발효조를 이용한 Monascus anka의 적색소와 황색소의 생산

  • In order to develop the method for mass production of natural food colorant from Monascus anka, optimum cultivation conditions for producing red and yellow pigments by cultiva- ting the mold in a jar fermenter and their color characteristics were investigated. The mold produced red and yellow pigments both intracellularly and extracellularly. These pigments showed unique light absorption characteristics with maximum absorption of 494, 380, 506, and 388 nm for extracellular red pigment (ERP), extracellular yellow pigment (EYP), intracellular red pigment (IRP), and intracellular yellow pigment (IYP), respectively. Optimum conditions for producing red pigments were found to be temperature 30$\circ$C, initial pH 6.0, rice powder 3-5%, peptone 0.05%, magnesium sulfate 0.25%, aeration rate 0.1 vvm. Optimum temperature for producing yellow pigments was around 35$\circ$C which is higher than that of producing red pigments. The initial pH and rice powder concentration for producing yellow pigments were the same as those of producing red pigments. The higher concentration of nitrogen source and inorganic salt, aeration rate, the more the yellow pigments were produced. The optimum agitation speed was 100 - 300 rpm for pigment production.

Optimum Design of a Pin-Fins Type Heat Sink Using the CFD and Mathematical Optimization

  • The shape of $7\times7$ pin-fins heat sink is optimized numerically to obtain the minimum pressure drop and thermal resistance. In this study, the fin height (h), fin width (w), and fan-to-heat sink distance (c) are chosen as the design variables and the pressure drop $({\Delta}P)$ and thermal resistance $(\theta_j)$ are adopted as the objective functions. To obtain the optimum design values, we used the finite volume method for calculating the objective functions, the BFGS method for solving the unconstrained non-linear optimization problem, and the weighting method for predicting the multi-objective problem. The results show that the optimum design variables for the weighting coefficient of 0.5 are as follows: W=4.653 mm, h=59.215mm, and c=2.667mm. The objective functions corresponding to the optimal design are calculated as ${\Delta}P=6.82$ Pa and $(\theta_j)=0.56K/W$. The Pareto solutions are also presented for various weighting coefficients and they will offer very useful data to design the pin-fins heat sink.

새송이버섯 biomass를 위한 최적배양 조건 (Culture Condition for Biomass of Pleurotus eryngii)

  • 새송이버섯의 biomass를 위한 최적조건을 규명한 결과, 균사체 spot 배양을 위한 최적 조건은 PDA배지를 사용하여 24$^{\circ}C$에서 18일 배양 시 최적인 것으로 판단하였으며, 새송이버섯 균사체 Biomass를 위한 bioreactor의 배양 최적조건은 PDMP배지를 사용하여 pH 5.5, 18$^{\circ}C$에서 27일 배양시 reactor의 반응은 최적의 상태를 나타낼 수 있을 것이라 판단되었다. 최적조건에서 bioreactor를 이용한 새송이버섯 균사체의 인공적인 대량 배양이 가능하였으며, bioreactor에서 대량 인공 배양된 균사체를 이용한 새송이버섯 재배 시, 자실체가 형성되어 새송이버섯의 대량생산을 위한 biomass가 가능하였다.

프로바이오틱 유산균 발효조건 탐색을 위한 다반응 최적화의 활용 (Applying Multi-Response Optimization to Explore Fermentation Conditions of Probiotics)

  • This review serves two purposes: first, to promote the use of improved optimization techniques in response surface methodology (RSM); and second, to enhance the optimum conditions for the fermentation of probiotics. According to research in dairy science, Lactiplantibacillus plantarum K79 is a candidate probiotic that has beneficial health effects, such as lowering blood pressure. The optimum conditions for L. plantarumK79 to produce peptides with angiotensin-converting enzyme (ACE) inhibitory activity were proposed, through modeling each of ACE inhibitory activity and pH as a function of the four factors that are skim milk concentration (%), incubation temperature (℃), incubation time (hours), and starter added amount (%). To estimate optimum conditions, the researchers employed a desirability-based multi-response optimization approach, utilizing third-order models with a nonsignificant lack of fit. The estimated optimum fermentation conditions for L. plantarum K79 were as follows: a skim milk concentration of 10.76%, an incubation temperature of 36.9℃, an incubation time of 23.76 hours, and a starter added amount of 0.098%. Under these conditions, the predicted ACE inhibitory activity was 91.047%, and the predicted pH was 4.6. These predicted values achieved the objectives of the multi-response optimization in this study.

응집제 생산 미생물의 분리 및 배양조건 (Isolation of Microorganism Producing Flocculant and Its Culture Conditions)

  • 상하수도 및 폐수처리에 많이 사용되고 있는 무기 및 유기합성 고분자응집제를 미생물 응집제로 대체 하기 위한 연구의 일환으로 토양으로부터 목적 미생물을 분리하여 형태학적, 생리학적 성질을 조사한 결과 Aspergillus sp. JS-42로 명 명 되 었다. 또한 Aspergillus sp. JS-42의 응집제 생산을 위한 최적배지 벚 최적 배양조건은 다음과 같이 설정하였다. 배지 조성은 3.0% glucose, 0.2% yeast ex ttract, 0.1 % $(NH_4)_2S0_4, 0.05% CaCI_2.2H_20$ 의 경우가 응집제 생산에 우수하였으며, lJ~양온도 및 초 기 pH는각각$25^{\circ}C$ 와pH 7.0의 경우가최적이였다. 이 배양조건 하에서 90시간 전후로 배양하였을 때 응집제 생산이 최대가 되었으며 이때는 screenmg시 의 기초배지에 비해 응집활성이 거의 8배 정도로 향 상되었다. 이 물질은 각종 기질에 대해 넓은 작용범 위와 강력한 응집능을 발휘하였으며 pH 및 열처리 에 대해 극히 안정한 물질이였다. 또한 이 물질은 점성이 강한 고분자성인 다당류의 일종인 것으로 판단 되였다.

맥주박을 이용한 알코올 발효사료의 최적 발효조건 규명 (Determination of Optimum Fermentation Condition for Alcohol-Fermented Feeds with Wet Brewer's Grains)

  • 본 시험은 맥주박의 알코올 발효가 발효특성 및 사료의 영양적 가치에 미치는 영향을 검토하기 위하여 실시하였다. 본 시험에 이용한 시험사료는 맥주박과 파쇄 옥수수를 50:50(풍건상태)의 비율로 혼합하여 혐기 발효에 의하여 발효사료를 제조하였다. 발효 조건은 당밀 첨가 수준(0, 2 및 5%), yeast 첨가 수준(0, 0.1, 0.5, 1.0 및 2.0%) 및 발효 온도(4, 10, 30, 40 및 50$^{\circ}C$)에 따른 변화를 검토하였으며, 각 조건에 의한 알코올 함량, live yeast cell number (LYCN) 및 pH 변화를 조사하였다. 당밀 첨가에 따른 알코올 함량은 첨가수준이 증가할수록 높게 생성되었다. 발효 24시간 동안에 yeast 0.1, 0.5, 1.0 및 2.0% 첨가에 따른 알코올 함량은 각각 1.9, 2.4, 3.1 및 3.1%로 나타나, yeast 첨가수준 1% 까지는 yeast의 첨가수준이 증가할수록 시험사료중의 알코올 생성량도 비례적으로 증가하였으나(P<0.05), yeast 첨가수준 1.0 이상에서는 일정한 양의 알코올 이 생성되었다. 발효 초기의 LYCN은 yeast 첨가수준이 높을수록 높게 나타났으나(P<0.05), 발효 48시간 후의 LNCN은 0.1% 이하에서는 유의적으로 낮게 나타났다(P<0.05). 발효 온도에 따른 알코올 생성량은 발효 온도 30 및 40$^{\circ}C$에서 유의적으로 높은 결과를 보였으며(P<0.05), 30$^{\circ}C$와 40$^{\circ}C$ 간의 알코올 생성량은 30$^{\circ}C$에서 높은 경향이었다. 발효 온도에 따른 pH는 발효 온도가 낮을수록 발효 24시간까지 급격히 감소하는 것으로 나타났다(P<0.05). 이상의 결과로 미루어 맥주박의 사료적 가치를 높이기 위해 yeast의 첨가량, 당밀(발효기질), 발효온도 및 발효시간 등의 요인을 고려하였을 경우, 본 시험의 조건하에서 원활한 알코올 발효와 발효사료의 품질을 향상시키기 위한 적정 발효조건은 당밀 5%와 yeast 1%를 첨가하여 30$^{\circ}C$에서 24시간 동안 발효시키는 것이 가장 바람직한 것으로 나타났다.

Optimization of Maillard Reaction in Model System of Glucosamine and Cysteine Using Response Surface Methodology

  • Sulfur-containing amino acids play important roles in good flavor generation in Maillard reaction of non-enzymatic browning, so aqueous model systems of glucosamine and cysteine were studied to investigate the effects of reaction temperature, initial pH, reaction time, and concentration ratio of glucosamine and cysteine. Response surface methodology was applied to optimize the independent reaction parameters of cysteine and glucosamine in Maillard reaction. Box-Behnken factorial design was used with 30 runs of 16 factorial levels, 8 axial levels and 6 central levels. The degree of Maillard reaction was determined by reading absorption at 425 nm in a spectrophotometer and Hunter's L, a, and b values. ${\Delta}E$ was consequently set as the fifth response factor. In the statistical analyses, determination coefficients ($R^2$) for their absorbance, Hunter's L, a, b values, and ${\Delta}E$ were 0.94, 0.79, 0.73, 0.96, and 0.79, respectively, showing that the absorbance and Hunter's b value were good dependent variables for this model system. The optimum processing parameters were determined to yield glucosamine-cysteine Maillard reaction product with higher absorbance and higher colour change. The optimum estimated absorbance was achieved at the condition of initial pH 8.0, $111^{\circ}C$ reaction temperature, 2.47 h reaction time, and 1.30 concentration ratio. The optimum condition for colour change measured by Hunter's b value was 2.41 h reaction time, $114^{\circ}C$ reaction temperature, initial pH 8.3, and 1.26 concentration ratio. These results can provide the basic information for Maillard reaction of aqueous model system between glucosamine and cysteine.

Culture conditions for mycelial growth of Poria cocos

  • This study was carried out to determine the basic mycelial culture conditions for Poria cocos growth. According to colony diameter and mycelial density, suitable media for mycelial growth were Malt yeast extract, Potato dextrose agar, Yeast extract agar, and Yeast malt agar. The optimum temperature for mycelial growth was between 25 and $35^{\circ}C$, and the optimum pH value was between 4 and 7. Carbon and nitrogen sources were fructose and yeast extract. The optimum C/N ratio was about 10 to 1 with 2% glucose. Other minor components for optimal growth were thiamine-HCl and nicotinamide as vitamins, acetic and lactic acid as organic acids, and $MgSO_4{\cdot}7H_2O$ and $FeSO_4{\cdot}7H_2O$ as mineral salts.

플라즈마 공정에 대한 소독 모델 적용 (Application of Disinfection Models on the Plasma Process)

  • The application of disinfection models on the plasma process was investigated. Nine empirical models were used to find an optimum model. The variation of parameters in model according to the operating conditions (first voltage, second voltage, air flow rate, pH) were investigated in order to explain the disinfection model. In this experiment, the DBD (dielectric barrier discharge) plasma reactor was used to inactivate Ralstonia Solanacearum which cause wilt in tomato plantation. Optimum disinfection models were chosen among the nine models by the application of statistical SSE (sum of squared error), RMSE (root mean sum of squared error), $r^2$ values on the experimental data using the GInaFiT software in Microsoft Excel. The optimum model was shown as Weibull+talil model followed by Log-linear+ Shoulder+Tail model. Two models were applied to the experimental data according to the variation of the operating conditions. In Weibull+talil model, Log10($N_o$), Log10($N_{res}$), ${\delta}$ and p values were examined. And in Log-linear+Shoulder+Tail model, the Log10($N_o$), Log10($N_{res}$), $k_{max}$, Sl values were calculated and examined.

Characterization of Aspartate Aminotransferase Isoenzymes from Leaves of Lupinus albus L. cv Estoril

  • Two aspartate aminoransferase (EC 2.6.1.1) isoenzymes (AAT-1 and AAT-2) from Lupinus albus L. cv Estoril were separated, purified, and characterized. The molecular weight, pI value, optimum pH, optimum temperature, and thermodynamic parameters for thermal inactivation of both isoenzymes were obtained. Studies of the kinetic mechanism, and the kinetics of product inhibition and high substrate concentration inhibition, were performed. The effect of some divalent ions and irreversible inhibitors on both AAT isoenzymes was also studied. Native PAGE showed a higher molecular weight for AAT-2 compared with AAT-1. AAT-1 appears to be more anionic than AAT-2, which was suggested by the anion exchange chromatography. SDS-PAGE showed a similar sub-unit molecular weight for both isoenzymes. The optimum pH (between 8,0 and 9.0) and temperature ($60-65^{\circ}C$) were similar for both isoenzymes. In the temperature range of $45-65^{\circ}C$, AAT-2 has higher thermostability than AAT-1. Both isoenzymes showed a high affinity for keto-acid substrates, as well as a higher affinity to aspartate than glutamate. Manganese ions induced an increase in both AAT isoenzymes activities, but no cooperative effect was detected. Among the inhibitors tested, hydroxylamine affected both isoenzymes activity by an irreversible inhibition mechanism.
