Neopestalotiopsis Leaf Blight, an Emerging Concern on Leatherleaf Fern in Indonesia

  • Leatherleaf fern (Rumohra adiantiformis) is an important ornamental plant in Indonesia and global. Green fern leaves with bold dark green color with long shelf-life, attract florists as decoration. Indonesia is one important leatherleaf fern exporters, however currently an outbreak of leaf blight decreased production significantly. Initial symptom was reddish brown spots from edge of leaf, which was gradually followed by dark-brown necrotic lesions causing leaf blight and dried. This is a study to do Koch-Postulate approach and molecular identification, to identify the pathogen of the "new emerging disease" reported. Based on multigene analysis using primers from ITS, β-tub and tef1-α gene markers, the pathogen was identified as Neopestalotiopsis sp. All sequences have been deposited in GenBank with accession number of OR905551 (ITS), OR899817 (ßtubulin) and OR899816 (TEF). This Neopestalotiopsis leaf blight causes an emerging concern in leatherleaf fern in Indonesia and global biosecurity because it infected an export commodity.

질소시비량이 자생 가는쇠고사리와 탐라별고사리의 생육에 미치는 효과 (Effect of Nitrogen Application Rate on Growth of Rumohra aristata and Cyclosorus dentatus of Korea Native Fern)

  • For the two species of native fern that have high development potential for interior landscape, Rumohra aristata and Cyclosorus dentatus, the physiological and growth characteries as nitrogen application rate such as $NaNO_3$ 0.0mg/L, $NaNO_3$ 0.5mg/L, $NaNO_3$5.0mg/L, $NaNO_3$ 50mg/L and $NaNO_3$ 500mg/L were surveyed. 1. A significant changes of Rumohra aristata, in top growth was not distinctive. However, as nitrogen application rate increased, the numbers of leaves declined and the content of chlorophyll and the number of wilted leaves increased. 2. As nitrogen application rate increased, the top growth of Cyclosorus dentatus and the content of chlorophyll was increased. However, its overall, ornamental value was reduced due to the rising number of wilted leaves and the declining number of new leaves.

Chlorophyll Fluorescence and Growth Response of Three South Korea Native Fern Species under In-door Light Intensity

  • This study investigated the growth of native ferns under indoor light intensities to identify the introduction possibility as in-door ornamental plants. Three evergreen perennial fern species used in this experiment were Coniogramme japonica (Thunb.) Diels, Woodwardia japonica (L. f.) Sm., and Cyclosorus acuminatus (Houtt.) Nakai ex H. Itô. The light intensities were adjusted to 10, 50, 100 and 200 PPFD (µmol·m-2·s-1) based on the measurement of the various indoor light quantities. The experiment was conducted for a total of 8 weeks, and the light period (12/12h), temperature (25±1℃), and humidity (55±3%) were maintained during the experiment. The control plant group was grown in glass greenhouse for the same period. As the result of the study, in door C. japonica showed better growth under light intensities of 100, 200 PPFD. However, withering of the plants were observed under all light intensities except the control, which causes an ornamental value decrease. This seems to be related to the increase of DIo/RC value in chlorophyll fluorescence parameters. In the W. japonica growth data, the plant height was not significantly different from the control but, the leaf number decreased more than a two-fold. Also, the formed leaves turned brown and showed a poor growth and SPAD value at 200 PPFD had decreased significantly. Growth data of C. acuminatus was not significantly different with the control at all light intensities however, withering was observed at 100 and 200 PPFD. In chlorophyll fluorescence parameters, significant decrease in Pi_Abs and increase in DIo/RC value at 200 PPFD impose that stress caused by the intense light might be the reason of the withering of the plants.

Optimal culture conditions for mass production of rock polypody (Polypodium vulgare L.)

  • This study aimed to develop a suitable method for inducing the proliferation of prothallus and producing sporophytes of rock polypody (Polypodium vulgare L.). The prothalli used in all experiments were obtained from spore germination and sub-cultured for 8-week intervals. The most appropriate media for prothallus propagation were investigated by culturing 300 mg of prothallus in MS ($1/4{\times}$, $1/2{\times}$, $1{\times}$, and $2{\times}$ strength) medium and in Knop medium for 8 weeks. Cultures were maintained at a temperature of $25{\pm}1^{\circ}C$, light intensity of $30{\pm}1.0{\mu}mol-m-2{\cdot}s-1$, and a photoperiod of 16/8 h (light/dark). Fresh weight of prothalli was 4.8 g on $1{\times}$ MS, 4.5 g on $1/2{\times}$ MS and 4.3 g on 1/4 MS medium. To select a suitable soil combination for sporophyte formation, 1.0 g of prothallus was ground with distilled water, spread in five combinations onto different soil substrates (decomposed granite, horticultural substrates, peat moss, and perlite), and then cultivated for 13 weeks. The sporophyte cultures were maintained at a temperature of $25{\pm}1^{\circ}C$, light intensity of $43{\pm}2.0{\mu}mol-m-2{\cdot}s-1$, humidity of $84{\pm}1.4%$, and a photoperiod of 16/8 h (light/dark). The results showed that a mixture containing a 2:1 (v:v) ratio of horticultural substrate and perlite, increased sporophyte formation to 462.5 sporophytes per pot (7.5 cm2). The other soil substrates produced from 314.5 to 405.3 sporophytes per pot. Therefore, our results will provide conditions suitable for mass production of Polypodium vulgare L.

배양토의 조성에 따른 자생 더부살이고사리의 생육과 생리에 미치는 영향 (Influence of Soil Mixture on the Growth and Physiological Characteristics of Polystichum lepidocaulon Native Fern)

  • 본 연구는 자생지환경에서 배합토에 따른 더부살이고사리의 생육을 살펴봄으로써 적정한 생육환경을 구명함은 물론, 실내조경용 소재로 활용성을 높이기 위해 수행하였으며 그 결과는 다음과 같다. 1. 더부살이고사리는 비침상조건보다 침상조건에서 생육이 좋았으며 배합토에 있어서는 밭흙 : 마사토 : 부엽토처리구가 마사토 : 부엽토처리구나 부엽토 단용처리구보다 초장, 소엽장, 소엽폭, 엽병, 관상가치가 높았다. 2. 엽체생중과 엽건조중은 전반적으로 침상조건보다는 비침상조건에서 높았으며 침상조건에서의 엽생체중과 엽건조중은 밭흙 : 마사토 : 부엽토처리구가 다른 배합토에 비해 높았다. 3. 포자엽수는 비침상보다 침상조건에서는 증가하는 경향을 나타내었으며 마사토 : 부엽토처리구가 밭흙 : 마사토 : 부엽토처리구나 부엽토 단용처리구보다 많았다. 4. 생리활성의 경우 기공전도도, $CO_2$이용효율은 밭흙 : 마사토 : 부엽토처리구> 마사토 : 부엽토처리구> 부엽토 단용처리구 순으로 되었으나 광합성률, $CO_2$흡수율, 수분이용효율은 부엽토 단용이 다른 두가지 배합토보다 높았다.

Practical Propagation Methods for Production of Prothalli and Sporophytes in Deparia pycnosora (Christ) M. Kato

  • Deparia pycnosora (Christ) M. Kato is a fern used as ornamental plant. In addition, it is called "Teol-go-sa-ri" in Korean name. The aim of this study was to develop a practical propagation method of D. pycnosora using tissue culture technique. Prothallus obtained from spore germination was the used as experiment materials. The prothalli (300 mg) used in all experiments were sub-cultured for 8-week intervals. The most suitable media for prothallus propagation were identified by culturing 300 mg of prothalli in $1/4{\times}$, $1/2{\times}$, $1{\times}$, $2{\times}$ MS medium and in Knop medium for 8 weeks. Also, the prothalli were cultured by chopping with a scalpel. In addition, sucrose, activated charcoal, and total nitrogen source were added in different concentrations based on the culture medium selected. Cultures were maintained at a temperature of $25{\pm}1^{\circ}C$, light intensity of $30{\times}1.0{\mu}mol-m-2{\cdot}s-1$, and a photoperiod of 16/8 h (light/dark) in in vitro. The results showed that optimum was achieved prothallus fresh weight and development in $1{\times}$ MS medium. When other components were added to the basic $1{\times}$ MS medium, prothallus propagation was maximized in $1{\times}$ MS medium supplemented with 2% sucrose, 0.2% activated charcoal, and 60 mM total nitrogen. To select a suitable soil mixture for sporophyte formation, 1.0 g of prothallus was blended with distilled water, spread on five combinations of different soil substrates (decomposed granite, horticultural substrates, peat moss, and perlite), and cultivated for 12 weeks. The sporophyte cultures were maintained at a temperature of $25{\pm}1^{\circ}C$, light intensity of $43{\pm}2.0{\mu}mol-m-2{\cdot}s-1$, humidity of $84{\pm}1.4%$, and a photoperiod of 16/8 h (light/dark). As a results, horticultural substrate alone, 2:1 (v:v) mixtures of horticultural substrate and perlite, and 2:1 mixtures of horticultural substrate and decomposed granite induced 208.0, 201.3 and 248.8 sporophytes per pot, respectively. Therefore, this result could provide a practical mass propagation method of D. pycnosora

실내 습도조건이 가는쇠고사리, 석위의 생육에 미치는 영향 (Effects of Indoor Relative Humidity Conditions on the Growth of Arachniodes aristata and Pyrrosia lingua in Native Ferns)

  • 자생 양치식물중 실내식물로서 개발가치가 높은 가는쇠고사리(Arachniodes aristata )와 석위(Pyrrosia lingua)의 실내 습도조건에 따른 생육 반응을 살펴보고자 수행하였으며, 그 결과는 다음과 같다. 1. 가는고사리와 석위는 상대습도 50%, 90% 보다 25%와 같은 저습도시 식물체내수분함량, 엽록소함량, 관상가치가 저하되었다. 2. 상대습도에 대한 용토별 생육에 있어 고습도인 90%일시 용토별 생육에는 큰 차이가 나타나지 않았으나, 중습도인 50%일 경우 일반 토양이 가장 좋은 것으로 나타났다. 저습도인 25%의 경우 가는쇠고사리는 인공배합토에서, 석위는 일반토양에서 생육이 좋은 것으로 나타났다.
