Enzymatic Hydrolysis for Effective Extraction of Antioxidative Compounds from Hizikia fusiformis

  • Hizikia fusiformis hydroysates by five carbohydrases (Viscozyme, Celluclast, Termamyl and Ultraflo) and five proteases (Protamex, Kojizyme, Neutrase, Flavourzyme and Alcalase) were investigated for their extraction efficacy (yield and total total polyphenolic content) and antioxidative activity (DPPH radical and hydrogen peroxide scavenging activity). Termamyl and Ultraflo of the carbohydrases and Flavourzyme and Alcalase of proteases were selected by their high eficacy of extraction and antioxidative activity. Selected enzymes were used to investigate the optimum enzymatic reaction time and dosage (enzyme/substrate ratio) suitable for hydorolysis. Optimum reaction time for the enzymatic hydrolysis was 3 days and optimum dosage of hydrolysis was observed as 5%. Simultaneously, Ultraflo of the two carbohydrases and Alcalse of the two proteases were selected as the most effective enzymes. Combination of Ultraflo and Alcalase under optimum hydrolysis conditions could intensify the extraction efficacy of antioxidative materials form H. fusiformis. The hydrolysate obtained by combining the enzymes was separated into four different molecular weight fractions (<1kD, 1-10 kD, 10-30 kD and >30 kD) and recorded the polyphenolic content distribution and respective antioxidative ability. The fraction <1kD was identified as less effective and those fractions > 1kD indicated comparatively higher antioxidative activities related to their polyphenolic content.

Hydrolysis of Lactose in Whey by the BetavD-Galactosidase (Beta-D-Galactosidase에 의한 유청에 함유된 유당의 가수분해)

  • The optimum condition for the developement of a whey beverage from the concentrated whey was studied. Reverse osmosis system was used to obtain concentrated lactose from cheese whey. The hydrolysis degree of lactose by $\beta$-D-galactosidase was determined using HPLC (high performance liquid chromatography). The order of hydrolysis degree was 1:1, 2:l and 3:l concentrated lactose. It resulted from the concentrated salt which slightly inhibited $\beta$-D-galactosidase with constant enzyme dosage. The optimum condition for enzyme dosage was 2% in non-concentrated lactose, 3% in 2:l and 3% in 3:l concentrated lactose after 4 hours of reaction. When the 3:l concentrated lactose was used, more than 70% was hydrolyzed by 3% enzyme dosage. Furthermore the change of fermented whey by lactic acid bacteria was investigated. Based on the result of sensory test, the most favorable response was obtained at pH 4.2 and titratable acidity of 0.7% about 6 hours of fermentation at $37^{\circ}C$ with 2%: thermophilic starter.

  • In the water purification plant, the raw water is promptly purified by injecting chemicals. The amount of chemicals is directly related to water quality such as turbidity, temperature, pH and alkalinity. At present, however, the process of chemical reaction to the turbidity has not been clarified as yet. Since the process of coagulant dosage has no feedback signal, the amount of chemical can not be calculated from water quality data which were sensed from the plant. Accordingly, it has to be judged and determined by Jar-Test data which were made by skilled operators. In this paper, it is concerned to model and control the coagulant dosing process using jar-test results in order to predict optimum dosage of coagulant, PAC(Polymerized Aluminium Chloride). The considering relations to the reaction of coagulation and flocculation, the five independent variables(turbidity, temperature, pH, Alkalinity of the raw water, PAC feed rate) are selected out and they are put into calculation to develope a neural network model and a fuzzy model for coagulant dosing process in water purification system. These model are utilized to predict optimum coagulant dosage which can minimize the water turbidity in flocculator. The efficacy of the proposed control schemes was examined by the field test.

Photocatalytic Degradation of Quinol and Blue FFS Acid Using TiO2 and Doped TiO2

  • The photodegradation of the model compounds Quinol, an aromatic organic compound and Acid blue FFS, an acid dye of chemical class Triphenylmethane was studied by using illumination with UV lamp of light intensity 250W. $TiO_2$ and $TiO_2$ doped with Boron and Nitrogen was used as catalyst. The sol-gel method was followed with titanium isopropoxide as precursor and doping was done using Boron and Nitrogen. In photocatalytic degradation, $TiO_2$ and doped $TiO_2$ dosage, UV illumination time and initial concentration of the compounds were changed and examined in order to determine the optimal experimental conditions. Operational time was optimized for 360 min. The optimum dosage of $TiO_2$ and BN doped $TiO_2$ was obtained to be 2 $mgL^{-1}$ and 2.5 $mgL^{-1}$ respectively. Maximum degradation % for quinol and Blue FFS acid dye was 78 and 95 respectively, at the optimum dosage of BN-doped $TiO_2$ catalyst. It was 10 and 4% higher than when undoped $TiO_2$ catalyst was used.

Effect of Coagulation and Homogenization on the Dissolved Air Flotation and Sedimentation of Bulking Sludge (응집과 균질화가 팽화슬러지의 용존공기부상과 침전에 미치는 영향)

  • The objective of this study is to examine the effect of the coagulation and homogenization in bulking sludge thickening of paper manufacturing plant using DAF(Dissolved Air Flotation) and gravitational sedimentation. The effects of parameters such as dosage of coagulant and homogenization time were examined. The results showed that DAF and sedimentation was affected aluminum sulfate and anion polymer coagulant differently. At the optimum dosage of aluminum sulfate, thickening efficiency of DAF and sedimentation process were increase 1.25 time and 2.02 time, respectively. At the optimum dosage of anion polymer coagulant, thickening efficiency of DAF process was increase 1.35 time, but thickening efficiency at sedimentation was 1.06 time. When anion polymer coagulant of 0.5 mg/l was added in DAF process, water content of sludge was decreased from 96.6% to 90.7% in dewatering process using Buchner funnel test device. After homogenization(20500 rpm, 10 min), Sauter mean diameter of sludge floc was decreased from 631 ${\mu}m$ to 427 ${\mu}m$, however increase of flotation efficiency by DAF was only 1.09 time.

Optimum PP Fiber Dosage for the Control of Spalling of High Strength Reinforced Concrete Columns

  • Spalling is defined as damages to concrete exposed to high temperature during fire, causing cracks and localized bursting of small pieces of concrete. As the concrete strength increases, the degree of damage caused by spalling becomes more serious due to impaired permeability. It is reported that polypropylene(PP) fiber has an important role in protecting concrete from spalling, and the optimum dosage of PP fiber is 0.2%. However, this study was conducted on non-reinforced concrete specimens. The high-temperature behavior of high-strength reinforced concrete columns with various concrete strength and various quantity of PP fibers is investigated in this study. The results revealed that the ratio of unstressed residual strength of columns increased as the concrete strength increased and as the quantity of PP fiber increased from 0% to 0.2%. However, the effect of PP fiber quantity on residual strength of column was barely above 0.2%.

The Experimental Study of Predicting Optimum Dosage of PAC Using Jar-Test Results (Jar-Test를 이용(利用)한 응집제(凝集劑) 주입율(注入率) 결정(決定)에 관한 실험연구(實驗硏究))

  • In this experimental study, it is concerned to develop a simple equation using jar-test results in order to predict the optimum dosage of coagulant, PAC(polyaluminum chloride). Considering the relationships with the reactions of coagulation and flocculation, the four independent variables (e.g. turbidity, temperature, pH and alkalinity) are selected out of many parameters and they are put into calculations to develop an equation by means of multi-regression method. As the result, the dosing rate of PAC is proportional to turbidity, pH and alkalinity, but in inverse to temperature. And the developed equation is as follow, $$D_c=\frac{3.2{\cdot}T^{0.37}{\cdot}A^{0.04}{\cdot}P^{0.5}}{t^{0.1}},\;(R^2=0.9443)$$ And also, comparing between the estimated value from the equation and the real dosing rate in the plant, Kwangam and Tdukdo, during 1988~1991, it is represented an agreement having a relative error of 16.4%, 17.8%, respectively.

  • As recently raw water quality has been polluted as well as its quality has been remarkably varied according to season and region, the precise control of coagulant dosage is being keenly required in water treatment plants. The amount of coagulant is closely related to raw water quality such as turbidity, alkalinity, water temperature, pH, electrical conductivity, etc. Since the optimum quantity of chemicals is not yet finalized, so dosage rate must be decided by using jar test that takes one or two hours. Hereupon, the output signal of stream current and multi-regression on historical data were proposed to be applied to the coagulant dosing control. In consequence of applying the scheme to automatic determination of the dosage rate, it was testified that the determination of dosage rate was very effective in case it is performed as to real-time sensing of water quality and the output signal of stream current.

  • In this study, fundamental properties of mortar with the variation of dosage of anti-rust agent has been tested to evaluate the effect of the anti-rust agent. Dosage of anti-rust agent was fixed as 0,3,6,9%, respectively. The variation of the flow, compressive strength and diffusion speed of chloride ion has been tested to evaluate the effect of the anti-rust agent. Results showed that flow has been smally increased. For the compressive strength, compressive strength increased with the increase of dosage of agent. For the diffusion speed of chloride, the speed decreased with the increase of dosage of the anti-rust agent. For the overall consideration, it could be identified that when the dosage of anti-rust agent was fixed as 3-6%, the mortar showed the optimum performances.

Automatic T-P Coagulation Control System using an EC in the MSBR Process - Full Scale Study - (MSBR 공정에서 전기전도도를 이용한 인 제거 자동제어시스템 - 현장 적용 중심 -)

  • Many sewage treatment plants have applied the advanced technology of chemical coagulant system to remove phosporus in Korea. However there are some problems for the injection of optimum coagulant dosage. In order to solve these problems, the research related to the more cost-effective automatic total phosphorus coagulation control system using an EC(Electrical Conductivity) have been in progress. This study was conducted by the same process and operation method as the Lab-scale for public small town sewage treatment plant. First, it confirmed the correlation among the EC, PO4-P and coagulant dosage in the Lab-scale MSBR(Membrane Sequencing Batch Reactor) process. Next, it analyzed that correlation coefficient of EC and the coagulant dosage was 0.92 in the Full-scale MSBR process. As a result, not only T-P removal efficiency was doubled but also it satisfied the effluent water quality standard in a stable manner. In addition, by applying the automatic control system using the EC, compared to the fixed coagulant injection system the coagulant dosage could be reduced by 28%.
