Inhibition of Compylobacter jejuni in Chicken by Ethanol, Hydrogen Peroxide, and Organic Acids

  • Growth inhibition of Compylobacter jejuni ATCC 33291 was observed in the presence of various preservatives at various temperatures. The addition of ethanol (0.5% to 5%), hydrogen peroxide (0.05%), acetic acid (1%), propionic acid, benzoic acid, and sorbic acid showed strong antibacterial activities against C. jejuni at pH 5.5 or 6.5. The addition of 1% acetic acid and lactic acid were most effective at $42{\circ}C, followed by $25{\circ}C$ and $4{\circ}C$. This indicated that the inhibitory effect was temperature dependent. In the chicken model system, the practical death rate of C. jejuni in the FBP-media with 1% acetic temperatures ($4{\circ}$C$, $25{\circ}$C, and $42{\circ}$). Therefore, precaution has to be taken in the use of organic acids as a disinfectant in the chicken slaughterhouse.

  • pKi values and sensitivity of acid-base indicators in the mixed solvent systems of water-alcohols and water-acetone at differential pH and in concentration media were determined by spectrophotometric method. When the concentration of the organic solvents were increased, the pKi values of sulfophthalein indicators, thymol blue and bromcresol purple, became larger whereas those of an axo indicator, methyl yellow, became smaller nad the pH values of the equivalent points of acids became smaller and the pH values of the equivalent points of acids became higher than those in the aqueous solution. potassium bicarbonate, a standard compound used in the organic solvent systems without a prerequisite procedure of expelling carbon dioxide by boiling.

  • Herbchoke solution produced by Choa pharmaceutical company contained pyroligneous liquid and extracts from artichoke leaves. Pyroligneous liquid is produced by process carbonizing Oak in 350∼400$^{\circ}C$. It contain water 96%, acetic acid 3% and organic compound 1%. There are 200 kinds of constituents including minerals, vitamin B-complex and organic acids in it. The organic acids of them were presumed as active materials. It is traditionally used for treatment of stress related disorder, hepatic disease, immune disorder, G-I disorder and inflammatory disease. Extracts from artichoke leaves are traditionally used for treatment of stress related disorder, that is, hepatic disease, dyspeptic disorder, hyperlipidemic disorder and diuretic disorder. The aim of this study was to investigate anti-stress effects of Hervchoke juice produced from Choa pharmaceutical company.

Controls of the Hydrolysis of Ginseng Saponins by Neutralization of Organic Acids in Red Ginseng Extract Preparations (홍삼의 가열추출 과정중 유기산 중화에 의한 사포닌의 가수분해 억제)

  • Glucosidic bonds at the C20 position of the sapogenins were hydrolyzed easily in the lower pH, higher temperatures and longer times to give prosapogenins and sugars. The glucosidic bond of saponin at the C3 of ginsenoside-Rb1, which is secondary carbon, was relatively stable due to the low electron density of -0.2. But the bond of saponin at the C20 position, which is tertiary carbon with the relatively high electron density of -0.3, was liable to be hydrolyzed even in weakly acidic solution by the increase of heating time. On the other hand, red ginseng contained 13.34 mg/g of citric acid, 8.78 mg/g of malonic acid, 3.70 mg/g of oxalic acid, 2.13 mg/g of malic acid and 0.44 mg/g of succinct acid. Ginseng saponins were very stable in ginseng extract neutralized with sodium carbonate or sodium bicarbonate corresponding to the equivalent amount of the total organic acid in the red ginseng.

  • The seasonal patterns of dissolved organic matter (DOM) in Lake Kasumigaura, a shallow, eutrophic lake, and serveral DOM sources in its catchment area were investigated. DOM was fractionated using three resin adsorbents into classes: aquatic humic substances (AHS=humic acid+fulvic acid), hydrophobic neutrals (HoN), hydrophilic acids (HiA), bases (BaS) and hydrophilic neutrals (HiN). The DOM produced significantly different fraction distributions depending on the origin of sample. AHS and HiA prevailed over AHS in the lake while AHS and HiA existed at almost the same concentration levels in the rivers. AHS seems to be a more dominant component in rever water than lake water. The dominance of organic acids was also observed in the DOM sources: forest stream (FS), plowed field percolate (PFP), domestic sewage (DS) and sewage treatment plant effluent (STPE).

Zinc Chloride Toxicity on Free Proline and Organic Acids in Germinating Rice Seed

  • The study was conducted to find the critical concentrations of zinc toxicity and to determine the changes of the contents of free proline and organic acids with treatment of different zinc chloride concentrations during rice germination and seedlings grown for seven days. The concentration of zinc chloride, 140 ppm, inhibited root elongation as much as 46 times compared with the control, and the germination rate was also decreased in all treatments of zinc chloride, showing that the germination rate decreased more with increasing concentrations of zinc chloride. Its rate was only 13% with treatment of 140 ppm zinc chloride. The content of free proline with treatment of zinc chloride, 140 ppm, was highest about 4,873 $\mu$M at 3 days compared with the control. Malic acid concentration with treatment of zinc chloride, 140 ppm, increased to approximately 4 times compared to the control. Citric and succinic acid content were also slightly increased in all treatments of zinc chloride.

  • As a preliminary experiment to investigate the effect of ozone sterelization on the ginseng powder, the changes of fatty acid composition and organic acid content and sensory properties in ginseng powder treated with ozone was investigated. Ginseng powder was treated with 0.5 ppm ozone for 24 hours and 48 hours, respectively. With increase in ozone treatment time, the content of crude lipid and increase. Contents of unsaturated fatty acid-linolenic acid, linoleic acid, and oleic acid, etc.-decreased with ozone treatment time, whereas saturated fatty acid or same number-stearic acid-increased odor, bitter taste, roasted taste and sweet taste were thiner with ozone treatment times, but pungent taste was thicker with those treatment. These changes of ordors and tastes of ginseng powder with ozone treatment were predicted by oxydation of lipids, flavor components and saponins.

  • 나뭇잎으로부터 발생되는 저분자량 유기산을 이온크로마토그래피를 이용하여 분석하였다. 이 방법은 자연상태의 나뭇잎 (needle litter)을 유기용매의 추출 없이 그대로 이용할 수 있어서 사용하기에 편리하다. 본 논문에서는 이온 크로마토그래피의 사용방법과 침엽수잎(Pacific Silver Fir)의 부후정도에 따른 유기산의 종류와 농도의 변이를 간략하게 소개한다. 초기의 침엽수잎 여과액으로부터는 $C_2$에서 $C_4$까지의 아세트산(acetic acid), 프로피온산(propionic acid), 옥살산(oxalic acid) 그리고 포름산(formic acid) 등이 주종을 이루지만 부후정도가 커짐에 따라 방향핵을 가진 방향핵 유기산(aromatic organic acid)이 발생한다.

  • The approximate rate and stoichiometry of the reaction of excess Thexylbromoborane-methyl sulfide, $ThxBHBr{\cdot}SMe_2,$ with selected organic compounds containing representative functional groups under standardized conditions (methylene chloride, $0^{\circ}C)$ were studied in order to characterize the reducing characteristics of the reagent for selective reductions. The selectivity of the reagent was also compared to the selectivity of thexylchloroborane-methyl sulfide. Thexylbromoborane appears to be a much milder and hence more selective reducing agent than thexylchloroborane. The reagent tolerates many organic functionalities. Thus, the reagent shows very little reactivity or no reactivity toward acid chlorides, esters, epoxides, amides, nitro compounds including simple olefins. However, this reagent can reduce aldehydes, ketones, carboxylic acids, nitriles, and sulfoxides. Especially the reagent reduces carboxylic acids including ${\alpha},{\beta}$ -unsaturated ones and nitriles to the corresponding aldehydes. In addition to that, thexylbromoborane shows good stereoselectivity toward cyclic ketones, much better than the chloro-derivative.

Hydrogen Gas Production by Fermentation from Various Sugars Using Clostridium butyricum NCIB 9576 (Clostridium butyricum NCIB 9576에 의한 당으로부터 혐기적 수소생산)

  • Clostridium butyricum NCIB 9576 evolved hydrogen gas and produced various organic acids from glucose, lactose, starch, and glycerol. Total amount of hydrogen gas produced from 1 and 2% glucose were 630 and 950ml $H_2$/l-broth, respectively, for the first 24 hrs of incubation and the maximum hydrogen production rates were 42 and 94ml $H_2$/hr/1-broth, respectively. Teh initial pH 6.8 decreased to 4.2~4.5 during the first 12~16 hrs of fermentation when the pH was not controlled, resulting in ceasing the cell growth and hydrogen evolution and in degradation of 82 and 40% glucose after 24hrs of incubation from 1 and 2% glucose, respectively. When pH was controlled to 5.5, glucose was consumed completely and resulted in increasing hydrogen production approximately 38~50% compared to the experiments without the pH control. C. butyricum NCIB 9576 produced hydrogen gas approximately 644, 1,700 and 3,080 ml $H_2$/l-broth with 0.5, 1 and 2% lactose, respectively and the maximum hydrogen production rates were 41, 141 and 179ml $H_2$/hr/l-broth, respectively. All of the lactose added was degraded completely during fermentation even though pH was not controlled. C. butyricum NCIB 9576 produced 183 and 709ml $H_2$/l-broth with 0.1 and 0.5% starch for 48 hrs, respectively, when pH was not controlled. The maximum rates of hydrogen gas production were 43 and 186ml $H_2$/l-broth, respectively and 80~100% of starch added was fermented. Approximately 107ml $H_2$/l-broth was produced using 1% glycerol by C. butyricum NCIB 9576 and the pH was maintained higher than 6.1 during fermentation without pH control. The degradation of glucose, lactose, starch and glycerol by C. butyricum NCIB 9576 were affected by the pH of fermentation broth and the organic acids released during fermentation. The pH of feremtntation broth dropped to 4.2~4.6 after 12~14 hrs incubation when glucose was used as a substrate while pHs were maintained above pH 5 under the same experimental conditions when lactose, starch and glycerol were used. The organic solvents and acids produced during glucose fermentation were mainly ethanol, butyrate, acetate and a little of propionate, while butyrate was the main organic acids during the lactose, starch, and glycerol fermentation by C. butyricum NCIB 9576.
