약용건조과실류의 잔류농약분석 (Analysis on Pesticide Residue in Medicinal Dried Fruits)

  • 2007~2009년 서울 강북지역에서 유통되고 있는 건조과실류 4종류에 대하여 전제 213건의 시료를 수거하여 잔류 농약을 분석하였다. 20종의 농약이 검출되었으며 66건(31.0%)의 시료에서 농약이 검출되었고, 8건(3.8%)의 시료에서 잔류 허용기준을 초과하였다. 농약성분별 검출 빈도는 cypcrmethrin, fenvalerate, endosulfan, chlorpyrifos, bifenthrin 순이었다. 농약계열별 분포로는 피레스로이드계, 유기인계, 유가염소계, 다카르복시미드계 농약 순으로 검출이 높았다. 작물별 구기자(53.0%), 대추(34.9%), 복분자(7.6%), 오미자(4.6%) 순으로 검출률을 보였다. 본 연구에서 검출농약이 잔류하는 건조대추의 섭취로 인체에 유입될 원일 섭취 추정량으로 평가된 위해지수는 각 농약별 위해성 평가는 %ADI 값이 모두가 0.0001~0.0081%로서 인제 위해도는 낮은 것으로 판단되었다.

Evaluation of Dermal Absorption Rate of Pesticide Chlorpyrifos Using In Vitro Rat Dermal Tissue Model and Its Health Risk Assessment

  • All pesticides must be assessed strictly whether safe or not when agricultural operators are exposed to the pesticides in farmland. A pesticide is commonly regarded as safe when estimated dermal absorption amount is lower than the acceptable operator's exposure level (AOEL). In this study, dermal absorption rate of chlorpyrifos, a widely used organophosphate insecticide, was investigated using rat dermal tissue model. Chlorpyrifos wettable powder solved in water (250, 500 and 2,500 ppm) was applied to freshly excised rat dermal slices ($341{\sim}413{\mu}m$ thickness) on static Franz diffusion cells at $32^{\circ}C$ for 6 hours. After exposure period of 6 hours, and then washing-at residual amount of chlorpyrifos was analyzed in dermal tissues, tape strips, washing solution, washing swabs of receptor bottles and receptor fluids at 1, 2, 4, 8 and 24 hours. Chlorpyrifos was only detected in dermal tissue but not found in receptor fluid at each concentration and time point, and the absorption rate of 250, 500 and 2,500 ppm was 2.36%, 1.96% and 1.69%, respectively. The estimated exposure level of chlorpyrifos was calculated as 0.012 mg/kg bw/day. The health risk for farmers in this condition is a level of concern because the estimated exposure level is 12 times higher than AOEL 0.001 mg/kg bw/day. However, actual health risk will be alleviated than estimated because absorbed chlorpyrifos is not permeated into internal body system and only retained in skin layer.

북방수염하늘소(Monochamus saltuarius) 성충의 살충제 처리에 따른 소화 효소의 활성 변화 (Changes in Esterase Isozyme Activity After Pesticides Treatment in Digestive Juice of Monochamus saltuarius (Gebler) Adult)

  • 북방수염하늘소(M. saltuarius) 성충 장(gut)내에 존재하는 소화액에 대한 에스테라제(esterase)의 활성 변화를 in vitro에서 조사하였다. ${\alpha}$-naphthyl acetate와 ${\alpha}$-naphthyl butyrate 기질을 이용한 실험에서는 4개의 밴드가 관찰되었다. Carbofuran과 methomyl을 비교 처리한 결과 methomyl에서 강한 효소활성 저해가 관찰되었다. 특히 Est1은 carbofuran과 methomyl에 의해 모두 저해되는 것으로 나타났다. Chlorpyrifos, methidation, phenthoate를 비교 처리한 결과 chlorpyrifos에 의해 모든 밴드들이 저해되는 것을 볼 수 있었다. 특히 Est1은 chlorpyrifos에 의해 효소활성이 완전히 저해되었고, methidation 처리시 약 70% 정도의 효소활성이 저해 되었다. Eserine을 처리한 결과 Est1은 약 70% 정도의 활성 저해를, Est2, Est3, Est4에서는 미미한 효소활성 저해가 나타났다. ${\alpha}$-pinene에서는 뚜렷한 효소활성 저해는 관찰할 수 없었으나 bornyl acetate와 camphor에서는 미미한 효소활성 저해가 관찰되었다.

  • Recently, we reported an improved technology for the degradation of organophosphate nerve agents using whole cells of genetically engineered Escherichia coli that anchored and displayed the enzyme organophosphorus hydrolase on the cell surface. In this paper we report the immobilization of these cells on highly porous sintered glass beads and the subsequent application of the immobilized cell in a continuous-flow packed bed bioreactor for the biodetoxification of a widely used insecticide, coumaphos.

많은 양을 음독한 급성 amitraz 살충제 중독 1례 (A Case of Amitraz Insecticide Intoxication after Ingestion of Large Amount)

  • Amitraz is used as farm-animal insecticide. Its side effects in humans are related to its pharmacological activity on alpha 2-adrenergic receptors. The case describes a previously healthy 46-year-old woman who intentionally ingested approximately 250mL of liquid amitraz. She presented with vomiting, altered mental status, miosis, dry mouth, hypopnea, metabolic and respiratory acidosis, hypotension, hypothermia, polyuria, metabolic acidosis, elevated serum aminotransferase and abdominal distension. Supportive treatments including mechanical ventilation, hydration, dopamine infusion, bicarbonate infusion and gastric decompression resulted in improvement. By hospital day 3, she recovered with resolution of abdominal distension. It is paramount to recognize amitraz poisoning when a pesticide-intoxicated patient presets with signs and symptoms consistent with organophosphate intoxicated patients but with greater alpha 2-adrenergic related symptoms such as decreased bowel motility and xerostomia.

작업장 공기 중 파라티온과 작업자 소변 중 p-니트로페놀의 시료채취 및 분석 (Sampling and Analysis of Parathion in the Air and Urinary p-Nitrophenol for Parathion Manufacturing Workers)

  • Although parathion is an organophosphate pesticide being legally applied for the purpose of agriculture and is being manufactured, parathion in the air and urinary p-nitrophenol, a metabolite of parathion, were not analysed in Korea. Air of the parathion manufacturing workplace was sampled by OVS-2 tubes using NIOSH 5600 and spot urine of workers was sampled at the end of shift. Parathion and urinary p-nitrophenol were analysed by GC/MS (5973 MSD connected with Agilent 6890 GC) and the protocol was included in this study. It was found that this protocol should be so sensitive that determining parathion in the air and urinary p-nitrophenol below level of ACGIH TLV and BEI be adequate. Another finding was that total sampling volume of air of NIOSH 5600 of 240 L should be adjusted to be less than 120 L due to breakthrough.

유기인계 농약의 연령에 따른 감수성 차이 (Difference of Age-Related Sensitivity to Organophosphates)

  • The potential for a given anticholinesterase pesticide to exhibit age-related toxicity is essential information for an accurate and proper risk assessment of that compound. This investigation was designed to study the age-related toxicity of active metabolites of four organophosphates using in vitro detoxification measurement. The blood samples were collected from 1 month and 18 months old rats. The $IC_{50}$ values of mouse brain recombinant AChE of chlorpyrifos-oxon, diazoxon, malaoxon and paraoxon were 10.35, 112.84, 151.28 and 18.43 nM, respectively. When the plasma of young rats, and $CaCI_2$were added, the $IC_{50}$ values of mouse brain recombinant AChE of chlorpyrfos-oxon, diazoxon, malaoxon and paraoxon were 31.89, 164.25, 139.94 and 16.36 nM, respectively. The $IC_{50}$ values of mouse brain recombinant AChE of chlorpyrifos-oxon, diazoxon, malaoxon and paraoxon were changed to 136.840, 1244.45, 654.54 and 52.66 nM by A-esterases In adult rats. These results suggest that four organophosphates have a potential toxicity to exhibit age-related sensitivity.

Determination of Parathion Metabolite, p-Nitrophenol in Urine of Parathion Factory Workers

  • Parathion is an organophosphate pesticide being legally applied for the purpose of agriculture and is being manufactured in Korea. A gas chromatography/mass spectrometric method was developed for the determination of parathion urinary metabolite, p-nitrophenol. p-Nitrophenol was extracted from weak acidic urine, and then measured by gas chromatography-mass spectrometry (selected ion monitoring). The recovery of pnitrophenol in the overall procedure was 88.2%. The detection limit of the assay was 1.0 $\mu$ g/L based upon assayed urine of 2.0 mL. The method was applied to the determination of p-nitrophenol in urine of workers of a parathion industry. Spot urines of workers of a parathion industry were sampled at the end of shift and pnitrophenol was analyzed using above developed method. p-Nitrophenol could be detected in all of the urine samples at concentrations varying from 3.0 to 681 $\mu$ g/L.

In vitro Screening of Oxime Reactivators on the Model of Paraoxon-inhibited Acetylcholinesterase-SAR Study

  • Acetylcholinesterase reactivators are crucial antidotes for the treatment of organophosphate intoxication. Standard in vitro test was chosen using a rat brain homogenate as the source of AChE. Screening of reactivation potency was performed with two concentration of reactivator (1000 ${\mu}M$ and 10 ${\mu}M$). Results were compared to established reactivators pralidoxime, methoxime, HI-6, trimedoxime and obidoxime. More than 30 novel reactivators performed equal or better reactivation ability of POX-inhibited AChE compared to currently used reactivators. The structure-activity relationship for reactivators of paraoxon-inhibited AChE was developed.

Presticide Resistance Menagement of Pest and Beneficial Arthropods and More Biologically-Based IPM on Apple

  • Resistance evolution to organophosphate-based pesticides in apple and pear inhabiting arthropods of western North America extends to many classes of pest and some beneficial species. Resistance management programs to minimize resistance in pests while exploiting it in natural enemies have met with mixed success. Among beneficials, resistances have been exploited mostly among predators of pest mites. Evolution of resistant mites, leafminers, leafhopper, aphids, leafrollers and some internal fruit feeders have led to development of new monitoring methods and means to delay or avoid resistance. But it is resistance to azinphosmethyl in codling moth (Cydia pomonella) that is changing the pest control system and moving it from chemical to biologically-based means. Newly merging IPM system will depend more on use of biological, cultural, behavior and genetic controls. But more selective pesticides also will be needed to augment pheromones, resistant host plants and genetically altered organisms. These more biologically-based tactics will be prone to resistance evolution in pests as well, if used too unilaterally and/or too extensively.
