3가지 약용 허브 추출물에 함유된 유기산 검색 및 조성 비교 (The Screening and Pattern Comparison of Organic Acids in 3 Kinds of Medicinal Herbal Extracts)

  • Gas chromatography를 이용한 profiling과 신속한 동정을 허브류에 존재하는 유기산의 분석에 적용하여 보았다. 흡착제로 Chromosorb P와 용출 용매로 diethyl ether를 사용하는 고체상 추출법을 이용하여 허브추출물로부터 유기산을 분리한 후, tert-butyldimethylsilyl 유도체로 만들어 극성이 다른 DB-5와 DB-17 컬럼으로 동시분석하였다. 두 컬럼에서 분리된 유기산 피크들을 dual capillary column system의 retention index(RI) library 탐색과 RI specta 비교로 신속하고 정확하게 동정할 수 있었으며 이들을 GC-MS로 확인하였다. 허브추출물들의 유기산 분석 결과 fatty acid, dioic acid, hydroxy acid, aromatic acid등 24가지의 유기산이 동정되었고 각 허브의 종류에 따라 특징적인 유기산 GC-profile을 나타내었다. 각 허브추출물의 GC profile의 유기산 조성만을 단순화시킨 RI spectra로 표현한 결과, 각 허브의 유기산 패턴의 분별이 매우 용이하였다.

로스팅 조건 변화에 따른 커피 추출액의 화학성분 및 관능 특성 (Chemical Composition and Sensory Attributes of Brewed Coffee as Affected by Roasting Conditions)

  • The objectives of this study were: 1) to examine the contents of chemical composition (chlorogenic acids, caffeine, free acids, and free sugars) and 2) to evaluate the sensory attributes (sourness, bitterness, and sweetness) of brewed coffee as affected by two roasting conditions such as varied in the roasting time with the same roasting temperature (RT) and with same color and yield (CY). Quantitative analysis of chemical components was performed using high-performance liquid chromatography (HPLC) system, and quantitative descriptive analysis (QDA) was conducted to analyze sensory attributes. Based on the results of chemical analysis, chlorogenic acids were significantly different (p<0.05) in the short term roasted samples (RT 240 and CY 240), but there was no significant difference in caffeine contents (p>0.05). Organic acid levels were different between RT and CY coffee samples. RT 240 coffee had the most level in organic acids and the longer the roasting time of coffee, the lesser the level of organic acids in coffee was found. However, there was no significant difference in CY coffee (p>0.05). The results of sensory evaluations show that the degree of roasting changed according to the roasting time despite of the roasting temperature. Long term (RT 80) coffee was relatively bitter while short term (RT 240) coffee was relatively sweeter. Also, there was no significant difference (p>0.05) in the sensory characteristics (bitterness and sweetness) of CY coffee although they were roasted at different temperatures. Therefore, the current study concluded that better understanding of proper roasting time and temperature improves the quality of brewed coffee.

소나무 수피의 페놀액화에 대한 다양한 산 촉매의 효과 (Effect of Various Types of Acid Catalysts on Phenol Liquefaction of Pinus densiflora Bark)

  • 소나무 수피의 페놀 액화시 다양한 종류의 산 촉매를 도입하여 그 효과를 검토하였다. 소나무 수피의 페놀 액화시 무기산 촉매로서 염산이 가장 효과적이었으나, 90%이상의 액화를 위하여 적어도 11 mmol이상의 염산의 첨가가 필요하였다. 사용된 카르본산류의 경우 triflouroacetic acid (TFA)가 액화에 가장 효과적이었으나 80%이상의 액화는 어려웠다. 그러나 유기설폰산 촉매류, 즉, p-toluenesulfonic acid (PTSA) 및 methanesulfonic acid (MSA)의 경우 소량의 첨가로도 낮은 액화온도에서 높은 액화율을 얻을 수 있었다. 특히 PTSA는 140℃의 낮은 액화 온도 조건에서도 92%의 액화율을 나타내어 수피 페놀액화 시 매우 효과적인 촉매로 판명되었었다.

  • The piperazine salts with 24 kinds of phenols and 6 kinds of organic carboxylic acids were prepared. The test of anthelmintic effect against Ascaris suilla, in vitro, for Hexachlorophene-piperazine salt, Bithionol-piperazine salt, Kainic acid-piperazine salt were also carried out compared with piperazine, Hexachlorophene, Bithionol and Kainic acid.

좀가시 오갈피나무의 성분연구 (Phytochemical Studies on the Barks of Acanthopanax senticosus forma intermis)

  • Chemical constituents of fruits, leaves and barks of Acanthopanax senticosus forma inermis were studied. Their fruits have higher contents of crude ash, crude protein, crude fat, fructose and glucose than those of other Acanthopanax species, and contained larger amount of glutamic acid and malic acid among amino acid and organic acid, respectively. The compounds identified from their barks and leaves, were $\beta$-sitosterol and stigmasterol, sesamin, savinin, syringaresinol diglucoside, oleanolic acid, chiisanoside and polyacetytene ($C_9H_{10}O_2$, mp. 62~63).

  • The changes of contents of organic acids in flue-cured tobacco leaves depending on stalk position and maturity viz immature, mature and overmature were studied in 1982-83. Among them, particularly, the leaves harvested at mature stage have the lowest amounts of oxalic and citric acids in smoking or cutter leaves but the acid contents has increased during the overmature. The linolenic acid content tended to be lower with increase of maturity.

Optimization of Headspace Sampling Using Solid Phase Microextraction For Volatile Organic Acids in Different Tobacco Types

  • A Solid-phase micro extraction(SPME) was evaluated as a tool for headspace sampling of tobacco samples. Several experimental parameters (sampling temperature, pH, and type of SPME fibers) were optimized to improve sampling efficiency in two aspects ; maximum adsorption and selective adsorption of volatile organic acids onto SPME fibers. Among four types of SPME fibers such as PDMS(Polydimethylsiloxane), PA(Polyacrylate), Car/PDMS (Carboxen/Polydimethylsiioxane) and PDMS/DVB(Polydimethylsiioxane/Divinylbenzene) which were investigated to determine the selectivity and adsorption efficiency. A variety of tobacco samples such as flue cured, burley and oriental were used in this study. The effect of these parameters was often dominated by the physical and chemical nature (volatility, polarity) of target compounds. This method allowed us to make important improvements in selectivity and sensitivity. The Car/PDMS fiber was shown to be the most efficient at extracting the 10 selected volatile organic acids. The parameters were optimized: $80^{\circ}C$ adsorption temperature, 30 min of adsorption time, $240^{\circ}C$ desorption temperature, 1 min of adsorption time.

된장 숙성중 정미성분의 변화에 관한 연구 (II) -유리당과 휘발성, 비휘발성 유기산 (Studies on the Changes of Taste Compounds during Soy Paste Fermentation(II))

  • 조미료로서 된장의 수용도를 높일 수 있는 자료를 제공하고자 황곡균으로 개량식 된장을 제조하고 이것을 180일 간 숙성시키면서 맛성분으로 중요한 역할을 하는 유리당, 휘발성 유기산, 비휘발성 유기산을 분석하였다. 그 결과 된장의 감미원인 유리당은 glucose>xylose> arabinose순서로 많이 존재하였다. 휘발성 유기산은 ace-tic, formic, butyric, propionic acid가 검출되었고 총량은 숙성 60일가지 증가하였다. 180일 숙성 후, 휘발성유기 산의 함량은 acetic>propioniclbutyric>formic acid순서 로 많이 존재하였다. 비휘발성 유기산은 담금 직후부터 succinic>glutaric>lactir acid순서로 많았으며 숙성 45일부터 tartaric>citric>malic이 뒤를 이은 순서로 생성되었다.

Microflora Management in the Gastrointestinal Tract of Piglets

  • The stressful physiological and environmental conditions around weaning often promote the proliferation of pathogens in the digestive tract of piglets resulting in diarrhoea and reduced daily weight gain. Typical dietary practices to maintain growth performance and health have led to an increased use of antimicrobial growth promoters. Due to the advanced ban of antibiotics in pig production, new concepts have been developed to secure animal health and growth performance, feed efficiency, and product quality as well. Several naturally occurring compounds seem to beneficially affect the composition and activity of the microflora in the gastrointestinal tract (GIT) of pigs. These are, among others, organic acids, probiotics, prebiotics, and enzymes. Organic acids are already widely used, especially in pigs, due to their positive effects on GIT health and growth performance. Probiotics have been shown to be effective against diarrhoea though effects may be dependent on diet composition and environmental conditions. Prebiotics may influence composition and activity of the intestinal microflora. Additionally, pre- and probiotics may exert positive influences on immune response, whereas enzymes may enhance feed digestibility by breaking down anti-nutritional factors. In the following, the focus will be directed to the role of organic acids, probiotics, prebiotics, and feeding enzymes as potential modulators of GIT health.

GC-MS 크로마토그램의 컴퓨터 자동해석을 이용한 유전성 대사질환의 진단법 개발 (Development of a GC-MS Diagnostic Method with Computer-aided Automatic Interpretation for Metabolic Disorders)

  • Purpose: A personal computer-based system was developed for automated metabolic profiling of organic aciduria and aminoacidopathy by gas chromatography-mass spectrometry and data interpretation for the diagnosis of metabolic disorders Methods: For automatic data profiling and interpretation, we compiled retention time, two target ions and their intensity ratio for 77 organic acids and 13 amino acids metabolites. Metabolites above the cut-off values were flagged as abnormal compounds. The data interpretation was a based on combination of flagged metabolites. Diagnostic or index metabolites were categorized into three groups, "and", "or" and "NO" compiled for each disorder to improve the specificity of the diagnosis. Groups "and" and "or" comprised essential and optional compounds, respectively, to reach a specific diagnosis. Group "NO" comprised metabolites that must be absent to make a definite diagnosis. We tested this system by analyzing patients with confirmed Propionic aciduria and others. Results: In all cases, the diagnostic metabolites were identified and correct diagnosis was founded to be made among the possible disease suggested by the system. Conclusion: The study showed that the developed method could be the method of choices in rapid, sensitive and simultaneous screening for organic aciduria and amino acidopathy with this simplified automated system.
