The bacteria B1-9 that was isolated from the rhizosphere of the green onion could promote growth of pepper, cucumber, tomato, and melon plants. In particular, pepper yield after B1-9 treatment on the seedling was increased about 3 times higher than that of control plants in a field experiment. Partial 16S rDNA sequences revealed that B1-9 belongs to the genus Pantoea ananatis. Pathogenecity tests showed non-pathogenic on kimchi cabbage, carrot, and onion. The functional characterization study demonstrated B1-9's ability to function in phosphate solubilization, sulfur oxidation, nitrogen fixation, and indole-3-acetic acid production. To trace colonization patterns of B1-9 in pepper plant tissues, we used $DRAQ5^{TM}$ fluorescent dye, which stains the DNAs of bacteria and plant cells. A large number of B1-9 cells were found on the surfaces of roots and stems as well as in guard cells. Furthermore, several colonized B1-9 cells resided in inner cortical plant cells. Treatment of rhizosphere regions with strain B1-9 can result in efficient colonization of plants and promote plant growth from the seedling to mature plant stage. In summary, strain B1-9 can be successfully applied in the pepper plantation because of its high colonization capacity in plant tissues, as well as properties that promote efficient plant growth.

We evaluated 209 Capsicum accessions from the National Agrobiodiversity of RDA, for their reaction against of anthracnose disease caused by Colletotrichum acutatum. Two hundred nine accessions of Capsicum were almost composed with Capsicum annuum var. annuum originated from 37 countries. The percentage of infection due to C. acutatum ranged from 11.1% to 100% on immature and matured pepper fruits at 21 days after inoculation. On immature fruits of pepper, one hundred seventy three accessions of tested pepper were found infected with C. acutatum. Out of the 173 accessions, eighty five accessions were susceptible to anthracnose with dark brown to black lesions and slowly increased in size at 21 days after inoculation. Only thirty six accessions showed resistance against C. acutatum. On mature fruits of pepper, one hundred ninety eight accessions were infected with C. acutatum at 21 days after inoculation. Twenty one accessions showed a resistance reaction against C. acutatum. The results of this work indicated that all of resistance accessions to C. acutatum were have a genetic potential for the resources of resistance can be further used in pepper breeding programme against anthracnose and also demand more detailed investigation in this.
This research investigated the application of semi-dried red pepper in Kimchi manufactured using semi-dried red pepper with seed ratios of 20, 40, 60 or 80%. Among the treatments, there were no significant changes in pH, acidity and lactic acid bacterial cell counts during the fermentation period. Kimchi made with a higher pericarp ratio had higher levels of vitamin C, ASTA value and capsainoids during fermentation. The organic acid level showed no significant difference with the various semi-dried red pepper seed ratios, and during fermentation the malic and succinic acid levels decreased while the acetic and lactic acid levels increased. In a sensory evaluation, the overall acceptance was highest for the control group, but the acceptance of Kimchi prepared with semi-dried red pepper at a seed ratio lower than 20% was not significantly different.
Previously, Pseudomonas plecoglossicida YJR13 and Pseudomonas putida YJR92 from a sequential screening procedure were proven to effectively control Phytophthora blight caused by Phytophthora capsici. In this study, we further investigated the anti-oomycete activities of these strains against mycelial growth, zoospore germination, and germ tube elongation of P. capsici. We also investigated root colonization ability of the bacterial strains in square dishes, including cell motility (swimming and swarming motilities) and biofilm formation. Both strains significantly inhibited mycelial growth in liquid and solid V8 juice media and M9 minimal media, zoospore germination, and germ tube elongation compared with Bacillus vallismortis EXTN-1 (positive biocontrol strain), Sphingomonas aquatilis KU408 (negative biocontrol strain), and MgSO4 solution (untreated control). In diluted (nutrient-deficient) V8 juice broth, the tested strain populations were maintained at >108 cells/ml, simultaneously providing mycelial inhibitory activity. Additionally, these strains colonized pepper roots at a 106 cells/ml concentration for 7 days. The root colonization of the strains was supported by strong swimming and swarming activities, biofilm formation, and chemotactic activity towards exudate components (amino acids, organic acids, and sugars) of pepper roots. Collectively, these results suggest that strains YJR13 and YJR92 can effectively suppress Phytophthora blight of pepper through direct anti-oomycete activities against mycelial growth, zoospore germination and germ tube elongation. Bacterial colonization of pepper roots may be mediated by cell motility and biofilm formation together with chemotaxis to root exudates.
In order to improve practical utility of agro-microorganisms (AMs) which had been cultured and disseminated to promote plant growth and to control crop diseases, 51 isolates of AMs were collected from 18 agricultural extension centers in local government and screened for multi-functions such as antifungal activity, activities of phosphorus solubilization, IAA and siderophore production, nitrogen fixation, and hydrolytic enzyme activity. Finally we selected one isolate showing good antifungal activity and multi-functions related to plant growth and disease control. The selected isolate, Paenibacillus polymyxa CW, showed good inhibitory effect against plant pathogens, Pyricularia gresea, Colletotrichum acutatum, Fusarium oxysporum, Phomopsis sp., Aspergillus niger, Rhizoctonia solani and Phytophthora capsici. Suppressive effect of P. polymyxa CW against the used plant pathogens except for R. solani was much higher than that of P. polymyxa AC-1 storing in National Academy of Agricultural Science. We found P. polymyxa CW isolate showed good activity in siderophore and IAA formation, and nitrogen fixation. With P. polymyxa CW isolate, siderophore formation activity was similar to that of P. polymyxa AC-1, but IAA formation and nitrogen fixation activity was much higher than that of P. polymyxa AC-1. However neither P. polymyxa CW nor P. polymyxa AC-1 showed hydrolytic enzyme (chitinase, pectinase and cellulase) activity. The treatment of P. polymyxa CW with culture suspension of different cell density ($10^8$, $10^7$. $10^6$ cfu/ml) showed that the highest density reduced incidence of red pepper powdery mildew by 68.3% after 10 days of application. As application density of P. polymyxa CW was decreased, its control efficacy was proportionally decreased. In addition, when P. polymyxa CW was treated to control tomato powdery mildew at the same concentrations and their control effects were investigated after 7 days of inoculation, disease incidence was 0.03, 19.5, 45.7%, respectively, compared to 56.3% that of untreated check. Like red pepper powdery mildew, increase of application density of P. polymyxa CW resulted in increase of its control efficacy proportionally. P. polymyxa CW showed a density-dependent control efficacy against red pepper and tomato powdery mildews. Therefore we think that mode of action of the antagonist for suppressing two powdery mildew diseases might be antibiosis and density of more than $10^8cfu/ml$ was needed to control effectively the two diseases. On this basis, we think that P. polymyxa CW can be a promising control agent for suppressing powdery mildews of red pepper and tomato.
Sufficient inorganic nitrogen supply for crop growth is crucial for economically sustainable organic farming. The effects of an application of cover crop biomass on crop growth, nitrogen utilization and carbohydrate composition were investigated during early stage. Short-term changes in soil nitrogen after incorporating fresh hairy vetch and rye shoots were measured. The inorganic nitrogen from cover crops reached the peak at 15 ($NH_4-N$) and 24 ($NO_3-N$) days after incorporation, and then decreased rapidly. The highest concentration of soil nitrate showed at 27 days of incorporation in hairy vetch and at 18 days in rye, and three fold differences exhibited between two treatments. Crop growth under hairy vetch or rye incorporation significantly differed. At 20 DAT, dry matter production in NPK and hairy vetch was about two fold greater than that in rye. Difference in decomposing rates of hairy vetch and rye had also influence on nitrogen status in leaves and roots of pepper plants. Total nitrogen was greater in NPK and hairy vetch than in rye until 20 DAT, whereas inorganic nitrogen (nitrate and nitrite) concentration was higher in rye. Temporal changes in soluble sugars and starch in pepper plants among treatments were similar, although difference in the amount existed. It was suggested that hairy vetch as an alternative nitrogen source promoted crop growth and mineral utilization during early growth stage, whereas an obvious effect in rye was not found.
BACKGROUND: In organic agriculture, various cover crops have been used to control weeds. In this study, we investigated the suppressive effects of Vicia tetrasperma (L.) Schred (Eolchigi wandu) on the occurrence of major insect pests and their natural enemies in pepper. METHODS AND RESULTS: To estimate the effect of cover-crop on arthropod diversities and occurrences of insect pest in pepper. V. tetrasperma was sowed as cover-crop plot October, 2008 and 2009. Control plot was kept bare in winter season and mulched with black plastic-film before transplanting red pepper seedlings. Pepper seedlings, Capsicum annuum, were transplanted on the 19th of May, 2009 and on the 20th of May, 2010, respectively. Five Yellow sticky traps were set and changed at 7 days interval. Densities of aphids and thrips and damaged fruit rates by oriental tobacco budworm, Helicoverpa assulta, were counted. CONCLUSION(S): Populations of aphids, thrips and braconid wasps were maintained high on V. tatrasperma before transplanting pepper seedlings and in early season of pepper. However, the densities of aphids on pepper in the control plot were much higher than in the cover-crop plot in the early stage of pepper. Damaged fruit rates by both of thrips and oriental tobacco budworm were not significantly different between the two experimental plots.
To determine the effect of winter green manure crops for tillage and no-tillage organic pepper cultivation in plastic film house, five different green manure crops were cultivated during winter season. In nutrition composition of green manure crops, total nitrogen contents were higher in Vicia hirsuta and Vicia angustifolia than any other crops. The average dry weights of green manure crops were 8.3 ton per ha in tillage and 7.0 ton per ha in no-tillage, among green manure crops that of Secale cereale was the highest. Fertilizer supply was depended on the biomass of the cultivated green manure crops and nutrition contents, total nitrogen supply of V. angustifolia was 226 kg and that of S. cereale was 251 kg per ha in tillage field. In no-tillage field, N-supply of V. angustifolia was 197 kg and that of S. cereale was 222 kg per ha. In yield components of red pepper, fruit numbers per plant were 55.5 in green manure crop tillage and 37.0 in no-tillage cultivation. Among green manure crops, the yield of dried red pepper was the highest for V. angustifolia in both green manure crop tillage and no-tillage cultivation.
Three sludge types from pharmaceutical byproducts and one sludge type from cosmetic waste-water sludge as raw materials of compost were used in a field based concrete pot ($4\;m^2$, $2\;m{\times}2\;m$) for investigating damage of red pepper cultivation. These sludges and pig manure (1 Mg/10a, dry basis) were incorporated into the upper of clay loam soil prior to transplanting with red pepper. Changes in concentration and properties of heavy metal for both of soil and plant were investigated 4 times during of red pepper growth. Plant height and stem diameter of red pepper in sludge treatments except to Pharmaceutical sludge 3 were poor than those of NPK treatment. This result were regarded as an effect of incompleted decomposition sludge which has a lot of organic matter concentration. Amount of total As was increased rapidly Jul. 8. in soil, total Zn Cu Pb Cd were in harvest time, and 1 N-HCl extractable Zn Cu Pb Cd As were in harvest at middle stage and then decreased. Amounts of nitrogen in plant (leaf and stem) were high in Phamaceutical Sludge 1 and fig Manure treatment in early and middle stage because of organic matter and nitrogen concentrations and characteristics. Amounts of Zn, Pb, and Ni in leaf and amount of Zn and Pb in stem were increased in harvest time so that we need to have a concern in detail. Total yield of red pepper was Pig Manure > Phamaceutical Sludge 3 > Phamaceutical Sludge 1 > NPK > Phamaceutical Sludge 2 and Cosmetic Sludge treatment was decreased considerably to compare to others. Amounts of Zn and Cu in green and red pepper in harvest time were higher than the other heavy metals. Finally these results can use to utilize that finding damage on crop for authorization and suitability estimation of raw material of compost.
The changes of the content of organic acids, carbon dioxide, alcohols and carbonyl compounds of the various Kimchis which were made of cabbage with green onion, garlic, ginger or red pepper and fermented at $12^{\circ}C-16^{\circ}C$ were investigated. Nonvolatile organic acids identified were lactic, succinic, fumaric and malic acid. Volatile organic acids identified were acetic, formic, propionic, butyric, valeric, n-caproic and n-heptanoic acid. Carbonyl compounds identified were acetaldehyde and acetone. The content of lactic acid was increased with fermentation, and higher in Kimchis containing red pepper, garlic and green onion. The content of acetic acid was increased with fermentation, especially in Kimchi containing garlic. The content of carbon dioxide was higher in Kimchi containing garlic. Alcohols identified in all Kimchis was only ethanol. Carbonyl compounds had no direct effect on off-flavor of Kimchi.
