한국어판 다낭성 난소 증후군 삶의 질 측정도구의 타당도와 신뢰도 (Validity and Reliability of a Korean version of Polycystic Ovary Syndrome Questionnaire)

  • Purpose: Polycystic Ovary Syndrome Questionnaire (PCOSQ) was developed to measure health-related quality of life of women who had polycystic ovary syndrome. The purpose of this study was to exam reliability and validity of the Korean version of PCOSQ. Methods: A sample of 101 women who had polycystic ovarian syndrome was recruited from outpatient gynecology clinics. Factor analysis was carried out to verify the composition of dimensions of the Korean version of PCOSQ. Concurrent validity was examined by assessing the correlation between the PCOSQ and health related quality of life, and convergent validity was evaluated by testing correlation between PCOSQ and depression. Reliability was assessed using Cronbach's ${\alpha}$. Results: The content validitywas satisfactory (CVI=0.85). Factor analysis identified 5 factors, and factor loadings of the PCOSQ ranged from .48 to .90. Correlation was found between the PCOSQ and health related quality of life (Physical component score: r=.33, p=.001; Mental component score: r=.44, p<.001) and between the PCOSQand depression (r=-.36, p<.001). The Cronbach's ${\alpha}$ of the PCOSQ was .93, indicating reliable reliability. Conclusion: These results demonstrate that the Korean version of PCOSQ is a reliable and valid instrument for evaluating Korean-speaking women with polycystic ovarian syndrome.

담배나방의 난소발생시 폴리아민 대사에 미치는 상승아치사온도의 효과 (Effects of Elevated Sublethal Temperature on Polyamine Metabolism during Ovarian Development of the Tobacco Budworm, Helicoverpa assulta)

  • To elucidate the effect of elevated sublethal temperature ($33\pm1^{\circ}C$) on polyamine metabolism and oogenesis, we investigated alterations in the major polyamines and ornithine decarboxylase (ODC) and arginine decarboxylase (ADC), and ovarian development during the pupal-adult development of the tobacco budworm, Helicoverpa assulta. Ovaries ODC activity under the elevated sublethal temperature ($33\pm1^{\circ}C$) were lower than those of the optimal rearing temperature ($25\pm1^{\circ}C$). whereas ovarian ADC activity was consistently higher than the optimal rearing temperature ($25\pm1^{\circ}C$). When the gonads were exposed to the higher temperature, ovarian putrescine showed somewhat suppressed levels throughout development, indicating a relatively high correlationship with the alteration aspects in ODC or ADC activity under elevated sublethal temperature. A somewhat precocious ovary was observed in an early stage of development at $33\pm1^{\circ}C$, but cellular abnormalities occurred in this ovary. The ovary developed under elevated sublethal temperature was observed the inhibitional effect of polyamine metabolism and the abnormal development of ovariole, which seem to be related to the sterility.

Factors Affecting Primary Cultures of Abalone Haliotis discus hannai Ovary-dissociated Cells and General Culture Aspects

  • We investigated factors affecting primary cultures of Pacific abalone Haliotis discus hannai ovary-dissociated cells to identify general aspects of their early-phase culture. Ninety-seven cell populations derived from 30 individuals were cultured in different media with varying compositions of medium supplements, and initial attachment, subculture, and survival for ${\geq}10$ weeks were assessed according to medium composition and individual. We also examined the time required for subculture and the rate of cell death according to both culturing period and passage number within 10 weeks. A lack of fetal bovine serum (FBS) and hemolymph significantly inhibited the growth of cultured cells, while we detected no significant effect of medium composition on initial cell attachment. Through data reallocation, with the omission of data from cell populations cultured in FBS-free and hemolymph-free media, we showed that growth inhibition was also affected by individual differences among the abalones used. During the culture, we observed four different types of cell morphology. Moreover, considerable time was required for subculture-18.4 and 19.5 days for first and second subcultures, respectively-and cell death did not occur within 30 days or for passage 0. Our results will provide valuable information for developing universal cell culturing guidelines in abalone species and suggest the feasibility of culturing abalone ovary-dissociated cells.

Localization (and profiles) of tyrosine-phosphorylated proteins in female reproductive organs of adult rats

  • Objective: Tyrosine phosphorylation is an essential process in many biological systems, including the male reproductive system. The presence of tyrosine-phosphorylated (TyrPho) proteins has been well documented in male reproductive organs, but research in fertile females is still limited. Methods: The ovary, oviduct, and uterus of adult female Sprague-Dawley rats in the estrus phase were used to localize TyrPho proteins using an immunohistochemical technique. These proteins were separated and their expression patterns were examined by sodium dodecyl sulfate-polyacrylamide gel electrophoresis and Western blot analysis, respectively. Results: TyrPho proteins were localized in the cytoplasm of the oocyte except the antral fluid; in the granulosa cells, theca cells, and stromal cells of the ovary; at the apical surface of oviductal epithelial cells; and in the basal epithelium and submucosa of the uterine wall. Moreover, we found that 72-, 43-, and 28-kDa TyrPho proteins were localized in the ovary, while 170-, 55-, and 43-kDa proteins were localized in the oviduct. In the uterus, we detected four major bands, corresponding to 61-, 55-, 54-, and 43-kDa TyrPho proteins. Conclusion: Given that these TyrPho proteins were found in major reproductive organs in the estrus phase, these proteins may play important roles in female fertility.

  • An abnormally enlarged right ovary and a mass in fat surrounding the right kidney were discovered in a dairy cow during routine postmortem examination at slaughter. The ovary was dark reddish and multinodular in shape. Numerous cystic structures were identified in the mass. Histopathologically, the ovary was completely replaced with large, uniform, polyhedral neoplastic cells containing vesicular nuclei and prominent nucleoli. The mitotic index was high. In the lymphatic vessels, tumor emboli were observed. Another mass in the fat surranding the right kidney had the same histological features as the ovarian mass. This animal was diagnosed with malignant dysgerminoma and metastasis to other peritoneal organs.

방사선 조사선량에 따른 생쥐 난소의 조직화학적 변화 (Study of Histochemical Changes of Mouse Ovary Under X-ray Irradiation)

  • 본 연구는 X-선을 생쥐 생체에 조사한 후, 방사선 조사선량에 따른 난소조직의 형태적 변화를 Masson's trichrome, 세망섬유 염색법 등의 조직화학반응을 확인한 결과 X-선을 조사한 난소조직은 방사선량이 증가함에 따라 성장 난포의 형태적 변화가 뚜렷하였으며, 특히 과립층세포의 변성에 의한 핵은 응축과 투명대의 변화가 현저하였다. 또한 Masson's trichrome 염색과 세망섬유 염색을 통한 조직화학반응의 결과 과립층세포의 변화와 난포기저막의 변형, 그리고 세망섬유의 비정상적 배열이 확인되었다.

고지사육유우의 번식장해 발생상태에 관한 조사연구 (Studies on the Occurence of Reproductive Disorders of Dairy Cattle Resident in High-land)

  • Breeding and infertility status of Holstein cows reared in pasture of High-land was investigated. The results obtained were summarized as follows; 1. Eight hundred and forty Holstein cows with heifers (12.4%), 3 years old cows (19.3), 4 years old cows (16.7%), 5 years old cows (12.5%), 6 years old cows (11.4%), 7 years old cows (13.2%), 8 years old cows (9.4%) and above 9 years old cows (5.1%) were investigated. 2. They were 72.4% of conceived cows, 8.2% of uncertain pregnancy, 7.7% of physiological empty and 11.7% of reproductive disorder. 3. The percentage of cow concepted with 1, 2, 3 and more than 4 times of A.I. was 49.2, 28.8, 14.6 and 7.4% respectively. 4. In the nutritional condition of infertile cows, excellent, good, fair and poor was 7.1, 30.6, 36.7 and 25.5%, respectively. 5. Among 98 infertile cows, distribution of reproductive disorder was 41.8, 37.8, 5.1, 5.1 and 11.2% in ovary, uterus, vagina, oviduct and others, respectively. Ovary showed higher percentage than any other reproductive organs. Among the ovarian syndromes, lutein cystic ovary, follicular cystic ovary, and persistent corpus luteum were 31.7, 26.8 and 19.5%, respectively. 6. Four years old cow showed highest distribution (16.4%) among the aged groups in disordered cows. In the syndromes of reproductive disorder, latent endometritis showed higher frequency (14.3%) than any others. 7. Infertile cows with complex syncrome of genital disease was 29.6%.

돼지 난소내 여포의 폐쇄에 따른 미세구조의 변화에 관한 연구 (A Study on the Fine Structural Changes of Porcine Ovarian Follicles during Atresia)

  • This experiment has been done in order to study the correlation between the ultrastructure changes and the atresia phenomenon of the follicles in porcine ovary. The ovaries were assorted according to the presence or absence of corpus luteum. Thereafter, the follicles were classified into normal, pyknotic, necrotic and cystic groups by atretic characteristics on the histological observation, and then their ultrastructures were examined with an electron microscope. The results were as followings. 1. In normal group, granulosa cells represented the ultrastructural characteristics of protein-synthesizing cells. Since the initiation of atresia, the line structure of granulosa cells showed many of the characteristic features of steroid-secreting cells, followed by gradual pyknosis. 2. In necrotic group of the ovary without corpus luteum, the theca interns became hypertrophic and displayed the ultrastructural features of active steroidsecreting cells. But this phenomenon was not seen in the follicles of the ovary with corpus luteum. 3. Degenerative changes of cumulus cells were similar to those of granulosa cells, and the degenerating oocytes showed the degeneration of cellular organelles, cytoplasmic vacuolization and disappearance of microvilli on the surface. The degeneration of granulosa cells tended to procede that of oocytecumulus complex in the follicles of the ovary having no corpus luteum, but this tendency was reversed in the case of presence of corpus luteum. 버섯파리(Coboldia fuscipes)의 난소발육 및 난황단백질에 관한 연구 (Ovarian Development and Yolk Protein of the Mushroom Fly, Coboldia fuscipes (Diptera: Scatopsidae))

  • Ovarian development and yolk protein (YP) of mushroom fly, Coboldia fuscipes, were characterized. C. fuscipes has a pair of ovaries, composed of 130∼140 ovarioles, respectively. Ovarian development begins at 1 day of pupa, and growth of the ovaries continued to become a matured shape at 1 day after emergence. The YP of C. fuscipes identified by SDS-PAGE analysis revealed that the protein is composed of three subunits, designated YP1 (61 kDa), YPS(50 kDa), and YP3 (47 kDa). These three subunits of YP gradually decreased during embryogenesis. The YP was first detected in the 3 day-old pupal ovary and was continually detected up to 2 day-old adult, but not in the hemolymph. Furthermore, Western blot analysis of male and female adult hemolymph and ovary revealed that the antibodies against YP1, YP2, and YP3 reacted with three YP bands in ovary and egg extract, respectively. However, this reactivity was not observed in the male and female hemolymps. Therefore, it is assumed that the UP of C. fuscipes is synthesized in the ovary at 3 days after pupation.
