Verifications of Resistance to Phytophthora spp. in 2-year-old Citrus junos Cultivars and Related Specie

  • Yuzu (Citrus junos) gummosis disease, caused by Phytophthora nicotianae, was first reported in 1997. As known in citrus, Phytophthora is the most fastidious soil-borne pathogen to control. In order to minimize its damage to Citrus spp., integrated pest management (IPM) approach, including fungicide chemicals and resistant cultivars, is necessary. Therefore, in this study we tried to evaluate tolerance of yuzu cultivars and its related species against yuzu Phytophthora. Trifoliate orange was evaluated as a susceptible host to yuzu Phytophthora by both mycelial growth onto extract media and immature fruit inoculation. However, in zoospores spray-inoculation on 2-year-old cuttings tree, trifoliate orange appeared to have a resistant property as showing less than 6% diseased leaf rate. Among yuzu cultivars only 'Namhae No. 1' appeared resistant property against both P. nicotianae and P. citrophthora. The 'Namhae No. 1' showed 5.7% and 10.6% diseased leaf ratio by P. nicotianae and P. citrophthora, respectively. Clearly, in order to reduce damages caused by two yuzu Phytophthora, we suggest that growers may utilize a trifoliate orange as a rootstock and 'Namhae No. 1' as a scion for fruit production.

Acridine orange-induced changes in the O-side chain lengths of lipopolysaccharides of Salmonella pullorum (Salmonella pullorum lipopolysaccharide의 acridine organe에 의한 O-side chain 길이의 변화)

  • The morphological hetergeneity of lipopolysaccharides(LPSs) and variation in the O-side chain lengths of LPSs of Salmonella pullorum, which was serially subcultured on the brain heart infusion agar containing $100{\mu}g/m{\ell}$ of acridine orange, was analyzed in Sephadex G-50 column chromatography and silver-stained polyacrylamide gels. The biochemical differences in LPS W and LPS A0150 were identified. Increases in the contents of O-polysaccharides of LPS A0150 than those of LPS W were reflected in the profiles of chromatography and silver-stained polyacrylamide gels. In summary, LPS molecules of S pullorum A0150 appeared to be enriched in the molecules with long O-polysaccharide chains than those of LPS W.

Micronucleus Frequencies in Human Umbilical Cord Blood by the Supravital Staining Method (인체에서의 초생체 염색법을 이용한 제대혈내 소핵 출현 빈도)

  • This study was conducted to quantify of micronucleus frequencies in human umbilical cord blood by supravital staining method with acridine orange, and to find some factors that affected on micronucleus frequncies in humans. In this study, we used umbilical cord blood of new born infants that have sufficient reticulocytes compared with adult peripheral blood. The cord bloods were taken after childbirth from 60 normal infants in industrial and coastal region in Korea. The total of 3 ${mu}ell$ cord blood was applied to slide coated with acridine orange, and micronuclei were observed under fluorescent microscopy. Demographic factors and independent variables were collected from mothers by questionnaire. The frequencies of micronuclei in umbilical cord blood of new born infants were 0-5 per 2,000 reticulocytes by supravital staining method, and mean value and standard deviation were 1.75$\pm$0.97. There were no significant difference by the regions, smoking habits of father or mother. However, age of mother showed significant positive correlation with frequencies of micronuclei (p<0.05). Smoking at home by fathers also was found as a significant variable by muliple regression analysis. Therefore, further studies would be needed for genotoxicological evaluation of new born infants by microneuli test using supravital staining method.

Studies on Transfer Printing of Cotton and Polyester/Cotton Blended Fabrics Treated with Water Soluble Polyurethane Resin (수용성 폴리우레탄 수지 처리된 면 및 폴리에스테르/면 혼방직물의 전사날염에 관한 연구)

  • In this study, selected cotton fabrics and polyester/cotton(P/C) blended fabrics are treated with a soluble polyurethane(SPU) and then, printed by heat transfer to determine the effect of SPU treatment on dye uptake of the samples. The results obtained are as follows: 1) In heat transfer, dye-uptake gets higher in Vopotion to temperature and time. The optimum printing temperature and printing time of C.I. Disperse Orange 3 and C.I. Disperse Violet 1 are $200^\circ{C}$ and 50sec. 2) Dye-uptake gets higher according to SPU concentration ; both cotton and P/C fabrics show the highest at $100g/\ell$. 3) In color, as temperature, time and SPU concentration increase, P/C fabrics show more yellowish orange color than cotton fabrics in case of C.I. Disperse Orange 3 and P/C fabrics show more reddish violet color than cotton fabrics in case of C.I. Disperse Violet 1. 4) All fastness of cotton and P/C fabrics treated with SPU are good, but color fastness to washing and water of cotton fabrics treated with SPU are not good.

Rapid Fenton-like degradation of methyl orange by ultrasonically dispersed nano-metallic particles

  • This study investigates methyl orange (MO) degradation by an ultrasonically dispersed nano-metallic particle (NMP) assisted advanced Fenton process. The NMPs were synthesized from the leachate of automobile-shredder residue. X-ray photoelectron spectroscopy and Fourier-transform infrared spectroscopy were performed for the prepared NMPs. Various parameters, such as the effects of the NMP dosage, the pH value of the solution, the initial concentration of MO, and the amount of $H_2O_2$ on the degradation efficiency of MO were studied. The MO degradation efficiency could be increased by approximately 100% by increasing the dosages of the NMPs and $H_2O_2$ to certain limits, after which in both cases the degradation efficiency was reduced when an excess amount was added. The MO degradation efficiency was found to be 100% at pH 2.0 and 2.5 with the 10 mg/L of initial concentration of the MO. The degradation of MO by ultrasonically dispersed NMPs was appropriate with the pseudo-first-order kinetics.

In vitro Propagation of Junos Orange (Citrus junos Sieb.) through Nucellar Polyembroid Cultures

  • An in vtro nucellar polyembryo propagation method was established with mature seed of the Citrus junos Sieb. 7-8 nucellar polyembryos per seed were induced on MS basal medium without plant growth regulators. The polyembryos developed to complete plantlets on teatment with IBA. These shoots grew further in MS medium without plant growth regulators. Rooting of shoots occurred on MS medium supplemented with IBA. These plantlets were successfully transplanted to small plastic pot containing soil mixture. Somatic embryos were induced from nucellar polyembryo and maturation occurred spontaneously from proliferating cultures on MS medium without growth regulators. Random Amplified Polymorphic DNA (RAPD) marker analysis of in vitro and in vivo grown junos orange showed identical polymorphism indicative of their genetic stability. The RAPD polymorphism produced revealed same banding pattern in each regenerant. Hence, propagaton of junos orange by nucellalr polyembryos was efficient and produced in genetically stable plants under in vitro conditions.

Simplified Bilayer White Phosphorescent Organic Light-Emitting Diodes

  • We report on highly efficient blue, orange, and white phosphorescent organic light-emitting diodes consisting only two organic layers. Hole transporting 4, 4,' 4"-tris (N-carbazolyl)triphenylamine (TcTa) and electron transporting 2-(diphenylphosphoryl) spirofluorene (SPPO1) are used as an emitting host for orange light-emitting bis(3-benzothiazol-2-yl-9-ethyl-9H-carbazolato) (acetoacetonate) iridium ((btc)2(acac)Ir) and blue light-emitting iridium(III)bis(4,6-difluorophenyl-pyridinato-N,C2') picolinate (FIrpic) dopant, respectively. Combining these two orange and blue light-emitting layers, we successfully demonstrate highly efficient white PHOLEDs while maintaining Commission internationale de l'eclairage coordinates of (x = 0.373, y = 0.443). Accordingly, we achieve a maximum external quantum, current, and power efficiencies of 12.9%, 30.3 cd/A, and 30.0 lm/W without out-coupling enhancement.

Comparison of the Effects of Blending and Juicing on the Phytochemicals Contents and Antioxidant Capacity of Typical Korean Kernel Fruit Juices

  • Four Korean kernel fruit (apple, pear, persimmon, and mandarin orange) juices were obtained by household processing techniques (i.e., blending, juicing). Whole and flesh fractions of each fruit were extracted by a blender or a juicer and then examined for phytochemical content (i.e., organic acids, polyphenol compounds). The antioxidant capacity of each juice was determined by ferric reducing antioxidant power (FRAP) and 2,2-diphenyl-1-picrylhydrazyl (DPPH) assays. Results revealed that juices that had been prepared by blending whole fruits had stronger antioxidant activities and contained larger amounts of phenolic compounds than juices that had been prepared by juicing the flesh fraction of the fruit. However, the concentration of ascorbic acid in apple, pear, and mandarin orange juices was significantly (P<0.05) higher in juice that had been processed by juicing, rather than blending. The juices with the highest ascorbic acid (233.9 mg/serving), total polyphenols (862.3 mg gallic acid equivalents/serving), and flavonoids (295.1 mg quercetin equivalents/serving) concentrations were blended persimmon juice, blended mandarin orange juice, and juiced apple juice, respectively. These results indicate that juice extraction techniques significantly (P<0.05) influences the phytochemical levels and antioxidant capacity of fruit juices.

The Investigation on Color Change of Dis-azo Acid Dye in Wool Dyeing (양모섬유의 염색시 디스아조계 산성염료의 변색현상 규명)

  • It is already known that the color of wool fabric dyed with disazo acid dyes could be changed in dyeing process and this is accelerated under alkaline condition. Focus was given to figuring out the mechanism of this color change, through the LC-MS analysis. In this study, no color change was seen in wool fabrics dyed with C. I. Acid Blue 113 under weak acidic, neutral and weak alkaline conditions for 1hour. However, the wool fabrics dyed under weak alkaline condition for a long time over 3 hours fumed reddish orange. When the wool fabrics dyed under weak acidic, neutral and weak alkaline conditions were treated with $0.5g/L\;Na_2C0_3$ solution, all of them turned reddish orange. On the other hand, the color of silk fabrics dyed with C. I. Acid Blue 113 were not changed after the same alkaline treatment. Wool contains cystine and cysteine, whereas silk does not. Due to the reversible reduction/oxidation process of cystine and cysteine in wool dyeing, the C. I. Acid Blue 113 of the dis-azo type is decomposed by reduction and consequently turned them into the reddish orange mono-azo types dye.

Effects of Mugwort, Dried Orange Peel and Duching on Lipid Metabolism in Hyperlipidemia Rats (쑥 , 진피, 두충이 고지혈증 흰쥐의 체내 지질대사에 미치는 영향)

  • This study was performed to investigate the influence of dietray food from the three CHinese medicinal herbs(Mugwort, Dried organge peel and Duchung) on lipi d metabolism in rats. Male rats of Sprague-Dawley strain were divided into four dietary groups(Cellulose (CL), Mugwort(MW), Dried organge peel9OP), Duchung(DC)) after three weeks of hyperlipidemic dieting and were fed ad libitum each of the experimental diets for six weeks. The fiber sources of the experimental diets were prepared by drying and milling mugwort, orange peel, duchung and cellulose. Each of the Fiber sources was mixed into the diet to achieve 10g% levels of total dietary fiber. Levels of apparent fat absorption exhibited a significant decrease in the MW and DC groups. The OP group also showed a decreasing trend, but to a lesser extent. The levels of serum total lipid were not significantly different among the four experimental groups. Total serum cholesterol levels were significantly lower in the OP grop that in both the CM and MW group. Serum TG and LDL-cholesterol levels were significantly higher in the DL group than other groups. Serum HDL-cholesterol levels were significantly levels were significantly lower in the CL group than in the other groups. Total lipid and total cholesterol in liver were significantly higher in the CL group than in the other groups. Total lipid and total cholesterol in feces displayed a significantly increase in the MW and DC groups when compared with the CL group. The OP group also showed an increase compared to the CL group.
