국내에서 인공재배되고 있는 느타리버섯 (Pleurotus ostreatus), 표고버섯 (Lentinus edodes), 양송이버섯 (Agaricus bisporus)의 유기산 및 지방산 조성을 GLC법에 의하여 분석하고 버섯의 종류, 크기, 부위에 따른 성분조성의 차이를 비교 검토하였다. 유기산은 3종의 버섯에서 lactic, oxalic, fumaric, succimic, malic, citric 및 pyroglutamic acid등이 확인되었고 malic, citric, pyroglutamic acid등이 많이 함유되어 있었다. 총 유기산 함량은 느타리버섯 $2.4{\sim}4.0%$, 표고버섯 $1.7{\sim}3.6%$, 양송이버섯 $1.9{\sim}3.1%$이었다. 부위별로는 갓부위가 자루부위보다 많았는데 느타리버섯에서는 oxalic과 pyroglutamic acid, 표고버섯은 malic과 citric acid, 양송이버섯은 fumaric acid가 특히 많았으며 크기별로는 크기가 클수록 총유기산 함량은 감소하는 경향이었고 특히 citric acid의 감소가 현저하였다. 지방산은 linoleic, palmitic 및 oleic acid가 주요 지방산이었으며 부위 및 크기별로 비교했을 때는 지방산 조성에 뚜렷한 차이가 없었다.
간단하고, 경제적이고, 다양한 응용이 가능한 시스템을 이용하여 거울상 이성질체를 분리 및 분취하였다. 소수성 아미노산 이성질체를 N-hexadecyl-L-proline이 코팅된 역상 원심 액체크로마토그래피로써 분리, 분취하였다. 아미노산 이성질체의 용리 및 분리에 영향을 주는 구리(Ⅱ) 농도, pH, 이동상의 유기용매 종류 및 농도, 원심 액체크로마토그래피의 원형 컬럼의 회전속도 등에 대하여 연구하였다. 수 mg의 소수성 아미노산을 분취하였다. 모든 다른 아미노산을 분리 및 분취하려면, 다양한 코팅 리간드에 대하여 연구하여야 한다.
A gas chromatography/mass spectrometric (GC/MS) method has been developed for the determination of trace mono-n-butyltin (MBT), di-n-butyltin (DBT), and tri-n-butyltin (TBT) compounds in sediments. Samples were extracted by 10% acetic acid in methanol containing 0.03% tropolone and were then derivatized for GC/MS analysis. Ethylation by sodium tetraethylborate and phenylation by sodium tetraphenylborate were evaluated as a derivatization reaction of the organotins in sample extract. n-Hexane was added into reaction media in the beginning of the reaction for the continuous extraction of derivatized organotins. Ethylation requires less than 2 hours to get proper derivatization yields for MBT, DBT, and TBT altogether and produces relatively low amounts of side reaction products. Compared to ethylation, phenylation requires much longer time but provides relatively lower yield and produces considerable amounts of side reaction products. Therefore, the ethylation reaction was applied for the analysis of organotin compounds in sediment. An isotope dilution mass spectrometric (IDMS) method based on GC/MS has been applied to the accurate determination of DBT compounds in the sediments. The IDMS results from the analyses of sediment samples showed a reasonable repeatability and a good agreement with the values obtained by IDMS based on liquid chromatography/induced coupled plasma/mass spectrometry.
This study analyzed the effect of organic solvent extractives on the classification of wood species via near-infrared spectroscopy (NIR). In our previous research, five species of Korean softwood were classified into three groups (i.e., Cryptomeria japonica (cedar)/Chamaecyparis obtuse (cypress), Pinus densiflora (red pine)/Pinus koraiensis (Korean pine), and Larix kaempferi (Larch)) using an NIR-based principal component analysis method. Similar tendencies of extractive distribution were observed among the three groups in that study. Therefore, in this study, we qualitatively analyzed extractives extracted by an organic solvent and analyzed the NIR spectra in terms of the extractives' chemical structure and band assignment to determine their effect in more detail. Cedar/cypress showed a similar NIR spectra patterns by removing the extractives at 1695, 1724, and 2291 nm. D-pinitol, which was detected in cedar, contributed to that wavelength. Red pine/Korean pine showed spectra changes at 1616, 1695, 1681, 1705, 1724, 1731, 1765, 1780, and 2300 nm. Diterpenoids and fatty acid, which have a carboxylic group and an aliphatic double bond, contributed to that wavelength. Larch showed a catechin peak in gas chromatography and mass spectroscopy analysis, but it exhibited very small NIR spectra changes. The aromatic bond in larch seemed to have low sensitivity because of the 1st overtone of the O-H bond of the sawdust cellulose. The three groups sorted via NIR spectroscopy in the previous research showed quite different compositions of extractives, in accordance with the NIR band assignment. Thus, organic solvent extractives are expected to affect the classification of wood species using NIR spectroscopy.
자소자를 무 무게에 대해 0, 0.25, 0.5, 0.75, 1.0%의 비율로 첨가하여 담근 동치미 중 자소자를 첨가하지 않은 대조구와 전보에서 가장 기호도에서 가장 좋은 평가를 받고 이화학적으로 가장 바람직한 결과를 나타낸 0.5% 첨가한 동치미의 유리당, 유리아미노산, 휘발성 유기산 및 향기성분을 알아본 결과 다음과 같다. 1. Glucose와 fructose의 함량은 발효가 진행되면서 감소하는 반면 glycerol의 경우에는 동치미 담금직후 및 적숙기에는 존재하지 않았으나 서서히 증가하여 발효 말기인 45일에 나타나는 경향이었다. 유리당 함량은 자소자를 0.5% 첨가한 경우가 자소자를 첨가하지 않은 대조구보다 약간 많았다. 2. 유리아미노산은 대조구의 경우 저장기간이 증가함에 따라 서서히 증가하였으나 0.5%의 자소자를 첨가하여 담근 동치미의 유리아미노산은 맛이 좋아지는 시기인 발효 11일까지는 다량의 아미노산을 함유하였으나 저장기간이 증가함에 따라 감소하는 경향으로 나타났다. 3. 확인된 비휘발성 유기산은 latic acid, fumaric acid, succinic acid, malic acid, tartaric acid, citric acid 였는데, 발효가 진행됨에 따라 lactic acid와 succinic acid만이 점차로 증가하였고, 나머지는 감소하였다. 4. 동치미의 향기성분은 담금 즉시에는 0%와 0.5%를 비교한 결과 두 처리구 모두 함황 화합물이 대부분을 차지하여 동치미의 주 향기성분이었는데, 발효가 진행되면서 함황 화합물의 수나 %area가 약간 감소하였고, 산류나 알코올류 등이 증가하였다 이에 반해 자소자 처리구의 경우는 산류의 큰 증가를 보이지 않았고, 또한 함황 화합물의 변화가 두드러지지 않았고, 거의 유지하는 수준이었다. 본 실험의 결과 자소자를 첨가하지 않은 대조구보다 0.5% 처리구가 유리당, 유리아미노산, 비휘발성 유기산의 함량이 다소 높게 나타나 전보에서 가장 기호도가 높게 평가된 자소자 0.5% 첨가한 것과 일치하는 결과를 보였다.
Ground-based measurements were conducted from January 6 to 12 of 2015 for understanding characteristics of nitrogen containing carbonaceous aerosols as 16 amino acids at the Mokpo National University, Korea. The detailed amino acid components such as Cystine ($(SCH_2CH(NH_2)CO_2H)_2$) and Methionine ($C_5H_{11}NO_2S$) and their sources were analyzed by High-Performance Liquid Chromatography with Fluorescence Detection (HPLC-FLD) for behavior of secondary products in particulate matter. In addition, organic carbon (OC) and elemental carbon (EC) based on the carbonaceous thermal distribution (CTD), which provides detailed carbon signature characteristics relative to analytical temperature, and water soluble organic carbon (WSOC) by total organic carbon (TOC) analyzer were used to understand the carbon compound behaviors. The backward trajectories were discussed for originations of carbonaceous aerosols as well. Different airmasses were classified with the amino acids and OC thermal signatures. The results can provide to understand the aging process influenced by the long-range transport from East Sea area.
An optically active diacid containing the L-histidine moiety was prepared by reacting pyromellitic dianhydride (1,2,4,5-benzenetetracarboxylic acid 1,2,4,5-dianhydride) 1 with L-histidine 2 in acetic acid, and was polymerized with several aromatic diamines 5a-g to obtain a new series of optically active poly(amide-imide)s (PAIs) using two different methods, such as direct polycondensation in a medium consisting of N-methyl-2-pyrrolidone (NMP)/triphenyl phosphite (TPP)/calcium chloride ($CaCl_2$)/pyridine (Py) and direct polycondensation in a tosyl chloride (TsCl)/pyridine (Py)/N,N-dimethylformamide (DMF) system as a condensation agent. The resulting new polymers 6a-g with inherent viscosity was obtained in good yield. The polymers were readily soluble in polar organic solvents, such as N,N-dimethyacetamide (DMAc), N,N-dimethyformamide (DMF), and dimethyl sulfoxide (DMSO). The obtained polymers were characterized by FTIR, specific rotation, elemental analysis as well as $^1$H-NMR spectroscopy and gel permeation chromatography (GPC). The thermal stability of the resulting PAIs was evaluated with thermogravimetric analysis techniques under a nitrogen atmosphere.
벤조산과 그 유도체들에 대한 음이온 교환분리를 여러 농도의 염화니켈-메탄올 용매 내에서 연구하였다. Amberlite CG-400, $Cl^-$ form에 대한 유기산들의 부피분배계수를 측정하고 이 분배계수로부터 제시된 적절한 농도의 $NiCl_2-MeOH$ 용액을 용리액으로 사용하여 몇 개의 혼합유기산을 정량적으로 분리하였다. 모든 유기산들의 농도는 자외선 분광광도계를 사용하여 정량하였다.
본 연구는 모과의 비휘발성 성분에 과한 조성을 규명하기 위한 기초 연구 과제로써 행하여졌다. 주요 비휘발성 성분은 유리 아미노산의 경우 valine, asparagin ${\gamma}-aminobutyric\;acid$, aspartic acid 와 serine 이 전체의 72%를 차지하고 arginine, tyrosine, methionine과 tryptophan은 거의 검출되지 않아다. Peptide 구성 아미노산으로는 주로 glutamic acid와 glutamine이었으며 cysteic acid, methione sulfone과 tryptophan은 검출되지 않았다. 핵산관련 물질의 분석 결과 cytosine, UMP, CMP는 소량 존재하셨으며 GMP, IMP, AMP는 검출되지 않았다. 당 분석결과 주성분은 glucose, sorbose, sucrose, fructose였으며 fructose 함량이 가장 높은 것으로 판명되었다. GC 및 GC-MS방법에 의해 총 11성분의 유기산이 동정 되었으며 이들중 tartaric acid및 ${\alpha}-ketoglutaric\;acid$가 주성분이었다. 정량된 총 비타민 C 함량은 386.6 mg%였으며 ascorbic-acid dehydroascorbic acid와 2,3-diketo-L-gulonic acid는 각각 28.8mg%, 154.5mg%, 197.3mg%였다. 무기성분으로는 칼슘과 인이 주요 성분으로 나타났으며 카드뮴, 구리, 납은 소량 존재하였다. 모과의 천연 및 합성 extract에 대한 관능검사의 결과 유리형, 아미노산, 당, 유기산, 비타민 C및 무기질의 맛의 주성분으로 판명되었다. 따라서 모과의 향미 성분 중 비휘발성 성분은 주로 상기 다섯 그룹에 의해서 나타남을 확인하였다.
