Modulation of Activator Protein-1 (AP-1) and MAPK Pathway by Flavonoids in Human Prostate Cancer PC3 Cells

  • In last couple of decades the use of natural compounds like flavonoids as chemopreventive agents has gained much attention. Our current study focuses on identifying chemopreventive flavonoids and their mechanism of action on human prostate cancer cells. Human prostate cancer cells (PC3), stably transfected with activator protein 1 (AP-1) luciferase reporter gene were treated with four main classes of flavonoids namely flavonols, flavones, flavonones, and isoflavones. The maximum AP-1 luciferase induction of about 3 fold over control was observed with $20\;{\mu}M$ concentrations of quercetin, chrysin and genistein and $50\;{\mu}M$ concentration of kaempferol. At higher concentrations, most of the flavonoids demonstrated inhibition of AP-1 activity. The MTS assay for cell viability at 24 h showed that even at a very high concentration $(500\;{\mu}M)$, cell death was minimal for most of the flavonoids. To determine the role of MAPK pathway in the induction of AP-1 by flavonoids, Western blot of phospho MAPK proteins was performed. Four out of the eight flavonoids namely kaempferol, apigenin, genistein and naringenin were used for the Western Blot analysis. Induction of phospho-JNK and phospho-ERK activity was observed after two hour incubation of PC3-AP1 cells with flavonoids. However no induction of phospho-p38 activity was observed. Furthermore, pretreating the cells with specific inhibitors of JNK reduced the AP-1 luciferase activity that was induced by genistein while pretreatment with MEK inhibitor reduced the AP-1 luciferase activity induced by kaempferol. The pharmacological inhibitors did not affect the AP-1 luciferase activity induced by apigenin and naringenin. These results suggest the possible involvement of JNK pathway in genistein induced AP-1 activity while the ERK pathway seems to play an important role in kaempferol induced AP-1 activity.

  • As one of the casting plans to improve the scientific inquiry ability of the students, the aims of this study are to develop and apply a basic inquiry program for the inquiry subject of Seogwipo fossil formation, and analyze the basis inquiry ability of the students to show in the inquiry activity process actually. The results obtained in 5th grade 48 elementary school students of Seogwipo-city are as follows; Students executed observing activity using an appropriate senses such as senses of vision and touch, and showed the tendency which tries to observe the form overall rather than the partial form of the fossil formation. But the ability to utilize appropriately for predicting and inferring with the facts which could depend on observing activity was low. And we found out that the misconceptions influence on inquiry activity. Therefore, to help understanding deeper for the students' basic inquiry element, it is thought that a study of the various educational guidance ways is necessary to this. The teachers also have to study the various ways to induce the scientific conception through the application of proper teaching-learning for correction of misconceptions because misconceptions have an influence on inquiry activity together. If various inquiry programs considering the regional-specific characteristics are developed to cause students' interest, students would come to participate in inquiry activity aggressively a little more.

A CLINICAL STUDY FOR CARIES ACTIVITY OF PRESCHOOL CHILDREN USING CARIOSTAT (Cariostat를 이용한 아동의 우식활동에 관한 임상적 연구)

  • The purpose of the study was to determine the ability of the caries activity test, 'Cariostat' to identify preschool children with dental caries. The subjects of this study were 76 children of 3 to 5 years of age. Oral examination was carried out to assess the caries experience using a mirror and explorer. In all subjects buccal plaque samples were obtained, incubated, and scored as the manufacturer's instructions for the Cariostat test. Statistical analyses were used with the Crosstabulation. The test group exhibited mean dft index 5.21, while 27.63% of subjects were caries free and had no restorations. The sensitivity of the Cariostat test was found to be 98.8% while the specificity was found to be 19.05% The results showed that caries activity measured by visual reading was highly significant to the Cariostat score and dft index(P<0.001). The Cariostat method has indicated the future possible occurrence of caries. The use of this method is meant to require in the caries prevention program.

Direction in Health Education of smoking Prevention for Elementary School Children (초등학생의 흡연예방 보건교육 방향에 관한 연구 - 초등학교 교사 의견을 기반으로 -)

  • In order to provide a desirable direction for the health education on preventing smoking of elementary school students, by understanding the views held by elementary school teachers on smoking prevention education, 529 teachers at elementary schools located in Seoul were responded by constructed questionnaires during the period of Nov. 15 to Dec. 18 1999. The results of the survey analysis were as follows: 1. 91.9% of the respondents said that schools should offer education programs on preventing smoking, and this view was more strongly shared by teachers with religions than non-religious ones. 2. 37.6% of teachers responded that the education should start with 4th-5th graders, while 36.0% opted for 6th grades. With regard to the appropriate class in providing such education, 41.1% selected physical education (PE), followed by extracurricular activity class with 28.8%, 45.1% of the respondents said discussion would be the most effective education method, while 39.9% recommended using experiments. fall in diseases and growth and developmental defect were pointed out as the most interesting topic for students by 33.2% and 28.1% of the surveyed, respectively. As for the most effective media for education, 57.2% recommended video, followed by computer games(19.2%) and cartoons (20.0%). 64.1% of the teachers picked school nurses as the most appropriate person in carrying out the education. The results of the finding show that it would be most desirable to provide health education on preventing smoking to senior elementary schoolers during the PE or extracurricular activity class by school nurses. The program would be structured to first show video clips on growth and developmental defect or fall in diseases resulting from smoking and to have discussions or experiments on related matters.

A Study on Multibiometrics derived from Calling Activity Context using Smartphone for Implicit User Authentication System

  • Current smartphone authentication systems are deemed inconvenient and difficult for users on remembering their password as well as privacy issues on stolen or forged biometrics. New authentication system is demanded to be implicit to users with very minimum user involvement being. This idea aims towards a future model of authentication system for smartphones users without users realizing them being authenticated. We use the most frequent activity that users carry out with their smartphone, which is the calling activity. We derive two basics related interactions that are first factor being arm's flex (AF) action to pick a phone to be near ones' ears and then once getting near ear using second factor from ear shape image. Here, we combine behavior biometrics from AF in first factor and physical biometrics from ear image in second factor. Our study shows our dual-factor authentication system does not require explicit user interaction thereby improving convenience and alleviating burden from users from persistent necessity to remember password. These findings will augment development of novel implicit authentication system being transparent, easier, and unobtrusive for users.

Hepatoprotective Effects of Waxy Brown Rice Fermented with Agrocybe cylindracea

  • The methanol extract of waxy brown rice fermented with Agrocybe cylindracea was prepared. The extract was then freeze dried and fed to rats at the level of 0.5, 1.0, 2.0 g/kg body weight for 14 days, followed by the treatment with carbon tetrachloride for three consecutive days to induce hepatotoxicity. After sacrificing the rats, the enzyme activity of aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP), lactate dehydrogenase (LDH), and ${\gamma}$-glutamyl transpeptidase (${\gamma}$-GTP) in serum was determined. Biochemical analysis on serum for albumin, total protein, triglyceride, and total as well as HDL-cholesterol were carried out along with a histopathological study of liver tissues. Based on these data, we suggest that the waxy brown rice cultured with A. cylindracea may exert hepatoprotective activity against hepatotoxicity caused by chemicals such as carbon tetrachloride.

Screening of Sterol Biosynthesis Inhibitors from Natural Products Using Recombinant Yeast Carrying Human Lanosterol Synthase

  • For the screening of inhibitors of sterol biosynthesis from natural products, a simple and rapid assay method was developed using recombinant yeast carrying human lanosterol synthase, main target of this assay method. Sterol biosynthesis inhibition activity was monitored only by the inhibition of growth of the recombinant yeast. By changing the substrate, this assay method can figure out which step is inhibited in the sterol biosynthesis by the test material. With this assay method total 102 plant samples were screened for their inhibitory activity of sterol biosynthesis. Among plant water extracts screened, 11 plant samples showed inhibitory activity on sterol biosynthesis in ergosterol (-) medium. For selection of the specific inhibitory materials, 11 plant samples were reassayed in ergosterol (+) medium. After all 5 plant samples, Abutilon avicennae Gaertn. (stem), Alnus japonica Steud. (stem), Amaranthus mangostanus L. (aerial part), Philadelphus schrenckii Pupr. (leaf) and Pimpinelia brachycarpa Nakai (aerial part), showed specific inhibitory activity.

The Effect of Perceiver's Personality on Visual Evaluation of Clothing (지각자의 성격특성이 의복의 시각적 평가에 미치는 영향)

  • The purpose of this study was to find out differences of visual evaluation according to perceiver's personality(introvert-extrovert) and clothing styles. Subjects were 120 women in age from 18 to 31 years in Seoul. The stimuli of 7 clothing styles were used for visual evaluation. Three dimensions of visual evaluation were derived by falters analysis: elegance, activity individuality, and fashionability. The pants suit was estimated low in activity individuality and fashionability, but the hip-hop style high in activity individuality. The skirt suit was rated high in elegance, but the 8th length pants low in elegance. Personality had significant influences on the evaluation of activity individuality. Extrovert women perceived all stimuli to be more active and individual than introvert. Fashionability factor had an interaction effect by personality and clothing expenses. The introvert women in low clothing expenses group evaluated the stimuli to be more fashionable than the introvert in high clothing expenses group. There were significant differences in preference of all clothing styles according to personality. Extrovert women liked 8th length pants more than introvert. The introvert women in low clothing expenses group liked the skirt suit less than the extrovert.

Antioxidative and Antitumor Activity of Extracts from Saussurea lappa (목향 추출물의 항산화 및 항암활성)

  • This study was carried out to investigate the antioxidative and antitumor activities of Saussurea lappa for the purpose of developing a functional food. The methanol extract of Saussurea lappa was fractionated with five solvents (hexane, chloroform, EtOAc, BuOH, water) and examined for antioxidative activities and xanthine oxidase inhibitory activity in addition to growth inhibitory activity of human cancer cell (HT-29, SNU-1, HeLa). Total phenol compound contents were higher in EtOAC fraction than other fractions. Also, electron donating ability was over 90% at $500{\mu}g/ml$ (93.1 %) and $1000{\mu}g/ml$ (94.3%). The hexane fraction revealed stronger nitrite scavenging ability than the positive control (ascorbic acid) and its abilities were 22.4% and 42.8% at $500{\mu}g/ml$ and $1000{\mu}g/ml$, respectively. Xanthine oxidase inhibitory activity had similar tendency to electron donating ability. The EtOAc fraction showed high inhibition to xanthien oxidase activities at $500{\mu}g/ml$(81.9%) and $1000{\mu}g/ml$(90.4%). In the antitumor activity test, the hexane fraction exhibited potent growth inhibition activity against HT-29, SNU-1 and HeLa cells. Therefore, we believe that Saussurea lappa can be developed into a functional food with antioxidant activity. Additional studies are required for the hexane and chloroform fractions of Saussurea lappa to develop them into therapeutic supplements for stomach cancer, colon cancer, and cervical cancer.

A Comparative Study on the Prevalence of Refractive Errors Between Urban and Rural Primary School Children (초등학생의 지역간 시력차이에 관한 연구 -대구 및 영천지역을 중심으로-)

  • The study was carried out to estimate the prevalence and possible causes of refractive errors in primary school children. At one primary school in urban Daegu, one in the medium-sized city of Yongcheon and 2 in rural areas, children wearing glasses and children with visual acuity of less than 0.7 were given our questions. 354 out of 378 children responded to our questions. The research was done from March 2000 to February 2001. The data were analyzed by percentage, x2 test, t-test, ANOVA and Scheff method. Children with eyesight problems were more prevalent in urban areas than rural areas. The lack of luminosity during reading (p=0.015), length of reading time (p=0.08) and posture which watching TV (p=0.023) appeared leading causes of progressing myopias. The visual impairment caused by refractive errors may affect the mental and physical activity of primary school children and lead to social isolation, loneliness and depression. However, little attention may be paid by professionals engaged in providing health care to the children. Once we have recognized the impact of visual impairment, we may be in a better position to prevent progressing refractive errors.
