Neuroprotective Effects of Korean Kiwifruit against t-BHP-induced Cell Damage in PC12 Cells (국내산 참다래 추출물의 신경독성 방어효과)

  • Oxidative stress induced by reactive oxygen intermediates has been implicated in a variety of human diseases including neurodegenerative disorders, cancer, cardiovascular and respiratory diseases, and mode of action of environmental toxicants. Tert-butylhydroperoxide (t-BHP) is an organic lipid hydroperoxide analogue, which is commonly used as a pro-oxidant for evaluating mechanisms involving oxidative stress in cells and tissues. In this study, the underlying mechanisms involved in the protective effects of Hwabuk 94 kiwifruit (Actinidia deliciosa cv. 'Hwabuk 94'), which is cultivated in Jeju, on the t-BHP-induced cytotoxicity in PC12 cell. The pretreatment of rat pheochromocytoma cell line PC12 with Hwabuk 94 extract ($1-100\;{\mu}g/ml$) resulted in a significant recovery from t-BHP-induced cell death and increased Bcl-2 and procaspase-3 expression, whereas the expression of Bax and cleaved PARP were decreased in a dose-dependent manner compared to the control. Furthermore, Hwabuk 94 inhibited the t-BHP-induced p38 MAP kinase and extracellular signal-regulated kinase 1/2, but not c-Jun N-terminal kinase activations. Finally, these findings suggest that Hwabuk 94 kiwifruit might attenuate t-BHP-induced PC12 cell cytotoxicity, at least in part, through the inhibition of signaling pathways mediated by the ERK1/2 and p38 MAP kinase.

  • The present experiment was conducted to investigate causes affecting the reduction of red pepper production in the continuous cultivation upland soil from 1985 to 1986 in Imsil Chonbuk Province. The results obstained are summarized as follows: 1. Area ratio of continuous cultivation 2 years was 12.7%, 3 years 6.8% and over 4 years 48.9%. 2. Soil hardness and volume of solid and liguid of red pepper continuous cultivation soil were higher than those of one year cultivation, while pH and content of organic matter of continuous cultivation soil were low. The exceeding optimum level of phosphorus and potassium appeared factors affecting and reduction of red pepper of continuous cultivation soil. 3. Microflora density in continuous cultivation soil was high but bacteria/fungi (B/F) and actinomycetes/fungi (A/F) ratio were low. 4. The density of soil nematodes in continuous cultivation soil were higher than that in one year cultivation soil, however, the steeper and better drainage soil lowered the density of nematodes. 5. Continuous cultivation over 4 years showed 14.3% of plants diseased by phytophthora while 0.7% in one year cultivation soil.

Effect of Nitrogen Level on Yield and Quality of Gyokuro Tea (질소비료 시용량에 따른 옥로차의 수량 및 품질)

  • The reasonable level of nitrogen fertilizer of Gyokuro tea which is producted fresh leaves plucked from shade tea plants is a key factor to increase crop yield and quality. Nitrogen were applied at the level to 60, 80, 100 and $120kg\;10a^{-1}$. Analytical results of yield and quality of Gyokuro tea are summarized as follows : The soil fertility of experimental field was higher in organic matter($59.8g\;kg^{-1}$) and available phosphate($1,285mg\;kg^{-1}$), but lower in pH(4.79) compared to the common field. As the amount of nitrogen fertilizer application was increased to $80kg\;10a^{-1}$, the yield of tea leaves, the content of total nitrogen, total amino acid, caffeine and chlorophyll were increased. However, the yield of tea leaves was not increased above $100kg\;10a^{-1}$ and the increased level of nitrogen fertilizer did not show any different in the content of tannin and vitamin C. When the amount of nitrogen fertilizer application was raised, the content of thiamine and arginine increased, but the content of aspartic acid, serine and glutamic acid decreased. The content of fatty acid was produced $2.850{\sim}3.012mg\;100g^{-1}$ Especially, the content of oleic acid, linoleic acid and linolenic acid was higher at the level of $80kg\;10a^{-1}$ application than other treatments. As nitrogen was applied at $80kg\;10a^{-1}$, sensory score was 0.2~4.6 point higher than other treatments. Consequently, $80kg\;10a^{-1}$ is considered to be the best level of nitrogen fertilizer both quality and crop yield.

  • As a basic research for developing a sludge application technique in forestry, this study was conducted to investigage a reasonable amount and period of sludge to be applied onto forest nursery of Acer palmatum Thunb. The paper mill sludge was used for the study, and ammonium nitrate was treated to some of them to reduce the C/N ratio lower than 40. The maximum amount of applied sludge was 7ton/ha/year, and they were spread on spring and on late summer. The sludge application did not stimulate early growth of the trees during a year. Since there was no significant changes in soil chemical properties, the amount of applied sludge was thought to be too little to result in a significant role for tree growth. However, the soil water showed significant increase of Cu when the activated sludges were applied during late summer. Thus, the time of application need to be determined cautiously with considering the risk of ground water contamination. In conclusion, if we set a goal from the view point of long-term concept such as forest productivity rather than to get a short-term product, the uncomposted sludge in small amount during growing season might be considered to be applied on forest nursery.

Soil and Leaf Nutrient Properties by Establishment Periods of Chesnut (Castanea crenata Sieb. et. Zucc.) Orchards in Sancheong-gun (경상남도 산청군 밤나무 재배지의 조성 기간에 따른 토양 및 잎 양분 특성)

  • Optimum soil management of chestnut orchards is important to ensure high quality and yields of chestnut. This study was conducted to evaluate soil and green leaf characteristics by establishment periods of chestnut (Castanea crenata Sieb. et. Zucc.) orchards in Sancheong-gun, Gyeongsangnam-do. Total 30 plots with the criteria of similar cultivation practices were chosen and classified into two establishment periods by over 20-year-old orchards (17 plots) and below 19-year-old orchards (13 plots). Soil bulk density was significantly higher (p<0.05) in over 20-year-old ($1.16g\;cm^{-3}$) than in below 19-year-old ($1.03g\;cm^{-3}$) plots. Soils in over 20-year-old plots were severely acidified with pH 4.56 compared to pH 4.73 in below 19-year-old plots. However, soil organic matter, total nitrogen, available phosphorus, and potassium were not significantly different (p>0.05) between both establishment periods. Leaf area, leaf mass, and nutrient (N, P, K, Ca, Mg) concentration of green leaves were also not significantly different (p>0.05) between both establishment periods. The results indicate that soil bulk density in chestnut orchards is dependent on the establishment periods, while soil chemical property and leaf nutrient concentration may be little influenced by the establishment periods.

Application of lactic acid bacteria producing antifungal substance and carboxylesterase on whole crop rice silage with different dry matter

  • Objective: This study was conducted to investigate effects of antifungal substance and carboxylesterase-producing inoculant on fermentation indices and rumen degradation kinetics of whole crop rice (WCR) silage ensiled at different dry matter (DM) contents. Methods: Dual-purpose inoculants, Lactobacillus brevis 5M2 and Lactobacillus buchneri 6M1, confirmed both activities of antifungal and carboxylesterase in the previous study. The WCR at mature stage was chopped, and then wilted to obtain three different DM contents consisting of 35.4%, 43.6%, and 51.5%. All WCR forages were applied distilled water (CON) or mixed inoculants with 1:1 ratio at 1×105 colony forming unit/g (INO), and ensiled into 20 L mini silo (5 kg) in quadruplicates for 108 d. Results: The INO silages had lower lactate (p<0.001) and butyrate (p = 0.022) with higher acetate (p<0.001) and propionate (p<0.001) than those of CON silages. Ammonia-N (p<0.001), lactate (tendency; p = 0.068), acetate (p = 0.030), and butyrate (p<0.001) concentrations of INO silages decreased linearly with increasing DM content of WCR forage. The INO silages presented higher lactic acid bacteria (p<0.001) with lower molds (p<0.001) than those of CON silages. Yeasts (p = 0.042) and molds (p = 0.046) of WCR silages decreased linearly with increasing DM content of WCR forage. In the rumen, INO silages had higher the total degradable fraction (p<0.001), total volatile fatty acid (tendency; p = 0.097), and acetate (p = 0.007), but lower the fractional degradation rate (p = 0.011) and propionate (p<0.001) than those of CON silage. The total degradable fraction (p<0.001), total volatile fatty acid (p = 0.001), iso-butyrate (p = 0.036), and valerate (p = 0.008) decreased linearly with increasing DM content of WCR forage, while the lag phase (p<0.001) was increased linearly. Conclusion: This study concluded that application of dual-purpose inoculants on WCR silage confirmed antifungal and carboxylesterase activities by inhibiting mold and improving rumen digestibility, while increase of wilting times decreased organic acids production and rumen digestibility.

A Study on the Characteristics of Coffee Ground(CG)-RDF by Using Different Drying Method (건조법에 따른 커피박 고형연료의 특성 고찰 연구)

  • In this study, the characteristics of coffee grounds were reviewed by making them from solid fuel through heat-drying and oil-drying method. The differences in the higher calorific power by each dried sample were compared. And industrial analysis using the thermogravimetric analyzer was considered for applicability to organic waste and oily samples. Before and after drying, the surface of the specimen was observed with SEM equipment and the ingredients were measured through the EDS equipment. As a result, no other hazardous substances, such as heavy metals, were measured. Next, The differences between thermal decomposition and combustion reactions were considered through the TG and DTG curves. As a result, it is that the oil-dried coffee grounds is longer to burn than the heat-dried coffee grounds. Finally, the combustion gases emitted through the thermogravimetric analyzer were collected and the carbon monoxide and carbon dioxide performed qualitative and quantitative analysis using GC over time.

Evaluation of Fluoride Removal Effect with Growth of Attached Microbial Community in Middle and Small Stream (중·소하천에서 부착미생물군집의 성장에 따른 불소 제거 효과 평가)

  • This study was conducted to understand the growth of attached microbial community in small and medium streams of Gyeonggi-do and the removal efficiency of fluoride with changes in growth. The sites monitoring were carried out for 12 weeks at the P1 and P2(P1 is the point where the discharge water flows. P2 is the downstream point of about 2 km flow), and investigated the water quality and the biomass analysis of the attached microbial community. The growth of the attached microbial community increased up to the 7th investigation, and it was observed that detachment phenomenon occurred. Influencing factors about growth of the attached microbial community were flow rate, velocity, and organic material(T-N & T-P). Meanwhile, fluoride content of attached microbial community also tended to increase until the 7th investigation, and decrease from the 8th. It is assumed that fluoride content has also be reduced with the detachment phenomenon of the attached microbial community. It is expected that this will contribute to the evaluation and management of the use of attached microbial community as a means of stream management. The application of techniques using the attached microbial community should include basic investigation of factors that may affect the growth of the attached microbial community and replacement of the attachment plate according to the time of removal.

Changes in Sediment Properties Caused by a Covering of Oyster Shells Pyrolyzed at a Low Temperature (저온 소성 굴 패각의 피복에 의한 연안 오염 퇴적물의 성상 변화에 관한 연구)

  • In this study, pyrolyzed oyster shells at a low temperature ($350^{\circ}C$) were applied for a mesocosm experiment to confirm resulting changes in the properties of sediment. After creating a covering of oyster shells, an increase in ORP and decrease in ammonia in the overlying water was observed in an experimental case. The decrease of TOC in this experiment was due to the dilution of organic matter due to the addition of inorganic matter (pyrolyzed oyster shells). The decrease in the concentration of AVS was observed due to the adsorption of AVS by the surface of the oyster shells. From the results obtained in this experiment, it has been concluded that pyrolyzed oyster shells at a low temperature can be used for remediation of polluted sediment.

Antibacterial effect of Zingiberaceae extracts mediated photodynamic therapy on Streptococcus mutans (Streptococcus mutans에 대한 생강과 천연추출물의 광역학 항균효과)

  • Purpose: This study evaluated the antibacterial effects of curcuma, ginger, and finger root extracts in water-soluble powder on planktonic Streptococcus mutans(S.mutans), which is known to cause dental caries, in order to confirm whether these extracts could perform as photosensitizers for the effects of photodynamic therapy (PDT). Methods: This study used the strain of streptococcus mutans ATCC 25175 distributed by the Korean Collection for Type Cultures of the Korea Research Institute of Bioscience & Biotechnology. Commercial edible curcuma, ginger and finger root were used as the natural extracts for the use of photosensitizer. To extract organic solvent, 3 g of each powder was mixed in $30m{\ell}$ of dimethyl sulfoxide (DMSO, VWR, Germany) before extraction. $1.8m{\ell}$ of the photosensitizer solution, manufactured in the concentrations of 5, 0.5, and $0.05mg/m{\ell}$, was mixed with $0.2m{\ell}$ of the S. mutans culture medium that had been cultured for 2 days. To induce the photodynamic reaction, Qraycam (AIOBIO, Seoul, Korea) equipped with 405 nm LED was used to expose light for 5 minutes to irradiate 59 nW energy for 300 seconds. Results: Compared with the case with no light, a higher photodynamic therapeutic effect was confirmed with $0.05mg/m{\ell}$ curcuma powder extract, the concentration of $0.5mg/m{\ell}$ and LED light of 405 nm wavelength (p=0.000, p=0.003). $0.05mg/m{\ell}$ of curcuma powder extract and the concentration of $0.5mg/m{\ell}$ showed 100% antibacterial effect when exposed to light, whereas the concentration of $5mg/m{\ell}$ showed 11.95% antibacterial effect. When exposed to light, $0.05mg/m{\ell}$ of ginger powder extract showed an antibacterial effect which didn't statistically decrease. The concentrations of $0.5mg/m{\ell}$ and $5mg/m{\ell}$ did not show any antibacterial effects. As a result of examining any photodynamic therapeutic effects of finger root powder extract on S. mutans, no statistically significant effect was found. Conclusion: The curcuma powder extract is expected to perform as a photosensitizer. Even though belonging to the same ginger family, ginger powder and finger root powder seem difficult to perform as photosensitizer.
