated. Methods: The preventive effect of SYR on skin aging was investigated in human HaCaT keratinocytes in the presence of H2O2, and the keratinocyte cells were treated with SYR (0-200 ㎍/mL). mRNA and protein levels of MMP-2 and -9 were determined by real-time PCR and Western blotting, respectively. Radical scavenging activity was researched by 2,2 diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) assays. LC3B level was assessed by Western blotting and confocal microscopy. Results: SYR significantly reduced gene expression and protein levels of MMP-9 and -2 in both H2O2-treated and untreated HaCaT cells. SYR did not show cytotoxicity to HaCaT cells. SYR exhibited DPPH and ABTS radical scavenging activities with an EC50 value of 10.77 and 10.35 ㎍/mL, respectively. SYR elevated total levels of endogenous and exogenous LC3B in H2O2-stimulated HaCaT cells. 3-Methyladenine (3-MA), an autophagy inhibitor, counteracted the inhibitory effect of SYR on MMP-2 expression. Conclusion: SYR showed antioxidant activity and up-regulated autophagy activity in H2O2-stimulated HaCaT cells, lowering the expression of MMP-2 and MMP-9 associated with skin aging. Our results suggest that SYR has potential value as a cosmetic additive for prevention of skin aging.

Formulation and Quality Evaluation of Chicken Nuggets Supplemented with Beef and Chicken Livers

  • This study explores the potential of utilizing meat byproducts, specifically chicken and beef liver, to enhance the nutritional value of processed foods like chicken nuggets. Proximate analysis was conducted on the livers, including moisture, ash, fat, and protein content, and degradation potential was observed. Antioxidant potential was analyzed through 2,2-diphenyl-1-picrylhydrazyl (DPPH). The total phenolic content (TPC), oxidative stability through peroxide value (POV), and free fatty acid (FFA) were performed to evaluate quality changes during seven-day storage. The radical scavenging activity showed that beef liver has excellent antioxidant capacity (61.55%- and 195.89- mM gallic acid equivalent for DPPH and TPC, respectively) compared to chicken liver and significantly increased the antioxidant potential of nuggets by 5%-10%. POV and FFA values increased with increased storage days for the liver and its incorporation in nuggets. However, the values remained under the 10 meq/kg threshold. Incorporating the livers into chicken nuggets led to a significant (p=0.000) improvement in nutritional content, particularly a 1.5%-2% increase in protein, with a similar increase in mineral content. Texture and sensory evaluations indicated favorable consumer acceptability for liver-enriched nuggets. Overall, this research shows the value of adding liver as a functional ingredient to enhance the nutritional profile of processed foods.

Effect of L-ascorbic acid on the degradation of aflatoxin $B_1$ (아스콜빈산에 의한 Aflatoxin $B_1$의 파괴에 관한 연구)

  • Large amount of aflatoxin $B_1(AFB_1)$ is disappeared in the presence of L-ascorbic acid(AA) in buffer solution at pH values from 1 to 7 during 5 days of Incubation at $37^{\circ}C$. $AFB_1$ was quite stable at pH's between 5 and 7 when AA was absent(control), however, $50{\sim}60%$ of APB, was degraded in its presence after 5 days. The rate of disappearance of $AFB_1$ increased with a decreasing of pH in the presence of AA, even though $AFB_1$ in the control degraded increasingly with the decrease in $pH(pH{\leq}4)$. The level of $AFB_1$, decreased as the reaction temperature increased when $AFB_1$ reacted with AA. The aflatoxin could not be detected at all after 3 days when the reaction occurred at $60^{\circ}C$, while the aflatoxin was stable at $5^{\circ}C$ thoughout the reaction period. $90{\sim}96%$ of $AFB_1$ was found to be degraded in a far when $AFB_1$ reacted with AA plus different concentrations of $CuSO_4{\cdot}5H_{2}O$, showing remarkably faster rate than the control; however, different concentrations of L-cysteine instead of $CuSO_4\;5H_{2}O$ protected the degradation of aflatoxin and no $AFB_1$ was degraded for a day and resulted in less $AFB_1$ disappeared than the control. The degradation of $AFB_1$ was dependent on AA concentration and the rate of disappearance as the concentration of AA decrease, but $AFB_1$ concentration did not influence the rate. The product formed when $AFB_1$ reacted with AA was identified to $AFB_{2a}$ by using HPLC chromatographic examinations, and by UV spectrum of $AFB_1$ reacted with AA. The disappearance of $AFB_1$ was correlated well in the appearance of $AFB_2a$. From the results, the degradation of $AFB_1$ in the presence of AA is probably due to one or more of the oxidative products of AA which was produced during the AA oxidation.
